Localization and expression of CSF-1 receptor in rat dental follicle cells.

Colony-stimulating factor-1 (CSF-1) accelerates tooth eruption in rats and is localized in the dental follicle, a loose connective tissue sac that is necessary for eruption to occur. CSF-1 enhances the cellular events that occur in the follicle prior to eruption--namely, an influx of monocytes into the follicle early post-natally to form the osteoclasts needed to resorb bone for the eruption pathway. Because CSF-1 levels are at a peak at day 3 post-natally, and because CSF-1 has an autocrine effect on its own gene expression, the question remains as to what causes the subsequent decline in CSF-1 protein and mRNA after day 3 post-natally. To determine if the autocrine effect is inhibited through the CSF-1 receptor, analysis of the CSF-1 receptor mRNA levels in cultured dental follicle cells reveals that high concentrations of CSF-1 reduce the gene expression of the CSF-1 receptor. Interleukin 1 alpha, a molecule that enhances CSF-1 gene expression, has no effect on CSF-1 receptor mRNA levels. Immunostaining for the CSF-1 receptor protein shows that it is present in the dental follicle early post-natally and is either absent or greatly reduced by day 10 post-natally. Earlier studies showed that the mRNA levels of the CSF-1 receptor also parallel this time course. Thus, the above results suggest that the feedback inhibition of the autocrine effect of CSF-1 on its own expression is through the effect of CSF-1 inhibiting the translation and transcription of its receptor. In turn, these molecular interactions possibly regulate the cellular events that occur in the follicle prior to and during eruption.

[Changes in contents of Zn, Cu, Fe, Ca, Mg in serum, urine and blister fluid after burn injury].

106 burned patients were divided into two groups: group L consisted of 57 patients with burns larger than 30% TBSA, and group S consisted of 49 patients with burns smaller than 30% TBSA. The contents of Zn, Cu, Fe, Ca, Mg in serum, urine and blister fluid were determined on 1, 2, 3, 7, 14, 21, 28 days after burn injury. The results indicated that all of the elements in serum were decreased except serum Fe on first day, and the levels were much lower in group L than in group S. The contents of Zn, Cu, Fe in urine were increased significantly and Ca, Mg in urine decreased. The contents of Zn, Fe, Ca in blister fluid were similar to normal values of serum, and Cu, Mg in blister fluid were less than that of serum. The studies indicated that the decrease in contents of Zn, Cu, Fe, Ca, Mg after burn injury was propably related to loss of these elements from urine and wound.

[Detection of p53 gene mutations in hepatocellular carcinoma].

This study screened 32 cases of hepatocellular carcinoma (HCC) from Chengdu detecting HBV DNA. HBsAg and p53 mutations by using Southern blot hybridization, immunohistochemistry and polymerase chain reaction/restriction enzyme digest methods, respectively. The results revealed all the cases had been infected by HBV; the frequency of HBV DNA integration into HCC cell was 72%, and the positive staining for HBsAg 96.3%(26/27). Of the 32 cases, 8 showed nuclear staining of p53 protein (25%), no mutation of p53 gene at all in 27 nontumorours liver tissues was identified. The resluts suggest the inactivation of p53 function may play a significant role in the genesis of HBV-associated HCC; however, the largely negative p53 mutation results in the current study conversely indicate that hepatocarcinogenesis may even involve other comprehensive mechanisms. Further studies are worth doing to evaluate the possible contribution of HBV to the p53 mutation in HCC.

[Differentiation-inducing effect of stepholidine and retinoic acid on human head and neck carcinoma].

To observe the differentiation-inducing effect of Stepholidine (ST) and Retinoic Acid (RA) on laryngeal and nasopharyngeal carcinoma, the expression of oncogene (C-myc, bcl-2) protein and tumor suppressor gene (p53) was done by using flow cytometry and ABC immunohistochemical methods combined with image analysis technique. The results indicated that after treated with ST and RA, the cells became well-differentiated, the cell growth was suppressed, contact suppress was partially recovered, colony forming was decreased, protooncogenes (C-myc bcl-2) protein was decreased, tumor suppressor gene (p53) protein was increased. No significant difference was observed between the cells treated with ST and TA. We believed that the Chinese medicine-Stephania might be expected to become a new prospective differentiation inducer in carcinoma of head and neck.

[Study of the molecular mechanism of human cytochrome P450 and genetic polymorphism of drug oxidative metabolizing enzyme].

Polymorphism has been detected in a variety of drug-metabolizing enzymes at both phenotypic and genotypic level. Human cytochrome P450 enzymes have been studied extensively in recent years, and the majority of mutations which give rise to a defective phenotype have now been identified(for example CYP2D6, CYP2C19, etc.). There are some groups of enzymes showing definite polymorphism at the phenotypic level but the exact genetic mechanisms are not yet clear(CYP1A1, CYP1A2, etc.). There are still other groups of enzymes, showing some indication of polymorphism at either the phenotypic or genotypic level yet have not been unambiguously demonstrated (for example CYP2A6, CYP2C9, CYP2E1, CYP3A4, etc.). The molecular mechanism of all these polymorphisms and possible polymorphisms is discussed, with particular reference to the effects of this variation on drug metabolism and on the susceptibility to chemically-induced diseases.

[Effects of HE particles on medicinal plant Agastache rugosus (Fisch. et Mey.) O. Ktze].

The biological effect of HE particles on the seeds of Agastache rugosus was probed in experiments on board a retrievable satellite. The result shows that the germination rate of the seeds pierced by HE particles radiation appears rather low. The seeds hit by HE particles (piercing radiation or not) start to germinate two days earlier than those in the control group, and the first leaf emerges four to five days earlier than that in the control group. The resultant seedlings grow markedly faster. Variations take place in the nuclear types of chromosome. The yield of essential oils becomes slightly higher. No marked changes have been observed in the major chemical components of these oils.

[The study of actinomyces viscosus adhere to teeth. III. The identification of mutants of A. viscosus and preparation of rabbit IgG against type 1 and type 2 fimbriae].

The authors prepared rabbit antisera against 5519 and 5951. The IgGs against type 1, type 2 fimbriae were purified from the antisera using sera absorbed with the other strain and IgG purification method. It was identified by SDS-PAGE and ELISA. This result suggested the purified proteins were the IgG against type 1 and type 2 fimbriae.

[The study of actinomyces viscosus adhere to teeth. IV. The study of salivary protein receptors of A. viscosus].

In order to determine the receptors of A. viscosus for adhesion, 7 kinds of main purified proteins from pellicle formed by whole saliva were tested. The result indicated that: 1. APRP, statherin, PRPG and SIgA were the receptors of standard and fresh isolated A. viscosus; 2. APRP and stather promoted the adherence of 5519 (1+2-) to HA, and SIgA promoted the adherence 5951 (1-2+) to HA; PRPG was the common receptor of the two fimbriae. It suggested that there were receptors not only type 1 fimbriae but also type 2 in pellicle. Two types of fimbriae were involved in the adherence of A. viscosus to pellicle. The difference existed in the receptor of adherence and the activity site of the same receptor.

[Effect of extremely low frequency (correction of frenquency) magnetic field on brain ischemic reaction in rats].

The effect of extremely low frequency magnetic field (ELMF, 15 Hz, 18 mT) on ischemic brain produced by ligation of general carotid artery in left side was studied in rats. The results showed that the degree of brain blood vessel dilation was higher in magnetic field group than that in control group at the 1st, 3rd and 6th hour after ligation and the extent of damage of neurocyte was apparently lighter in magnetic field group than that in control group at the 3rd and 6th hour after ischemia. It indicates that ELMF stimulation improves the brain blood circulation after brain ischemia and alleviates the brain ischemic reaction.

Comparison of effects of 5 and 20 Hz magnetic field on brain responses.

Effects of 5 and 20 Hz magnetic field (MF) on brain response were compared in 22 normal subjects during selective response (SRP) and selective mental arithmetic (SMA). It was found that significant changes in event-related potentials and EEG power spectra were induced by 5 Hz MF, but changes induced by 20 Hz MF were only in EEG coherence during SMA. The features of brain response changes suggest that the brain is in a more well being state after 5 Hz MF stimulation.

[A study on immune efficacy in children immunized by hepatitis B vaccine in disease surveillance points of Fujian].

A survey was carried out for immune effect among 1-9 years old children who had been immunized with Hepatitis B vaccine from normal population in the disease surveillance points in Fujian province. According to the unified programme of the national hepatitis sero-epidemiological survey, 758 children were investigated. The samples were tested by RIA kit. The results showed as follows. Compared with the HBsAg positive rate (23.16%) among the nonvaccinated children, the HBsAg positive rate among the vaccinated children was declined to 4.79%, and the effectiveness was 79.23%. Anti-HBs positive conversion rate (S/N > or = 2.1) was from 89.29% at the 1st year to 66.67% at the 6th year of vaccination. The results indicate that the Hepatitis B vaccination possesses a good protective efficacy.

[A gas chromatographic study on the characters of adsorbents in the enrichment of organic compounds from air samples].

A gas chromatographic method for the measurement of enriching characters of adsorbents used in preconcentration of organic compounds from air samples was studied. The character parameters suggested in this study include adsorption parameters (breakthrough volume or maximum sample volume) and desorption ones (minimum flashing volume and the lowest desorption temperature). The parameters of some low-molecular-mass compounds such as alcohols, saturated hydrocarbons, aromatics, esters and halohydrocarbons on GDX-301 were measured by using elution GC method. The maximum sample volumes of all the 20 compounds (with exception of methanol) got from these experiments are not less than 0.8 L at 35 degrees C for sampling tube (filled with 2 g GDX-301) and 0.3 L at 0 degrees C for focus tube (with 0.14 g GDX-301). The corresponding minimum flashing volumes of less than 177 mL and 13 mL respectively are needed to complete desorption at 180 degrees C, and the lowest desorption temperatures of not over 180 degrees C and 157 degrees C at flashing volume of 180 mL and 30 mL are lower than the maximum allowable operating temperature (250 degrees C) of GDX-301. The results showed that GDX-301 is a good adsorbent for enrichment of the organic compounds mentioned above. The following operating conditions are recommended: using the above sampling tube and focus tube, actual sample volume < or = 0.8 L, adsorption temperature < or = 35 degrees C and focus temperature 0 degrees C, desorption for 6 min from sampling tube and 1 min from focus tube at 180 degrees C with flashing rate 30 mL/min. Good correlations between the parameters and boiling point of the investigated compounds were found. It can be used to estimate the parameters of other organic compounds not measured in this study.

[Analysis of hexamethylene bisacetamide in biological fluids and tissues by RP-HPLC].

This paper describes the analysis of anticancer drug hexamethylene bisacetamide (HMBA) in biological fluids and tissues by RP-HPLC. The samples of plasmas, urine and tissues from cancer mice and rabbits were first used with methanol for pretreatment to remove protein impurity, then analyzed by RP-HPLC on an ODS column. The mobile phase was methanol/water (70/30, V/V), UV detection is employed at 210 nm. 153 samples were analyzed and good results were obtained. The recoveries of 93.3%-104% with a relative standard deviation of 0.04%. The regression equation is: Y = 0.32X - 1.07, with a linear correlation coefficient of 0.998 and the linear range of HMBA is 0 to 1000 mg/L. A sample analysis can be completed within 13 minutes. This method could be satisfactorily applied not only for the analysis of HMBA but probably also for the analysis of other drugs in biological fluids and tissues.

Comparison of two enzyme-linked immunosorbent assay tests for diagnosis of Helicobacter pylori infection in China.

An ELISA based on a pool of United States strains of Helicobacter pylori was compared with a newly developed ELISA based on a pool of Chinese strains. Both assays were tested using sera from 132 Chinese study subjects with biopsy-proven H. pylori infection. Using cutpoints designed to yield equal specificities of 94.9% in an uninfected control population, the sensitivity of the Chinese assay was 100.0%, compared to 97.7% for the United States assay (P = 0.25 by McNemar test). These results suggest that a H. pylori assay based on pooled antigens from United States strains will perform as well in the rural Chinese population as one based on antigens from Chinese strains.

[Study on particle contamination produced by syringe needles and rubber stoppers for pharmaceutical solutions].

The contamination of particles produced by syringe needles and rubber stoppers for pharmacal solution of venous transfusion was studied. The results showed that the contamination of particles was positive correlation with the usage frequency of syringe needles, so the syringe needles for single use were suggested. To compare two kinds of syringe needles with different structure (side hole and incline), the result was that the difference of particle contamination between them was similar and without statistic significance. The particles contamination produced by primary and used rubber stoppers which was protected by polyester film was studied. It was shown that the difference between them was no significant statistically. The results of this study supplyed a scientific evidence about repeated use of rubber stoppers.

Effects of high intensity impulse noise on ionic concentrations in cochlear endolymph of the guinea pig.

OBJECTIVE: To investigate the effect of acoustic overstimulation on concentrations of cations in cochlear endolymph and analyze the relationship between the ionic changes in endolymph and the hearing loss. METHODS: The endocochlear potentials (EP), K+, Na+ and Ca2+ concentration in cochlear endolymph were examined in vivo for normal and 167 +/- 2 dB SPL impulse noise exposed groups of guinea pig bored on time course by means of double-barreled ion-selective microelectrodes. Brain stem auditory evoked potential (BAEP) was used to evaluate the auditory function. Ca(2+)-ATPase activity was demonstrated cytochemically in the lateral cochlear wall as induced by Ando et al with slight modification. RESULTS: The K+ and Ca2+ concentration exhibited significant changes in 8-hour groups (P < 0.05 for K+ and P < 0.01 for Ca2+). Then the K+ concentration was eventually resumed to the initial levels in accordance with the EP recovery in 7 days, while during the same period Ca2+ concentration was always significantly higher than that in control group (P < 0.01). The BAEP threshold shifts were correlated well with changes in ionic concentrations, especially Ca2+ (P < 0.001), in the endolymph induced by acoustic trauma. Although the normal positive EP was observed 7 days after noise exposure, the function of the vascular stria was not completely restored as revealed by the fact that the Ca(2+)-ATPase was diffused to the apical membrane surface. CONCLUSIONS: Endolymph compartment intrinsic mechanism for maintaining ionic composition is seriously deteriorated after high impulse noise stimulation. The changes of the unique environment of endolymph may play an important role in the mechanism of sensorineural hearing loss induced by acoustic trauma.

[Postsurgical changes of the calculated wall shear stress in autogenous interpositional venous grafts by blood flow waveform analysis: clinical and experimental studies].

This investigation was designed to evaluate the postsurgical changes of autogenous, interpositional venous grafts in terms of the calculated wall shear stress by blood flow waveform analysis. In a model of tethered elastic tube, mean wall shear stress (tau) was obtained by the following equation: tau = 32 microQ/(pi.D3). Twenty-four grafts in repairing limb arterial injuries from 24 patients were examined with Doppler ultrasonic image instrument (7.5 MHz) for follow-up studies. Three mongrel dogs were selected for experimental studies. A segment of femoral vein (4-5 cm) was excised, reversed and interpositionally grafted into the femoral artery. The mean wall shear stress was calculated at 1 hour, 24 hours, 1 week, 4 and 12 weeks after implantation. Two occluded grafts were demonstrated in one femoral and one popliteal artery. The value of mean wall shear stress was higher in upper limb group (over 1.1 N/m2) than in lower limb group (five over 1.1 N/m2 and seven below 1.1 N/m2). The value of mean wall shear stress of grafts with follow-up time of more than 24 weeks was over 1.1 N/m2 in 6 grafts including one from lower limb while that was below 1.1 N/m2 in 6 grafts (all from lower limbs). No significant difference was found between the mean flow velocity values in upper and lower groups. The results from the experiment revealed that the value of mean wall shear stress decreased at one hour after operation, and increased and reached a fixed value beyond 1 week postsurgery. The vein graft in upper limb has better early and long-term result than that in lower limb because wall shear stress is correlated with the intimal hyperplasia, then the outcome of autogenous vein graft. In conclusion, we propose that wall shear stress be applied for follow-up studies on vascular grafts in limb arterial injuries and hemodynamic factors be considered for arterial reconstructive procedures.

Evidence from Earthquake Data for a Partially Molten Crustal Layer in Southern Tibet

Earthquake data collected by the INDEPTH-II Passive-Source Experiment show that there is a substantial south to north variation in the velocity structure of the crust beneath southern Tibet. North of the Zangbo suture, beneath the southern Lhasa block, a midcrustal low-velocity zone is revealed by inversion of receiver functions, Rayleigh-wave phase velocities, and modeling of the radial component of teleseismic P-waveforms. Conversely, to the south beneath the Tethyan Himalaya, no low-velocity zone was observed. The presence of the midcrustal low-velocity zone in the north implies that a partially molten layer is in the middle crust beneath the northern Yadong-Gulu rift and possibly much of southern Tibet.
