The aim of the present work is to investigate in vivo cytokine production during chemohyperthermia. 11 patients suffering from gastric adenocarcinoma (n = 6), ovarian adenocarcinoma (n = 4) or malignant mesothelioma (n = 1) were studied. Patients received 60 mg mitomycin or 100 mg cisplatin per square meter during 2 h in 6 liters of a heating solution (temperature 42 degrees C, flow rate 200 ml/min in a closed circuit) after previous surgical resection. Tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) were measured at 0, 30, 45, 60 and 90 min, both in the blood stream and in the heating solution circulating intraperitoneally. We observed a slight increase in plasma IL-6 occurring as soon as 30 min, a dramatic rise in IL-6 in the heating solution. TNF-alpha values were only slightly augmented. In addition, the importance of various factors in the induction of IL-6 and TNF-alpha production during chemohyperthermia (temperature, mitomycin C, cisplatin) were studied using a whole blood ex vivo model.
PKH dyes were initially developed by Horan et al. to provide appropriate probes for in vitro and in vivo cell tracking. It has been reported for many cell types that PKH bind irreversibly to the cell membrane without significantly affecting cell growth. Thus, these probes provide an opportunity for long-term cell monitoring and the identification of cells of interest among a heterogeneous cell population. An important feature is that upon cell division, the probe is partitioned equally between each daughter cell, making it possible to quantify tell fluorescence by flow cytometry. In this situation. the flow cytometric study of PKH67 characteristics shows that this probe does not affect the main cell-functions such as viability or proliferation. Moreover, the intracellular distribution of PKH67 is demonstrated by following its kinetics of internalization by confocal microscopy. These results present PKH67 as a probe suitable for dynamic analysis of cell proliferation as well as the study of intracellular localization and membrane recycling mechanisms.
Cell growth inhibitions on human cancer cell cultures were determined for the indole sulphur-containing phytoalexins cyclobrassinin, brassilexin (previously isolated from vegetables of the Cruciferae family) and their synthetic analogues 5-methoxybrassilexin and homocyclobrassinin. The most biologically active of these products is brassilexin (LD50 = 8 micrograms/ml).
A therapeutic study was conducted by practitioners working in a group practice, to evaluate the activity of tiapride in treating headaches, over a period of 1 year. The results were satisfactory in 60.6% of cases, especially in patients with headaches of psychogenic or digestive origin. The dosage was 150 to 200 mg orally, and the product was well tolerated apart from somnolence which was noted in about 1 case out of 4.
The number of our findings is still to small to draw definite conclusions. They however show that this technique is very sensitive and highly specific for the diagnostic of tissular amebiasis. Moreover, it significantly reduces the reading difficulty, because the morphologic differences between ameba and the other elements of the antigen is easy to see. The other advantages of the method are: the use of usual optic microscope, which allows the serologic diagnostic in moderatly fitted out laboratories, the use of electron microscope which allows the study of internal structure of the parasite, the preservation of the slides for several months, permitting therefore comparative studies.
