Kynurenine metabolism-related gene signature for prognostic stratification in hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) remains a major global health burden with high mortality rates and limited therapeutic options. The identification of reliable biomarkers for early diagnosis and prognosis prediction is urgently needed. Kynurenine metabolism, a critical pathway in immune regulation and tumor progression, has been implicated in various cancers. However, its prognostic value in HCC has not been fully elucidated. This study aimed to develop a prognostic risk model based on kynurenine metabolism-related genes (KMRGs) for HCC patients. METHODS: Transcriptomic and clinical data of HCC patients were retrieved from The Cancer Genome Atlas (TCGA) and the International Cancer Genome Consortium (ICGC) databases. A prognostic risk model was established using least absolute shrinkage and selection operator (LASSO) and Cox regression analyses. Survival analysis and functional enrichment analysis were conducted to validate the predictive performance of the model and to investigate the underlying mechanisms. ALDH8A1 was ultimately identified as a target gene based on survival analysis, and its impact on tumor cell migration was assessed using the HCC cell line. RESULTS: A prognostic model based on seven KMRGs was established. The high-risk group exhibited significantly worse overall survival compared to the low-risk group. Functional enrichment analysis in high-risk patients highlighted significant enrichment in core biological processes, including spliceosome assembly and ribonucleoprotein complex biogenesis. Furthermore, a nomogram integrating the risk score and clinical pathological features was developed, demonstrating moderate predictive performance for HCC prognosis. CONCLUSIONS: This study successfully constructed a prognostic risk model based on seven KMRGs, providing a valuable tool for predicting clinical outcomes in HCC patients. These findings highlight the potential role of kynurenine metabolism in HCC progression and offer new insights for future therapeutic strategies.

Leukemia stem cells.

Cancer is a multifaceted disease in which cell proliferation is no longer under normal growth control. Accumulating data have suggested the existence of cancer stem cells, a minor population of tumor cells that possess the stem cell property of self-renewal and that are responsible for the initiation and maintenance of cancer. The knowledge of cancer stem cell biology is most advanced in research on the hematopoietic cancer, leukemia. With the identification of leukemia stem cells (LSCs) capable of repopulating nonobese diabetic/severe combined immunodeficient (NOD/SCID) mice, this body of research has led to conclusive proof for cancer stem cells. This review focuses on the biological characterization of LSCs for each type of leukemia, which has provided key insights into leukemogenic pathology and LSC-targeted therapies.

[Observation on therapeutic effect of rolling needle therapy on insomnia].

OBJECTIVE: To assess the effect-increasing action of rolling needle therapy on insomnia. METHODS: Sixty-four cases were randomly divided into a rolling needle treatment group and a control group, 32 cases in each group. The control group were treated with oral administration of clonazepam 4 mg, once each night, for 4 consecutive weeks. The rolling needle therapy group were treated with the same treatment as the control group, plus the rolling needle stimulation of the back. The therapeutic effect was assessed by effective rate of sleep improvement and Pittsburgh sleep quality index (PSQI). RESULTS: After treatment there was a significant difference between the two groups in the score of PSQI (P < 0.01). CONCLUSION: The rolling needle therapy can increase the improving action of clonazepam on insomnia, and increase life quality of the patient.

[A survey of the therapeutic reason of orthodontic patients in Luzhou].

OBJECTIVE: To investigate the therapeutic reason of orthodontic patients in Luzhou. METHODS: A questionnaire investigation was made for the following randomly chosen people: 452 teenage patients from orthodontic department of stomatology hospital affiliated to Luzhou Medical College were brought into treatment group. 600 students from Luzhou No. 2 Middle School were brought into non-treatment group. The questionnaire investigation was conducted to two groups. The results were statistically analysed with SPSS 10.0. RESULTS: (1) The main aim for orthodontic treatment was to align the teeth. (2) 37.39% of the patients accepted orthodontic treatment for parent's demand. (3) 76.1% of the patients expected to align the dentition. (4) Most people in treatment group could accept the time and expenses in the orthodontic treatment, but most people of non-treatment group could not accept the expenses in the orthodontic treatment. (5) Most people thought appearance was important. CONCLUSION: Aesthetic is the main reason for orthodontic patients.

[Role of AcSDKP on collagen synthesis and degradation in cultured rat cardiac fibroblast].

OBJECTIVE: To investigate the role of AcSDKP on collagen synthesis and degradation in cultured rat cardiac fibroblasts. METHODS: Neonatal rat cardiac fibroblasts were isolated and stimulated by PDGF. The cell proliferation was observed by (3)H-TdR incorporation assay. The synthesis of collagen was measured by (3)H-proline incorporation assay. The expression of type I and type III collagen and MMP-1 protein were measured by Western blot. The MMP-2 and MMP-9 activity was evaluated with zymography assay. RESULTS: PDGF stimulated cardiac fibroblasts proliferation with increased collagen synthesis and type I and type III collagen protein expressions as well as MMP-2 and MMP-9 activities and MMP-1 expression. AcSDKP inhibited cardiac fibroblasts proliferation induced by PDGF and reduced collagen synthesis and type I and type III collagen protein expression. AcSDKP also further up-regulated MMP-2 and MMP-9 activities and MMP-1 expression in cardiac fibroblasts induced by PDGF. CONCLUSION: AcSDKP inhibited proliferation and collagen synthesis and up-regulated matrix metalloproteinases activity or expression induced by PDGF, which was possibly related with the effect of AcSDKP anti-fibrosis.

[The prevalence and its screening methods of primary open angle glaucoma in defined population-based study of rural and urban in Beijing].

OBJECTIVE: The purpose of the study is to determine the prevalence of primary open-angle glaucoma (POAG) in persons aged 40 or above in Beijing, China. METHODS: From June 2001 to October 2001, the screening population was identified by a door-to-door census in five metropolitan resident areas in the north of Beijing and three villages in a county south to Beijing. The screening included visual acuity, frequency doubling perimetry (FDP, C-20 screening program), non-contact tonometry, slit lamp microscopy, anterior chamber depth (Van Herick method), and fundus photography. The suspect glaucoma and definite glaucoma patients were asked to have an examination of Octopus 1-2-3 perimetry (G1X TOP threshold program), repeat tonometry, gonioscopy and fundus stereo-photography at Beijing Tongren Hospital. RESULTS: There were 4451 subjects who were examined at the study sites. The response rates in rural and urban were 79.58% and 87.13%, respectively. In this 40 years-old or above population, the prevalences of POAG were 1.97% in rural men, 2.07% in urban men, 1.04% in rural women and 1.42% in urban women. In this study, 92.30% POAG patients in rural and 87.30% POAG patients in urban were new diagnosed cases. The prevalence of POAG increased with age and the change was exponential. In 50% POAG patients first IOP measurement was less than 21 mm Hg (1 mm Hg = 0.133 kPa). The prevalence of monocular eye blindness was 15.40% and 10.90% in rural and urban, respectively. CONCLUSIONS: Owing to use fundus photography and integrated evaluation of optic disc, it is possible to diagnose in earlier stage of POAG. This study identifies more patients with POAG than any previous population-based studies of China, and is similar to other studies of Asia such as in India and Singapore. The reason of lower POAG prevalence in rural women than in urban may be that the anterior chamber depth of rural women is shallower than that of urban women. It may cause difficult to differentiate the chronic primary angle closure glaucoma from POAG at screening sites, so the part POAG patients may be included in primary angle closure glaucoma patients.

Development and Preliminary Application of a Peptide Mass Fingerprinting Technique in Proteome Research.

Proteome means the total proteins expressed by the genome in a cell or tissue. Two-dimensional electrophoresis (2-DE) is now the most powerful separating technique and the key separation method used in proteome. Peptide mass fingerprinting (PMF) is becoming a widely used and vastly developed technique for protein identification in 2-D gels. In this research, a systematic method to identify the proteins in polyacrylamide gels by PMF was developed. Proteins were digested in-gel by enzyme and the masses of generated peptides were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). The data obtained from PMF were used in protein database search and the protein identification. The proteins from human lung cancer cells isolated by 2-DE were subjected to identification by the PMF method developed in this work. Three spots of proteins in gel were identified as G3P2_HUMAN, UBL1_ HUMAN and TPIS_HUMAN.
