A new putative fimbrial colonization factor, CS19, of human enterotoxigenic Escherichia coli.

A gene probe derived from the colonization factor antigen I (CFA/I) operon cross-hybridized at very low stringency to plasmid DNA from coli surface antigen 17 (CS17)-producing enterotoxigenic Escherichia coli (ETEC) and from the ETEC strain F595C, which was negative for previously described CFAs, CSs, and putative colonization factors (PCFs). A 16-kDa protein was identified in sodium dodecyl sulfate-polyacrylamide gel electrophoresis of heat extracts prepared after growth of strain F595C at 37 degrees C on CFA agar containing bile salts. Transmission electron microscopy revealed bile salt- and temperature-dependent expression of fimbriae with a diameter of 7 nm. After transformation with a recombinant plasmid harboring the cfaR gene, which encodes a positive regulator of several CFAs, PCFs, and CSs, the 16-kDa protein was hyperexpressed. Polyclonal antibodies raised against this protein bound to the fimbriae and inhibited the adhesion of F595C bacteria to tissue-cultured Caco-2 cells. Nucleotide sequence determination of the gene encoding the 16-kDa fimbrial subunit revealed a high degree of amino acid sequence homology to the CFA/I, CS1, CS2, CS4, CS14, and CS17 polypeptides. The term CS19 is proposed for the new fimbria.

Agglutination Tests

Genotyping by PCR, of Staphylococcus aureus strains, isolated from mammary glands of cows.

A total of 71 Staphylococcus aureus strains isolated from bovine mammary glands were identified and subtyped. The methods used to differentiate between the S. aureus isolates were the DNA polymorphism pattern after amplification with a Polymerase Chain Reaction using several primer combinations and phage typing. The DNA fingerprinting technique using RAPD, ERIC1R and ERIC primers proved to be useful in differentiating isolates of S. aureus. Differentiation of isolates using phage typing gave no additional information compared to the DNA technique. The outbreak of S. aureus in the herd studied was mainly caused by one S. aureus strain. Other strains were only found on three occasions, twice in subclinical infections and once from a case of clinical mastitis. In the latter case the dominant strain was isolated from a different quarter of the same cow. Four of the ten cows studied suffered from clinical mastitis. From those four cows, three remained infected with the same S. aureus strain despite antibiotic treatment.

Isolation and characterisation of dog uropathogenic Proteus mirabilis strains.

Proteus mirabilis strains isolated from the urine of dogs with urinary tract infections, were characterised with respect to the production of haemolysin and fimbriae. In contrast to healthy dogs, P. mirabilis was also isolated in high numbers from the faeces of dogs suffering from recurrent urinary tract infections. Production of fimbriae was demonstrated by electron microscopy and the presence of genes for two different types of major fimbrial subunits (MR/P-like or UCA-like) was demonstrated by Southern hybridisation. These genes were absent in the Proteus vulgaris, Providentia rettgeri and Morganella morganii strains tested. All but one P. mirabilis strains were haemolytic and most strains produced fimbriae albeit in different amounts. The UCA fimbrial subunits from dog and human isolates have identical molecular weights and N-terminal sequences and are immunologically cross reactive. It was concluded that dog uropathogenic P. mirabilis strains are very similar to human uropathogenic P. mirabilis strains.

Amino Acid Sequence

Phylogenetic position of Taylorella equigenitalis determined by analysis of amplified 16S ribosomal DNA sequences.

The 16S ribosomal DNA sequence of Taylorella equigenitalis (formerly Haemophilus equigenitalis), the causative organism of contagious equine metritis, was determined. A phylogenetic analysis of this sequence revealed a phylogenetic position of T. equigenitalis in the beta subclass of the class Proteobacteria apart from the position of Haemophilus influenzae, which belongs to the gamma subclass of Proteobacteria. A close phylogenetic relationship among T. equigenitalis, Alcaligenes xylosoxidans, and Bordetella bronchiseptica was detected; Spirillum volutans and Chromobacterium fluviatile (Iodobacter fluviatile) were in the same group but slightly removed. This relationship is surprising in view of the considerable differences in the G + C contents of the genomes of these bacteria.

The transmandibular implant: a 12-year follow-up study.

A clinical study of the use of the transmandibular implant for reconstructing severely atrophic mandibles in preparation for an implant-borne dental prosthesis was performed. This study included 368 patients with a mean height of the mandible of 10 mm. They were examined clinically, radiographically, and by means of a questionnaire. The follow-up period for 185 patients was 5 years or more, and 55 were followed 10 years or more. Removable dentures were made for 350 patients, and fixed bridges for 18 patients. Forty-three patients had reversible complications, one patient had unilateral hypesthesia, and six patients had their implant removed. Reversible complications of infection, gingival hyperplasia, loading of individual posts, and fenestration of threads were controlled without deleterious effects on the gingival, mucosal, or osseous tissues. The transmandibular implant system was shown to successfully restore function of the severely atrophic mandible in a more predictable manner than augmentation procedures.

Adult

Visor osteotomy and vestibuloplasty--a one-stage procedure. A preliminary report.

The visor osteotomy as described by HARLE gives a considerable increase of the absolute height of the atrophic mandible, between the mental foramina. After 6 weeks a total vestibuloplasty with mucosal grafting (and a lowering of the floor of the mouth) is performed. A method is described in which the visor osteotomy and the vestibuloplasty are performed in one stage.

Treatment of mandibular fractures by means of stable internal wire fixation.

Treatment according to the principles of stable internal fixation offers great advantages for fractures of the mandibular body. Most authors believe that a stable internal fixation can be achieved only by means of bone plating or screw fixation. At Groningen a wire connection was designed by BRONS which could be considered to be stable. The application of this stable internal wire fixation and the results of treatment in 272 fractures of the mandibular body between 1967 and 1976 are discussed.

Operative treatment of arthrosis deformans of the temporomandibular joint: a preliminary report.

If patients with arthrosis deformans of the temporomandibular joint are treated without success by conservative means, operative therapy may be indicated. The type of operation depends on the complaints: a)resection of the auriculotemporal nerve and superficial temporal artery; or b) the same operation in combination with condylar shaving. Twenty-four joints in 14 patients underwent a denervation. Thirty joints in 19 patients were also shaved. The results were very satisfactory.
