[Study on transfer of T-1982 (cefbuperazone) into female genital organs].

One gram of T-1982 (cefbuperazone), a new cephamycin antibiotic, was given intravenously to a total of 28 patients prior to abdominal total hysterectomy for uterine myoma or benign ovarian tumor. Bilateral uterine arteries were clamped at 0.25, 0.5, 1, 2, 4 hours or 6 hours after administration, and serum samples and uterine tissues were taken for the measurement of T-1982 concentration by bioassay method. A little difference was found in the serum concentration between cubital venous and uterine arterial serum, the half lives being 1.88 hours and 1.74 hours, respectively. The initial concentrations were estimated to be 99.9 micrograms/ml and 103.2 micrograms/ml, respectively. The peak concentrations were obtained at 30 minutes in the endometrium, myometrium, oviduct and ovary, and at 1 hour in the cervix uteri and portio vaginalis. These were 45.2 micrograms/g, 22.4 micrograms/g, 36.1 micrograms/g, 19.9 micrograms/g, 27.6 micrograms/g and 23.4 micrograms/g, respectively. The tissue concentrations thereafter decreased in the same manner as the serum concentrations. Judging from its favorable transfer into the uterine tissues and MICs against clinical isolates often seen in the field of obstetrics and gynecology, T-1982 was evaluated to be clinically useful in the treatment of obstetrical and gynecological infections.

Isolation of chloroquine-resistant Chinese hamster V79 cell variants that are also resistant to ammonium chloride.

Chloroquine-resistant (CQr) clones (CQ-21 and CQ-22) have been isolated from mutagenized hamster lung V79 cells by exposing the cells to a high dose of chloroquine. CQ-21 and CQ-22 showed about 3-fold higher resistance to chloroquine than the parental V79 cells, and they showed specific cross-resistance to another amine, NH4Cl, which is also concentrated in lysosomes. CQr clone showed no cross-resistance to other unrelated agents. Chloroquine-induced inhibition of [125I]ricin internalization was observed in both cell lines at neutral pH, but the inhibition of uptake was less in the variant. Also, the degradation of endogenous protein was slowed in the mutant; further, treatment of cells with 30 micrograms/ml of chloroquine inhibited the degradation of endogenous proteins in the parental V79, but not in CQ-22 cells. Similar levels of acid phosphatase, beta-glucuronidase and cathepsin D were observed in V79 and CQ-22 cells, but the level of cathepsin B was lower in the mutant. Electron microscopy showed an increased number of electron-dense bodies, possibly autophagosomes/lysosomes, in the mutant cells grown for 4 days with 5 micrograms/ml of chloroquine. Similar aberrant structures were observed in the parental V79 cells treated for only 3 h with 5 micrograms/ml of chloroquine.

[Experimental studies on regeneration of myoepithelial cells--the rat submandibular gland autografted in the tongue].

The regeneration process of myoepithelial cells in the injured salivary gland was studied. Fragments of the rat submandibular gland autografted in the tongue were examined from 12 hours to 14 days after implantation by means of the methods of autoradiography, histochemistry and electron microscopy. Two days after implantation, the central area of the fragments showed necrosis and parenchymal regeneration was, on the other hand, revealed at the remaining periphery. It was demonstrated by [3H] thymidine autoradiography that the regeneration of the intralobular parenchyma took place at the remaining intercalated and striated portions of the duct, but never did at the secretory endpieces. Initially, solid cell clusters were composed of regenerating uniformly immature polygonal epithelial cells. Four days after implantation, intracytoplasmic filaments running parallel to the plasma membrane of the cell, measured 50-70 A in diamter, were detected in some cells of the cluster. These filaments were accompanied by dense bodies characteristic of those of smooth muscle cells, and were thereby identified with myofilaments. Based on the findings, the regenerating cells with myofilaments were recognized as immature myoepithelial cells. Showing mosaic arrangement, they intermingled with the other kind of regenerating epithelial cells in the cluster. Each regenerating cell cluster formed gradually tubular structure with epithelial stratification. Ten days after implantation, most tubular structures were consisted of the single layer of columnar or cuboidal cells. Fourteen days after implantation, alkaline phosphatase activity characteristic of mature myoepithelial cells was observed in the spindle cells located between tubular epithelium and basal lamina. Ultrastructurally these spindle cells were identified with mature myoepithelial cells because of furnishing myofilaments with dense bodies. The results of this experiment suggest that, in case of tissue injury, the epithelial regeneration in the intralobular portion of the salivary gland is initiated at the intercalated and/or striated portions of the duct, and followed by myoepithelial differentiation of some of the regenerating epithelial cells.

[An electromyographic study on the canine gastric motility following segmental gastrectomy with selective proximal vagotomy].

In order to clarify the gastric motility after segmental gastrectomy (SG) with selective proximal vagotomy (SPV), seventeen canines equipped with four bipolar electrodes on the anterior wall of the stomach underwent four types of operation: 1-group, SG (middle corpus) with SPV-6 dogs; 2-group, SG (middle corpus) with SPV and pyloroplasty-5 dogs; 3-group, SG (upper corpus) with SPV-3 dogs; 4-group, SG (middle corpus) with antral vagotomy (AV)-3 dogs. 1-group: In the preanastomosis, the basic electrical rhythm (BER) frequency was not altered significantly, but the propagation velocity of the BER decreased by about 2 to 20% in the late postoperative period. In the postanastomosis, the BER frequency was markedly reduced, but progressively recovered to show the synchronization with that in the preanastomosis following 15 to 30 days postoperation. The propagation velocity of the BER decreased by about 3 to 25% in the late postoperative period. In the postanastomosis, dysrhythmias were observed to occur in high incidence in the early postoperative period, ut were transient occurrence. 2-group: The high-frequency-dysrhythmias (about 10 cycles/min) were observed. Pyloroplasty increased the frequency of dysrhythmias due to deranging the electric insulator of the pyloric ring. 3-group: In the late postoperative period, the BER frequency didn't synchronize with that in the preanastomosis in fasting. 4-group: Dysrhythmias were observed to occur in the late postoperative period. It was suggested that the gastric motility of the SG with SPV recovered in the late postoperative period, therefore pyloric ring can be preserved.

Dose-dependent excretion of DDE (1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene) in rats.

Dose-dependent excretion of p,p'DDE in rats was investigated. p,p'DDE itself was the major excreta in rats. But some o,p'isomer of DDE was detected in feces by GC-MS analysis. The excretion of p,p'DDE after a single administration was modified by its dose level. The time pattern of p,p'DDE excretion agrees well with the modified Hill equation. The value of the equilibrium constant (K') increases in proportion to time t after p,p'DDE administration. Using the modified Hill equation and the linear K' equation, the excretion rate of p,p'DDE during the experimental time t can be estimated. The estimated p,p'DDE excretion rate in feces agrees well with the measurements.

Studies on the effect of dietary protein and fat content upon DDT metabolism in rat liver.

Rats were supplied with 25 kinds of food, which were divided into 5 classes of protein and 5 classes of fat content, to examine the effect of dietary protein and fat on the metabolism and retention of DDT in the liver. The results suggested that dietary protein and fat changed DDT and its metabolites concentration in liver. The concentration of DDT decreases according to the increase of dietary protein content. The concentration of DDT and its metabolites in liver increases when the dietary fat content increases. Polynomial and multiple regression analyses were carried out to confirm the effect of dietary protein and fat on DDT metabolism. The results suggest that the residual concentration of DDT and its metabolites (DDD and DDE) is a function of dietary protein and fat content, and can be represented in equation form. The estimation of the concentration of DDT and its metabolites from the equation agrees well with the measured concentration in liver.

Subcellular localization of the methylcitric-acid-cycle enzymes in propionate metabolism of Yarrowia lipolytica.

The subcellular localization of the four characteristic enzymes of the methylcitric acid cycle was studied with glucose-grown as well as n-alkane-grown cells of Yarrowia lipolytica. Microsomes and peroxisomes showed no cycle enzyme activities. The four cycle enzymes were constitutively localized in mitochondria, with the exception of the dual localization of the fourth enzyme, 2-methylisocitrate lyase, in mitochondria and cytoplasm, where the lyase may function to supply pyruvate (the end-product of the catabolism of the propionate residue) to various reactions.

Angiocardiographic study of coarctation of the aorta--morphology and morphogenesis.

Angiocardiographic morphology and associated cardiac anomalies of the coarctation of the aorta (COA) were analysed in the light of recently advanced theory of its morphogenesis. Analysis of 154 clinical cases of COA revealed the presence of associated cardiac anomalies which could obstruct fetal blood flow to the ascending aorta in many cases. Systolic left ventricular outflow tract was significantly narrow in cases of ventricular septal defect (VSD) associated with COA in comparison with the isolated VSD (p less than 0.01). Good quality aortograms obtained from 41 cases of COA were analysed. Ages ranged from one month to 20 years and major cardiac anomalies were associated in 25 cases. Age-matched 37 cases were studied as the controls. In COA, the isthmus was narrower than the controls in 88% of the cases, and in the remaining it was low in the normal range. Severely hypoplastic isthmus in early infancy became the adult type after 5 years in one of the 4 cases which had repeated aortography. Localized stenosis at the distal end of isthmus was present in 89%, and absent in 11% (4 cases). In these 4, isthmal narrowing was associated with major intracardiac anomalies, large patent ductus arteriosus (PDA) and pulmonary hypertension (PH). The anterior ostium to the descending aorta was obstructed by constricted ductus arteriosus (PDA) and pulmonary hypertension (PH). The anterior ostium to the descending aorta was obstructed by constricted ductus arteriosus in some infants with COA. These angiographic findings are compatible with the following working hypothesis: ischemic hypoplasia results from prenatal decrease of blood flow to the ascending aorta, and localized stenosis is related to abnormal distribution of the ductal tissue to the descending aorta and post-natal constriction of the ductus arteriosus. In 3 infants COA was associated with persistent fifth aortic arch.

Inhibitory effect of acephate (N-acetyl O, S-dimethyl thiophosphoramide) on serum cholinesterase--effect of acephate on cholinesterase.

The Lineweaver-Burk plot of the activity of human serum cholinesterase against the concentration of butyrylthiocholineiodide was shown by two intersecting lines. The Hill plot of cholinesterase activity was linear over the entire range of the substrate concentration. The n value, an interaction coefficient, was less than 1.0 (about 0.8). These results suggest that cholinesterase has multiple substrate binding sites. Acephate, one of the organophosphorous insecticides, inhibited the activity of cholinesterase. Acephate at concentration under 1.25 mM (about 230 ppm in serum) did not inhibit the activity of cholinesterase. The minimum concentration of acephate inhibition of cholinesterase activity was at 2.5 mM. An equilibrium constant(K) can be used as an indicator of inhibitory effect on cholinesterase. The serum cholinesterase activity of workers who were exposed to acephate is not affected when the concentration of acephate in serum is less than 200 ppm. This result suggests that the activity of serum cholinesterase is not an accurate indicator of the exposure of the low toxic insecticides, e.g. acephate. The inhibitory effect of acephate on cholinesterase decreased after the incubation with S-9 mixture. This result suggests that a part of acephate is metabolized to inactive substances in the liver.
