Research of blastocyte-like structure in chicken.

The chicken embryo is a classic model used to investigate embryonic development, gene expression, and tissue differentiation, and is also an important research tool in studying the animal functional genomics. The whole blastoderms of fresh unincubated eggs from White Leghorn chickens were collected with a paper ring, mechanically broken into small pieces and cultured in medium. Then the small pieces would develop into blastocyte-like structures (BLS), which could be facilitated by an addition of fetal bovine serum (FBS) to the primary culture and their diameter was nearly doubled from 12 to 24 h. The additional yolk had no positive effect on the development in the first 12 h but encouraged the BLSs attaching and inner cells differentiating instead in 24 h. The inner cells of the BLS showing a high alkaline-phosphatase activity similar to those in mouse embryonic stem (ES) cells and also expressing a large amount of the specific stage embryonic antigen-1 (SSEA-1) on the surface, which was known to be the characteristic of non-differentiated mouse and avian ES cells, could finally differentiate into nerve-like cells, fibroblast cells and so on in the medium. Leukemia inhibitory factor (LIF) facilitated the cells' proliferation and prevented differentiation in the suspended culture of the BLSs. So we drew the conclusion that the BLS obtained from broken blastoderm can be used to amplify avian ES cells so as to initiate a new method of producing transgenic chickens.

Software reliability prediction using recurrent neural network with Bayesian regularization.

A recurrent neural network modeling approach for software reliability prediction with respect to cumulative failure time is proposed. Our proposed network structure has the capability of learning and recognizing the inherent internal temporal property of cumulative failure time sequence. Further, by adding a penalty term of sum of network connection weights, Bayesian regularization is applied to our network training scheme to improve the generalization capability and lower the susceptibility of overfitting. The performance of our proposed approach has been tested using four real-time control and flight dynamic application data sets. Numerical results show that our proposed approach is robust across different software projects, and has a better performance with respect to both goodness-of-fit and next-step-predictability compared to existing neural network models for failure time prediction.

A novel approach for short-term load forecasting using support vector machines.

A support vector machine (SVM) modeling approach for short-term load forecasting is proposed. The SVM learning scheme is applied to the power load data, forcing the network to learn the inherent internal temporal property of power load sequence. We also study the performance when other related input variables such as temperature and humidity are considered. The performance of our proposed SVM modeling approach has been tested and compared with feed-forward neural network and cosine radial basis function neural network approaches. Numerical results show that the SVM approach yields better generalization capability and lower prediction error compared to those neural network approaches.

[Studies on chemical constituents from herb of Hemiphragma heterophyllum].

OBJECTIVE: To study the chemical constituents from Hemiphragma heterophyllum. METHOD: The constituents were isolated and purified by chromatographic technology and their structures were elucidated by physico-chemical evidence and spectral data. RESULT: Eight compounds were isolated and identified as hemiphroside C (I), plantainoside E (II), hemiphroside A (III), acetoside (IV), globularin (V), 10-(Z)-cinnamoyl-catapol (VI), iso-scrophularioside (VII), cinnamic acid (VIII). CONCLUSION: Compound I was a new phenylethanoid glucoside and compounds II and VI were isolated for the first time from this plant.

Purification, crystallization and preliminary X-ray studies of a p-nitrophenylphosphatase from Bacillus stearothermophilus.

Thermostable p-nitrophenylphosphatase from Bacillus stearothermophilus has been expressed in Escherichia coli, purified and crystallized. The crystals belong to space group C(2), with unit-cell parameters a = 67.17 A, b = 57.84 A, c = 62.49 A and alpha = 90.0 degrees, beta = 95.4 degrees, gamma = 90.0 degrees. Diffraction data were collected to 1.40 A resolution with a completeness of 94.7% (96.6% for the last shell), an R(fac) value of 0.074 (0.341) and an I/sigma (I) value of 30.1 (2.67).
