[Effects of soil moisture on photosynthetic characteristics of different specialized end-uses winter wheat at their later growth stages and on their yields].

In this study, pot experiments with various cultivars of specialized end-uses winter wheat were carried out at Henan Agricultural University Research Station during 2001-2003. The results showed that as for cultivar Yumai 34, the values of its chlorophyll meter (SPAD), PSII activity (Fv/Fo) and maximal PSII light energy transformation efficiency (Fv/Fm) were the highest when soil moisture content was at 60% of field capacity (FC), but the photochemical quenching coefficient (qP), non-photochemical quenching coefficient (qN), apparent photosynthetic electron transport rate (ETR) and the ratio of PSII photochemical quantum yield (phi2) were all increased at 80% of FC. For high-yielding cultivar Yumai 49, its SPAD, qP, qN and ETR were higher at 80% of FC, but Fv/Fo, Fv/Fm and phi2 were less affected by FC. For cultivar Yumai 50, the values of its photosynthetic characteristics were the highest at 80% of FC, except that the qN value was the lowest. The yield and the relative content of grain protein of cultivar Yumai 34 were also increased at 60% of FC. Furthermore, there was a significant difference among grain yields. The relative contents of grain protein of cultivars Yumai 49 and 50 were improved at 40% of FC, while the highest yields were obtained at 80% of FC. There were significant differences in the grain yields of Yumai 49 and 50 among different soil moisture treatments.

[Construction of Bacillus thuringiensis labeled recombinant strain and horizontal transfer of its cry1Ac10 gene].

A recombinant plasmid pBMBZGC10 was obtained by the ligation of gfp-cry1Ac10 fusion gene and vector plasmid pAD4412, which was then introduced by gene pulser into acrystalliferous strain CryB, and a recombinant strain CryB(pBMBZGC10) was obtained. Different fermentative solutions of recombinant strain were used for multi-spraying on Brassica pekinesis, Ipomoea aquatica and Lycopersicon esculentum leaves. The results of fluorescent detection and PCR amplification revealed that cry1Ac10 gene did not transfer into indigenous bacteria, actinomyces and fungi in test soil, and could not be detected in roots, stems and leaves of test plants.

[Preparation of anti-morphine monoclonal antibodies with complete cross reactivity with heroin].

OBJECTIVE: To prepare and characterize anti-morphine monoclonal antibodies (mAbs) with complete cross reactivity with heroin. METHODS: Morphine-6-succinyl was conjugated with Blue carrier (BC) in the presence of carbodi imide. BALB/c mice were immunized with the holoantigens (M-6-S-BC) and Freund adjuvant, and hybridoma was generated with mouse spleen cells and Sp2/0 myeloma cells. After 3 fusions and cloning, 4 hybridoma cell lines secreting monoclonal antibodies to morphine were obtained and the mABs purified from the mouse ascites followed by characterization with enzyme-linked immunosorbent assay (ELISA). RESULTS: Four hybridoma cell lines secreting anti-morphine mAbs were established and designated as G1, G3, H4 and B1, respectively. All the mAbs belonged to IgG1 isotype and could bind to heroin better than to morphine and codeine, which were identified by competitive inhibition ELISA. CONCLUSION: The anti-morphine mAbs can bind with the opioids, especially heroin, and can be useful for sensitive detection of opioids.

[Genotypic antiretroviral resistance testing and phylogenetic analysis of protease and reverse transcriptase in antiretroviral drug-naïve AIDS patients in Henan province].

OBJECTIVE: Frequency, type and clinical implications on protease and reverse transcriptase drug resistance mutations were investigated and phylogenetic analysis in antiretroviral drug-naïve AIDS patients was carried out in Henan province. METHODS: 45 plasma samples were separated from the anticoagulatory whole blood, from which reverse transcription-polymerase chain reaction technique was used to amplify the partial pol gene. The sequences were analysed for genotypic antiretroviral resistance and phylogenetic relation through landing the websites http://hivdb.stanford.edu and http://hiv-web.lanl.gov, under BioEdit and DNAClub software. RESULTS: Partial pol sequences of 36 samples were successfully amplified. The major mutation rate of resistance to protease was 8.3% (3/36), including types D30A, V32A, G73C and V82A. Minor mutation rate of resistance was 100%, including types of L63PS (36/36), I93L (35/36), V77IL (34/36), A71IVT (10/36) and D60E (2/36). The mutation rate of resistance to reverse transcriptase was 38.9% (14/36). Mutation-scoring and clinical implication clewed drug resistance rates were 5.6% (2/36) and 22.2% (8/36) to protease inhibitors and reverse transcriptase inhibitors respectively, while 1 sample was potentially low-level resistant to all of the protease inhibitors and 3 samples to part of the reverse transcriptase inhibitors. Phylogenetic analysis revealed that the pol gene of 36 samples were highly homologous and having a near relative to B.US.83.RF ACC M17451. 36 samples seemed to have the same infection source while their resistance mutations were not due to drug-resistant virus infection but to the evolving of virus in vivo. CONCLUSION: Most of the antiretroviral drug-naïve AIDS patients in Henan province were sensitive to the currently available antiviral medicine, but antiviral treatment must be in accordance with the strict procedure and to keep better adherence, to avoid the epidemics caused by drug-resistant virus.

[The diagnostic value of dynamic CT in solitary pulmonary nodules: a prospective study].

OBJECTIVE: To evaluate imaging features of benign and malignant solitary pulmonary nodules (SPN) using dynamic computed tomography (dCT) to improve the accuracy of radiological diagnosis. METHODS: Fifty-one patients with SPN were studied by dCT. In this procedure, a bolus of 100 ml contrast medium was administrated intravenously at a rate of 4 ml/sec. The same-located dynamic scans were carried out from 15 sec to 120 sec following the injection. Time-attenuation curves (TAC) were created according to circular or oval ROI drawn over nodules. Histopathological diagnosis was considered as the golden standard in all patients. Double-blind examination and evaluation were carried out and the data were analyzed statistically with Mann-Whitney U test. RESULTS: Thirty eight cases were diagnosed to be malignant SPN (mSPN) and 13 cases to be benign SPN (bSPN). The benign SPN were further divided into two groups, bSPN(1) consisting 6 cases with chronic pneumonitis, nodular tuberculosis or sclerosing hemangioma and bSPN(2) consisting 7 cases with tuberculoma, pulmonary cyst, pulmonary sequestration or mycosis ball. There were statistically significant differences between mSPN and bSPN(2) in peak heights of enhancement (87.6 HU vs. 57.8 HU, P < 0.01), enhancement values (peak heights- unenhanced CT values, 59.6 HU vs. 11.1 HU, P < 0.01). However, no statistically significant differences of those two values existed between mSPN and bSPN(1). TAC of mSPN reached to peak height more rapidly and remained a plateau. TAC of bSPN(1)s showed similar changes to that of mSPN despite a delayed reach to a peak height or even a descending branch. TAC of the bSPN(2)s was lower and flatter without peak height. If a threshold of 20 HU was taken for dCT, the lesions with < or = 20 HU were diagnosed as benign, and the lesions with > 20 HU were diagnosed as malignant, with a sensitivity of 100%, a specificity of 54.0% and an accuracy of 88.4%. CONCLUSION: (1) Absence of the marked enhancement (< or = 20 HU) in dynamic CT is strongly predictive of benignancy. (2) The peak height and enhancement value of dCT are helpful to differentiate malignant SPNs from benign ones. (3) The TAC configuration is helpful in differentiating malignant SPNs from benign ones. Descending branches could be found in some benign lesions, but not in the malignant ones. The TAC of tuberculoma and mycosis ball is usually relatively low and flat without any peak height.

Breastfeeding experiences of Japanese women living in Perth, Australia.

The aim of this study was to document the breastfeeding practices of Japanese-Australian mothers living in Perth. A cross-sectional survey of mothers who had delivered babies in Japan or Australia or both was carried out on a sample of 163 mothers recruited through Japanese social and cultural groups in Perth and by a 'snowball' technique. Factors involved in the decision to breastfeed were analysed using multivariate regression analysis. The main outcome measures were the initiation and duration of breastfeeding and cultural beliefs about breastfeeding. Breastfeeding initiation rates of the Japanese-Australian mothers in Japan and in Australia were higher than for other Australians and are consistent with breastfeeding rates in Japan. In australia, 65% of Japanese-Australian mothers were still breastfeeding at six months. The most common reason for the decision to cease breastfeeding was 'insufficient breastmilk'. The significant factors in breastfeeding duration were 'the time the infant was introduced to infant formula', 'the time when the feeding decision was made', 'doctors support breastfeeding' and 'the mother received enough help from hospital staff'; these were positively associated with the duration of breastfeeding. Japanese mothers take a lot of notice of advice given by health professionals about infant feeding practices.

Detection of FLT3 gene and FLT3/ITD mutation by polymerase chain reaction-single-strand conformation polymorphism in patients with acute lymphoblastic leukemia.

OBJECTIVE: To analyze Fms-like tyrosine kinase 3 (FLT3) gene and FLT3 internal tandem duplication (ITD) mutation in acute lymphoblastic leukemia (ALL) patients of different immunological subtypes. METHODS: Polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) was used to detect FLT3 gene and FLT3/ITD mutation in 63 ALL cases. RESULTS: Among the 63 ALL cases, FLT3 gene was detected in 41 (61.5%) cases. The positivity rate of FLT3 gene in pre-pre B-lineage ALL, pre-B-ALL, B-lineage ALL and T-lineage ALL cases were 93.3% (14/15), 77.8% (14/18), 41.7% (5/12) and 28.6% (4/14), respectively. The positivity rate of FLT3 gene was significantly higher in pre-pre B-ALL/pre B-ALL subtypes (84.8%) than in B-ALL subtypes (41.7%, P<0.005), and the rate was significantly higher in B-ALL subtypes (73.3%) than in T-ALL subtypes (28.6%, P<0.001). Two cases (3.2%) were found to have FLT3/ITD mutation, which were also positive for myeloid antigen expression and diagnosed as acute mixed-lineage leukemia, showing leukocytosis and high percentage of bone marrow blast cells with poor prognosis. CONCLUSIONS: FLT3 gene can be detected in both B-and T-lineage ALL patients, but more frequently in the former. In B-lineage ALL patients, FLT3 gene is more frequent in cases with undifferentiated than those with differentiated blast cells. FLT3/ITD is rarely detected in ALL patients and FLT3/ITD mutation detection might be helpful to identify the genotypes and evaluate the prognosis of acute leukemia.

[Effects of extract from Cornus officinalis on nitric oxide and NF-kappaB in cortex of cerebral infarction rat model].

OBJECTIVE: To observe the change of nitric oxide (NO) and expression of nuclear factor-kappa B (NF-kappaB) in the cortex of cerebral infarction rat induced by photochemical reaction, and study the effect of extract from Cornus officinalis (whose main ingredient is iridoid glycoside) in the course of disease. METHOD: After rats were fed with experimental drugs for 7 days, the model of cerebral infarction was induced. Spectrophotography and immunohistochemistry were used to detect the change of the content of NO, NOS and the expression of NF-kappaB in the cortex. RESULT: Compared with control group, distinct infarction was visible in the model group, and the content of NO, the activity of NOS and the positive cell number of NF-kappaB were increased obviously. Compared with model group, the extract of C. officeinalis decreased the area of infarction, the content of NO, the activity of NOS and the positive cell number of NF-kappaB. CONCLUSION: The iridoid glycoside of C. officinalis may have therapeutical effect on cerebral infarction through regulating the content of NO and NF-kappaB.

[Expression of PKD1 and PKD2 transcripts and proteins and its significance in different types of kidney tissues and kidney lines].

OBJECTIVE: To investigate the expression and function of PKD1 and PKD2 in different kidney tissues and cell lines. METHODS: Immunoprecipitation, Western blotting, In situ hybridization and immunohistochemical staining methods were used to observe the expression of PKD1 mRNA and PKD2 mRNA and their protein abundance in different kidney tissues and cell lines. RESULTS: Coordinate expressions of PKD1 and PKD2 were found in all kidney tissues and cell lines. Distribution of PKD1 mRNA and PKD2 mRNA and their protein polycystin-1 and polycystin-2 in normal human adult kidney tissue were mainly expressed in the medullary collecting ducts and distal tubules. Positive staining was also found in the majority of cyst-lining epithelial cells of PKD1 cystic kidney tissue, PKD1 cyst-lining epithelia cell line and LLC-PK1. The expression level of them in cystic epithelia of ADPKD kidney tissue was much higher than that in adult renal tubules (P < 0.01). CONCLUSIONS: Similar expression pattern of PKD1 and PKD2 and their different tissue distribution in different kidney tissues show that the molecular mutuality of PC-1 and PC-2 might be the base of their functional correlation. Polycystins might play an important role in the maintenance of tubular architecture.

[A study on the influenza surveillance program in Tianjin, 2004].

OBJECTIVE: To explore the trend of influenza epidemics, predominate strains of the virus in Tianjin city and to analyze the economic impact of vaccine-based interventions for the prevention and control of influenza. METHODS: Data on epidemiological studies and on virus surveillance was gathered. Monte Carlo mathematical simulation modelwas used for data analyses. RESULTS: From 29, December, 2003 to 2, January, 2005, the proportion of influenza-like infection cases was accounted for 8.93% of the total number of patients from the outpatient departments of 4 general hospitals in Tianjin. The proportion reached its peak from November to February and it was accounted for 9.39% in epidemic period, which was significantly different from that in the non-epidemic period (u = 15.53, P< 0.05). There was 56 strains of influenza virus isolated in which 45 were indentified as A(H3N2) and 11 as B with a total positive rate as 13.21%. Predominant strain was found bing type A(H3N2). The positive rate was 15.41% in the epidemic period, which was significantly different from that in the non-epidemic period (u = 2.519, P < 0.05). The cost per visit ranged from 475.93 to 581.69 Yuan (RMB) with an average cost of 528.81 Yuan. When the attack rate increased to 30 percent, the positive mean net returns would have been 24 million Yuan among the 0-19 age group. CONCLUSION: Influenza did not seem to be prevalent in Tianjin from 29, December, 2003 to 2, January, 2005. The main type of influenza was type A(H3N2). It is necessary to pay attention to the all-year round surveillance program due to the transformation of influenza type A and the slight increase of incidence in summer.

Genome-wide linkage scan identifies a novel genetic locus on chromosome 5p13 for neonatal atrial fibrillation associated with sudden death and variable cardiomyopathy.

BACKGROUND: Atrial fibrillation (AF) is the most common sustained cardiac arrhythmia, and patients with AF have a significantly increased risk for ischemic stroke. Approximately 15% of all strokes are caused by AF. The molecular basis and underlying mechanisms and pathophysiology of AF remain largely unknown. METHODS AND RESULTS: We have identified a large AF family with an autosomal recessive inheritance pattern. The AF in the family manifests with early onset at the fetal stage and is associated with neonatal sudden death and, in some cases, ventricular tachyarrhythmias and waxing and waning cardiomyopathy. Genome-wide linkage analysis was performed for 36 family members and generated a 2-point logarithm of the odds (LOD) score of 3.05 for marker D5S455. The maximum multipoint LOD score of 4.10 was obtained for 4 markers: D5S426, D5S493, D5S455, and D5S1998. Heterozygous carriers have significant prolongation of P-wave duration on ECGs compared with noncarriers (107 versus 85 ms on average; P=0.000012), but no differences between these 2 groups were detected for the PR interval, QRS complex, ST-segment duration, T-wave duration, QTc, and R-R interval (P>0.05). CONCLUSIONS: Our findings demonstrate that AF can be inherited as an autosomal recessive trait and define a novel genetic locus for AF on chromosome 5p13 (arAF1). A genetic link between AF and prolonged P-wave duration was identified. This study provides a framework for the ultimate cloning of the arAF1 gene, which will increase the understanding of the fundamental molecular mechanisms of atrial fibrillation.

Genomewide analysis of gene expression associated with Tcof1 in mouse neuroblastoma.

Mutations in the Treacher Collins syndrome gene, TCOF1, cause a disorder of craniofacial development. We manipulated the levels of Tcof1 and its protein treacle in a murine neuroblastoma cell line to identify downstream changes in gene expression using a microarray platform. We identified a set of genes that have similar expression with Tcof1 as well as a set of genes that are negatively correlated with Tcof1 expression. We also showed that the level of Tcof1 and treacle expression is downregulated during differentiation of neuroblastoma cells into neuronal cells. Inhibition of Tcof1 expression by siRNA induced morphological changes in neuroblastoma cells that mimic differentiation. Thus, expression of Tcof1 and treacle synthesis play an important role in the proliferation of neuroblastoma cells and we have identified genes that may be important in this pathway.

Apoptosis pathway of liver cells in chronic hepatitis.

AIM: To study the pathway of apoptosis in chronic liver disease and the role of mitochondria in programmed cell death. METHODS: Liver biopsy specimens from 72 cases of chronic hepatitis and 29 cases of post hepatitis cirrhosis were studied. The pro-apoptotic protein Fas, FasL, Bax and the anti-apoptotic protein Bcl-2, Bcl-x(L), Bcl-2 alpha were studied immunohistochemically by SP method. Specimens from 15 cases of chronic hepatitis and post hepatitis cirrhosis were examined for their ultramicrostructures with special attention to their mitochondrial changes. Specimens from 3 normal adults (demised in traffic accidents) were used as control. RESULTS: The expression of proapoptotic proteins (Fas, FasL, Bax) in hepatocytes was significantly higher in the chronic hepatitis group than in the cirrhosis group (P<0.001). In the study of ultramicrostructure 364 hepatocytes were examined, from 12 cases of chronic hepatitis (including 10 mild cases, 1 moderate case and 1 severe case). Out of 364 hepatocytes 40 (11.0%) hepatocytes were found with various kinds of destruction in their mitochondria. Rupture of the outer membrane of mitochondria and the leakage of matrix from the intermembrane space were definitely demonstrated. The ultramicrostructural changes of mitochondria in the chronic hepatitis group were statistically higher than that in normal adults control group (chi(2)=4.32, P<0.05). CONCLUSION: The result of the study was in support of the current view that the apoptotic process in chronic hepatitis patients were largely along the intrinsic pathway (mitochondrial pathway), given that the intrinsic and extrinsic pathways could interlinked (converged) at some point on their progression, also it is impossible at present to exclude the possibility that the two pathways could be chosen by hepatocytes in parallel simultaneously.
