T lymphocyte subpopulations in Graves' disease: relationship with clinical conditions.

T lymphocytes were fractionated according to their receptors for IgG (TG) or IgM (TM) and scored in 37 patients with Graves' disease (17 hyperthyroid and untreated. 10 euthyroid on antithyroid drugs, 10 in long-term remission after radioiodine therapy). TG percentages were very low both in untreated and in drug-treated patients. By contrast, normal TG levels were observed in patients in long-term remission. These data are consistent with the hypothesis of a defective suppressor cell activity in Graves' disease.

Human T-lymphocyte subpopulations in Hashimoto's disease.

An inherited defect os suppressor T-lymphocytes has been hypothesized in Hashimoto's thyroiditis. To assess this hypothesis, human T-lymphocyte subsets (TG, T lymphocytes with surface receptors for the Fc fragment of immunoglobulin G; TM, T lymphocytes with Fc receptor for immunoglobulin M) have been studied in nine patients affected by the disease. To cells have been previously shown to be suppressors in the pokeweed mitogen-stimulated B-cell differentiation and have proved abnormal in several autoimmune or immunodeficiency disorders. The number of TG lymphocytes in the patients did not differ from that in normal controls. It is possible that 1) suppressor T-lymphocytes are not involved in the pathogenesis of Hashimoto's disease or 2) antigen-specific suppressor T-cells are involved, but too are low in number with respect to total TG.

[Methodological problems concerning the assay of sex hormone binding globulin with DEAE-cellulose].

Since 1966 a carrier protein for the sex hormones, testosterone and estradiol, has been identified. Mickelson and Petra measured SHBG concentration by adsorption method in 1974. The SHBG contained in serum is incubated with DHT-3H and adsorbed to a cellulose filter. In this study we have reviewed this method. The method showed good reproducibility with a coefficient of variation of 8,1% for low concentration of SHBG and 11,6% for high concentration. No effects of serum dilution was observed. Normal values, expressed as micrograms DHT bound in 100 ml serum, resulted: females, 0,88 +/- 0,08 (ES); males, 0,58 +/- 0,04; pregnancy, 2,94 +/- 0,4. The mean value for males was significantly lower than the mean value for normal females (P < 0,005); pregnancy had a mean value statistically different from the normal females (P < 0,001).

[Unusual clinical aspects of hyperthyroidism. Apathic hyperthryroidism].

The principal data in the literature regarding apathic hyperthyroidism are summarised and commented on 3 personal cases are then reported. The nosography of these forms is discussed and emphasis laid on pathogenetic hypotheses and diagnostic and therapeutic aspects. Diagnosis should be precise and early so as to prevent the extremely serious course which is a characteristic of this rare, but not exceptional variant of hyperthyroidism.

Transmembrane signaling via both CD3 and CD2 human T cell surface molecules involves protein kinase-C translocation.

The activation of T lymphocytes by appropriate pairs of anti-CD2 monoclonal antibodies has been shown to involve phospholipase-C and phosphoinositide hydrolysis. In this paper we show that the stimulation of the human cloned leukemic T cell line Jurkat by anti-CD2 as well as anti-CD3 monoclonal antibodies induces translocation from cytosol to cell membrane of protein kinase-C (PKC), which is dependent on the formation of 1,2-diacylglycerol from inositol 4,5-diphosphate. PKC translocation is rapid and transient: the kinetics of enzyme redistribution are similar for CD2 and CD3. These results further stress that CD2 and CD3 T cell activation pathways use similar signal transducing mechanisms.

Anti-human thyroglobulin autoantibodies assayed by an enzyme-linked immunosorbent assay.

A rapid and simple enzyme-linked immunosorbent assay (ELISA) for anti-thyroglobulin IgG antibodies is described. The method is based on a 'sandwich' using purified human thyroglobulin adsorbed to polystyrene microplates, human serum and anti-human IgG antiserum conjugated to alkaline phosphatase. The sensitivity of the method is about 8 ng/ml, as evaluated with a purified anti-thyroglobulin antibody preparation. High concentrations of antibodies were observed, as expected, in autoimmune thyroid disease; however, the majority of normal subjects have detectable, although very low, antibody levels. We conclude the method is suitable for current clinical use.

Twenty-four hour profile of integrated TSH concentration in puberty.

The 24-h pattern of integrated serum TSH concentrations has been investigated in 12 healthy adult males and 14 pubertal boys by means of continuous blood flow sampling. For every subject the integrated values and the mean concentrations, corresponding to the entire 24 hours, to the daytime (08:00-20:00) and to the nighttime (20:00-08:00) were calculated. A circadian rhythmicity has been shown in boys, as well as in adults. The mean TSH concentrations were significantly higher in boys with respect to adults. The nyctohemeral maximum was mostly recorded in pubertal boys after sleep and in adults before the onset of sleep. Mean and integrated concentrations corresponding to the nighttime period were significantly higher than those corresponding to the daytime in both groups. In conclusion, the 24-h TSH secretion seems different in boys with respect to adults both in the increased concentrations and in the time (generally after the onset of sleep) of the maximum.

Dissociation between early and late events in T cell activation mediated through CD28 surface molecule.

The regulation of early and late events of T cell activation via the CD28 molecule has been investigated, using as an indicator system the differentiated leukemic T cell line Jurkat. Both CD3 and CD28 mAbs induced an increase in (Ca2+)i in Jurkat cells, although with different kinetics, the latter being slower than the former. CD28-mediated (Ca2+)i mobilization was highly sensitive to cholera toxin (ID50 25 ng/ml, vs 300 ng/ml for CD3 stimulation). The inhibitory action of cholera toxin was neither merely due to the increase in intracellular cAMP concentrations, nor to decrease in cell surface expression of the CD28 molecule. To evaluate the effects of cholera toxin on late events of Jurkat cell activation induced by CD28 and CD3 mAbs, the action of cholera toxin and cAMP and CD3- and CD28-mediated IL-2 secretion was analyzed. CD3-induced IL-2 secretion was highly sensitive to cholera toxin (ID less than 5 ng/ml); on the other hand, CD28-induced IL-2 secretion was poorly sensitive to cholera toxin, in sharp contrast to (Ca2+)i mobilization. On the basis of these data, it is hypothesized that the CD28 pathway could be associated with at least two distinct transduction mechanisms, one responsible for the (Ca2+)i rise in Jurkat cells and highly sensitive to cholera toxin, and the other, whose second messenger is unknown, resistant to cholera toxin and responsible for IL-2 secretion.

ICAM-1 on epithelial cells in allergic subjects: a hallmark of allergic inflammation.

Allergen-specific challenge was first shown to induce ICAM-1 expression on epithelial cells (ECs) of conjunctiva in allergic patients. The data have also been confirmed in the nose. We then checked for the presence of ICAM-1 consequent to natural exposure to allergens. For both conjunctivitis and rhinitis due to pollen we confirmed, during the pollen season, the presence of ICAM-1 on ECs. We demonstrated the endogenous source of ICAM-1 by in situ hybridization both in vitro and in vivo. ICAM-1 could be an activation marker on ECs, or could, enhanced EC susceptibility to bind offending cells such as eosinophils (LFA-1+). ICAM-1 is also a receptor for the vast majority of rhinoviruses, which are known to provoke, mainly in children, asthma attacks. Since we found that mite allergens can induce ICAM-1 on ECs, even during clinical latency, allergy may be considered as a primary event leading to asthma (through rhinovirus infection) and non-specific hyperreactivity.
