Rapid separation and determination of carbamate insecticides using isocratic elution pressurized capillary electrochromatography.

An isocratic elution pressurized CEC (pCEC) system was used to separate and determine ten carbamate insecticides. It was found that introduction of the electrical field, supplementary pressure, and SDS in the proposed method greatly improved the speed, column efficiency, selectivity, and repeatability for separation and determination of carbamates. On a capillary column of 75 microm ID packed with 3 microm octadecyl silica, baseline separation and detection of ten analytes was performed by using a mobile phase consisting of 30% v/v ACN and 70% v/v of 5 mmol/L ammonium acetate (pH 6.5) containing 1 mmol/L SDS and 0.01% triethylamine (TEA). Under the optimum conditions ten carbamate insecticides could be completely separated within 20 min. For the real vegetable samples, an SPE procedure for the cleanup of matrices was carried out prior to pCEC analysis. The detection limits of 0.05-1.6 mg/kg for ten carbamates and mean recoveries of 51.3-109.2% for eight kinds of vegetable samples at different concentrations of carbamates with RSD less than 11.4% were obtained, respectively. The proposed method has been proved to be effective in the rapid analysis of carbamate residues in vegetables.

Effects of antisense oligodeoxynucleotide to follicle-stimulating hormone receptor on the expression of proliferating cell nuclear antigen and vascular endothelial growth factor in primary culture cells derived from human ovarian mucinous cystadenocarcinoma.

The effects of antisense oligodeoxynucleotide (antisense ODN) to follicle-stimulating hormone receptor (FSHR) and follicle-stimulating hormone (FSH) on the expression of proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) were studied in primary culture cells derived from human ovarian mucinous cystadenocarcinoma (OMC). The primary OMC cells were cultured with the enzyme digestion method, and the expression of pan Keratin protein and FSHR mRNA was detected for identification of the cells. OMC cells were co-cultured with antisense ODN, nonsense ODN and FSH with different concentrations for 48 h and 72 h. The expression of PCNA and VEGF was detected by using SP immunohistochemistry. Compared with that in the control group, the PCNA and VEGF expression was increased obviously in FSH groups (P < 0.05 or P < 0.01), while decreased significantly in antisense ODN groups (P < 0.05 or P < 0.01) and unchanged in nonsense ODN groups, respectively. Meanwhile, antisense ODN could antagonize the increased expression of PCNA and VEGF caused by FSH significantly (P < 0.01). It was suggested that FSH might promote the development of OMC to some extent. Antisense ODN could inhibit the proliferative activity of OMC cells and the promoting proliferative activity enhanced by FSH.

Sickle cell leg ulcers: associations with haemolysis and SNPs in Klotho, TEK and genes of the TGF-beta/BMP pathway.

Cutaneous leg ulcers are common in sickle cell anaemia and their risk might be genetically determined. Sickle cell anaemia patients were studied to examine the relationship of leg ulcers with haemolysis and with single nucleotide polymorphisms (SNPs) in candidate genes that could affect sickle vasoocclusion. Leg ulcer patients had lower haemoglobin levels and higher levels of lactate dehydrogenase, bilirubin, aspartate transaminase and reticulocytes than did control patients with sickle cell anaemia but without leg ulcers. Age-adjusted comparisons showed that sickle cell anaemia-alpha thalassaemia was more frequent among controls than cases. These results strongly suggested that the likelihood of having leg ulcers was related to the intensity of haemolysis. 215 SNPs in more than 100 candidate genes were studied. Associations were found with SNPs in Klotho, TEK and several genes in the TGF-beta/BMP signalling pathway by genotypic association analyses. KL directly or indirectly promotes endothelial nitric oxide (NO) production and the TEK receptor tyrosine kinase is involved in angiogenesis. The TGF-beta/BMP signalling pathway modulates wound healing and angiogenesis, among its other functions. Haemolysis-driven phenotypes, such as leg ulcers, could be improved by agents that reduce sickle erythrocyte density or increase NO bioavailability.

Feature scaling for kernel fisher discriminant analysis using leave-one-out cross validation.

Kernel fisher discriminant analysis (KFD) is a successful approach to classification. It is well known that the key challenge in KFD lies in the selection of free parameters such as kernel parameters and regularization parameters. Here we focus on the feature-scaling kernel where each feature individually associates with a scaling factor. A novel algorithm, named FS-KFD, is developed to tune the scaling factors and regularization parameters for the feature-scaling kernel. The proposed algorithm is based on optimizing the smooth leave-one-out error via a gradient-descent method and has been demonstrated to be computationally feasible. FS-KFD is motivated by the following two fundamental facts: the leave-one-out error of KFD can be expressed in closed form and the step function can be approximated by a sigmoid function. Empirical comparisons on artificial and benchmark data sets suggest that FS-KFD improves KFD in terms of classification accuracy.

Dehydroepiandrosterone inhibited the bone resorption through the upregulation of OPG/RANKL.

The plasma level of dehydroepiandrosterone (DHEA) decreases gradually along with aging. The beneficial effects of DHEA as an anti-aging steroid, such as the stimulatory effect on immune system, anti-diabetes mellitus, anti-atherosclerosis, anti-dementia, anti-obesity and anti-osteoporosis have been demonstrated in experiment both in vitro and in vivo. It is important to investigate the effective mechanism of DHEA in therapeutics for postmenopausal osteoporosis. Having isolated and cultured osteoblasts (OBs) and osteoclasts (OCs), we analysed the effect of DHEA on osteoblastic viability, regulation of DHEA on the expression of osteoprotegerin (OPG)/receptor activator of NF-kappaB ligand (RANKL) mRNA in OBs, and then observed the action of DHEA on bone resorption of OCs in the presence or absence of OBs. The results showed that DHEA improved viability of OBs within the concentration range of 0.01-1 microM, especially at the concentration of 0.1 microM. DHEA could apparently increase the ratio of OPG/RANKL mRNA in OBs. In the presence of OBs, DHEA could decrease the number and area of absorption lacuna of specula. We concluded, therefore, only in the presence of OBs, DHEA could inhibit the bone resorption of OCs, which may be mediated by OPG/RANKL of OBs.

[Gene mutation analysis of coagulation factor VIII from a female patient with hemophilia A].

Hemophilia A affects male, whereas females are carriers and generally spared from this disease. However, we here reported a 65-year-old female with Hemophilia A while screening the gene mutation of coagulation factor VIII. The female went to hospital because of tripping to lead her right chest to be injured with subcutaneous hematoma. She had historically a hemorrhagic diathesis. The physical examination discovered her hip limited to bend and move, but no discrepancy length between her two legs. The initial laboratory tests showed that the activated partial thromboplastin time (APTT) was 61. 3 seconds (20-40 seconds), and the APTT corrected by mixing with normal plasma was 41.3 s, but the levels of PT, FIB and TT were normal. The plain radiographs revealed the hip joints to suffer from the acetabular dysplasia and osteoarthritis. The level of FVIII:C was 2%, F IX:C 200%, vWF:Ag 120%, vWF:Rcof 100%, vWF:CBA 128%, and the F VIII binding assay to vWF was normal. The primers for exon 14 of F VIII gene were designed according to the NM - 000132 gene sequence. DNA was abstracted from the patient blood. PCR were carried out and the DNA sequence was followed. A new mutation of 4111A-->C was discovered, which caused the amino acid sequence changed (T 1314 P). The mutation of T 1314 P may be the cause of this female patient to get the hemophilia A. This mutation was a novel one which has never been reported before.

[Autologous peripheral blood stem cell transplantation for the treatment of lower extremity arterial occlusive disease].

OBJECTIVE: To evaluate the safety and feasibility of autologous peripheral blood mononuclear cells (PBMNCs) implantation after granulocyte-colony stimulating factor (G-CSF)-induced mobilization in patients with lower extremity arterial occlusive disease (AOD). METHODS: A total of 12 patients with AOD were enrolled in this study. Following administration of rhG-CSF (150 microg/d) for 5 days, PBMNCs were harvested and injected intramuscularly in the diseased extremities (3 x 10(9) per limb). RESULTS: One patient received left leg amputation due to uncontrolled ulcer 15 days post PBMNCs transplantation and the symptoms and signs were improved significantly in 9 patients and the symptoms and signs remained unchanged in another 2 aged patients (> 70 years). Doppler ultrasonography measurement showed that peak systolic velocity in diseased extremities was significantly increased post transplantation [(44.55 +/- 4.13) cm/s vs. (21.32 +/- 0.63) cm/s, P < 0.01]. Contrast lower limb angiogram showed increased collateral vessels post transplantation. One aged patient (80 years) who did not respond to autologous PBMNCs received heterologous PBMNCs transplantation (PBMNCs was harvested from a young relative of him) 3 months post autologous PBMNCs transplantation and observed for another 3 months and all observed parameters improved significantly. CONCLUSION: Implantation of autologous PBMNCs collected after G-CSF administration might offer a simple, safe, and effective therapy for the AOD patients.

[Allogeneic hematopoietic stem cell transplantation for the treatment of mucopolysaccharidosis type 1: a case report].

Mucopolysaccharidosis type I (MPS-I) is an inborn error of metabolism with progressive multisystem involvement. Hurler syndrome is the most severe form of MPS-I that causes progressive deterioration of the central nervous system with ensuing death. This study reported the therapeutic effect of allogeneic hematopoietic stem cell transplantation (allo-HSCT) on Hurler syndrome in one case. The patient was a 25-month-old boy. He underwent allo-HSCT. The donor was his elder sister whose HLA-B locus was not matching. The reduced-intensity of BuCy conditioning regimen in allo-HSCT for this patient was as follows: busulfan 3.7 mg/kg daily at 9 to 6 days before transplantation, cyclophosphamide 42.8 mg/kg daily at 5 to 2 days before transplantation, and rabbit antithymocyte globulin 3.5 mg/kg daily at 1, 3, 5, and 7 days before transplantation. Human granulocyte colony-stimulating factor (G-CSF)-mobilized peripheral blood stem cells (CD34+ cells 12.8 x10(6)/kg) were infused and cyclosporine (CSA), short-course methotrexate, daclizumab and mycophenolate mofetil (MMF) were administered to prevent graft-versus-host disease (GVHD). Complete donor-type engraftment was confirmed by Short Tandem Repeat-Polymerase Chain Reaction (STR-PCR) on day 14 after transplantation. Neutrophil and platelet engraftment occurred on days 11 and 19 after transplantation respectively. Only grade I regimen-related toxicity of live and gastrointestinal tract occurred. GVHD and graft failure were not observed. After transplantation, the clinical symptoms and the neurocognitive function were greatly improved in this patient. It was concluded that allo-HSCT was effective for the treatment of MPS-I. The reduced-intensity conditioning regimen was helpful to decrease the regimen-related toxicity. Sufficient immunosuppressive therapy and adequate hematopoietic stem cells infusion may be beneficial to the donor cell engraftment and reducing the incidence of graft failure and GVHD.

[Studies on absorptive mechanism of lipophilic components of danshen from its hydroxypropyl-beta-cyclodextrin inclusion complex].

Taking Tanshinon IIA as the target and using reversed phase high performance liquid chromatography,we have developed a rapid and sensitive assay for lipophilic components of Danshen (LCD) in perfusate. The model of intestinal absorption was used to determine the concentration of Tanshinon IIA in rats as an in situ model. The change of concentration of Tanshinon IIA was separately calculated according to Michaelis-Menten and the Fick's equation to investigate the absorptive limit step of the LCD. It was concluded that the limit step to absorption of LCD was at the dissolution, by assessing the fitting index or correlative index. On the basis of the conclusion, the experiment was designed to prepare the inclusive complex of the LCD with hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and to study its absorptive mechanism. With the increase of dosage of complex, its absorption did not show saturated phenomenon in gastro-intestinal tract in rats and the constant Ka did not show significant difference, suggesting that the transport of mechanism in vivo is similar to passive transport.

[Interaction of bovine serum albumin with luteolin and apigenin].

AIM: To study the interaction mechanism of bovine serum albumin (BSA) with luteolin and apigenin. METHODS: Fluorescence quenching method and non-radioactive energy transfer theory were used. RESULTS: The binding constants at different temperature were determined and the quenching mechanism of them were suggested as static quenching. The transfer efficiency of energy and distance between BSA and luteolin or apigenin were investigated according to the mechanism of the Förster energy transference. CONCLUSION: The interaction between them seems to be strong and the binding force were mainly hydrophobic force. B(3')-OH,B(4')-OH strengthened the interaction of flavonoids and BSA.

[The effect of CACNA1H gene G773D mutation on calcium channel function].

OBJECTIVE: To study the effect of CACNA1H gene mutation G773D on calcium channel function. METHODS: By the overlap extension PCR we introduced G773D mutation into a human Cav3.2acDNA for constructing the mutant. And then using whole cell clamp technique, we studied the alterations of channel behavior in transfected HEK-293 cells. RESULTS: There were no difference in kinetics of activation and inactivation of calcium channel between wild type and mutant. However comparing with the wild-type Cav3.2 channel, G773D mutant could increase the calcium current density significantly. CONCLUSION: CACNA1H gene G773D mutation is able to increase calcium current and neuronal excitability.

[DHEA inhibits apoptosis of murine OB through MAPK signaling pathways independent of either ARs or ERs].

Dehydroepiandrosterone (DHEA) is a promising agent for the treatment of post-menopausal osteoporosis (PMO), but the molecular mechanisms and signaling pathways by which this steroid modulates apoptosis of osteoblasts (OB) are still poorly understood. In this study, the OBs were cultured in vitro by the enzyme-digested method, treated with DHEA (10(-7) mol/L) for 0h, 24h, 48h, 72h, respectively. The expressions of ER alpha, ER beta and AR mRNA in OB were analyzed by RT-PCR. After the primary OBs were deprived of serum for a further 24h, and then pretreated with 1 micromol/L ICI 182,780 (an estrogen receptor antagonist), 10 micromol/L Flutamide (an androgen receptor antagonist) or 100 micromol/L U0126 (a specific inhibitor of MAPK pathway) for 1h, they were treated with a series of concentrations of DHEA (10(-10) - 10(-5) mol/L) in serum-free medium for 72h,the apoptotic cells were analyzed by FCM with the Annexin-V-FITC/PI dual labeling technique. The OBs were incubated in 1 micromol/L ICI 182,780 or 10 micromol/L Flutamide for 25 minutes, and then treated with different concentrations of DHEA for the further 10 minutes. The phosphorylation status of ERK1/2 was analyzed by Western blot. After the OBs were incubated with DHEA (10(-7)mol/L) for 24h, 48h or 72h, respectively, its ERbeta and AR mRNA level were increased (P<0.05, P<0.01, respectively), but the ER alpha mRNA level had no change. The 10(-9) - 10(-6) mol/L of DHEA inhibited OBs early apoptosis induced by the serum deprivation (P<0.05, P<0.01, respectively). The inhibiting effect, moreover, could be blocked by the specific inhibitor of MAPK pathway, U0126. The effects of DHEA were neither blocked by the steroid hormone antagonist ICI 182,780 nor by Flutamide. Western blot showed that neither receptor antagonist ICI 182,780 nor Flutamide could block the DHEA-induced ERKs phosphorylation in OBs,which was similar to the apoptosis. DHEA inhibits apoptosis in OBs presumably via a DHEA-specific receptor that involves mitogen activated protein kinase (MAPK) signal pathway, phospho-pERK1/2, independent of either ARs or ERs.

[Urokinase-type plasminogen activator and urokinase-type plasminogen activator receptor in the seminal plasma and sperm of fertile and oligoasthenozoospermia males].

OBJECTIVE: To find the difference between the levels of urokinase-type plasminogen activator (uPA) and urokinase-type plasminogen activator receptor(uPAR) in the seminal plasma and sperm of fertile and oligoasthenozoospermia men, and to understand their correlation with male fertility. METHODS: The levels of uPA in the seminal plasma and sperm of 22 normospermic males and 44 oligoasthenozoospermia patients were measured by ELISA. RESULTS: (1) The average level of uPA in the seminal plasma and sperm of the normospermic group, ([4803.69 +/- 602.78] mU/L) and ([30.29 +/- 3.16] mU/10(6) sperm) were higher than those of the oligoasthenozoospermia group, ([4061.35 +/- 736.23] mU/L), and ([20.51 +/- 4.2] mU/10(6) sperm) (P < 0.01). (2) The average level of uPAR in the sperm of the normospermic group ([12.97 +/- 3.11] mU/10(6) sperm) was significantly higher than that of the oligoasthenozoospermia group, ([6.09 +/- 1.45] mU/10(6) sperm) (P < 0.01). (3) The levels of uPA and uPAR in the sperm and the content of uPA in the seminal plasma were positively correlated with sperm motility and viability. CONCLUSION: Urokinase-type plasminogen activator is related with male fertility and the levels of uPA and uPAR vary in the seminal plasma and sperm of fertile and oligoasthenozoospermia males.

[Study on degradation kinetics or potassium dehydroandrographolidi succinas].

OBJECTIVE: To investigate the hydrolytic degradation of the kinetic characteristics of Potassium Dehydroandrographolidi Succinas (DAS-K) in aqueous solution. METHODS: The HPLC method was used to determine the degradation kinetic parameters of DAS-K aqueous solution at different initial concentration, different pH, different ionic strength. various temperatures and in different buffer solutions. RESULTS: DAS-K hydrolytic degradation followed first-order kinetics as measured by HPLC. From pH 8, the hydrolytic degradation rate of DAS-K markedly increased with pH. DAS-K was unstable in alkaline pH solution. The species of buffer solutions seem to have different impact on the catalytic process. The ionic strength did not have significant effect on the stability of the drug. According to the Arrhenius plot, the dependence of the decomposition on temperature was a determining factor, the activation energy was estimated to be 95.68 KJ/mol in phosphate buffer solution at pH 8 and temperature from 60 to 90 degrees C. CONCLUSION: It was found that the hydrolytic degradation of DAS-K complied with first-order kinetics. The rate of hydrolytic degradation of DAS-K depended on the pH of solution, the buffer concentration, the buffer species and the temperature. Especially, pH value was an important factor in determining the rate of the hydrolytic degradation of the drug.

Retinoschisis at macular area in highly myopic eye by optic coherence tomography.

PURPOSE: To improve our understanding of macular schisis in highly myopic eyes. METHODS: From 2002.6-2003.12, the highly myopic eyes (> 6D) which has macular schisis found by OCT at our hospital were included in the study. Each eye then received further examinations including visual acuity, refractive error and ultrasonography. Then the results were analyzed to find the characteristic of the entity. RESULTS: During that period, OCT found macular schisis in 38 highly myopic eyes from 36 patients. Their vision was low, and their average axial length and refractive error were 29.6mm and -10.5D respectively. The appearance and the area of the schisis are different among the eyes. But the schisis can be largely divided into two types: inner and outer schisis. They can exit independently or concurrently in the same eye. And the schisis was often accompanied by other macular findings, like retinal detachment, macular hole. CONCLUSION: Macular schisis in highly myopic eyes is not uncommon in the clinic. OCT is useful in its diagnosis.

Ablation of mouse phosphomannose isomerase (Mpi) causes mannose 6-phosphate accumulation, toxicity, and embryonic lethality.

MPI encodes phosphomannose isomerase, which interconverts fructose 6-phosphate and mannose 6-phosphate (Man-6-P), used for glycoconjugate biosynthesis. MPI mutations in humans impair protein glycosylation causing congenital disorder of glycosylation Ib (CDG-Ib), but oral mannose supplements normalize glycosylation. To establish a mannose-responsive mouse model for CDG-Ib, we ablated Mpi and provided dams with mannose to rescue the anticipated defective glycosylation. Surprisingly, although glycosylation was normal, Mpi(-/-) embryos died around E11.5. Mannose supplementation even hastened their death, suggesting that man-nose was toxic. Mpi(-/-) embryos showed growth retardation and placental hyperplasia. More than 90% of Mpi(-/-) embryos failed to form yolk sac vasculature, and 35% failed chorioallantoic fusion. We generated primary embryonic fibroblasts to investigate the mechanisms leading to embryonic lethality and found that mannose caused a concentration- and time-dependent accumulation of Man 6-P in Mpi(-/-) fibroblasts. In parallel, ATP decreased by more than 70% after 24 h compared with Mpi(+/+) controls. In cell lysates, Man-6-P inhibited hexokinase (70%), phosphoglucose isomerase (65%), and glucose-6-phosphate dehydrogenase (85%), but not phosphofructokinase. Incubating intact Mpi(-/-) fibroblasts with 2-[(3)H]deoxyglucose confirmed mannose-dependent hexokinase inhibition. Our results in vitro suggest that mannose toxicity in Mpi(-/-) embryos is caused by Man-6-P accumulation, which inhibits glucose metabolism and depletes intracellular ATP. This was confirmed in E10.5 Mpi(-/-) embryos where Man-6-P increased more than 10 times, and ATP decreased by 50% compared with Mpi(+/+) littermates. Because Mpi ablation is embryonic lethal, a murine CDG-Ib model will require hypomorphic Mpi alleles.

[Treatment of rabbits bearing advanced VX2 tumors in the mammary gland with nano-sized liposomal adriamycin administered by various routes].

OBJECTIVE: To investigate the therapeutic effect of nano-sized liposomal adriamycin (NLADR) administered by various routes on rabbits bearing advanced breast tumors. METHODS: NLADR with a mean diameter of 120 nm was prepared by pH gradient-driven drug encapsulation method. VX(2) tumor mass suspension was injected into the breast tissues of 50 female New Zealand White rabbits. Six weeks after the inoculation 38 surviving tumor-bearing rabbits were randomly divided into 5 groups: group A (control group), receiving a sham treatment; group B, receiving subcutaneous injection of NLADR with a dose of 1 mg/kg into the areas adjacent to the implanted tumor; group C, receiving intravenous injection of NLADR with a dose of 1 mg/kg; group D, receiving NLADR with a dose of 1 mg/kg administered by subcutaneous injection combined with intravenous injection with half of the whole dose for both routes; and group E, receiving intravenous injection of free adriamycin (FADR). The treatment was repeated every 48 hours. The rabbits were killed 48 hours after the third treatment. The breast tumors, axillary lymph nodes, and all of the metastatic tumors anatomically detected in distant organs were collected. The sizes of tumors and axillary nodes before and after treatment were measured. Necrosis of tumor tissue was assessed pathologically. The mRNA expression of proliferating cell nuclear antigen (PCNA) was determined using RT-PCR. Apoptosis was identified and quantified as apoptosis index (AI) using TUNEL method. RESULTS: The average growth rate of tumor was the highest in group A (1.58) and the lowest in group C (1.33). The average growth rate of axillary lymph nodes was the highest in group A (3.70), significantly higher than in any other groups (all P = 0.00); and was the lowest in group B, significantly lower than groups A, C, and E (all P < 0.01), however without a significant difference between groups B and D (P = 0.148). The PCNA mRNA expression level of the implanted tumor in groups C was the lowest, significant lowest then those in group A and B (both P < 0.01). The sequence of PCNA mRNA in the axillary lymph nodes was group B < group D < group C < group E < group A with significant differences between group B and groups A, C, and E. The sequence of PCNA mRNA in the mediastinal lymph nodes was group B < Group E < group D < group C < group A. The PCNA mRNA expression level was the lowest in group D, significantly lower than that in group A (P = 0.011). Necrosis of implanted tumor, metastatic foci in lung and liver, and lymph nodes was obvious in groups C, D, and E. Necrosis of implanted tumor and metastatic foci in lung and liver was significantly obvious in group C than in group D (P = 0.000 and P = 0.022). Necrosis of the implanted tumor was significantly more obvious in group C than in group F (P = 0.033). Necrosis of axillary lymph nodes was significantly more obvious in group than in groups C and E (P = 0.000 and P = 0.000). The values of AI of the implanted tumor in groups C and D were 16.74% and 18.04%, both significantly higher than those in group E (P = 0.02 and P = 0.04), and those of group E were significantly higher than those in groups A and B (both P < 0.01). The AI value of the axillary lymph nodes was 21.73%, significantly than those in groups A, C, and E (all P < 0.01). The average AI values of the metastatic foci were 16.52%, 15.77%, and 14.50%, all significantly higher than that in group B (all P < 0.01), and that of group B was significantly higher than that in group A (P = 0.039). CONCLUSION: NLADR, especially intravenous administration combined with subcutaneous administration, is effective in treatment of advanced breast carcinoma with lymphatic and distant metastases.
