Regulation of ion and water transport across the eel intestine: effects of acetylcholine and serotonin.

Both acetylcholine (ACh) and serotonin (5-HT) lowered the serosa-negative transepithelial potential difference (PD) and the short-circuit current (Isc), accompanied by a decrease in NaCl and water absorption across the eel intestine. These inhibitory effects of ACh and 5-HT were blocked by atropine, a muscarinic receptor antagonist, and ICS-205930, a 5-HT3 receptor antagonist, respectively. Even after blocking the ACh receptor with atropine, 5-HT inhibited the PD and Isc, and ACh lowered them after blocking the 5-HT receptor with ICS-205930, indicating that ACh and 5-HT act independently. Similar inhibition in the PD and the Isc was observed after electrical field stimulation (EFS) which is expected to release endogenous regulators. These effects of EFS were reduced by 70% after simultaneous addition of atropine and ICS-205930. Since atropine and ICS-205930 block ACh and 5-HT receptors, respectively, these results suggest that endogenous ACh and 5-HT are released by EFS.

Study on suspended particulate matter and polycyclic aromatic hydrocarbons in indoor and outdoor air.

To study changes in the concentration of suspended particulate matter (SPM) in indoor and outdoor air, a new portable sampler (AND sampler) was designed that could separately collect particles exhibiting aerodynamic diameters of greater than 10 microns, 2-10 microns, and less than 2 microns. The relationship between the airborne particle concentration in indoor air and that in outdoor air varied with the aerodynamic diameter of the particles. The concentration of SPM in the indoor air increased in proportion to that in the outdoor air. The concentration of polycyclic aromatic hydrocarbons (PAH) in SPM also varied with the aerodynamic diameter of the particles. Fine particles exhibiting an aerodynamic diameter of less than 2 microns contained high concentration of PAH. The concentrations of benzo(k)fluoranthene, benzo(a)pyrene, and benzo(ghi)perylene in indoor air increased in proportion to those in outdoor air.

Magnetic resonance imaging of the brain in congenital rubella virus and cytomegalovirus infections.

Two children with congenital rubella virus and six with cytomegalovirus (CMV) infections, were examined by magnetic resonance (MR) and CT. Cranial MR imaging (MRI) with T2-weighted spin-echo (SE) and inversion recovery (IR) sequences demonstrated the following: periventricular hyperintensity (4), subcortical hyperintensity (5), delayed myelination (4), oligo/pachygyria (2), cerebellar hypoplasia (2). This study showed that the more-disabled children had more marked abnormal MRI findings. MRI was more effective in the detection of parenchymal lesion than was CT, although intraventricular calcification was better visualized with CT.

Magnetic resonance imaging in a case of mumps postinfectious encephalitis with asymptomatic optic neuritis.

A 5-year-old male patient with asymptomatic optic neuritis and mumps postinfectious encephalitis or acute disseminated encephalomyelitis is reported. Magnetic resonance imaging (MRI) with a short inversion time inversion recovery sequence was valuable in detecting clinically silent lesions of the unilateral right optic nerve in addition to visual evoked potentials. Evidence of concurrent optic neuritis was useful for detecting more extensive neurological involvement, leading to the diagnosis of mumps postinfectious encephalitis. A systematic MRI study should be performed in children with mumps encephalitis, regardless of appreciable clinical deficits.

Hypoplastic left heart syndrome with rhabdomyoma of the left ventricle.

A case of primary cardiac tumor with hypoplastic left heart syndrome (HLH) is presented. Sonographic examination at 32 weeks' gestation revealed a large tumor in the left ventricle of the fetal heart. The newborn died of congestive heart failure at 11 days of age. Autopsy demonstrated a large tumor in the left ventricle obliterating most of the left ventricular cavity, aortic atresia, and hypoplastic ascending aorta. Microscopically, the cardiac tumor showed "spider-cells" characteristics of rhabdomyoma.

Difference in the phenotypes of bronchoalveolar lavage lymphocytes in patients with summer-type hypersensitivity pneumonitis, farmer's lung, ventilation pneumonitis, and bird fancier's lung: report of a nationwide epidemiologic study in Japan.

We performed a nationwide epidemiologic study of hypersensitivity pneumonitis (HP) in Japan by questionnaire and found that 835 cases of HP were recognized during the 1980s; 74.4% were summer-type HP, 8.1% farmer's lung, 4.3% ventilation pneumonitis, 4.1% bird fancier's lung, 2.3% other types, such as chemical worker's lung, and 6.8% of unknown causative agent. It was found that the CD4/CD8 ratios of bronchoalveolar lavage (BAL) lymphocytes were significantly different with the type of disease. The ratio was 0.6 +/- 0.1 (mean +/- SEM) in summer-type HP (N = 271), 4.4 +/- 0.7 in farmer's lung (N = 22), 1.6 +/- 0.3 in ventilation pneumonitis (N = 19), and 2.0 +/- 0.5 in bird fancier's lung (N = 19). In farmer's lung, the CD4/CD8 ratio in smokers was 6.2 +/- 1.9 (N = 6) in contrast with 3.4 +/- 0.7 for nonsmokers (N = 16) (p less than 0.05). It has been generally considered that the phenotypes of BAL lymphocytes in patients with HP are predominately CD8 cells. Our present results, however, indicate that the phenotypes of BAL lymphocytes vary with the type of HP, probably depending on factors such as causative agent, smoking, or staging of the disease.

Stimulation by acetylcholine and inhibition by norepinephrine of transglutaminase activity in superior cervical ganglia excised from adult rats.

Very rapid changes in activity of transglutaminase (TG), a calcium-dependent enzyme contributing to cross-linkage formation of intracellular polypeptide chains, were observed in vitro in rat superior cervical ganglion (SCG) and nodose ganglion (NG) following application of cholinergic or adrenergic agonists and antagonists. In SCG, a tissue rich in synapses, the depolarizing agent acetylcholine (ACh, 0.1 mM) produced an 8.7-fold increase in TG activity within 5 min that lasted for 30 min and returned to control levels by 2 h. In contrast, the ACh-induced increase in TG activity in NG, a tissue containing neuronal cell bodies with few synapses, was more gradual and of smaller magnitude, reaching a peak of approximately 2.4 times control by 30 min that was maintained for at least 2 h. In both tissues the ACh-stimulation was effectively blocked by the nicotinic antagonist, hexamethonium (0.1 mM), whereas the muscarinic antagonist, atropine (0.1 mM), partially blocked the ACh effect in SCG and was without effect in NG. Addition of the hyperpolarizing adrenergic agonists norepinephrine (NE, 50 microM), isoproterenol (0.2 mM) or dopamine (0.1 mM) produced an inhibition of TG activity in SCG but had no effect in NG. The inhibitory effects of the adrenergic agonists in SCG were blocked by the beta-adrenergic antagonist, propranolol (10 microM) and alpha 2-adrenergic antagonist, yohimbine (10 microM). A kinetic study revealed that the ACh-induced stimulation of TG activity in SCG and NG was a result of decrease in apparent Km and an increase in Vmax value, whereas the NE-induced inhibition of SCG enzyme activity was a result of an increased Km and decreased Vmax. 45Ca2+ influx into excised SCG or NG was significantly reduced by the application of either ACh or NE. The ACh inhibition was effectively blocked by either hexamethonium or atropine. The NE inhibition was more effectively blocked by yohimbine than by propranolol. These results suggest that the rapid alterations of TG activity in SCG produced by cholinergic and adrenergic neurotransmitters are attributable to the processes of receptor-mediated depolarization and hyperpolarization, respectively, via modulation of nerve-impulse-induced Ca2+ fluxes during synaptic activity.

Rupture of the extensor digitorum communis tendons caused by occupational overuse.

Spontaneous rupture of the extensor pollicis longus tendon in excessive exertion is well-recognised, but similar rupture is rare in the common extensor tendons of the fingers. Two cases caused by occupational overuse are reported. Mechanical attrition at the distal edge of the extensor retinaculum produced by excessive use was thought to be responsible for the rupture.

Effect of maturation on activities of various proteases and protease inhibitors in the muscle of Ayu (Plecoglossus altivelis).

1. Increases in activities of muscle muticatalytic proteinase, modori-inducing proteinase (latent trypsin-like proteinase), cathepsin B and L-like proteases and cathepsin D were observed more markedly for male fish than female fish, in the spawning stage. 2. Decreases in inhibitory activities of muscle serine and cysteine protease inhibitors were observed more markedly for male fish than female fish in the spawning stage.

Gonadotropin-releasing hormone agonists induce meiotic maturation and degeneration of oocytes in the in vitro perfused rabbit ovary.

The present study was undertaken to assess the effects of gonadotropin-releasing hormone agonists (GnRH-a, buserelin and leuprolide acetate [LA]) on ovulation, oocyte maturation and degeneration, and steroid and prostaglandin production in the perfused rabbit ovary preparation. Ovulation did not occur in any of ovaries treated with buserelin or LA (10(2) to 10(4) ng/mL) in the absence of gonadotropin. Gonadotropin-releasing hormone agonists were associated with the resumption of meiosis in follicular oocytes in a dose-related manner. Furthermore, the addition of GnRH-a to the perfusate significantly increased the percentage of follicular oocytes that showed evidence of degeneration compared with contralateral untreated or human chorionic gonadotropin-treated controls. Prostaglandin E2 and prostaglandin F2 alpha production by the perfused rabbit ovaries were stimulated significantly by GnRH-a treatment. Exposure to GnRH-a failed to increase either progesterone or estradiol production by the perfused rabbit ovaries. These data demonstrate that GnRH-a act directly in the rabbit ovary to trigger meiotic maturation in oocytes within the follicles, concomitantly increasing oocyte degeneration.

Treatment of uterine leiomyomata with a luteinizing hormone-releasing hormone agonist: the possibility of nonsurgical management in selected perimenopausal women.

OBJECTIVE: To evaluate the efficacy of luteinizing hormone-releasing hormone agonist (LH-RH-a) in the treatment of leiomyomata. DESIGN: A retrospective randomized trial. SETTING: Hospital department of obstetrics and gynecology. PATIENTS: Twenty-five women, ages 36 to 54 years with symptomatic uterine leiomyomata, were divided into two groups according to the responsiveness to LH-RH-a: group A patients reached menopause after LH-RH-a, whereas resumption of menstruation occurred within 12 weeks after cessation of therapy in group B. INTERVENTIONS: Luteinizing hormone-releasing hormone agonist was administered intranasally three times a day with 150 micrograms insufflation of one spray in each nostril (total dose: 900 micrograms/d). MAIN OUTCOME MEASURES: Efficacies of treatment were assessed in terms of uterine volume, hemoglobin concentrations, serum levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol (E2), and bone density during and after treatment. RESULTS: In both groups, hemoglobin concentrations increased significantly after 16 weeks of treatment. A significant reduction in uterine volume was observed in both groups. After completing therapy, there was no further significant change in uterine volume in group A, whereas uterine volume in group B returned to pretreatment values. Serum LH and FSH concentrations were suppressed during treatment, but those gonadotropins in group A increased significantly up to the menopausal levels after treatment. Serum E2 concentrations in both groups showed consistent suppression by the end of the first treatment cycle. After cessation of therapy, serum E2 levels on group A remained in the castrate range, whereas E2 in group B returned to pretreatment levels, concomitant with the return of normal ovulation. CONCLUSIONS: Intranasal administration of LH-RH-a was successful in significantly decreasing uterine volume and increasing hemoglobin concentration in premenopausal women with leiomyomata.

Lumbosacral sympathetic trunk as a compensatory pathway for seminal emission after bilateral hypogastric nerve transections in the dog.

Seminal emission after transection of canine bilateral hypogastric nerves was investigated. Stimulation of severed hypogastric nerves caused full volume seminal emission (greater than four drops) in all six dogs examined. In contrast, stimulation of sympathetic trunks of other six dogs at the level of caudal mesenteric artery caused no emission in three dogs and a limited volume of seminal emission (less than one drop) by pressing the ampulla in the remaining. When hypogastric nerves were transected one month prior to the experiment, however, stimulation of sympathetic trunks in the same manner caused full volume of seminal emission in all six dogs. Distal dissection of sympathetic trunks demonstrated nerve fibers to the seminal tract via the pelvic splanchnic nerves. The results indicate that the seminal tract receives at least two sympathetic pathways; one from hypogastric nerves and the other from lumbosacral sympathetic trunks, and that the latter compensatorily generates seminal emission after hypogastric nerve transection.

Autoantigens for IgA anti-intercellular antibodies of intercellular IgA vesiculopustular dermatosis.

A new disease characterized by the presence of in vivo bound and/or circulating IgA anti-intercellular (IC) antibodies has recently been identified. We propose the term intercellular IgA vesiculopustular dermatosis (IAVPD) for this entity, which seems to be divided clinicopathologically into at least two distinct subtypes: intraepidermal neutrophilic IgA dermatosis (IEN type) and subcorneal pustular dermatosis-like cases (SPD type). Using immunoblot technique, we examined the antigen substances for the IgA anti-IC antibodies in the sera from one Japanese patient with IEN type of IAVPD and three Japanese patients with SPD type. A serum from a patient with IEN type reacted exclusively with a 120-kD protein in both the normal human skin extract and the bovine desmosome sample. Sera from three patients with SPD type reacted specifically with a doublet of 115-kD and 105-kD proteins, which appeared to be identical to desmocollins I and II, well known desmosomal core proteins, in the bovine desmosome sample. IgA antibody from our patients with IAVPD bound to neither pemphigus vulgaris antigen nor pemphigus foliaceus antigen. From these results, we suggest that IAVPD is different from pemphigus and is heterogeneous in terms of the antigens to which IgA autoantibodies bind.

Higher frequency of point mutations in the c-K-ras 2 gene in human colorectal adenomas with severe atypia than in carcinomas.

Human colorectal carcinomas may be induced from adenomas or they may occur de novo. To examine which is the main pathway, we analyzed point mutations at codon 12 in the c-K-ras 2 gene in 73 colorectal carcinomas, 13 metastatic tumors, 72 adenomas and 30 normal tissues. The c-K-ras 2 codon 12 mutation frequency was 0/30 in normal tissues, 0/17 in adenomas with mild atypia, 3/37 (8.1%) in adenomas with moderate atypia, 15/18 (83.3%) in adenomas with severe atypia, 19/73 (26.0%) in primary carcinomas and 3/13 (23.1%) in metastatic tumors. The mutation frequency in adenomas with severe atypia was much higher than that in carcinomas. These results indicate that many colorectal carcinomas may not be induced through adenomas with severe atypia.

Seminal emission by electrical stimulation of the spermatic nerve and epididymis.

The spermatic nerve and epididymis were stimulated electrically in dogs to elucidate the possibility of artificial seminal emission after bilateral transection of the hypogastric nerves and sympathetic trunks. Before transection, electrical stimulation of a distal end of the severed spermatic nerve caused a trace amount of emission in two dogs and no emission in the remaining four. In contrast, 1 month after the transection, stimulation of a distal end of the severed spermatic nerve caused seminal emission in all six dogs examined, with full seminal volume in four dogs and partial volume in the remaining two. Anatomically, sympathetic nerves originating from the upper portion of the lumbar sympathetic ganglia descended along the spermatic arteries to the testes as spermatic nerves. The present results indicate that spermatic nerves have the potential to generate seminal emission as a compensatory pathway after bilateral transection of the hypogastric nerves. Both direct and percutaneous electrical stimulation of epididymal tails resulted in a full volume of seminal emission in all dogs with transection of both hypogastric nerves and lumbosacral sympathetic trunks as well as in unoperated controls, while high voltage (8 V vs 40-80 V) was required to cause seminal emission by electrical stimulation on the skin surface. Direct stimulation of epididymal tails in men undergoing orchidectomy as treatment for prostatic carcinoma or during biopsy of the contralateral testis in a patient with a testicular tumour, resulted in seminal emission in all five epididymides examined either from the end of the severed vas deferens or in the posterior urethra if the vas deferens was not severed.

Stimulation of transglutaminase activity by GM1-ganglioside and alpha-sialylcholesterol in superior cervical and nodose ganglia excised from adult rat.

Changes in transglutaminase (TG) activity in superior cervical ganglia (SCG) and nodose ganglia (NG) excised from adult rats were examined following application of selected membrane transport-altering agents, including GM1-ganglioside (GM1) and alpha-sialylcholesterol (alpha-SC). Although TG activity of freshly dissected SCG and NG was relatively low, it increased gradually during 30 min of incubation, and it stayed at this elevated level for 2 h. Addition of alpha-SC at its maximal effective concentration, 20 microM, stimulated TG activity more than eightfold in SCG and more than twofold in NG by 30 min. Addition of GM1 at its most effective concentration, 5 nM, had similar effects, but of lesser magnitude. Cycloheximide, a potent inhibitor of protein biosynthesis, did not affect the GM1- or alpha-SC-evoked increases in ganglionic TG activity, suggesting that enzyme activation rather than synthesis of new enzyme was occurring. The stimulation of TG activity in both ganglia caused by either GM1 or alpha-SC was associated with a decrease in Km and an increase in Vmax values. Addition of cholera toxin B, which specifically masks the oligosaccharide chain of GM1, reduced the GM1-induced increase in TG activity by approximately 60% in SCG and 88% in NG. The alpha-SC-induced increase in TG activity was only partially mimicked by free cholesterol. Although application of either dibutyryl cyclic AMP or dibutyryl cyclic GMP produced little change in TG activity of either ganglion, phorbol ester clearly inhibited the enzymic activity. Because TG is a calcium-dependent enzyme, we measured 45Ca2+ influx into either ganglion, and found that it was reduced by GM1 and alpha-SC in SCG and by alpha-SC in NG.(ABSTRACT TRUNCATED AT 250 WORDS)

Loss of response to thyroid hormone in developing nervous tissue correlates with enhanced synthesis of a 25-kilodalton stress protein.

Directly following transfer to in vitro conditions, lumbar dorsal root ganglia (DRG) from triiodothyronine (T3)-treated premetamorphic tadpoles showed an approximately three fold increase in incorporation of bath-applied radiolabeled amino acid relative to DRG from vehicle-treated tadpoles. By contrast, at 1 h after transfer, no increased incorporation was detected. At this time, examination of in-vitro-synthesized [35S]proteins by two-dimensional gel electrophoresis revealed a selective enhancement of a slightly acidic 25-kilodalton (kD) polypeptide. Addition of 1 M glycerol to the incubation medium prevented the enhanced synthesis of the low-molecular-weight polypeptide and preserved the T3 response. When DRG were exposed in situ, the T3 response - monitored following intraganglionic injection of [35S]methionine-was detected after 2 h, but not after 4 h. Four hours were also the earliest time at which an elevated level of the 25-kD polypeptide was observed. The correlation between loss of hormonal responsiveness and appearance of a putative stress protein is of general importance as a cautionary note for in vitro studies of endocrine effects on developing tissues.
