Changes in the arrangement of the microtubule system of murine peritoneal cells after an antigenic stimulation.

We determined two characteristic morphological patterns typical for the organization of microtubule systems of murine peritoneal macrophages and lymphocytes. A small number of both cell types formed uropods with an axial bundle of microtubules. This number remains practically unchanged during 48 h of cultivation in non-antigenic environment but grows rapidly in the presence of an antigen.

A new approach to in vitro studies of antigenic response in earthworms.

A method for in vitro cultivation of earthworm gut wall tissue explants is described. Tissue explants are able to respond to protein antigen by secretion of antigen-binding protein into the culture medium. Large irregular cells and round-shaped cells are released from tissue explants during the cultivation. This method represents an advantageous tool for in vitro studies providing the possibility of parallel assays from the same donor.

Resistance of macrophages to 5-fluorouracil treatment.

The effect of 5-fluorouracil (5-FU) on murine monocytes and macrophages was the subject of this study. This drug strongly decreased the levels of all types of peripheral blood cell, but had no effect on the composition and cellularity of peritoneal exudate or the expression of Ia antigens or phagocytic activity of peritoneal macrophages. The pinocytic activity of normal macrophages was unchanged, but the pinocytosis of thioglycollate-stimulated macrophages was significantly reduced. The expression of Fc receptors was influenced only after the first day after 5-FU injection. The clearance and organ distribution of radioactive labeled antigen was also not changed. Our findings suggest that tissue macrophages are resistant to 5-FU, but the results obtained with stimulated macrophages suppose differential susceptibility of macrophages. We presume that the immunosuppressive action of 5-FU results from selective depletion and suppression of T and B lymphocytes rather than from changes in immunoregulating functions of macrophages.

Peptide fragments induce a more rapid immune response than intact proteins in earthworms.

The effect of in vivo proteolytic processing of protein antigen was studied in Eisenia foetida earthworms. Parenteral administration of the protein antigen induces elevated levels of an antigen-binding protein (ABP) which recognizes the protein used for stimulation. When the protein antigen is administered simultaneously with nontoxic serine proteinase inhibitor, ABP levels remain close to background. On the other hand, the in vivo adaptive response of earthworms to peptide fragments obtained by coelomic fluid digestion of the foreign antigen occurs even in the presence of proteinase inhibitor and, moreover, is significantly faster as compared to the response to intact antigen. These findings confirm the role of proteolytic processing in earthworms. MALDI mass spectrometric analysis of the fragments after coelomic fluid digestion has revealed the presence of the peptide fragments with molecular weights in the mass range 700-1100 Da.
