Jaw cysts with special regard to keratocyst recurrence. A long-term follow-up.

From an original material of 50 patients with 55 jaw cysts, 39 patients were followed up 32--129 months after surgery (mean: 92 months). In 21 of 55 cysts the primary lesion was histologically classified as a keratocyst. All keratocysts had a follow-up period of at least 6 years (72 months). The keratocyst recurrence rate was 50%. Seven of eight recurrences in the total material were keratocysts. There was no difference between males and females concerning the incidence of keratocysts or age at diagnosis.

Ultrastructure and elemental composition of the stapedius muscle in guinea pig.

Muscules stapedius in guinea pig was investigated with regard to morphology and elemental composition. The muscle fibers are multinucleated, contain a high amount of ribosomes, and a centriole is often found adjacent to the cell nucleus. Intramitochondrial inclusion bodies occur adjacent to cristae mitochondriales. Both type I and type II fibres occur, the latter with all three subtypes. Elemental analysis from the myofibrillar area revealed unexpected low concentration of K and high concentrations of NA, P, and Ca.

Biochemical studies on the embryonic development of the mammalian inner ear in organ culture.

Adenylate cyclase activity and phospholipid labeling were compared during embryonic development of the mouse inner ear in vivo and in vitro. Inner ears were explanted on the 16th gestational day and cultured in vitro for 3-12 days. The gestation time in vivo is 21 days. During the 1st week in vitro there is very little growth of the inner ear with regard to total protein content. In contrast, the labyrinth increases its protein content threefold during the corresponding period of time in vivo. The activity of adenylate cyclase develops parallel in vivo and in vitro until the 19th gestational day whereafter the specific activity of the enzyme in vitro surpasses that of the enzyme in vivo three- to fivefold suggesting a lack of control mechanisms in organ culture. Phospholipids are labeled by 32P in an essentially similar quantitative relationship in vivo and vitro, while some quantitative differences exist. According to the present study the usefulness of the organ culture for the investigation of inner ear development appears limited to a culture period corresponding to an age prior to birth.

Hypernephroma metastasis in the pituitary gland. A case report.

In a 59-year-old man with a hypernephroma tumour of the right kidney, a metastasis in the pituitary gland of this neoplasm was diagnosed 9 years after removal of the kidney. However, metastases were also found in the left kidney. It cannot be established whether the metastasis in the pituitary gland came form the original tumour or from the other diseased kidney. To the best knowledge, a hypernephroma metastasis to the pituitary gland has not been reported previously.

Some new aspects on damages in the organ of Corti after pure tone exposure.

(1) The loss of hair cells after pure tone exposure at the critical level of intensity shows an exact correlation to the frequency, the time of sound exposure, and time of recovery as regards the quality and quantity. (2) The total extent of the damage of sensory cells has to be estimated higher than the hair-cell loss as observed under the light microscope. Although an improvement of the function of the organ of Corti was determined electrophysiologically from the 5th to the 10th day of recovery, the hair-cell degeneration was progredient during this period. (3) The hair-cell damage is apically more strongly marked from the area of damage than basically. (4) Depending on the experimental conditions there are considerable degenerative alterations in the nerve fibres and in the connective apparatus of the organ of Corti besides hair-cell losses and damages.

The energy dispersive X-ray microanalysis technique in the study of fluids and tissues of the brain and the inner ear.

The temporal bone from the fetal and adult mouse and brain from fetal and adult rat were cryosectioned in a conventional cryostat and -30 degrees C using 16 micrometers thick sections. The elemental composition was analyzed in the fluid-filled spaces of the membranous labyrinth and ventricular cavities of the brain. After drying these spaces containing small crystal-like aggregations of material which morphologically differed depending on their elemental composition. This changed during embryonic maturation. The qualitative X-ray analytical microscopy seems to be a reliable method to follow changes in the elemental composition of fluids.

Ototoxicity or teratogenicity. An analysis of drug-induced effects on the early development of the mammalian otocyst.

The 13th gestational day inner ear (otocyst stage) is particularly vulnerable to toxic influence from the environment during its further development. Ethacrynic acid in a concentration of 1 and 10 micrograms/ml caused complete arrest in development, and this was not overcome when withdrawing the drug after in vitro exposure to ethacrynic acid during 3 days. Gentamicin exposure at 1 and 10 micrograms/ml caused dysmorphogenesis but did not inhibit cytodifferentiation which, however, often was poor. Higher concentrations of gentamicin (100 micrograms/ml) had a more generalized toxic effect not only on epithelial but also on mesenchymal tissue.

Aneuploid DNA pattern in human pituitary adenomas.

The DNA contents of 29 histologically and ultrastructurally benign pituitary tumours were analyzed with flow-cytofluorometry.--Cytogenetic malignancy, i.e., aneuploidy, was found in 12 cases, independent of endocrinological type of tumour. In 17 cases a diploid DNA content occurred. In aneuploid tumours varying percentage of cells in S-phase was found indicating different growth patterns. Aneuploid DNA pattern was often found in young patients with a short case history. Our findings indicate that analysis of cellular DNA content may reflect the biological properties of pituitary tumours, independent of morphologic characteristics and type of hormone secretion.

Elemental analysis of the early postnatal tectorial membrane.

The analytical electron microscope has revealed that the elemental composition of the early postnatal tectorial membrane is principally the same before and after the development of endolymph. The elemental composition of the tectorial membrane does not give absolute information on the permeability of the membrane to ions such as potassium and chlorine from the endolymphatic space. Such a permeability is, however, likely when comparing the relative peak intensities of chlorine and potassium before and after the development of endolymph.

Adenylate cyclase activity in the fetal and the early postnatal inner ear of the mouse.

The adenylate cyclase activity was analyzed in fetal, early postnatal and adult inner ears of the CBA/CBA mouse and also in approximately one month old inner ears from Shaker -1 and Shaker -2 mice. A comparison was made with the maturation of potassium levels in endolymph as investigated with the X-ray energy dispersive technique. Adenylate cyclase activity in the developing normal inner ear shows two significant periods of increases: from the 16th to the 19th gestational day in both the cochlear and vestibular parts of the labyrinth, and from birth to day 6 after birth in the lateral wall tissues of the scala media. During the first period the anatomical boundaries of the secretory epithelia are developing. The postnatal rise in adenylate cyclase activity correlates with the morphological maturation of stria vascularis at the cellular and subcellular levels and the rise in potassium content of endolymph. The rise of enzyme activity in the cochlear during the maturation of endolymph supports a link between adenylate cyclase and the control of inner ear fluids. Adenylate cyclase activity in stria vascularis/spiral ligament of Shaker -1 and Shaker -2 mice were at normal levels and correlated better with the rather normal morphology of the tissues than the abnormal composition of endolymph in these mutants.

Clinical and morphological findings in two cases of bromocriptine-treated prolactinomas.

The morphology of two cases of prolactin (PRL) secreting pituitary tumours was analyzed at the light and electron microscopic level. Prior to surgery both patients had been treated with bromocriptine. In both tumors, groups of cells revealed considerable morphological changes but these were not confined to the whole adenoma. In fact, most cells still appeared ultrastructurally intact. Pieces from both adenomas were explanted to an in vitro system. These specimens secreted mainly PRL though smaller amounts of growth hormone (GH) were also produced. The cell morphology of the cultured specimens often showed a large number of preserved cells.

[Organ culture as a model for human pituitary tumour research (author's transl)].

The organ culture technique based on material obtained from individual human pituitary tumours represents a model for the preservation of tissue in organotypic differentiation in which the tissue can be studied in the absence of vascular, hypothalamic and other neurogenic influences. In the organ culture system, four criteria are fulfilled: (1) preserved morphology at the light and electron microscopic levels, (2) preserved synthesis/secretion of those hormones as found in vivo (3) the same reactivity in vitro to physiological stimulators/inhibitors (TRH, LH-RH and dopamine) as in vivo (4) preservation of the same cell phase in the cell cycle and the same DNA content in cells in vitro as in vivo.

Early morphological changes following gamma irradiation. A comparison of human pituitary tumours and human acoustic neurinomas (schwannomas).

The in vitro system was used to study early ultrastructural effects following gamma irradiation of human pituitary adenomas and neurinomas with 30 and 70 Gy single doses, respectively. The acoustic neurinoma cells showed a large number of intracellular myelin figures and vesiculation of cytoplasm. The pituitary tumour cells showed vacuolization of cytoplasm and cell organelles. Occasionally, electron-dense cytoplasmic inclusion bodies with a fibrillar substructure occurred. The differences between the two types of tumours in irradiation-induced morphological changes may indicate differences in their biochemical structure and metabolic activities. The morphological findings after irradiation of specimens from all four neurinomas were rather similar after the same length of time but the structural alterations of pituitary cells differed considerably between individual tumours. The 70 Gy dose caused more rapidly-occurring morphological alterations than the 30 Gy single dose.

99TcmO4 as an indicator for specialized cell function in organ cultures of the human submaxillary gland. A pilot study.

Salivary gland tissue was cultured in vitro and viable cells were present during 14 days. The specific glandular cell function was estimated from the capacity to accumulate 99TcmO4 from the culture medium. The morphology of the cultured specimens was examined by light and electron microscopy. Structural changes and loss of the 99TcmO4 accumulation capacity increased with increasing culture time in vitro. A correlation was observed between the loss of 99TcmO4 accumulation capacity and the disappearance of zymogen granules from the cultured cells. However, the causative link between these two phenomena requires further analysis.
