[System review of the Chinese medicine bushenhuoxue for treating benign prostatic hyperplasia].

OBJECTIVE: To make a system review of the effects of Bushenhuoxue (kidney-tonifying and blood-activating prescription), a category of compound Chinese medicines, on benign prostatic hyperplasia (BPH) and its side effects. METHODS: All the research articles about Chinese medicines treating BPH from January 1978 to February 2003 were retrieved using the methods of international evidence-based medicine (EBM), and their qualities were evaluated based on JADAD standard and concealment of research allocation. Then the included articles went through META-analysis with Revman 4.2 software. RESULTS: The efficacy of Bushenhuoxue on BPH was better than Qianliekang but not statistically different from finasteride. The combined use of Bushenhuoxue with surgery had no statistical difference from mere surgical treatment. Four articles reported the side effects of this compound Chinese medicine including upset and pain in the abdomen, nausea, diarrhea and dryness of the nose. CONCLUSION: Massive, multi-center and randomized controlled clinical trials should be conducted to find out more effective methods for treating BPH with Chinese medicines based on the improvement of measurable symptom evaluation method and for evaluating their side effects.

[Effects of high level Zn intake on metabolism in man].

OBJECTIVE: To observe the effects of high level Zn intake on zinc (Zn) copper (Cu), lipids metabolism and antioxidation function in man so as to provide scientific basis for Zn supplementation. METHODS: 40 rural healthy men were chosen and tablets of 50 mg Zn (as 0.2 g of zinc glucose) per day was taken by them for eight weeks. All subjects were checked up for five times: at week 0, 2, 4, 8 during the period of Zn supplementation and at week 12 and their serum, RBC, hair, and 24h-urine were collected for assays at the same time. RESULTS: (1) Zn contents in serum, RBC and hair increased significantly after 2 to 4 weeks supplementation and decreased at 4 weeks after stopping supplementation, but they were still higher than those before supplementation. Zn contents in 24h-urine increased significantly after supplementation and decreased at 4 weeks after stopping supplementation to the same level as before supplementation. (2) Cu contents in serum, RBC and hair increased significantly after 4 weeks supplementation and decreased at 4 weeks after stopping supplementation. Cu contents in 24h-urine did not change significantly during experiment. (3) RBC superoxide dismutase (SOD) activities were decreased persistently after 2 weeks supplementation and were not recovered at 4 weeks after stopping supplementation. RBC glutathione peroxidase (GPX) activities increased significantly after 4 weeks supplementation. The content of lipid peroxide (LPO) increased significantly after 2 weeks supplementation. (4) Total cholesterol (TC), triglyceride (TG), Low density lipoprotein cholesterol (LDL-C) and apolipoprotein B100 (ApoB100) increased significantly, high density lipoprotein cholesterol (HLD-C) and apolipoprotein A1 (ApoA1) decreased after supplementation. CONCLUSION: When healthy men received 50 mg Zn supplement per day, it might interfere Zn, Cu and lipids metabolism and inhibit antioxidation process.

[Study on solid-phase-extraction spectrophotometric determination of silver with p-sulfobenzylidene-rhodanine].

The color reaction of silver with p-sulfobenzylidene-rhodanine (SBDR) was studied. Based on the color reaction of SBDR with silver (I) and the solid phase extraction of its colored complex with Waters Porapak Sep-Park-C18 cartridge, a new method for the determination of micro-amount of silver (I) was studied. In the presence of pH = 2.8 citric acid-sodium hydroxide buffer solution and Tween-80 medium, SBDR can react with silver (I) to form a stable 2:1 complex. The colored complex can be extracted by C18 cartridge and eluted by ethanol (containing 5% acetic acid), and then can be determined by spectrophotometry. The molar absorptivity is 7.53 x 10(4) L x mol(-1) x cm(-1) at 520 nm. Beer's law is obeyed in the range of 0-1.2 microg x (25 mL)(-1). This method can be applied to the determination of silver in water with satisfactory results.

Biological nitrogen removal with enhanced phosphate uptake in (AO)2 SBR using single sludge system.

Simultaneous biological phosphorus and nitrogen removal with enhanced anoxic phosphate uptake via nitrite was investigated in an anaerobic-aerobic-anoxic-aerobic sequencing batch reactor ((AO)2 SBR). The system showed stable phosphorus and nitrogen removal performance, and average removals for COD, TN and TP were 90%, 91% and 96%, respectively. The conditions of pH 7.5-8.0 and temperature 32 degrees C were found detrimental to nitrite oxidation bacteria but favorable to ammonia oxidizers, and the corresponding specific oxygen uptake rates(SOUR) for phase 1 and 2 of nitrification process were 0.7 and 15 mgO2/(gVSS x h) in respect, which led to the nitrite accumulation in aerobic phase of (AO)2 SBR. Respiratory tests showed that 40 mgNO2-N/L did not deteriorate the sludge activity drastically, and it implied that exposure of sludge to nitrite periodically enabled the biomass to have more tolerance capacity to resist the restraining effects from nitrite. In addition, batch tests were carried out and verified that denitrifying phosphorus accumulation organisms (DPAOs) could be enriched in a single sludge system coexisting with nitrifiers by introducing an anoxic phase in an anaerobic-aerobic SBR, and the ratio of the anoxic phosphate uptake capacity to aerobic phosphate uptake capacity was 45%. It was also found that nitrite (up to 20 mgNO2-N/L) was not inhibitory to anoxic phosphate uptake and could serve as an electron acceptor like nitrate, but presented poorer efficiency compared with nitrate.

[Ultrastructure study on the RPE, Bruch's membrane and choroid after laser photocoagulation of the retina].

OBJECTIVE: To observe ultrastructural changes and repair process of retina after laser photocoagulation of the retina, in order to provide histopathologic findings of laser treatment for RPE and choroidal diseases. METHODS: In seven patients with orbital malignant tumor needed exenteration, the normal retina was coagulated (using I, II, or III grade photic spot) 1, 3, or 7 days before the exenteration after signing consent by the patients. The retina and the choroid from these enucleated eyeballs were observed by electron microscope. RESULTS: One day after photocoagulation (I grade photic spot), some RPE cells were swollen and necrotic, with partial disappearance of microvilli. The number or RPE was reduced. Bruch's membrane was intact. Endothelial cells of choroidal capillaries were swelled with occlusion in a few of the lumens. In III grade photic spot, many RPE cells were lysed. Vacuoles were found in Bruch's membrane. Many choroid capillaries were occluded. In III grade photic spot, numerous RPE cells were disappeared and Bruch's membrane was destroyed partially. Middle size vessels in the choroid were damaged. Three days after photocoagulation, edema of RPE cells and choroid was relieved. Phagocytes appeared in the damaged area RPE cells and fibrous tissue in choroid started to proliferate. Seven days after photocoagulation, damage area was covered by proliferative RPE. Choroid vessels were decreased in number and fibrosis was appeared in the stroma. CONCLUSIONS: Photocoagulation causes edema and necrosis of RPE and various degrees destruction of Bruch's membrane and choroidallvessels. Subsequently, the damaged area is covered by proliferative RPE cells and fibrous tissue. Diseases of the RPE and choroid can be treated by laser at various degrees of energy.

[Influence of pacing site on myocardial transmural dispersion of repolarization in intact normal and dilated cardiomyopathy dogs].

In order to verify the hypothesis that left ventricular epicardial (LV-Epi) pacing and biventricular (BiV) pacing unavoidably influence the myocardial electrophysiological characters and may result in high risk of malignant ventricular arrhythmia, we calculated, in both normal mongrel dogs and dog models with rapid-right-ventricular-pacing induced dilated cardiomyopathy congestive heart failure (DCM-CHF), the monophasic action potential duration (MAPD) and the transmural dispersion of repolarization (TDR) in intracardiac electrogram together with the QT interval and T(peak)-T(end) (T(p(-T(e)) interval in surface electrocardiogram (ECG) during LV-Epi and BiV pacing, compared with those during right ventricular endocardial (RV-Endo) pacing. To prepare the DCM-CHF dog model, rapid right ventricular pacing (250 bpm) was performed for 23.6+/-2.57 days to the dog. All the normal and DCM-CHF dogs were given radio frequency catheter ablation (RFCA) to His bundle with the guide of X-ray fluoroscopy. After the RFCA procedures, the animals were under the situation of complete atrioventricular block so that the canine heart rates could be voluntarily controlled in the following experiments. After a thoracotomy, ECG and monophasic action potentials (MAP) of subendocardial, subepicardial and mid-layer myocardium were recorded synchronously in 8 normal and 5 DCM-CHF dogs during pacing from endocardium of RV apex (RV-Endo), epicardium of LV anterior wall (LV-Epi) and simultaneously both of the above (biventricular, BiV), the later was similar to the ventricular resynchronization therapy to congestive heart failure patients in clinic. The Tp-Te) meant the interval from the peak to the end of T wave, which was a representative index of TDR in surface ECG. The TDR was defined as the difference between the longest and the shortest MAPD of subendocardial, subepicardial and mid-layer myocardium. Our results showed that in normal dogs, pacing participating of LV (LV-Epi, BiV) prolonged MAPD of all the three layers of the myocardium (P<0.05) with the character that mid-layer MAPD was the longest and subepicardial MAPD was the shortest following subendocardial MAPD. At the same time, TDR prolonged from 26.75 ms at RV-Endo pacing to 37.54 ms at BiV pacing and to 47.16 ms at LV-Epi pacing (P<0.001). Meanwhile in surface ECG, BiV and LV-Epi pacing resulted in a longer Tp-Te) interval compared with RV-Endo pacing (P<0.01), without parallel QT interval prolongation. Furthermore, all the DCM-CHF model dogs showed manifestations of congestive heart failure and enlargement of left ventricles. Based on the lengthening of mid-layer MAPD from 257.35 ms to 276.30 ms (P<0.0001) and increase of TDR from 27.58 ms to 33.80 ms (P equals;0.002) in DCM-CHF model due to the structural disorders of myocardium compared with the normal dog, LV-Epi and BiV pacing also led to the effect of prolonging MAPD of three layers of the myocardium and enlarging TDR. From these results we make the conclusions that prolongation of MAPD of subendocardial, subepicardial and mid-layer myocardium and increase in TDR during pacing participating of LV (LV-Epi, BiV) may contribute to the formation of unidirectional block and reentry, which play roles or at least are the high risk factors in the development of malignant ventricular arrhythmia, especially in case of structural disorders of myocardium. These findings must be considered seriously when ventricular resynchronization therapy is performed to congestive heart failure patients.

A study of the chemiluminescence behavior of myoglobin with luminol and its analytical applications.

A chemiluminescence signal at 425 nm was observed when ferric state myoglobin was mixed with luminol in alkaline medium. Because the signal was remarkably enhanced in the presence of Fe(CN)6(4-), analytical applications were investigated in a flow-injection system. The increase in chemiluminescence was linearly dependent on myoglobin concentration in the range 0.1 to 100 nmol L(-1), and the limit of detection was 0.04 nmol L(-1) with relative standard deviation 3.2% (3 sigma). It was also found that binding of Mb with the ligands CN-, SCN-, and F- significantly inhibited the chemiluminescence reaction. The linear dynamic ranges for the ligands were 1.0-300.0, 0.1-3.0, and 0.5-100.0 nmol L(-1), and the limits of detection (S/N=3) 0.4, 0.04, and 0.2 nmol L(-1), for F-, CN-, and SCN-, respectively. The relative standard deviations were 5.32%, 6.13%, and 3.38% for 0.1 nmol L(-1) CN-, 0.5 nmol L(-1) SCN-, and 1.0 nmol L(-1) F-, respectively. At a flow rate of 2.0 mL min(-1) the assay could be accomplished in 1 min, including sampling and washing. The method has been successfully applied to the determination of myoglobin in human urine and F- in water samples. A possible mechanism of chemiluminescence production by myoglobin and luminol is presented.

Interferon gamma repression of collagen (COL1A2) transcription is mediated by the RFX5 complex.

Interferon gamma (IFN-gamma) plays an important physiological role during inflammation by down-regulating collagen gene expression and activating major histocompatibility II (MHC-II) complex. The activation of MHC-II by IFN-gamma requires activation of a trimeric DNA binding transcriptional complex, RFX5 complex, containing RFXB (also called RFXANK or Tvl-1), RFXAP, as well as RFX5 protein. Previously, we demonstrated that RFX5 binds to the collagen transcription start site and represses collagen gene expression (Sengupta, P. K., Fargo, J., Smith, B. D. (2002) J. Biol. Chem. 277, 24926-24937). In this report, we have examined the role of RFXB and RFXAP proteins within the RFX5 complex to regulate collagen gene expression. The data show that all three RFX5 complex proteins are required for maximum repression. Expression of proteins with mutations known to be important for RFX5 complex formation does not repress collagen promoter activity. Two mutated forms of RFX5 act as dominant negative proteins activating collagen expression and reversing IFN-gamma down-regulation of collagen expression in human lung fibroblasts. IFN-gamma increases expression and nuclear translocation of RFX5. RFXB has a naturally occurring splice variant isoform (RFX SV). Interferon increases expression of the long form of RFXB and decreases expression of RFX SV with the same kinetics as collagen gene expression. Overexpression of the splice variant form reverses the IFN-gamma induced collagen repression in human lung fibroblasts. Finally, all three RFX5 complex proteins increase at the collagen transcription start site with IFN-gamma treatment using chromatin immunoprecipitation analysis. Thus, these studies suggest an important role for RFX5 complex in collagen repression.

Loss of the expression of the tumor suppressor gene ARHI is associated with progression of breast cancer.

PURPOSE: Ductal carcinoma in situ (DCIS) is a preinvasive-stage breast carcinogenesis that accounts for approximately 20 approximately 25% of mammographically detected breast cancers. A significant fraction of untreated DCIS will evolve into invasive cancer. ras homologue I (ARHI) is an imprinted tumor suppressor gene that is expressed in normal breast epithelial cells but absent or down-regulated in breast cancer cells. This study investigated the relationship of ARHI expression to the progression of breast cancer. EXPERIMENTAL DESIGN: We analyzed ARHI expression in DCIS, invasive breast carcinoma, and adjacent normal breast epithelium from 64 formalin-fixed, paraffin-embedded DCIS specimens by both immunohistochemistry and in situ hybridization. We also analyzed the correlation between ARHI expression and progression of breast cancer, as well as the correlation of ARHI expression and cyclin D1 and p21(WAF1/CIP1) expression in DCIS. RESULTS: Normal breast epithelium was found in all of the specimens and invasive breast carcinoma was found in 23 specimens. ARHI mRNA and protein were detected in all of the normal breast epithelia. ARHI expression was detected mainly in cytoplasm and rarely present in the nucleus. By histochemical analysis, ARHI expression was down-regulated in 41% (26 of 64) of DCIS and 70% (16 of 23) of invasive carcinomas comparing the specimens with adjacent normal breast epithelium. When DCIS and invasive cancer were present in the same sample, ARHI was further down-regulated in 26% (6 of 23) of invasive carcinoma. In four cases [4 (17%) of 23] of invasive carcinoma, ARHI protein expression was totally lost. Consistent results were obtained with an in situ hybridization assay for ARHI at the mRNA level. Higher levels of expression of cyclin D1 and p21(WAF1/CIP1) were observed in DCIS than in the adjacent epithelia. The expression of cyclin D1 and p21(WAF1/CIP1) was inversely correlated with that of ARHI. CONCLUSIONS: Our results indicate that ARHI expression is markedly down-regulated in DCIS, and a further decrease in ARHI expression is associated with progression of breast cancer.

Different antigens trigger different Th1/Th2 reactions in neonatal mononuclear cells (MNCs) relating to T-bet/GATA-3 expression.

Neonates are known to have poor cellular immunity, especially poor Th1 response. We investigated how neonatal mononuclear cells raised different Th1/Th2 reactions in response to different antigens. Employing Dermatophagoides pteronyssinus (Der p) extract and varicella zoster virus (VZV) as antigens, we assessed Th1/Th2 reactions as demonstrated by IL-4/IFNgamma production and mRNA expression, and transcriptional factors T-bet/GATA-3 mRNA expression in mononuclear cells from human umbilical cord blood (CBMC). Results showed that VZV induced a dramatic increase of IFNgamma production by adult peripheral blood mononuclear cells (PBMC), whereas VZV did not drive CBMC to release significant IFNgamma production (1614.7+/-362.0 vs. 49.0+/-29.3,p<0.005). However, Der p induced higher IFNgamma production by CBMC than VZV (298.1+/-171.8 vs. 49.0+/-29.3, P=0.047). In contrast, VZV did not induce significant IL-4 production either by CBMC or by PBMC. Der p induced a comparative IL-4 production by CBMC and PBMC (2.58+/-0.84 vs. 2.04+/-0.37, p>0.05). A real-time RT-PCR analysis of IL-4 and IFNgamma mRNA expression showed that VZV induced a significantly higher IFNgamma, but not IL-4, mRNA expression in PBMC than CBMC. Der p did not induce significant difference of IFNgamma or IL-4 mRNA expression in PBMC and CBMC. VZV enhanced Th1-related transcription factor T-bet mRNA expression, in association with later down-regulation of Th2-related GATA-3 mRNA expression in PBMC. However, VZV did not up-regulate T-bet or down-regulate GATA-3 expression significantly in CBMC. In contrast, Der p induced an early GATA-3 expression and later T-bet expression in CBMC. These results suggest that different antigens trigger various Th1/Th2 reactions in PBMC and CBMC resulting from kinetic changes of T-bet/GATA-3 expression.

Differential association of the auxiliary subunit Kvbeta2 with Kv1.4 and Kv4.3 K+ channels.

Kvbeta2 subunits associate with Kv1 and Kv4 K+ channels, but the basis of preferential association is not understood. For example, detergent resistance suggests stronger auxiliary subunit association with Kv4.2 than with Kv1.2, but Kvbeta2 preferentially localizes with the latter channels in brain. Here we examine the interaction of Kvbeta2 with two native binding partners in brain: Kv4.3 and Kv1.4. We show that the auxiliary subunit binds more efficiently to Kv1.4 than to Kv4.3 in mammalian cells. However, preexisting Kvbeta2 complexes with Kv1.4 and Kv4.3 have similar detergent sensitivity. Thus, preferential steady state binding may reflect a difference in initial association rather than stability. We also find that that the cytoplasmic C-terminus of Kv4.3 inhibits Kvbeta2 association. Apparently, a region proximal to the Kv4.3 pore contributes to the inefficient auxiliary subunit interaction that produces preferential binding of Kvbeta2 to Kv1 channels.

Aberrant methylation and silencing of ARHI, an imprinted tumor suppressor gene in which the function is lost in breast cancers.

ARHI is a maternally imprinted tumor suppressor gene that maps to a site on chromosome 1p31 where loss of heterozygosity has been observed in 40% of human breast and ovarian cancers. ARHI is expressed in normal ovarian and breast epithelial cells, but ARHI expression is lost in a majority of ovarian and breast cancers. Expression of ARHI from the paternal allele can be down-regulated by multiple mechanisms in addition to loss of heterozygosity. This article explores the role of DNA methylation in silencing ARHI expression. There are three CpG islands in the ARHI gene. CpG islands I and II are located in the promoter region, whereas CpG island III is located in the coding region. Consistent with imprinting, we have found that all three CpG islands were partially methylated in normal human breast epithelial cells. Additional confirmation of imprinting has been obtained by studying DNA methylation and ARHI expression in murine A9 cells that carry either the maternal or the paternal copy of human chromosome 1. All three CpG islands were methylated, and ARHI was not expressed in A9 cells that contained the maternal allele. Conversely, CpG islands were not methylated and ARHI was expressed in A9 cells that contained the paternal allele of human chromosome 1. Aberrant methylation was found in several breast cancer cell lines that exhibited decreased ARHI expression. Hypermethylation was detected in 67% (6 of 9) of breast cancer cell lines at CpG island I, 33% (3 of 9) at CpG island II, and 56% (5 of 9) at CpG island III. Hypomethylation was observed in 44% (4 of 9) of breast cancer cell lines at CpG island II. When methylation of CpG islands was studied in 20 surgical specimens, hypermethylation was not observed in CpG island I, but 3 of 20 cases exhibited hypermethylation in CpG island II (15%), and 4 of 20 cases had hypermethylation in CpG island III (20%). Treatment with 5-aza-2'-deoxycytidine, a methyltransferase inhibitor, could reverse aberrant hypermethylation of CpG island I, II and III and partially restore ARHI expression in some, but not all of the cell lines. Treatment with 5-aza-2'-deoxycytidine partially reactivated ARHI expression in cell lines with hypermethylation of CpG islands I and II but not in cell lines with partial methylation or hypomethylation of these CpG islands. To test the impact of CpG island methylation on ARHI promoter activity more directly, constructs were prepared with the ARHI promoter linked to a luciferase reporter and transfected into SKBr3 and human embryo kidney 293 cells. Methylation of the entire construct destroyed promoter activity. Selective methylation of CpG island II alone or in combination with CpG island I also abolished ARHI promoter activity. Methylation of CpG I alone partially inhibited promoter activity of ARHI. Thus, hypermethylation of CpG island II in the promoter region of ARHI is associated with the complete loss of ARHI expression in breast cancer cells. Other epigenetic modifications such as hypermethylation in CpG island III may also contribute to the loss of ARHI expression.

Polymorphisms of IL-4, IL-4R alpha, and AICDA genes in adult allergic asthma.

The relationship between 3 polymorphisms sites [interleulin-4 (IL-4), IL-4 receptor (IL-4R) alpha chain and activation-induced cytidine deaminase (AICDA)] and adult allergic asthma in China was studied. By using case-control method, DNA and clinical data were obtained from allergic asthmatic patients and compared with those in the control subjects. The subjects were genotyped for the IL-4 C-589T promoter polymorphism, the IL-4R alpha chain Q576R and the AICDA C8408T by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The results showed that the IL-4 C-589T was not associated with adult allergic asthma in China. However, the IL-4R alpha chain 576R/R and AICDA 8408T/T frequency was significantly increased in allergic asthma group as compared with that in the control group [odd ratio (OR) = 3.797 and 9.127, respectively; P < 0.01)] and was correlated with the increased plasma total IgE. These data suggested that the IL-4R alpha chain 576R/R and AICDA 8408T/T genotypes confer genetic susceptibility to adult allergic asthma in China.

Suppression of angiogenesis and tumor growth by adenoviral-mediated gene transfer of pigment epithelium-derived factor.

Pigment epithelium-derived factor (PEDF) was identified from retinal pigment epithelial cells and has been shown to display neurotrophic effects. In addition it has been found to induce a potent inhibition of angiogenesis. In this study we have explored whether overexpression of PEDF by a gene transfer approach can block tumor angiogenesis and reduce tumor growth. We found that cells infected with an adenovirus encoding PEDF under the control of the CMV promoter (AdPEDF) secreted PEDF protein into the medium that exhibited strong inhibitory effects on migration and tube formation of endothelial cells cultured in the presence of vascular endothelial growth factor. Moreover, the systemic administration of AdPEDF was able to inhibit angiogenesis in Matrigel assay in vivo, and treatment with this adenovirus of established hepatocellular carcinoma tumor in nude mice resulted in strong suppression of tumor growth. This anti-tumor effect could also be seen in a mouse lung carcinoma model by systemic administration of vector. In that model, treatment of tumor by intratumoral injection of AdPEDF also caused significant inhibition of tumor growth. The anti-tumor effect was related to a decrease in density of microvessels in tumors after treatment with AdPEDF. These data suggest that the antiangiogenic properties of PEDF can be exploited to inhibit the establishment of tumor neovasculature and reduce tumor growth.
