An immunostimulatory oligodeoxynucleotide containing a cytidine-guanosine motif protects senescence-accelerated mice from lethal influenza virus by augmenting the T helper type 1 response.

The SAM-P1 strain of senescence-accelerated model mice shows an impaired T helper type 1 (Th1) immune response upon infection with influenza virus, which results in high susceptibility to the virus. Treatment of spleen cells from SAM-P1 mice with an immunostimulatory oligodeoxynucleotide containing a cytidine-guanosine motif (CpG ODN) in vitro increased the ratio of the titre of IFN-gamma to that of IL-4. Administration of CpG ODN to SAM-P1 mice generated satisfactory virus-specific cytotoxic T-lymphocyte responses and natural killer cell activation and the virus-specific immunoglobulin (Ig) isotype switched from IgG1 to IgG2a. Virus growth in the lungs of CpG ODN-treated SAM-P1 mice was cleared quickly and mice survived the lethal influenza virus infection. It could be inferred that a possible mechanism of CpG ODN for normalization of senescence-associated dysregulation of the Th1/Th2 balance involves the upregulated expression of CD154 and CD40 molecules on immune-competent cells. These results suggest that CpG ODN could contribute to the development of a protective strategy against infectious diseases, especially among immunocompromised elderly persons, by stimulating Th1 immune responses.

[Low intensity anticoagulation therapy after mechanical heart valve replacement].

OBJECTIVE: To investigate the proper anticoagulation intensity in patients after mechanical heart valve replacement in china. METHODS: The anticoagulation therapy intensity and the complications in 480 patients after mechanical heart valve replacement were studied. RESULTS: The follow-up rate was 89.17%, the total patient-years (Pty) was 2,110.04 years, the mean oral import warfarin dosage was (2.81 +/- 0.95) mg/day, and native warfarin dosage (2.38 +/- 0.46) mg/day. The mean PTR value of 2 116 samples was 1.43 +/- 0.26, and the INR value of 1 195 samples was 1.63 +/- 0.49. The total hemorrhage rate was 4.60% Pty, and the hemorrhage death rate was 0.38% Pty. The PTR and INR values were higher in the hemorrhage group than in the no-hemorrhage group. (t = 1.816, P < 0.05; t = 2.407, P < 0.01). The thromboembolism rate was 0.66% Pty, and the thromboembolism death rate was 0.05% Pty. There were 15 pregnancies in 14 women patients and no malformed newborns were found. CONCLUSIONS: The most important complication of anticoagulation therapy after mechanical heart valve replacement is hemorrhage in china; The proper anticoagulation intensities of this group are INR 1.5-2.0 and PTR 1.3-1.5. It is beneficial to adopt the low intensity anticoagulation therapy for decreasing the death rate from hemorrhage, protecting pregnant women and newborns from hemorrhage and malformation, and improving the life qualities of the patients.

Total synthesis of desferrisalmycin B.

The first total synthesis of a naturally occurring siderophore antibiotic, desferrisalmycin B, is described, and the configuration of the unknown stereocenter is assigned. The synthesis features a synthetic strategy of constructing the novel amino-heptopyranoside component by stereoselective dihydroxylation followed by a Bose-modified Mitsunobu reaction. Through this convergent approach, other members of salmycins should also be synthetically accessible.

Hydroxyfullerene as a novel coating for solid-phase microextraction fiber with sol-gel technology.

Hydroxyfullerene (fullerol) as a novel coating for solid-phase microextraction (SPME) fiber was first prepared by a sol-gel technology. The coating procedure involving sol solution composition and conditioning process was presented. A fullerene polysiloxane surface-bonded porous coating on the fused-silica fiber surface was obtained and confirmed by IR spectra and scanning electron microscopy. The coating has stable performance at high temperature (even to 360 degrees C) and solvents (organic and inorganic) because of the properties of fullerene and the chemical binding between the coating and the fiber surface. The extraction properties of the new coatings to less volatile organic compounds, such as polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons and polar aromatic amines were investigated using headspace SPME coupled with GC-electron-capture detection and GC-flame ionization detection. In addition, compared with commercial SPME stationary phases, the new coatings showed higher sensitivity, faster velocities of mass transfer for aromatic compound, and possessed planarity molecular recognition for PCBs. Moreover, this fiber was firm, inexpensive, durable and can be prepared simply. The fiber-to-fiber reproducibility was very good.

Total synthesis of exochelin MN and analogues.

The first total synthesis of exochelin MN is described along with rationally designed analogues. The required L-threo-beta-hydroxyamino acid components were constructed using either Sharpless asymmetric aminohydroxylation reactions or an aldol reaction of imidazolidinone 19. A new concise procedure for the preparation of the constituent six-membered cyclic hydroxamate was developed. In addition, a plausible mechanism for exochelin MN-mediated iron(III) transport was proposed. Biological studies of these compounds will be used to evaluate this hypothesis.

In vitro effects of STI 571-containing drug combinations on the growth of Philadelphia-positive chronic myelogenous leukemia cells.

BACKGROUND: Chronic myelogenous leukemia (CML) is characterized by a molecular aberration, a fusion BCR-ABL gene encoding for aberrant tyrosine kinase activity, which is crucial in the pathogenesis of CML. In vitro, inhibition of BCR-ABL protein tyrosine kinase activity by a tyrosine kinase inhibitor, Imatinib mesylate (STI571; formerly CGP57148B), successfully suppressed proliferation/survival of the BCR-ABL positive clones. In clinical studies, hematologic and cytogenetic remissions have been achieved in most patients with chronic phase CML; in accelerated and blastic phases of CML, STI571 appeared less effective. In the current study, the authors tested combinations of STI571 and cytarabine and homoharringtonine (HHT), drugs with documented activity in CML. METHODS: The single agents and their combinations were studied for in vitro effect on proliferation of BCR-ABL positive cell lines KBM5 and KBM7 by 3(4,5-dimethylthiazol-2yl)-2,5 diphenyl-tetrazolium bromide assay and on primary patient-derived BCR-ABL cells by clonogenic assays. The in vitro additive, synergistic, or antagonistic effects of cytarabine and HHT with STI571 were then investigated by computer-assisted analysis using the CalcuSyn software. RESULTS: STI571 consistently suppressed BCR-ABL positive cell proliferation with a dose-effect correlation. In the model system used, STI571/cytarabine and STI571/HHT combinations were more effective in inhibiting KBM5 and KBM7 cell growth than each drug as single agent. These results were also verified in primary CML-derived clonogenic cells in semisolid cultures. CONCLUSIONS: In this experimental system, our studies documented additive or synergistic effects with STI571 plus cytarabine or HHT, supporting the future use of STI571 combinations in clinical trials in patients with Philadelphia chromosome-positive leukemias.

Proteomic analysis of differential protein expression in human nasopharyngeal carcinoma cells induced by NAG7 transfection.

Nasopharyngeal carcinoma (NPC) is a commonly occurring tumor in southern China and south east Asia. A genetic factor has now been recognized to be associated with this cancer. A new gene, named NAG7, was cloned from the common minimal deletion region in 3p25.3-26.3. In order to investigate the function of NAG7 gene, proteomic methods were used to find and identify the differential proteins and expected to elucidate the mechanism of NAG7. The NAG7 eukaryotic expression vector was constructed and transfected into NPC cell line HNE1 with liposome. Twenty-two differential protein spots in transfected cells were found significant and reproducible using high-resolution two-dimensional electrophoresis. Nine proteins that were up-regulated and seven proteins that were down-regulated were identified by matrix assisted laser desorption/ionization time of flight mass spectrometry and database analysis. These proteins included growth arrest specific protein, DNA binding protein, caspase 6, pinch protein and ras-related protein rab-36, which are involved in cell cycling, transcription regulation, signaling pathways and apoptosis. NAG7 may exert its functions by mediating differential expression of these proteins.

Effect of magnesium lithospermate B on calcium and nitric oxide in endothelial cells upon hypoxia/reoxygenation.

AIM: To investigate the effect of magnesium lithospermate B (MLB) on hypoxia/ reoxygenation (H/R)-induced elevation of intracellular calcium concentration ([Ca2+]i) and nitric oxide (NO) release in endothelial cells. METHODS: The cultured human umbilical vein endothelial cells (ECV304) were exposed to hypoxia for 30 min under 95 % N2 and 5 % CO2, then reoxygenation for 30 min under air and 5 % CO2. Cell injury was evaluated by dye exclusion test, superoxide dismutase (SOD) assay, and molondialdehyde (MDA) assay. [Ca2+]i was determined by Fura 2-AM. NO content was examined by a NO assay kit. Endothelial nitric oxide synthase (eNOS) and inducible nitric oxide synthase (iNOS) mRNA expressions were measured by semi-quantitative RT-PCR. RESULTS: Cell viability was decreased from (93.1+/-1.2) % in normoxia to (88+/-3) % in H/R (P < 0.01), and SOD activity was also decreased from (0.24+/-0.07) kNU/L to (0.18+/-0.03) kNU/L in H/R (P >0.05), but MDA production was increased from (1.12+/-0.06) mmol/L in normoxia to (3.78+/-0.03) mmol/L in H/R (P < 0.01) in ECV304 cultured under calcium conditions. MLB 2.5, 5, and 10 mg/L increased cell viability and SOD activity, and inhibited MDA formation in ECV304. H/R increased [Ca2+]i (F340/F380 from 1.65+/-0.16 to 1.89+/-0.28), NO release [from (7.5+/-1.3) micromol/L to (16+/-5) micromol/L], and eNOS mRNA expression, but decreased iNOS mRNA expression in ECV304 (P <0.05). However, it did not affect them under calcium-free conditions. MLB inhibited H/R-induced increases in [Ca2+]i and eNOS mRNA expression, stimulated NO release and iNOS mRNA expression (P <0.05). CONCLUSION: MLB attenuates H/R-induced cell injury and increases NO release in ECV304. This increase of NO production is possibly associated with preventing cell injury induced by H/R in MLB-treated ECV304.

[Effect of shengji huayu recipe and its disassembled formulae on type I and III collagen synthesis in wound healing fibroblasts].

OBJECTIVE: To explore the mechanism of Shengji Huayu Recipe (SJHYR) and its 3 disassembled formulae in accelerating wound healing toward skin restoration. METHODS: Using in vitro cultured fibroblasts from granulation tissue of wound and compared with the normal skin fibroblast of suckling rat, who were treated separately by drug serum containing high and low dose SJHYR and its disassembled prescriptions (Shengji formula and Huayu formula). The type I and III collagen contents in the fibroblasts were determined with immunocytochemical ABC method. RESULTS: Shengji formula could increase the levels of type I and III collagens in fibroblasts, which was significantly higher than those in model cells and control (P < 0.01). Huayu formula lowered them to the levels below those in model cells (P < 0.01), while in comparing the levels in the cells treated by high or low dose SJHYR with those in control, no significant difference was shown(P > 0.05). CONCLUSION: SJHYR might accelerate wound healing toward skin restoration through regulating the ratio of collagen type I and III, to adjust their metabolism.

[Injury by electrical forces: pathophysiology, biophysics and pathogenesis].

As the technologic sophistication of generation and distribution of electrical energy has grown, so has the general concern about the effects of electric fields on human health. There can be no doubt that the significance of electrical trauma will continue to grow with our increasing use of power. It is apparent that our understanding of the various forms of electric trauma must increase, while we continue to promote safety near electrical hazards and develop effective medical therapies. Tissue damage as a result of electrical injury occurs by two mechanisms which are summative in action and have a variable degree of contribute to the ultimate damage produced. Thermal tissue damage occurs as a result of heat generated within the tissue (which offer an electrical resistance) secondary to the passage of the electrical current. High temperatures can also lead to cell membrane components, e.g., phospholipids, to dissolve. Electroportation damage is the tissue damage induced secondary to the strong electric field. Transmembrane potentials caused by electrical current result in the formation of pore in the phospholipid component of the cell membrane resulting in loss of function of the cell membrane with consequent cell death.

Peroral vaccination with a temperature-sensitive mutant of parainfluenza virus type 1 protects mice against respiratory challenge infection.

Peroral vaccination for preventing respiratory infectious diseases was investigated in a murine model using a temperature-sensitive (ts) mutant of parainfluenza virus type 1. The ts mutant virus administered perorally in drinking water neither multiplied nor caused lesions in the respiratory tract or the central nervous system. However, ts virus antigen-positive cells appeared in oropharyngeal lymphoid tissues. This type of antigenic stimulation was capable of inducing both humoral and cellular immune responses, together with an augmentation of interferon production and natural killer cell activity, making it possible to protect the mice against challenge infection with a virulent wild-type virus. These results suggest that the oral cavity, a constituent member of the common mucosal immune system, is a candidate organ applicable as a vaccine route against virus respiratory diseases.
