[C-AMP concentration in various organs of female rats and in human ovaries with aging (author's transl)].

The change in 3',5'-cyclic adenosine monophosphate (c-AMP) concentration was observed in various organs of rats in gonadal cycle in adult group and with aging (30, 70, 100, 120 weeks), and in human ovaries with aging. 1) The average c-AMP concentration of ovaries of rats showed a significant change with estrus cycle and was higher in the following sequence: proestrus, diestrus II, diestrus I and estrus phase. This tendency was also seen in hypothalamus and pituitary, but was not statistically significant, 2) The average c-AMP concentration in tissues began to decline significantly from 70 weeks in cerebral cortex and hypothalamus, and from 80 weeks in ovaries. However, on the other hand the concentrations in pituitary, liver and adrenal declined markedly from 100 weeks. 3) The c-AMP in ovaries of 80 weeks rats by pregnant mare serum (PMS) road increased by 0.5-fold in concentration, and by 0.6-fold in whole tissue relative to that of 30 weeks rats. 4) A significant difference in serum LH and FSH level between ovarian artery and vein was not found in cycling mature group, non-cycling climacteric group and post-menopausal group of women. 5) Both average concentrations and total values of c-AMP in ovaries of non-cycling climacteric and post-menopausal women were lower than those of mature cycling women. This fact may imply a different response by ovarian tissues such as corpus luteum, follicle and other tissues to gonadotropin. From these results of c-AMP in tissues, it is concluded that the decline of ovarian function with aging of rats was relatively earlier than pituitary, although being delayed compared with hypothalamus, and were the ovarian function in humans declined in the premenopausal period.

Experimental studies on the influence of male alcoholism on fetal development.

To study the effect of paternal chronic ethanol consumption on fetal development, an experimental rat model was established and compared to the fetal alcohol syndrome. Male and female Wistar rats were divided into 30% ethanol (E) and control groups. Before mating and during pregnancy the ethanol group and control group received E and water, respectively. Pregnancies were terminated on gestational day 21. The body weight, liver weight, blood glucose, serum insulin and cerebral CNPase activity were decreased in alcoholic males. The adverse effect of maternal chronic ethanol on fetal development was shown clearly and was not related to paternal ethanol. The adverse effect of paternal alcoholism on the fetus was shown in decreased litter size, or decreased body weight, cerebral weight and cerebral DNA, RNA and leucine incorporation into protein without a decrease in the litter size. The former finding was observed in the fetuses of aged male and female rats and latter in the fetuses of young female rats. In conclusion, both observations in our study indicate the adverse effect of paternal alcoholism on the fetal development.

Frequency of tuberous sclerosis in San-in district (Japan) and birth weight of patients with tuberous sclerosis.

Fifty-two patients with tuberous sclerosis including 44 propositi were collected from 11 hospitals and 20 institutions in the San-in district. Of the 52 patients 44 were alive at the time of the study and the prevalence of tuberous sclerosis among the general population of the San-in district in October 1979 was estimated to be one in 31,000 (3.2 x 10(-5)), which was the highest value among the surveys in certain geographical areas. The frequency of tuberous sclerosis in live-births was estimated to be one in 24,000 (4.0 x 10(-5)). The frequency of the sporadic cases in live-births was one in 31,000 (3.2 X 10(-5) and the mutation rate was directly estimated to be 1.6 X 10(-5) per gene per generation. A difference between the parental age or birth order of the sporadic cases and those of controls was not detected. The mean birth weight for the tuberous sclerosis group was 2,834 g which was slightly less than the 2,985 g for their normal sibs. The frequency of low birth weight, less than 2,500 g, among the children with tuberous sclerosis was 30% (7/23), which was 4 times that in the general population (P less than 0.005). These facts suggest that the intrauterine growth is affected in a considerable number of the patients.

tuberous sclerosis: hydroxyproline content in urine and tissues.

To elucidate the nature of an overabundance of collagen seen on microscopic examination in tuberous sclerous (TS), the hydroxyproline content in tissues and urine was determined. TS tissues of 5 patients were obtained on necropsy or plastic surgery. Urine was collected from 10 patients with TS and 19 controls. Tumors in kidney, pancreas, lung and heart but not brain contained more hydroxyproline than the surrounding tissues. In brain with the lowest hydroxyproline content, the tumor showed reduced hydroxyproline compared to normal. Collagen sheet in TS skin and shagreen patch showed the same hydroxyproline content as control skin. Th urinary hydroxyproline: creatinine ratios of the patients with TS were all higher than those in age-matched controls with or without anticonvulsant treatment except for congenital muscular dystrophy. The higher content of hydroxyproline in several affected tissues and urine of patients with TS might indicate that an increase in collagen synthesis occurs in TS.

The fetal alcohol syndrome in Japan.

In this retrospective study, the clinical findings of 26 cases borne by alcoholic mothers in Japan are summarized. The information on maternal alcohol drinking and abnormal offspring was gathered by a questionnaire survey. Twenty cases, mostly in institutions for mental retardation, were checked from all areas of Japan. Six children from another group were followed during health examinations of children in a selected area with many heavy drinkers. Growth deficiency was found in half of these cases with an increased rate of low birth weight babies (42% of cases) including small-for-date babies (23% of cases). More than 90% of these cases were in the retarded range of intelligence, 50% of which had IQ scores of 51-75. Gross motor development was also delayed. In all of the 6 follow-up cases, craniofacial anomalies such as hypoplastic nose and philtrum, narrow lip vermilion and short palpebral fissures were observed, although these facial features were mild. An aberrant palmar crease was seen with high frequency. CT of 3 cases did not show constant findings. The mothers were said to have drunk about 0.41 or more of 'sake' or whisky daily (average 110 ml of absolute alcohol) throughout pregnancy. Although typical cases of the fetal alcohol syndrome are less than one-third, this report provides evidence for the presence of mentally retarded children borne by alcoholic mothers in Japan and should alert many medical professionals to this problem.

Studies on structural lipids in the stratum corneum.

To determine the mode of existence of structural lipids in the stratum corneum, lipids induced in the thick horny cell layers of guinea pig skin by n-hexadecane treatment were analyzed. Total lipids in the stratum corneum were extracted by Folch's solution and the ratio of phospholipids to free cholesterol was 1:8. Lipids extracted by non-ionic surfactant treatment appeared to originate from both the cell surface and intercellular space of the stratum corneum and the ratio of free cholesterol to phospholipids in this lipid fraction was much higher than that in the total lipids of the stratum corneum extracted by Folch's solution.

Renal cell carcinoma having heterogeneous histological appearance and homogeneous enzymatic property.

A case of renal cell carcinoma (RCC) presenting various histologic growth patterns, including papillary, tubular and solid arrangements, and an area resembling sarcoma are described. In order to identify RCC of high versatility of renal carcinosarcoma, electrophoretic studies of tissue alkaline phosphatase (Al-P) and gamma-glutamyl transpeptidase (gamma-GTP), as well as histochemical studies of these enzymes, were conducted. The Al-P was proved to be bond type alone and the gamma-GTP corresponded with the novel isozyme associated with RCC tissue. Moreover, these two enzymes were found to be distributed in every part of different histologic appearance. It was concluded from these results that the histologic pleomorphism of the case herein reported was due to morphologic transformation of the neoplastic cells sharing essentially identical enzymatic nature.
