Paradoxical effects of gastrin releasing peptide on gastrin release and gastric secretion in the rat.

The effects of gastrin releasing peptide (GRP) on gastrin release and gastric secretion were studied in anesthetized rats. Intravenous infusion of GRP (1-16 micrograms/kg/hr) caused a dose-dependent increase in serum gastrin level, however, it had no effect on basal gastric secretion in the lumen-perfused stomach preparation. Furthermore, GRP inhibited gastric secretion stimulated by pentagastrin or histamine dose-dependently, but not by carbachol. Simultaneous infusion of GRP and a beta adrenergic blocking agent, propranolol, an inhibitor of somatostatin release, did not alter the inhibitory effect of GRP on pentagastrin-stimulated gastric secretion. These results suggest that the inhibitory effect of GRP on gastric secretion in a stimulated condition is mediated via peptide hormones coreleased by GRP, and not via beta-adrenergic pathways.

Cholinergic effects of histamine-H2 receptor antagonists partly through inhibition of acetylcholinesterase.

The effects of histamine H2-receptor antagonists on acetylcholinesterase and pseudocholinesterase activity were studied. All H2-antagonists tested inhibited both enzyme activities dose-dependently. The potency of inhibitory activity of H2-antagonists on acetylcholinesterase estimated from median inhibitory dose were in the following order of decreasing activity: ranitidine greater than TZU-0460 greater than cimetidine greater than YM-11170, whereas that on pseudocholinesterase were TZU-0460 greater than ranitidine greater than cimetidine greater than YM-11170. As the effects derived from the inhibition of acetylcholinesterase by H2-antagonists may affect intestinal motility, we studied ileal muscle contractions. Ranitidine had the most potent stimulating effect on contraction, the pattern of which was similar to physostigmine and was blocked by atropine and morphine. YM-11170 had a weak action on muscle contraction and cholinesterase activities.

Effect of teprenone on the content of phospholipids in gastric secretion in man.

The phospholipid content of basal and pentagastrin-stimulated gastric secretion was measured in 9 healthy volunteers. One week oral administration of teprenone did not alter the gastric acid and pepsin secretion. The phospholipid content was increased by teprenone. Changes in subclasses of phospholipids in gastric mucus by teprenone treatment, namely, increased phosphatidylcholine and decreased lysophosphatidylcholine, were observed.

Effect of intracerebroventricular injection of vasoactive intestinal polypeptide on systemic blood pressure and pancreatic and gastric secretions in the rat.

In order to examine the role of vasoactive intestinal polypeptide (VIP) in systemic blood pressure and pancreatic and gastric secretions in rats, the peptide was given to rats either intravenously (i.v.) through the femoral vein or intracerebroventricularly (i.c.v.) through the lateral ventricle. VIP, when given i.v., showed a systemic, hypotensive effect and a stimulatory effect on pancreatic secretion in urethane-anesthetized rats. When VIP was given i.c.v., there was no change in systemic blood pressure and pancreatic secretion in urethane-anesthetized rats, nor in gastric secretion both in urethane-anesthetized, stomach-perfused rats and in conscious, chronic gastric fistula rats. More research is needed to determine the central effect of VIP in digestive secretion.

Effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on interleukin 1 production by macrophages.

The effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on the in vitro functions of guinea pig macrophages were studied. A high dose (1 mg/ml) of EHDP inhibited interleukin 1 (IL 1) production by oil-induced peritoneal macrophages stimulated with muramyl dipeptide (MDP), lipopolysaccharide (LPS), phorbol myristic acetate (PMA), heat-aggregated IgG2 or calcium ionophore A23187. On the other hand, low doses (less than 0.125 mg/ml) of EHDP augmented the MDP induced IL 1 production by macrophages. This biphasic effect was also observed when macrophages were exposed to EHDP at 37 C for 24 hr and then stimulated with IL 1 inducers. Superoxide anion generation induced by formyl peptide or PMA was not affected by preincubation of the macrophages with doses of EHDP up to 1 mg/ml. Adherence and spreading of macrophages was inhibited by EHDP in a dose dependent manner without affecting cell viability. These results demonstrated that EHDP acted on macrophages directly and modulated IL 1 production in vitro.

Effect of intracerebroventricular injection of vasoactive intestinal polypeptide on systemic blood pressure, and pancreatic and gastric secretions in the rat.

In order to examine the central effects of VIP (vasoactive intestinal polypeptide) on systemic blood pressure, and pancreatic and gastric secretions in rats, the peptide was given to rats intracerebroventricularly (i.c.v.) through the lateral cerebroventricle. VIP, when given i.c.v., showed no effects on systemic blood pressure, and pancreatic and gastric secretions in rats. The present results indicate that VIP affects these functions not centrally but peripherally. Physiological role of VIP in the brain remains to be elucidated.

Immunohistological study with peroxidase-antiperoxidase staining in a case of generalized prepubertal periodontitis.

A case of prepubertal periodontitis was observed and examined immunohistologically with peroxidase-antiperoxidase staining. The patient was an 11-year and 7-month-old Japanese girl, well-developed and well-nourished. Her parents were first cousins. Her chief complaint was the loosening and loss of the permanent teeth. There was a similar history of primary dentition. Her remaining permanent teeth were loosened with severe alveolar bone loss, but calculus deposit was minimal. Significantly, there was no palmar-plantar hyperkeratosis. General examination showed normal data except for the increase of the immunoglobulin concentrations. In neutrophil function tests chemotaxis was depressed, although phagocytosis, random migration and superoxide production were within normal limits. Histologically, neutrophils were seen in the gingival tissue and other findings were also similar to those of adult periodontitis. In immunohistological examination, IgG-bearing cells which mostly consisted of plasma cells predominated in the lesion. Considering the past history, the immunodeficiency and the absence of palmar-plantar hyperkeratosis, the case was diagnosed as prepubertal periodontitis.
