Artocarpus integrifolia lectin(s): use and applications in chromosome studies of lymphocyte cultures.

A simple procedure for extracting a lectin from the seeds of the jackfruit, Artocarpus integrifolia, is described. The extracts had an average protein concentration of 13.3 mg/ml, and were strongly erythroagglutinating with a 10(6) titre. Erythroagglutination was shown to be non-specific for A1-B-0 erythrocytes. Lymphoblastic transformation and mitogenic stimulation occurred in lymphocyte cultures both in man and Rhesus monkey, and chromosome preparations were obtained. Such preparations were of good quality and were adequate for chromosome analysis with chromosome banding techniques. In man, the response to the mitogenic stimulation did not vary significantly between individuals. In Rhesus monkey, where Phaseolus vulgaris phytohaemagglutinin produced a weaker mitogenic stimulation than in man, jacalin proved to be very effective.

ABO aggulutinins from Biomphalari straminea snails.

Extracts from B. straminea spawn are active against A and B normal red cells. A1 and A2 subgroups may be differentiated with trypsin-, papain- and pronase-treated cells. 0 cells treated with papain, pronase and neuraminidase react weakly to the extracts.

Agglutinins and hemolysins from Biomphalaria tenagophila snails.

A saline anti-A and incomplete anti-A,B agglutinin were found in spawn extracts of Biomphalaria tenagophila snails. Significance differences after papain and pronase treatments were also found for A1 and A2 subgroups. The hemolysin titers are high, and unspecific.

Construction of a normalized cDNA library for the Trypanosoma cruzi genome project.

Sequencing of the Trypanosoma cruzi genome is underway. Expressed sequence tags, obtained from cDNA libraries, facilitate mapping and gene discovery. The efficiency of large-scale generation of such tags is increased when using normalized cDNA libraries, where the frequency of individual clones is brought within a narrow range. Repetitive sequencing of abundant clones is therefore minimized. We constructed a normalized cDNA library from epimastigotes of clone CL Brener, and the efficiency of normalization of representative clones was assessed and shown to be adequate. The normalized cDNA library has been distributed to several groups and large-scale sequencing is currently in progress.
