Short-term frequency-dependent plasticity at recurrent mossy fiber synapses of the epileptic brain.

The recurrent mossy fiber pathway of the dentate gyrus expands dramatically in human temporal lobe epilepsy and in animal models of this disorder, creating monosynaptic connections among granule cells. This novel granule cell network can support reverberating excitation but is difficult to activate with low-frequency stimulation. This study used hippocampal slices from pilocarpine-treated rats to explore the dependence of synaptic transmission in this pathway on stimulus frequency. Minimal electrically evoked EPSCs exhibited a high failure rate ( approximately 60%). Stimulus trains delivered at a frequency of <1 Hz depressed synaptic transmission, as evidenced by an increase in response failures. Conversely, stimulus trains delivered at higher frequencies reduced the percentage of response failures and increased the amplitude of compound EPSCs, including pharmacologically isolated NMDA receptor-mediated EPSCs. Short-term frequency-dependent facilitation was of modest size compared with mossy fiber synapses on other neuronal types. Facilitation depended on the activation of kainate receptors by released glutamate and was inhibited by feedback activation of type II metabotropic glutamate receptors. These results suggest that the recurrent mossy fiber pathway may be functionally silent during baseline asynchronous granule cell activity in vivo attributable, in part, to progressive transmission failure. The pathway may synchronize granule cell firing and may promote seizure propagation most effectively during the brief periods of high-frequency granule cell firing that occur during normal behavior, during the periods of hypersynchronous fast activity characteristic of epileptic brain and, most importantly, during the period of increasing granule cell activity that precedes a spontaneous seizure.

[Thermo-sensitive period and critical temperature of fertility transition of thermo-photo-sensitive genic male sterile wheat].

The thermo-sensitive period and the critical temperature of fertility transition of C49S, a principal thermo-photosensitive genic male sterile line in two-line hybrid wheat, was studied in the growth chambers for controlling temperature and photoperiod. The seeds were sown on different time for some years. The results showed that the thermo-sensitive period in fertility expression of C49S was from PMC formation stage to mature pollen stage, and there were two most sensitive stages to temperature on fertility expression. One was the PMC meiosis stage, and the other was the middle microspore stage. The critical temperatures evoking a complete male sterility were the mean minimum temperature at PMC meiosis stage (Tmin1), the mean temperature at microspore stage (T2) and the mean minimum temperature at microspore stage (Tmin2) lower than 8.5 degrees C, 13.5 degrees C and 10.5 degrees C, respectively. The critical temperatures keeping a nearly normal male fertility Tmin1 and T2 and Tmin2 were higher than 11.5 degrees C, 15.0 degrees C and 12.5 degrees C, respectively. The value as well as the conditions and the risks of thermo-photo-sensitive genic male sterile line of wheat applied to hybrid wheat were evaluated in this paper.

[Biological characteristics of human umbilical artery smooth muscle cells cultured in vitro and the preestablishment of immortalized cell line].

OBJECTIVE: To establish the best primary and propagated culture approach of the human umbilical artery smooth muscle cells (HUASMC) and immortalized the primary cells. METHODS: The primary culture of HUASMC was performed using the primary-explant method, and the effects on the cells' growth characteristic by different culture mediums were conducted. Primary cultures of HUASMC derived from normal human umbilical arteries were transfected with recombination plasmid containing human telomerase reverse transcriptase (hTERT) open reading frame. The transfected HUASMC was analyzed for cell biological characteristics, and growth characteristics, and specific alpha-actin. RESULTS: Highly purified primary HUASMC were gained and could be propagated about 10 population doublings (PD). After PD 4, the cells lost the proliferation ability gradually. However, the transfected HUASMC could be propagated at least 20 population doublings and could express specific alpha-actin. CONCLUSION: The hTERT transfected HUASMC could keep the basic biological characteristic of smooth muscle cells and prolonged life span at least 20 PD. It might be a good cell model for the study of cardiovascular disease, myoma of uterus and chronic graft disfunction. It might also bring the hope to engineered vascularization in the future.

Factors contributing to the incompatibility between simplified-step adhesives and chemically-cured or dual-cured composites. Part III. Effect of acidic resin monomers.

PURPOSE: This study examined the polymerization kinetics of acid-contaminated light- and chemically-cured resins with the use of differential scanning calorimetry. MATERIALS AND METHODS: Light-cured and chemically-cured versions of an experimental bis-GMA/TEG-DMA resin at the ratio (w:w) of 62:38 were prepared. Four acidic resin monomers with either carboxylic (DSDM and MAA) or phosphoric functional groups (MP and 2MP) were added at a 10 wt% concentration to these resins to simulate the intermixing of resin composites and unpolymerized acidic monomers along the adhesive-composite interface of simplified-step dentin adhesives. Different concentrations of the most acidic monomer 2MP (10 to 50 wt% for the light-cured resin, and 2 to 4 wt% for the chemically-cured resin) were also added to examine their contribution to reducing the extent and rate of polymerization of these resin mixtures. The effect of sodium benzene sulphinate on the polymerization of the chemically-cured resin that was contaminated with 10 wt% 2MP was also investigated. RESULTS: At 10 wt% concentration, all acidic monomers had a limited effect on the polymerization of the light-cured resin. The rate and extent of polymerization in the chemically-cured resin was substantially reduced by the carboxylic acid monomers DSDM and MAA, while polymerization was completely inhibited in the presence of the organophosphate monomers MP and 2MP. Substantial reductions in the rate and extent of polymerization of the light-cured resin occurred only in the presence of high concentrations (30 to 50 wt%) of 2MP. More acute polymerization inhibition was observed when the chemically-cured resin was contaminated with 2 to 3 wt% 2MP, with polymerization completely inhibited at 4 wt%. Addition of sodium benzene sulphinate to 2MP-contaminated chemically-cured resin revived the previously uncured resins, but their rate and extent of polymerization were still inferior to that of uncontaminated chemically-cured resin. CONCLUSION: When the influence of adhesive permeability was excluded with the use of neat, water-free resins, deactivation of the tertiary amine utilized in the chemically-cured resin by even very low concentrations of acidic resin monomers accounts for the reported incompatibility between simplified-step dentin adhesives and chemically-cured composites. The polymerization of light-cured resin is only affected by much higher concentrations of acidic resin monomers.

Effect of the iron chelator desferrioxamine on manganese-induced toxicity of rat pheochromocytoma (PC12) cells.

Alterations in iron levels are likely to influence the biological actions of Mn in PC12 cells, because both metals are transported via the divalent metal transporter 1 (DMT1; also Nramp2 or DCT1). Studies were performed to determine the effect of the iron chelator desferrioxamine (DfO) on Mn-induced PC12 cell death and neuronal differentiation. Cell death almost doubled when PC12 cells were exposed for 24 hr to both DfO (10 microM) and Mn (0.3 mM) as opposed to Mn alone. DfO also stimulated Mn-induced neuronal differentiation by enhancing the phosphorylation of both ERK1 and 2 and also attenuated the increase in caspase 3-like activity induced by 0.3 mM Mn by approximately 50%, indicating that caspase activation, as reported previously, does not contribute to Mn-induced PC12 cell death. DfO also affected Mn-induced suppression of mitochondrial function as indicated by an additional 16% loss of ATP formation in PC12 cells cotreated with 0.3 mM Mn. Because sequestration of iron by DfO would be expected to lead to increased transport of Mn, studies were performed to determine whether iron inhibited Mn transport in PC12 cells. Iron inhibited 54Mn transport with an IC50 of approximately 20 microM. In addition, coincubation of DfO with Mn in PC12 cells resulted in increased expression of both the iron response element-positive and the iron response element-negative forms of DMT1. Taken together, these results demonstrate that iron status is likely to have a direct effect on the uptake and biological actions of Mn and probably other divalent metals that are transported by DMT1.

Analysis of oxysterol binding protein homologue Kes1p function in regulation of Sec14p-dependent protein transport from the yeast Golgi complex.

Oxysterol binding proteins (OSBPs) comprise a large conserved family of proteins in eukaryotes. Their ubiquity notwithstanding, the functional activities of these proteins remain unknown. Kes1p, one of seven members of the yeast OSBP family, negatively regulates Golgi complex secretory functions that are dependent on the action of the major yeast phosphatidylinositol/phosphatidylcholine Sec14p. We now demonstrate that Kes1p is a peripheral membrane protein of the yeast Golgi complex, that localization to the Golgi complex is required for Kes1p function in vivo, and that targeting of Kes1p to the Golgi complex requires binding to a phosphoinositide pool generated via the action of the Pik1p, but not the Stt4p, PtdIns 4-kinase. Localization of Kes1p to yeast Golgi region also requires function of a conserved motif found in all members of the OSBP family. Finally, we present evidence to suggest that Kes1p may regulate adenosine diphosphate-ribosylation factor (ARF) function in yeast, and that it may be through altered regulation of ARF that Kes1p interfaces with Sec14p in controlling Golgi region secretory function.

A real-time fluorogenic phospholipase A(2) assay for biochemical and cellular activity measurements.

A fluorogenic analog of the PLA(2) substrate PC, named Dabcyl-BODIPY-PC, or simply DBPC, was synthesized with a fluorescence quencher (Dabcyl, 4-[(4-[N,N-dimethylamino]phenyl)azo]benzoic acid) in the sn-1 acyl chain and a BODIPY fluor in the sn-2 acyl chain. DBPC was recognized by sPLA(2) from each of the four sources examined (bee venom, human synovial fluid, cobra venom, and bovine pancreas). A dramatic and quantifiable fluorescence enhancement of DBPC occurred upon phospholipase digestion both in the presence and absence of excess PC. Both real-time and endpoint assays for PLA(2) were sensitive, consistent, and rapid. Thus, DBPC can be used as a sensitive fluorogenic probe for in vitro high-throughput screening assays for PLA(2) activation and inhibition and would expedite studies of PLA(2) in cellular signaling, in vitro screening for drug discovery, and subcellular localization of enzyme activity.
