Squamous cell carcinoma and mammary abscess formation through squamous metaplasia in Smad4/Dpc4 conditional knockout mice.

Smad4 is a central mediator for TGFbeta signals, which play important functions in many biological processes. To study the role of Smad4 in mammary gland development and neoplasia, we disrupted this gene in mammary epithelium using a Cre-loxP approach. Smad4 is expressed in the mammary gland throughout development; however, its inactivation did not cause abnormal development of the gland during the first three pregnancies. Instead, lack of Smad4 gradually induced cell proliferation, alveolar hyperplasia and transdifferentiation of mammary epithelial cells into squamous epithelial cells. Consequently, all mutant mice developed squamous cell carcinoma and/or mammary abscesses between 5 and 16 months of age. We demonstrated that absence of Smad4 resulted in beta-catenin accumulation at onset and throughout the process of transdifferentiation, implicating beta-catenin, a key component of the Wnt signaling pathway, in the development of squamous metaplasia in Smad4-null mammary glands. We further demonstrated that TGFbeta1 treatment degraded beta-catenin and induced epithelial-mesenchymal transformation in cultured mammary epithelial cells. However, such actions were blocked in the absence of Smad4. These findings indicate that TGFbeta/Smad4 signals play a role in cell fate maintenance during mammary gland development and neoplasia.

[Development of the human/rat chimera model with neonatal rats].

The purpose of this study was to transplant neonatal rat with human cord blood Lin(-) cells to test the possibility of this xenograft model. The Lin(-) cells were purified from human cord blood (CB) using negative selection strategy based on different lineage-specific antigens. The Lin(-) cells were injected into the liver of neonatal rats using a microinjector at an average of 5 x 10(5) cells for each. Peripheral blood (PB) and spleen were collected at 2,4 and 8 weeks after injection. Flow cytometry was performed to detect human cells in the rat PB, PCR was used to detect human cells in PB as well as spleen. The results showed that a definite proportion of human cells existed in peripheral blood of chimeric rat and the human specific beta2 microglobulin gene fragments were detected in spleen genomic DNA of chimeric rat. It is concluded that human/rat chimera model can be developed with neonatal rats. Human/rat xenograft model may provide a useful and convenient method for human hematopoietic stem cell assay in vivo.

[In vitro fluoroquinolone resistance in ocular bacterial isolates].

OBJECTIVE: To assess the in vitro resistance of ocular bacterial isolates to fluoroquinolone that are commonly used in the treatment of ocular infection. METHODS: Ocular bacterial isolates (n = 347) from Beijing Institute of Ophthalmology were tested for their resistance to ofloxacin, ciprofloxacin and norfloxacin. In vitro susceptibility testing was determined by the Kirby-Bauer disk diffusion method. RESULTS: The total number of ocular bacterial isolates was 347 from January, 1999 to December, 2000 including gram-positive cocci 192 (55.3%), gram-negative cocci 16 (4.6%), gram-positive bacilli 44 (12.7%), gram-negative bacilli 89 (25.7%), Nocardia 6 (1.7%). The percentage of resistance in total ocular bacterial isolates to ofloxacin, ciprofloxacin and norfloxacin was 28.5%, 25.9% and 34.9% respectively. The resistance of streptococcus species to ofloxacin (5.9%) and ciproloxacin (11.8%) was significantly lower than gentamycin (52.9%) and tobramycin (70.6%) (P < 0.01), and to norfloxacin (23.5%) was significantly lower than tobramycin (P < 0.01). Pseudomonas showed significantly lower resistance to ciprofloxacin (6%) ofloxacin (14.0%) and norfloxacin (14.0%) than gentamycin (42.0%) (P < 0.01 and P < 0.05). The percentage of resistance in staphylococcus species to norfloxacin (46.8%) was higher than gentamycin (30.8%) and tobramycin (27.6%) (P < 0.05 and P < 0.01 separately). The resistance of gram-positive bacilli to ofloxacin, ciproloxacin and norfloxacin. was equivalent (P > 0.05). CONCLUSIONS: The fluoroquinolones, ofloxacin and ciprofloxacin have higher overall in vitro efficacy on most of the ocular bacteria than gentamycin and tobramycin, but ofloxacin and ciprofloxacin are equivalent to gentamycin and tobramycin in efficacy against staphylococcus species.

Influence of patients' age on functional recovery after transplantation of olfactory ensheathing cells into injured spinal cord injury.

OBJECTIVE: To evaluate the restoration of function after spinal cord injury (SCI) in patients of different ages who have underwent intraspinal transplantation of olfactory ensheathing cells (OECs). METHODS: One hundred and seventy-one SCI patients were included in this study. Of them, 139 were male and 32 were female, with age ranging from 2 to 64 years (mean, 34.9 years). In all SCI patients the lesions were injected at the time of operation with OECs. According to their ages, the patients were divided into 5 groups: </= 20 years group (n = 9), 21 - 30 years group (n = 54), 31 - 40 years group (n = 60), 41 - 50 years group (n = 34) and > 51 years group (n = 14). The spinal cord function was assessed based on the American Spinal Injury Association (ASIA) Classification System before and 2 - 8 weeks after OECs transplantation. One-way ANOVA and q test were used for statistical analysis, and the data were expressed as mean +/- SD. RESULTS: After surgery, the motor scores increased by 5.2 +/- 4.8, 8.6 +/- 8.0, 8.3 +/- 8.8, 5.7 +/- 7.3 and 8.2 +/- 7.6 in 5 age groups respectively (F = 1.009, P = 0.404); light touch scores increased by 13.9 +/- 8.1, 15.5 +/- 14.3, 12.0 +/- 14.4, 14.1 +/- 18.5 and 24.8 +/- 25.3 respectively (F = 1.837, P = 0.124); and pin prick scores increased by 11.1 +/- 7.9, 17.2 +/- 14.3, 13.2 +/- 11.8, 13.6 +/- 13.9 and 25.4 +/- 24.3 respectively (F = 2.651, P = 0.035). Restoration of pin prick in > 51 years group was better than other age groups except 21 - 30 years group. CONCLUSION: OECs transplantation can improve the neurological function of spinal cord of SCI patients regardless of their ages. Further research into the long-term outcomes of the treatment will be required.

[Kinetics of P-chlorophenol wastewater treatment by UV/H2O2 oxidation].

The photodegradation of p-cholorophenol in a hydrogen peroxide-aided photolysis process was investigated. The reaction was influenced by the original concentration of p-chlorophenol and H2O2 addition and carrier gas. With the aid of dioxygen, the removal of phenol and CODCr were reached about 96% and 50% respectively if the original concentration of H2O2 added was only half of stoichiometric calculation. A pseudo-first order kinetic model was adopted to represent the reaction.

An event-related potential study on visual perceptual learning under short-term and long-term training conditions.

The present study investigated learning-induced changes of event-related potentials in a visual discrimination task, focusing on making a comparison between short-term and long-term learning perceptual processes. Event-related potentials were recorded from two groups of human adults. One group (short-term training group) was given 1.5 h of training in a single day, and another group (long-term training group) was given the same training procedure (1.5 hour of training) on each of 3 consecutive days. The results demonstrated that for short-term training group, along with the reduction of reaction times, the amplitudes of N1 and N2 negativities over the central/parietal areas decreased during the training. For long-term training group, however, after long-term training, the N2 effect disappeared and the N1 effect occurred over the posterior areas. In addition, the amplitudes of N2 for long-term training group were less than those for short-term training group. Our results suggest that neural activity depends not only on perceptual mechanisms and on the parameters of the physical stimuli but also on the extent of the observer's previous learning.

[Industrial hand injuries by punch machines: a case-control study].

OBJECTIVE: To identify factors associated with industrial hand injuries by punch machines. METHODS: From September, 1999 to June, 2000, 108 patients of industrial hand injuries by punch machines were collected. For each patient, 2 controls were randomly selected. One was from the same industry; the other was from a big industry with thousands of punch machines. They matched on gender, job and age. In-person interviews were conducted to collect data on demographics, work history, work characteristics, work injuries, work organization, health behaviors, anthropometric and psychological factors. The Eysenck Personality Questionnaire (EPQ) was selected to evaluate the psychological performance. Risk factors were examined via multivariate logistic regression techniques. RESULTS: There were four important factors affecting hand injuries by punch machines: The first was the length of service index (odds ratio [OR] = 0.972, 95% confidence interval [CI] = 0.957, 0.988). The second index was neuroticism (OR = 1.649, 95% CI = 1.264, 2.151). A safety equipment index (OR = 0.260, 95% CI = 0.123, 0.547) and a safety operation criterion index (OR = 0.136, 95% CI = 0.064, 0.286) were also significant factors. CONCLUSIONS: Results suggest that the accident-prone population are those who have shorter service length and neurotic psychological performance. It may reduce the incidence of industrial hand injuries by punch machine to strengthen the appliance of safety equipment and enforcement of safety operation criterion, especially to the accident-prone population.

CpG-oligodeoxynucleotide rejection of a neuroblastoma in A/J mice does not induce a paraneoplastic disease.

Oligodeoxynucleotides containing CpG motifs (CpG-ODN) are powerful immunostimulating agents that are currently entering clinical trials in various human diseases. Concerns exist about potential auto-immune diseases triggered by such treatment. We thus investigated whether tumor rejection induced by CpG-ODN treatment could lead to a harmful auto-immune reaction against the nervous system (neurological paraneoplastic disease) at the time of acute tumor rejection, or in long-term surviving animals. Mice bearing established neuroblastomas were treated with intra-tumoral injections of CpG-ODN, resulting in tumor inhibition and tumor rejection in one-third of the animals. Immunocytochemistry and Western blot studies revealed no specific anti-neuronal antibodies. None of the animals developed neurological disabilities and histological studies of the nervous system were normal. CpG-ODN can therefore trigger neuroblastoma rejection without inducing neurological paraneoplastic disease.

Skin-stage schistosomula of Schistosoma mansoni produce an apoptosis-inducing factor that can cause apoptosis of T cells.

Skin-stage schistosomula of Schistosoma mansoni were found to secrete molecules that are pro-apoptotic for skin T lymphocytes as measured by annexin V staining, caspase-3 activity, caspase-8 activities, and DNA fragmentation. Caspase-8 activities in lymphocytes peaked approximately 8 h and caspase-3 activity peaked approximately 16 h after exposure to the parasite secretions. Subset analysis showed that mainly CD4(+) and CD8(+) cells (but not B cells) were susceptible to the parasite-induced pro-apoptotic effect. In situ staining confirmed the presence of apoptotic T cells around challenge parasites in the skin of naive or immunized animals. Analysis of T cells to identify the potential molecular pathway of the parasite-induced apoptosis showed increases in the expression of Fas, FasL, and the Fas-associated death domain. Blocking of FasL with a fusion protein reversed the parasite-induced apoptosis, suggesting a role for the Fas/FasL-mediated pathway in the parasite-induced T cell apoptosis. Subsequent analyses of the secretions of skin-stage schistosomula identified the pro-apoptotic activity as being associated with a protein of approximately 23 kDa. This protein was termed S. mansoni-derived apoptosis-inducing factor.

Cloning and characterization of a calcium-binding, histamine-releasing protein from Schistosoma mansoni.

A homologue of the mammalian translationally controlled tumor protein (TCTP) was cloned from the human parasite Schistosoma mansoni (SmTCTP). Sequence analysis showed that SmTCTP differed from other reported TCTPs in having only one signature sequence. Subsequently, SmTCTP was cloned in a T7 expression system and expressed as a histidine-tagged fusion protein. Recombinant SmTCTP (rSmTCTP) has a molecular mass of approximately 23 kDa with the histidine tag. Further analysis showed that SmTCTP transcripts and protein are expressed in all life cycle stages of the parasite within the vertebrate hosts. Interestingly, antibodies to SmTCTP were present in the sera of mice 9 weeks after infection with S. mansoni. Characterization studies showed that rSmTCTP is a calcium-binding protein that can cause histamine release from basophil/mast cells and induce eosinophil infiltration. These findings suggest that SmTCTP may have an important role in the development of allergic inflammatory responses associated with schistosomiasis and may be a target for new drug development.

A comparison of whole-genome shotgun-derived mouse chromosome 16 and the human genome.

The high degree of similarity between the mouse and human genomes is demonstrated through analysis of the sequence of mouse chromosome 16 (Mmu 16), which was obtained as part of a whole-genome shotgun assembly of the mouse genome. The mouse genome is about 10% smaller than the human genome, owing to a lower repetitive DNA content. Comparison of the structure and protein-coding potential of Mmu 16 with that of the homologous segments of the human genome identifies regions of conserved synteny with human chromosomes (Hsa) 3, 8, 12, 16, 21, and 22. Gene content and order are highly conserved between Mmu 16 and the syntenic blocks of the human genome. Of the 731 predicted genes on Mmu 16, 509 align with orthologs on the corresponding portions of the human genome, 44 are likely paralogous to these genes, and 164 genes have homologs elsewhere in the human genome; there are 14 genes for which we could find no human counterpart.

[Stereotactic biopsy for intracranial deep lesions].

OBJECTIVE: To investigate the histological diagnostic efficacy and accuracy of stereotactic biopsy for intracranial deep lesions under computerized tomography (CT) and magnetic resonance imaging (MRI) guidance. METHODS: CT or MRI-guided stereotactic biopsy was performed in 605 cases with intracranial deep lesions. All the procedures were done under local anesthesia with Leksell stereotactic system. CT or MRI film was used to determine the coordinate in 450 cases and the computer-assisted planning for neurosurgery (CAPN) in 155 cases. RESULTS: Brain tumors, inflammatory lesions, and abnormal but nonspecific lesions were diagnosed pathologically in 537 cases (88.75%), 30 cases (4.96%), and 13 cases (2.15%) respectively. In 20 cases (3.31%) the biopsy procedure failed to provide useful pathological evidence. The total positive rate of biopsy was 96.69%. Complications associated with biopsy occurred in 18 patients (2.98%), and 2 patients (0.33%) died following the procedure. CONCLUSION: Stereotactic biopsy using MRI-guided stereotactic approach or CT-guided technique is a safe and reliable method for histological diagnosis of intracranial deep lesions.

Schistosoma mansoni, S. haematobium, and S. japonicum: early events associated with penetration and migration of schistosomula through human skin.

Migratory pattern of schistosomula of Schistosoma mansoni, S. haematobium, and S. japonicum through human skin were analyzed in skin organ cultures. These studies showed that the schistosomula of S. mansoni and S. haematobium has similar migratory patterns through human skin. During the first 24h after infection nearly 90% of S. mansoni and S. haematobium schistosomula were present only in the epidermis. Majority of the schistosomula were found in the dermis only after 48h and they appear to reach the dermal vessels around 72h after infection. Migratory pattern of S. japonicum on the other hand was significantly different from the other two species in that over 90% of the parasites had already reached the dermis within the first 24h and schistosomula were present in the dermal vessels within 2h after infection. Analysis of the cytokine pattern at 8h after infection by a macro gene array and RT-PCR analysis showed that out of 24 different cytokines analyzed only IL-1ra, IL-10, and TNF-alpha were increased in the human skin following infections with S. mansoni and S. haematobium, whereas, after infection with S. japonicum there was significant increases in IL-1beta, IL-1ra, IL-2, IL-6, IL-8, IL-10, IL-15, IL-18, and TNF-alpha. Immunohistochemical analysis of epidermal sheets showed focal accumulation of HLA-DR(+) cells in areas where schistosomula of S. mansoni had entered the human skin.

Structure of a TonEBP-DNA complex reveals DNA encircled by a transcription factor.

Tonicity-responsive enhancer binding protein (TonEBP), also known as NFAT5, is a unique member of the NFAT family of transcription factors that regulates gene expression induced by osmotic stress in mammalian cells. Unlike monomeric members of the NFAT family, TonEBP exists as a homodimer and binds asymmetric TonE DNA sites; furthermore, the affinity of TonEBP for DNA is much lower than that of other NFAT proteins. How TonEBP recognizes the TonE site and regulates the activation of hypertonicity response genes has not been clear. Here we show that TonEBP adopts a NF-kappaB-like structure upon binding to DNA, providing a direct structural link between the NFAT and NF-kappaB family of transcription factors. We also show that TonEBP completely encircles its DNA target and present biochemical evidence that the DNA encirclement may lead to increased kinetic stability of the TonEBP-DNA complex. Thus, the list of proteins that bind DNA by encirclement is now expanded to include sequence-specific transcription factors.
