Distinct surface phenotypes of B cells responsible for spontaneous production of IgM and IgG anti-DNA antibodies in autoimmune-prone NZB x NZW F1 mice.

Autoimmune-prone NZB x NZW F1 (B/W F1) mice produce a high titer of anti-DNA antibodies, In vivo and in vitro studies showed that in the early life of these mice, the immunoglobulin isotype of these antibodies almost exclusively belongs to IgM class, however, IgG anti-DNA antibodies begin to develop when the mice are about 5-6 months old and the titer exceeds that of IgM antibodies from age 7 months on. We asked whether or not the B cell population responsible for IgM and IgG antibody production belongs to the same lineage. The surface phenotypes of B cell populations responsible for the spontaneous production of either IgM or IgG anti-DNA antibodies were examined using panning and sorting methods with several monoclonal antibodies to B cells, including CD5 (Ly-1) and Lp-3; the latter defines a unique B cell differentiation antigen. We obtained evidence that surface phenotypes of B cells secreting IgM anti-DNA antibodies belong to CD5+ Lp-3- and those of B cells secreting IgG anti-DNA antibodies which occur only in old B/W F1 mice belong to CD5- Lp-3+ subpopulations. The majority of peritoneal B cells were CD5+ Lp-3+ throughout the life span of the mice and anti-DNA antibody production was never evidenced. These findings were discussed in relation to age-associated changes of B cell populations in the spleen of this strain of mice.

Spontaneous resolution of multiple hemangiomas of the brain. Case report.

A case of multiple intracerebral hemangiomas is reported. The lesions were angiographically hypervascular and accompanied by marked perifocal brain edema. After removal of the main lesion and postoperative steroid administration, the rest of the lesions resolved spontaneously. Histologically, the lesion was composed of capillaries and fibromyxoid stroma. The distinctive clinical, radiological and histological features of this lesion are discussed in contrast to several other types of intracranial vascular tumors.

Mineral and trace element content of human transitory milk indentified with inductively coupled plasma atomic emission spectrometry.

Mineral and trace element content of the human transitory milk collected on days 6 to 9 postpartum in Tokyo was measured by inductively coupled plasma atomic emission spectrometry (ICP-AES). Ten elements (Na, Mg, P, S, K, Ca, Fe, Cu, Zn, and Sr) were detected in ten-fold diluted milk sample with this method. With the stepwise multiple regression analysis, maternal and infants' biological attributes, such as weight, stature, sex, or age, were noted as factors contributing to the interindividual variation in Na, Mg, P, S, and Sr. Biological mechanism responsible for this selection cannot be found. Days postpartum for the milk collection was selected as a highly significant (p less than 0.001) variable with negative coefficient to explain milk S variation.

[An endoscopy-guided plan of radiotherapy for detecting esophageal cancer using a lugol staining].

The Lugol staining method has lately come to play an important role in the detection of superficial esophageal cancers which are extremely difficult to detect by using only a barium double-contrasted esophagiogram and conventional esophagioscopy. Thus we have tested an endoscopy-guided plan of radiotherapy for detecting an esophageal cancer using Lugol stain. This method was found useful in detecting intraepithelial small lesions, such as intramural metastasis or multiple origin cancers. Additionally, it has been found useful in determining the definite shape of superficial malignant lesions by evaluating biopsy specimens taken from the border of the non-stained regions.

Central sympathoinhibitory action of a new type of alpha-1 adrenoceptor antagonist, YM-617, in rats.

The structurally new type of alpha-1 adrenoceptor antagonist, YM-617 [R-(-)-5-[2-[(2-(o-ethoxyphenoxy)ethyl]amino)propyl]-2- methoxybenzenesulfonamide hydrochloride], is a phenethylamine derivative which is similar structurally to the catecholamines. The present study was undertaken to elucidate the effect of YM-617 on sympathetic nerve activity and baroreceptor afferent nerve activity in anesthetized rats. Intravenous administration of YM-617 (2, 10 and 50 micrograms/kg) produced a dose-dependent reduction in mean arterial pressure accompanied with bradycardia. YM-617 caused dose-dependent decreases in renal sympathetic nerve activity along with this hypotension. Cardiac sympathetic nerve activity, preganglionic adrenal nerve activity as well as aortic depressor nerve activity was decreased by YM-617. When an equihypotensive dose of YM-617 and a centrally acting antihypertensive drug, clonidine, were compared, the sympathoinhibitory potency of YM-617 was less than that of clonidine. These findings suggest that YM-617 might possess a central sympathoinhibitory action which could play a role in its antitachycardic or bradycardic effect and could be partially responsible for its hypotensive action.

Inhibitory effect of 2,3-dibenzylbutane-1,4-diol; a mammalian lignan, on the CA2+ channel in rabbit femoral artery.

2,3-Dibenzylbutane-1,4-diol (DBB) is a mammalian lignan derived from human urine. To investigate the pharmacological actions of mammalian lignans, the effects of DBB on high K(+)-induced contraction in rabbit femoral artery were studied. High K+ induced a transient contraction followed by sustained contraction, and both depended on extracellular Ca2+. DBB inhibited transient contraction as well as sustained contraction. Verapamil or nifedipine inhibited sustained contraction more effectively than transient contraction. DBB inhibited the transient contraction remaining in the verapamil- or the nifedipine-pretreated preparation in a concentration-dependent manner. Moreover, DBB inhibited sustained contraction elicited by Bay K 8644 (Ca2+ channel activator). These results indicate that DBB effects not only the sensitive Ca2+ channels but also the insensitive channels.

A case of fibrous hamartoma of infancy: an immunohistochemical study.

To clarify the histogenesis and immunohistochemical nature of fibrous hamartoma of infancy, we have studied a typical case of this disease using an immunohistochemical technique with a large panel of antibodies. It was found that immature-appearing stellate and spindle-shaped cells were mesenchymal in nature, but it was not ascertained whether those cells were differentiated into various specialized connective tissues corresponding with embryonic stages of connective tissue differentiation. These results reveal the immunohistochemical nature of tissue components of fibrous hamartoma but poorly support the previously described concept that fibrous hamartoma of infancy is a hamartomatous disease.

[A case of Becker muscular dystrophy with schizophrenic symptoms].

A 23-year-old male patient with Becker muscular dystrophy (BMD) who showed schizophrenic symptoms was reported. He tumbled easily and was poor at running since age at 8 years. He had difficulty in climbing stairs and was idle away all day long since age at 21 years. Although his premorbid personality was not schizoid, he showed auditory hallucinations and delusions without any psychogenetic moment at the age of 23. At first, he seemed to be schizophrenic, but after the treatment with antipsychotics, he always had an insight into his disease and exhibited natural emotional communication. He showed no autism and character changes. According to the Wechsler Adult Intelligence Scale (WAIS), intellectual impairment was notified (total IQ58). Neurological examinations revealed weakness and atrophy of muscles in the proximal part of his lower extremities, and pseudohypertrophy of calves. In the serum enzyme, serum creatine kinase (CK) level was elevated (700 U/L). Abnormal Q waves appeared in the leads, II, III, aVF, V5, V6 on the electrocardiogram (ECG), and the finding of the echocardiography suggested dilated cardiomyopathy. The electroencephalogram (EEG) revealed the basic rhythm of 9-10 Hz with 0 activities of 6-7 Hz which were predominant in frontparietal and central leads. The electromyogram (EMG) revealed a myopathic pattern with low voltage and short duration. A muscle biopsy from right biceps brachii disclosed the abnormal immunofluorescent staining pattern of dystrophin which is consistent with BMD patient, i.e., "patchy," discontinuous and faint immunoreaction at surface membrane of the fiber. Both molecular weight (380 kd: n = 400) and amount (30%; n = 100) of dystrophin were reduced.(ABSTRACT TRUNCATED AT 250 WORDS)

Fine structure of the inner enamel in human permanent teeth.

Using SEM after EDTA etching, the mid-coronal inner enamel of human permanent teeth was classified into three regions of the 1st, 2nd, and 3rd zones. The 1st zone showing a highly negative birefringence was the innermost 10 - 15 microns enamel. This zone consisted of arcade and circular initial prisms, and the succeeding arcade prisms only. These initial prisms arising perpendicularly to the dentine surface resembled pseudoprisms because these prisms showed a somewhat centripetal arrangement of crystallites and indistinct prism boundaries. The succeeding prisms were frequently bent following a faint slit within the prism. The 2nd zone adjacent to the 1st zone measured 20 - 40 microns in thickness. This zone was mainly composed of horseshoe-shaped prisms with EDTA-insoluble prism sheaths in the deep-etched prism boundaries, but the inner-half layer had dotted irregular prisms including circular, double marginal, and spiral shapes with the prism sheaths. Prismless structures were rarely seen in the 2nd zone. The 3rd zone was mainly occupied by horseshoe-shaped prisms without EDTA-insoluble prism sheaths in the deep-etched prism boundaries, although tuft prisms in the 3rd zone contained a large amount of EDTA-insoluble substances in the prisms, interprismatic regions, and the boundaries.

A new immunization protocol for the production of monoclonal antibodies specific for VL subgroups of human immunoglobulins.

Hybridomas producing monoclonal antibodies (Mabs) against the variable (V) region of human immunoglobulin light (L) chains have been difficult to establish in most cases. After fusion of myeloma cells with spleen cells obtained from mice immunized with VL fragments or L chains by conventional protocol using Freund's complete adjuvant (CFA), no hybridomas secreting Mabs specific for the VL subgroup have been obtained. In order to produce such Mabs, we have developed a novel immunization protocol consisting of the use of monophosphoryl lipid A and trehalose dimycolate for primary and secondary immunization courses and an intrasplenic injection for a final booster. A number of specific Mabs were successfully obtained and examples of clinical application for such Mabs was demonstrated by flowcytometry.

[Microflora around the bracket by direct bonding system].

The purpose of the present study is to clarify the influence of microflora in plaque of the brackets edge by direct bonding system and the cause of caries that is induced around the bracket during orthodontic treatment. The subjects were selected from the patients undergoing direct bonded orthodontic treatment and from the non-orthodontic patients. They were divided into four groups: those having no orthodontic treatment (control group), those having no caries around the bracket (group A) and those having caries around the bracket (groups B and C). Sample plaques were collected from the initial tooth surface with control group, from the tooth surface around the bracket with group A, from the carious cavity around the bracket with group B and from the non-carious tooth surface around the bracket with group C. The isolation rates of microflora in dental plaque of the four groups were compared, and the following conclusions were obtained. 1. Group A had almost same level with control group as to the proportion of each bacterium examined by shape and gram stain of microbiota. Group B had higher level of gram positive cocci than the other groups. 2. Group A and control group had no difference as to the isolation rates of genera Streptococcus, Actinomyces and Lactobacillus. 3. Group A had low level of S. mutans, was the same as control group. 4. The isolation rate of each genus: 1) The isolation rate of genus Streptococcus was almost same in groups A, C and control group. Group B had higher than the other groups. 2) The isolation rate of genus Actinomyces was the highest in all groups. 3) The isolation rate of genus Lactobacillus was exceedingly low. 4) Groups B and C had significantly higher level of S. mutans group than group A and control group. Group B had significantly lower level of S. mitior than group A and control group. 5. All four groups showed high level of biotype I, and biotype IV was observed in groups B and C. Biotype I was identified as S. mutans serotype c, biotype IV was observed as S. sobrinus serotype d.

[Radiographic staging procedures and radiotherapy of malignant lymphoma].

Radiographic studies play an important role in the clinical staging of malignant lymphoma (ML). Conventional procedures are plain chest X-ray, lymphangiography, gastrointestinal series, and 67Ga scintigraphy. Gastro-intestinal series is essential in non-Hodgkin's lymphoma (NHL) of Waldeyer's ring and thyroid. 67Ga scintigraphy is a non-invasive procedure useful in the screening of ML. Recently marked improvements have been made in the diagnostic modalities. CT scan and NMR have made it very easy to diagnose ML of the central nervous system and detect the extension of tumors at all sites. In some cases with Hodgkin's disease (HD), staging laparotomy is necessary, but not in cases with NHL. If tumors are localized, subtotal or total nodal irradiation is performed for patients with HD, and generous involved field is employed for NHL with or without combined chemotherapy at our department. Good local control is obtained except for NHL of the central nervous system at a total dose of 40-50 Gy. NHL is prone to relapse outside the radiation field. Therefore we consider that combined chemotherapy is indicated for NHL such as T-cell lymphomas, those with high grade histology classified according to the Working Formulation, and with involvement of lymph nodes larger than 6 cm in diameter.

Frequency of micronuclei in hepatocytes following X and fast-neutron irradiations--an analysis by a linear-quadratic model.

The usefulness of the micronucleus assay for investigating the radiation response of hepatocytes was examined. The frequency was defined as the ratio of the total number of micronuclei to the number of hepatocytes examined. The dose-response curves were curvilinear after X rays and linear after neutrons. These dose-response curves were analyzed by a linear-quadratic model, frequency = aD + bD2 + c. The a/b ratio was 3.03 +/- 1.26 Gy following X irradiation. This value is within the range of the alpha/beta ratios reported by others using the clonogenic assay of hepatocytes. While the a/b value for neutrons was 24.3 +/- 11.7 Gy, the maximum relative biological effectiveness of neutrons was 6.30 +/- 2.53. Since the micronucleus assay is simple and rapid, it may be a good tool for evaluating the radiation response of hepatocytes in vivo.

[Hypoxic cell radiosensitizer present status and problems from viewpoint of clinical radiotherapy].

The results of clinical trials of hypoxic cell radiosensitizers were reviewed in order to clarify the present status and problems in this field. Most trials failed to show an effectiveness of the combination, however, some ENT tumors and bladder cancers were improved in local tumor responses and survivals. A selection of the patients suitable for the combination and development of simple method for the selection are indispensable. SERs necessary for clinical radiotherapy are considered to be larger than 1.2 at 2 Gy fraction level. Present hypoxic cell radiosensitizers have no significant effects on alpha values. Since radiation responses at low radiation dose ranges are dominated by alpha values, the chemicals having an effects on alpha values have to be developed.

[Clinical assessment of thermoradiotherapy of breast cancer and cancer of the urinary bladder].

Non-randomized clinical trials of thermoradiotherapy for breast cancer and carcinoma of the urinary bladder were reported. Thermoradiotherapy was useful for breast cancer treatment in term of (1) increased local control for locally advanced tumors and (2) possibility of reducing dose of irradiation for recurrent tumors following radiotherapy. Tumor response to thermoradiotherapy was dependent on tumor volume and maximum or minimum tumor temperature. Thermoradiotherapy was also effective as a preoperative-treatment for urinary bladder cancer. The incidence of down-staging and degeneration of tumors was significantly higher in thermoradiotherapy than in radiotherapy. These effects were temperature dependent. Patients with T 3-4 NOMO or Grade 3 tumors treated with thermoradiotherapy showed a tendency of increased survival compared with those treated with radiotherapy alone.
