Approaches to define the role of SF-1 at different levels of the hypothalamic-pituitary-steroidogenic organ axis.

Targeted gene disruption has produced knockout mice globally deficient in the orphan nuclear receptor steroidogenic factor 1 (SF-1). These SF-1 knockout mice lacked adrenal glands and gonads and consequently exhibited adrenocortical insufficiency and sex reversal of their internal and external genitalia. They also had impaired expression of gonadtropins in the anterior pituitary gonadotropes and agenesis of the ventromedial hypothalamic nucleus (VMH), confirming roles of SF-1 at all three levels of the hypothalamic-pituitary-steroidogenic organ axis. Ongoing experiments are directed at using evolving techniques for tissue-specific gene inactivation to define the roles of SF-1 within discrete sites of the hypthalamic-pituitary-steroidogenic organ axis.

[Study of expression of hepatitis C virus antigens and viral replication in extrahepatic tissues].

OBJECTIVE: To explore the state of hepatitis C virus (HCV) infection and replication in extrahepatic tissues and its significance in hepatitis C patients. METHODS: Immunohistochemical assay and reverse transcriptase polymerase chain reaction (RT-PCR) were used respectively to detect HCV antigens (NS3, NS5 and CP10) and HCV RNA positive-and negative-strand in kidney, heart, pancreas, and intestine from 9 hepatitis C patients. RESULTS: Positive staining for HCV NS3, NS5 and CP10 in extrahepatic tissues was found in 6 of the 9 (66.7%) patients. The expression of HCV antigens was observed only in plasma of cells. The amount of positive-staining cells was less and there were a few differences between the kind, amounts and distribution of HCV antigen-positive-staining cells in different extrahepatic tissues. HCV RNA was detected in extrahepatic tissues from all the patients by RT-PCR. The detection rates of HCV RNA positive-and negative-strand were 64.5% and 35.5% in 31 extrahepatic tissues specimens, respectively. The positive of HCV RNA positive-and negative-strand detection was coincident with that of HCV antigens detection in these tissues (Kappa>0.40). CONCLUSIONS: These results suggest a possible presence of HCV infection and replication in extrahepatic tissues. Low-level of HCV extrahepatic infection and replication is possibly a major cause of a low positive rate and inconsistent results of HCV detection in extrahepatic tissues.

Two hundred and seventeen cases of winter diseases treated with acupoint stimulation in summer.

217 cases of chronic bronchitis and asthma were clinically treated and analyzed for the effects of combining electric stimulation with topical application of drug on acupoints. The results suggested that the combined therapy was superior to unitary therapy (P < 0.05). It is indicated that the combined therapy has a good curative effect in both short- and long-terms.

Clinical observation on treatment of non-parvicellular carcinoma of the lung with jin fu kang oral liquid.

Jin Fu Kang Oral Liquid ([symbol: see text]), made of traditional Chinese drugs for supplementing qi and nourishing yin, was developed according to the common symptoms in lung carcinoma with deficiency of both qi and yin. Of the 96 cases in the Jin Fu Kang group, 1 case got complete remission (CR) after treatment, 8 cases partial remission (PR), 52 cases no change (NC), PR + NC covering 63.5%. Of the 52 cases in the group of Jin Fu Kang plus chemotherapy, 11 cases got PR after treatment, 26 cases NC, PR + NC covering 71.2%. Of the 25 cases in the chemotherapy group, 4 cases got PR after treatment, 11 cases NC, PR + NC covering 60.0%. The results show that the therapeutic effectiveness in the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy was better than that in the chemotherapy group. The one-year survival rate and the two-year survival rate after treatment in the Jin Fu Kang group were 67.3% and 67.3% respectively; 66.7% and 66.7% in the group of Jin Fu Kang plus chemotherapy; and 40.3% and 0.0% in the chemotherapy group. The improvement of clinical symptoms, increase of body weight and improvement of health situation (KPS marks) after treatment in both the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy were better than that in the chemotherapy group. Some indicators of immunology and hemogram after treatment were greatly improved in the Jin Fu Kang group, worse in the chemotherapy group, but no obvious improvement in the group of Jin Fu Kang plus chemotherapy.

[Sequence analysis of phosphoprotein genes of respiratory syncytial virus field strains isolated in China].

OBJECTIVE: To get information about phosphoprotein (P) genes of RSV field strains isolated in China. METHODS: The complete nucleotide sequences of the P protein genes from six respiratory syncytial virus strains were determined with cDNA clones which had been amplified by RT-PCR and cloned into PTZ18R plasmid vector and compared with those of previously published counterparts of A2 and CH18537 strains. RESULTS: The data indicated that there had high identities (>95%) at nucleotide and amino acid levels among the strains within the same subgroup; there were more divergence among strains (<85%) from different subgroups at nucleotide and amino acid levels of P genes, especially in the 3'-noncoding regions and the divergent domains. The analysis of hydrophobicity indicated that there had difference from 50 to 100 amino acid of P proteins between A and B subtypes. CONCLUSIONS: The analysis of the P genes confirm that there existed variability among RSV field strains and strains A2 and CH18537. The data described above revealed the characteristics of the P genes of RSV field strains isolated in China.

[EEG synchronization index spectrum: definition, calculation and the changes related to visual selective response].

OBJECTIVE: EEG synchronization is usually estimated in terms of coherence amplitude, but the estimation is with apparent uncertainty because of the poor resolution both in time and frequency. The purpose of this work is to surmount this problem. METHOD: A new definition of EEG synchronization index spectrum [SynI(f)] was developed basing on the distribution characteristics of EEG coherence phase which was calculated by a tested algorithm with 1 s and 1 Hz resolution. The basic feature and its changes during visual selective responses were calculated in 25 normal subjects. EEG signals were recorded from 9 locations in two conditions: looking at the central LED only(VC) and making switch response to the target LED flash signals (T) differentially (DR): switch to left or right for Ts from left (LVF) or right (RVF) visual field, repectively but making no response to the non-target ones (NT). RESULT: (1) The frequency dependency of SynI varied with brain location, e.g., SynI was higher in alpha range than others at frontal locations but the situation reversed at posterior locations. (2) The SynIs between midline and right brain locations were higher than that between mid line and left ones. (3) As compared with VC, SynI was increased in DR condition at central and posterior brain locations but not frontal ones, it happened mainly in 7-23 Hz at central locations but in 1-4 Hz at posterior ones. (4) The augmentation of SynI in DR was greater for T than for NT signals. CONCLUSION: The results indicated that the SynI(f) was meaningful for studying the frequency-spatial feature of EEG synchronization change among brain locations related to cognitive activities.

Unsymmetrical response features of left and right brain to signals from left and right visual fields at different cognitive levels.

OBJECTIVE: To study the response characteristics of left and right brain to signals from left (LVF) and right (RVF) visual fields during cognitive activity. METHOD: ERPs at 9 locations to LVF and RVF signals were compared in 23 normal subjects in 3 task conditions: looking forward only (VC); making switch response to target signals (T) only (SR); making switch response to T differentially (DR). RESULT: (1) Significant difference in ERPs was found between that induced by LVF and RVF signals on left brain especially at frontal location (F5), which appeared as slow negative deflection induced by T and NT from RVF in SR and DR; (2) Condition-dependent unsymmetrical features between left and right brain were found: the slow potential at F5 was significantly more negative in T and contralateral spatial relation (i. e., left brain for RVF) but more positive in NT and ipsilateral relation than that at F6. CONCLUSION: Signals with psychological meaning in RVF might cause more psychological load as suggested by the results obtained in this study.

[Study on respiratory impedance of patients with extratracheal tumour].

OBJECTIVE: To determine the manifestation of impulse oscillometry (IOS) in variation of respiration impedance on patients with extratracheal tumour. METHODS: The IOS indices in 30 normal persons and 28 patients with extracheal tumour were measured. The ratio of forced expiratory volume after 1 second to forced vital capacity (FEV1/FVC) was evaluated and the maximal expiratory flow-volume curve (MEFV curve) was examined. The above values with t-test of SPSS statistics software were analyzed. RESULTS: Compared with the normal group, in the extratracheal group the ratio of actual value of resistance to the predict from 20 Hz to 35 Hz increased obviously and this only occurred with the reactance at 20 Hz (P < 0.01). The resistance curve showed clear rise after the 20 Hz and the reactance curve peaked at 20 Hz and was leveled to normal at 35 Hz. CONCLUSION: IOS is capable of presenting the distinctive indices for the study of respiration impedance on patients with extratracheal tumour.

[Influence of multiple coats of bonding agent on the bond strength of amalgam bonding].

OBJECTIVE: To evaluate the effect of multiple coats of bond on the bond strength of amalgam bonding. METHODS: One or two layers of Scotch-bond multi-purpose plus dental adhesive agent were applied on the dental walls of two groups of teeth respectively prior to amalgam condensation. The third group was set for control which only had amalgam filling. After one week, thermocycling, shear bond strength were tested for all specimens with 1195-Instron testing machine. Data were analyzed by SPSS 8.0. The SEM observation were made in other five modals with one or two layers bonding. RESULTS: The shear strengths of two or one layer bond and control one were 25.77, 20.69 and 13.05 MPa separately. The difference among them was statistical significant (P < 0.01). CONCLUSIONS: The microlocking was more obvious in specimens of two bonding layers.

[NOS activity in vascular smooth muscle cells of hypertension patients and the effect of AVP on NO synthesis].

OBJECTIVE: To investigate the changes of nitric oxide (NO) and nitric oxide synthase (NOS) in vascular smooth muscle cells (VSMCs) of the essential hypertension (EH) patients, determine the effect of arginine vasopressin (AVP) on NO synthesis, and evaluate their significance in the pathogenesis of hypertension. METHODS: VSMCs of EH and normotensives (NT) were isolated by compound collagenase method and cultured in vitro. We detected the NO concentration and the activity of NOS in cell culture medium and determined the effects of AVP on NO synthesis and NOS activity. RESULTS: NO concentration and the activity of NOS in VSMCs of EH were 0.84 nmol/2.5 x 10(6) cells +/- 0.08 nmol/2.5 x 10(6) cells and (0.23 nmol/min +/- 0.08 nmol/min per 2.5 x 10(6) cells respectively, which were significantly lower than in VSMCs of NT (2.48 nmol/2.5 x 10(6) cells +/- 0.19 nmol/2.5 x 10(6) cells, 0.37 nmol/min +/- 0.06 nmol/min per 2.5 x 10(6) cells, respectively, both P < 0.01). Under AVP induced conditions, NO concentration 3.89 +/- 0.33 nmol/2.5 x 10(6) cells and NOS activity 1.81 nmol/min 2.5 x 10(6) cells +/- 0.09 nmol/min 2.5 x 10(6) cells) in VSMCs of EH were significantly lower than in VSMCs of NT (9.09 nmol/2.5 x 10(6) +/- 0.75 nmol/2.5 x 10(6) cells), 2.80 nmol/min 2.5 x 10(6) cells +/-0.21 nmol/min 2.5 x 10(6) cells, respectively, both P < 0.01). NO concentration and NOS activity in VSMCs of both EH and NT group increased in a culture-time-dependent and AVP-concentration-dependent manner (both P < 0.01). NO concentrations in VSMCs of EH in different culture time increased with the enhancement of NOS activity, and there was significant positive relevance between them (4 h r = 0.940, P < 0.01; 8 h r = 0.993, P < 0.01. CONCLUSION: NOS-NO system of EH, with attenuated responsiveness to AVP, is probably abnormal in structure or function, and is involved in the pathogenesis of essential hypertension.

[Effect of combined operation including splenorenal shunt as the main technique for portal hypertension in children].

OBJECTIVE: To assess the effect of combined operation including splenorenal shunt as the main technique for portal hypertension in children. METHODS: 22 patients were reviewed and followed up from 1980 to 1998 (13 male, 9 female). The average age was 9.45 years. Intrahepatic type was shown in 16 patients and prehepatic type in 6. All patients had hematemesis, hematochezia or severe esophageal varices. Liver function was classified Child A in 14 patients, B in 7, and C in 1. Combined operation was performed in all patients. RESULTS: There was no operative death in this group. 21 patients (95.5%) were followed up, with the longest time for 19 years (average 9.58 years). Six patients had hemorrhage from the esophageal varices after operation, and two died of hemorrhage. The survival rates of < 3, -5, -10, -15, > 15 years were 95.2%, 100%, 93.8%, 100%, 100% respectively. No hepatic encephalopathy was noted. CONCLUSION: In treating and preventing hemorrhage from the esophageal varices, combined operation including splenorenal shunt as the main technique is the first choice for the portal hypertension in children. Attention must be paid to the patients who had hemorrhage from the esophageal varices after operation.

[Effects of oral calcium supplementation on blood pressure in population].

OBJECTIVE: To assess the effects of oral intake of calcium lactate tablet on blood pressure and serum levels of sodium, potassium, calcium and phosphors. METHODS: In a rural area with natural population with higher average blood pressure and prevalence of hypertension, and with high salt and low calcium intake in diet in Yu County, Shanxi Province, 112 volunteers aged 30 to 64 years were selected based on a mass screening for hypertension, and randomized into trial and control groups with double blinding, taking calcium lactate tablets (800 mg of calcium daily) and placebo respectively for five weeks. Ninety-eight of them complied with the medical order completely, 39 males and 59 females, and were followed-up for systolic (SBP) and diastolic blood pressure (DBP), and their fasting venous blood specimen and 8-hour nocturnal urine specimens were collected. Twenty-four-hour diet recall was obtained for three consecutive days from all of them. RESULTS: Reduction of 4.7 mm Hg (0.62 kPa) and 2.7 mm Hg (0.36 kPa) in systolic and diastolic blood pressure, respectively in the trial group was observed, as compared with that in control one, with P-values of 0.027 and 0.074, respectively, after 35 days of treatment. The higher their baseline BP level, the greater reduction of their BP after treatment. Urinary excretion of calcium increased and serum level of phosphorus reduced after oral administration of calcium. CONCLUSION: In an area with higher prevalence of hypertension and with high salt and low calcium intake in diet, change in diet composition or supplementation with calcium could reduce their average blood pressure and prevent them from hypertension for those with mild and moderate hypertension and in high risk with higher blood pressure.

[The effects of zinc supplementation on postburn nutritional status of zinc-deficient scalded rats].

OBJECTIVE: To investigate the effects of zinc supplementation on postburn nutritional status of zinc-deficient scalded rats. METHODS: Weanling Wistar rats were fed with zinc-deficient diets for one week to induce zinc-deficient status. The rats were inflicted with 20% TBSA deep partial thickness burn and thereafter fed with diets of low, normal and high zinc contents. The scalded rats fed with normal zinc-containing diet before and after injury were taken as normal control group. The changes of body weight, cumulative nitrogen balance and the contents of DNA and protein in jejunum mucous, liver, kidneys, spleen and testis were employed to represent the nutritional status of the rats. RESULTS: The rats fed with diets with normal and high contents of zinc were superior to those with low content of zinc in terms of body weight, food conversion rate, cumulative nitrogen balance and organ protein contents. The serum zinc level, alkaline phosphatase (ALP) activity and the DNA contents in jejunum mucous, kidneys, spleen and testis increased significantly after zinc supplementation. CONCLUSION: Zinc supplementation was helpful to improve postburn nutritional status in scalded rats. And relative higher amount of zinc supplementation might be beneficial to raise the DNA content in testis and kidneys when compared to that with normal zinc supplement.

[The clinical and experimental study on postburn metabolic characteristics of zinc and its influence on copper, iron and calcium].

OBJECTIVE: To observe postburn metabolic characteristics of zinc and its influence on copper, iron, calcium. METHODS: (1) To analyze the levels of Zn, Cu, Fe, Ca in the serum and urine of burn patient on different postburn days. (2) To replicate a rat model of zinc deficiency and to observe the symptoms and the effects of zinc supplementation in different dosage after scalding in rats with zinc deficiency. (3) To assay the levels of Zn, Cu, Fe, Ca in rat wound tissues following supplementation of zinc in diet and to wounds in rats after scald. RESULTS: (1) The levels of serum Zn and Cu decreased markedly on 1 similar 3 days in major burn patients, whereas the urine Zn and Cu increased obviously, reaching the peak value on the 14th day. The serum and urine Fe increased rapidly on PBD2 and then descended steeply. The serum and urine Ca kept lower levels all along. (2) In zinc deficient rats growth retardation appeared first and other symptoms emerged subsequently. Serum Zn decreased and serum Cu, Fe, Ca increased. (3) The levels of Zn, Cu, Fe, Ca in wound tissue of scalded rats showed a continuous postburn increase. The Ca contents in wound tissue increased about 50 -- fold. Zn content in wound was more obviously increased than Cu, Fe and Ca when Zn was supplemented through wound compared with that with diet supplementation. CONCLUSION: Serum Zn level was lowered after scald and it could be elevated after supplementing zinc by diet or through wound, Changes in serum Cu, Fe, Ca occurred corresponding to the change in serum Zn.

[Longitudinal biomechanical effect on repair options of artery injury].

OBJECTIVE: To investigate the relationship between the different defect length of vessels and the options of vascular repair, and to compare the different options of repair because of the longitudinal biomechanical effect. METHODS: A clinical analysis was undertaken to evaluate the major arterial and venous injuries in human extremities repaired by end-to-end anastomoses or venous autograft(177 cases, 185 vessels). Compared the defect length of the same kind of vessels repaired by different options (Student-t test). Evaluated the defect length to repair arterial injuries between by end-to-end anastomoses and by vein graft by means of 95% confidence interval. RESULTS: There was significant difference between the defect length of brachial artery repaired by end-to-end anastomosis and femoral artery and popliteal artery repaired by autogenous vein graft (P < 0.01). The upper limit of confidence interval in the defect length of brachial artery, femoral artery and popliteal artery was 3.17 cm, 2.81 cm and 2.44 cm respectively by end-to-end anastomosis by means of 95% confidence interval. The lower limit of confidence interval in the defect length of brachial artery, femoral artery and popliteal artery was 2.82 cm, 2.41 cm and 2.17 cm respectively by vein graft by means of 95% confidence interval. The defect length of brachial artery, femoral artery and popliteal artery repaired by vein graft was linear correlation with the length of graft. CONCLUSION: Because of the longitudinal biomechanical difference of arteries and veins in human extremities, different options of repair are necessary to different arterial injuries.

[Systemic aging and replicative senescence: in vivo and in vitro].

Cultured cells lose the ability of DNA synthesis, mitosis, and finally the ability of cell proliferation after they have undergone a finite number of population doublings in vitro, though the cells still maintain the basic metabolic process. This is termed replicative senescence. We review the prevalence of replicative senescence, summarize the features of senescent cells, and then focus on the links between systemic aging and replicative senescence. The present knowledge, albeit still incomplete, proposes that replicative senescence is a reflection of systemic aging at cell level, and it fully confirms replicative senescence as a good model for the research of systemic aging.

[Information technology of modern NIR spectral analysis].

The main points, flowdiagram and the levels of optimization and building models of modern NIRS analysis are described in this paper. The strategy of how to apply and develop the technology of Near-Infrared spectroscopy analysis in our country is also described. In order to develop the technology of NIRS, the Industrialization and Democratization should combined.

[Changes of trace elements in cells of lung cancer and cervix cancer before apoptosis and after apoptosis].

The sensitivity of total reflection X-ray fluorescence analysis (TXRF) is higher than normal X-ray fluorescence analysis (XRF). The cells of lung cancer and cervix cancer were researched before apoptosis and after apoptosis employed the method of TXRF on new transformed synchrotron radiation XRF experimental station of new 3W1A beam line which built on Beijing Synchrotron Radiation Facility (BSRF) of BEPC (new detector and spectrometer systems). We acquired their fluorescence spectrum of various elements before apoptosis and after apoptosis. We discovered there were interesting results that changes even remarkable changes of some elemental contents had occurred in these cells before apoptosis and after apoptosis. These alterations of elemental contents have served an inspiration for us: if their regularities of change are found, we will hope that some contributions would be done at the treatments of cancer.
