Sugar interaction with metal ions. The crystal structure and Raman spectra study of SmCl3-galactitol complex.

The crystal structure of 2SmCl3.galactitol.14H2O has been determined. The crystal system is triclinic, space group: P-1. The unit cell dimensions: a = 9.683(2) A, b = 10.341(2) A, c = 7.990(2) A; alpha = 108.01(3) degrees, beta = 92.71(3) degrees, gamma = 88.42(3) degrees. Each Sm atom is coordinated to nine oxygen atoms, three from the alditol and six from water molecules, with Sm-O distance from 2.417 to 2.520 A. The seventh water molecule is hydrogen-bonded by the hydroxy hydrogen on O-3 (O(3)-H(13)...O(10), 2.635 A). After forming complexes the peaks have shifted and the relative intensities have changed in the IR and Raman spectra, which are corresponding to the changes in bond distances and bond angles of the structures. The IR and Raman spectra of Pr-, Nd- and Sm-galactitol complexes are similar, which show that the three metal ions have the same coordination mode.

Corrections to moving chemical reaction boundary equation for weak reactive electrolytes under the existence of background electrolyte KCl in large concentrations.

In this report, the moving chemical reaction boundary (MCRB) was formed by the weak reaction electrolytes of NH3.H2O and CH3COOH under the existence of background electrolyte KCl in large concentrations, the experiments were compared with the predictions by the moving chemical reaction boundary equation (MCRBE) for weak reactive electrolytes. It was found that the experimental results are far from the predictions with the MCRBE. So the MCRBEs must be corrected under the given experimental conditions. The corrected MCRBEs are given for the MCRB formed with weak reactive electrolytes coupled with KCl at high concentrations.

Autocrine TGFbeta signaling mediates vitamin D3 analog-induced growth inhibition in breast cells.

In this study, we address whether TGFbeta signaling mediates vitamin D3 analog-induced growth inhibition in nonmalignant and malignant breast cells. Normal mammary epithelial cells (184), immortalized nonmalignant mammary epithelial cells (184A1 and MCF10A), and breast cancer cells (early passage MCF7: MCF7E) were sensitive to the inhibitory effects of vitamin D3 analogs (EB1089 and MC1288) while late passage MCF7 breast cancer (MCF7L) cells were relatively resistant. A similar pattern of sensitivity to TGFbeta was observed with these cells. Thus, the sensitivity to the vitamin D3 analogs correlated with the sensitivity to TGFbeta. MCF7L TGFbetaRII-transfected cells, which have autocrine TGFbeta activity, were more sensitive to EB1089 than MCF7L cells. TGFbeta neutralizing antibody was found to block the inhibitory effects of these analogs. These results are consistent with the idea that autocrine TGFbeta signaling mediates the anti-proliferative effects of the vitamin D3 analogs in these cells. The expression of TGFbeta isoforms and/or TGFbeta receptors was induced by the analogs in the vitamin D3 and TGFbeta sensitive cells. Vitamin D3 analogs did not induce TGFbeta or TGFbeta receptor expression in the resistant MCF7L cells. Therefore, EB1089 induces autocrine TGFbeta activity through increasing expression of TGFbeta isoforms and/or TGFbeta receptors. In addition, EB1089 induced nuclear VDR protein levels in the sensitive 184A1 cells but not in the resistant MCF7L cells. 184A1 cells were more sensitive to EB1089-induced VDR-dependent transactivation than MCF7L cells as measured by a luciferase reporter construct containing the VDRE, indicating a defect of VDR signaling in MCF7L cells. Smad3, a TGFbeta signaling mediator, coactivated VDR-dependent transactivation in 184A1 cells but not in MCF7L cells. These results indicate that Smad3 coactivates VDR to further enhance TGFbeta signaling and vitamin D3 signaling in the sensitive 184A1 cells. The results also indicate that Smad3 is not of itself sufficient to coactivate VDR in TGFbeta/vitamin D3 resistant MCF7L cells and other factors are required. We found that the PI 3-kinase pathway inhibitor LY29004 inhibited the synergy of TGFbeta and EB1089 on VDR-dependent transactivation activity. This indicates that the crosstalk between TGFbeta and vitamin D signaling is also PI 3-kinase pathway dependent.

Further evidence for mitochondrial dysfunction in progressive supranuclear palsy.

Recent data from our laboratory have identified a role for mitochondrial dysfunction in the pathogenesis of progressive supranuclear palsy (PSP). To extend this finding, we measured key parameters of mitochondrial function in platelet-derived cytoplasmic hybrid (cybrid) cell lines expressing mitochondrial genes from patients with PSP. We observed significant decreases in aconitase activity, cellular ATP levels, and oxygen consumption in PSP cybrids as compared to control cybrids, further suggesting a contributory role of impaired mitochondrial energy metabolism in PSP, possibly due to genetic abnormalities of mitochondrial DNA.

Audiogenic seizure susceptibility is induced by termination of continuous infusion of gamma-aminobutyric acid or an N-methyl-D-aspartic acid Antagonist into the inferior colliculus.

The inferior colliculus (IC) is strongly implicated in seizure initiation in a genetic form of audiogenic seizures (AGS) and in AGS observed during ethanol withdrawal (ETX). Ethanol is known to block the actions of excitatory amino acids (EAA) and enhance the actions of gamma-aminobutyric acid (GABA) in several brain areas, including the IC. The present study investigated the effects on susceptibility to AGS following withdrawal from continuous blockade of N-methyl-D-aspartic acid (NMDA) receptors or continuous activation of GABA receptors in the IC. This involved infusion of GABA (1 M) or a competitive NMDA antagonist, DL-2-amino-7-phosphonoheptanoic acid (AP7, 1 mM), at 0.25 microl/h for 7 days using an Alzet osmotic minipump. Following abrupt termination of the infusion, AGS susceptibility began at 30 min. The incidence of AGS was 38.9 and 56.3% following GABA and AP7 withdrawal, respectively. The AGS behaviors observed during withdrawal, which included wild running and bouncing clonus, were very similar to those evoked by acoustic stimuli during ETX. AGS susceptibility lasted for several hours and in 13% of animals persisted for up to 6 months. The current results support diminished GABAergic and elevated glutamatergic function in the IC as the critical mechanisms and sites for AGS initiation. The present study, coupled with previous evidence that chronic ethanol exposure reduced GABA-mediated inhibition and enhanced EAA-mediated excitation, suggests that these amino acid receptor-mediated alterations in the IC are key elements in initiating AGS during ethanol withdrawal.

Assessing pollution prevention program by QUAL2E simulation analysis for the Kao-Ping River Basin, Taiwan.

Wise and sustainable uses of water resources are essential for an effective river-basin-management planning. Recent management strategy further addresses the fact that quantity and quality of water are closely interrelated, and both must be considered simultaneously for all water resources and water quality management practices. The aim of this paper is to explore the impacts of water resources redistribution and pollution prevention actions between and within river basins simultaneously in South Taiwan. Much emphasis will be placed on assessing the impacts of water transfer over natural boundary to satisfy the needs of industrial development in the Tseng-Wen River system and its resultant influence on the water quality in the downstream area of the Kao-Ping River system where the pollution prevention program is to be implemented. The Kao-Ping River was further characterized hydraulically and environmentally, based on a full investigation of discharges and withdrawals in the river reaches. QUAL2E was successfully calibrated and validated using data collected between 1998 and 1999, and the model was capable of predicting the concentrations of biochemical oxygen demand, dissolved oxygen, total phosphate-phosphorus, and ammonia-nitrogen (NH3-N) for the entire river system. With the aid of QUAL2E simulation model, it shows eliminating the pig farming activities and constructing the sewer systems in the upstream area of Kao-Ping River cannot guarantee the full compliance with water quality standards in the downstream area and water transfer in the upstream area further increases negative impacts on the water quality in the wet season. The predicted situation of water quality in the dry season may even present worse condition. Additional water pollution control policy, such as the use of economic instruments, for controlling and reducing the waste-load of biochemical oxygen demand and ammonia-nitrogen is needed in the Kao-Ping River system in the long run.

Apolipoprotein C-I expression in the brain in Alzheimer's disease.

The H2 allele of apolipoprotein (apo) C-I is associated with Alzheimer's disease (AD). However, this association is potentially confounded by the linkage disequilibrium of H2 with the epsilon2 and epsilon4 alleles of apoE and of H1 with the epsilon3 allele. To establish plausibility for a direct role for apoC-I in AD, we compared apoC-I and apoE protein and mRNA levels in postmortem specimens of frontal cortex and hippocampus from AD patients with levels in nondemented controls. In H2-allelic individuals (usually also epsilon4 carriers), apoC-I mRNA levels were strikingly lower with AD (by 65%, P < 0.05), but apoC-I protein levels in AD were significantly higher (by 34%, P < 0.05). The opposite direction of the apoC-I mRNA and apoC-I protein level changes in AD in the epsilon4/H2 genotype may reflect decreased clearance of CNS lipoproteins associated with apoE4. In H1/H1 (usually epsilon3/epsilon3) individuals, both apoC-I protein and mRNA were lower in AD. ApoC-I protein levels in hippocampus were nearly twice those in frontal cortex. Immunohistochemistry of hippocampus revealed colocalization of apoC-I protein with the astrocytic marker GFAP. In addition, cultured human astrocytes expressed the mRNA for apoC-I. This study confirms apoC-I expression in the CNS and identifies astrocytes as the source of apoC-I. In addition, it has revealed differences in apoC-I expression based on site, genotype, and disease status that may reflect a role for apoC-I in the pathogenesis of AD.

Speciation of rare earth elements in soil by sequential extraction then HPLC coupled with visible and ICP-MS detection.

The distribution of rare earth elements (REE) in a pooled soil sample collected from Zhangzhou, Fujian Province, China, was screened by a five-step sequential extraction procedure coupled with ICP-MS determination after preconcentration of REE and removal of the matrix by extraction with 1-phenyl-3-methyl-4-benzoyl-5-pyrazolone (HPMBP). The results showed that the distribution of REE in the different fractions of the pooled soil sample studied followed the order soluble species (46.76%) > species bound to organic matter (22.08%) > species in the residue (16.77%) > species bound to Fe-Mn oxides (2.02%). An effective method for speciation of REE, which utilized weak cation-exchange HPLC separation hyphenated with post-column derivatization and visible or on line ICP-MS detection, was, moreover, developed and successfully applied to the speciation of REE in the soluble extract of the pooled soil sample. The stability of known complexes of lanthanum during the HPLC separation was investigated with fluoride, citrate, and ethylenediamine tetraacetic acid (EDTA) chosen as ligands modeling those in the soil. REE in the soluble extract of the pooled soil sample were subsequently classified into three types of species--< or = +1 charged complexes (negatively charged, neutral, and +1 charged), +2 charged complexes, and "free" REE species. This method is expected to be useful for identification of bioavailable (or toxic) species of REE in environmental samples.

Derivative hydride generation atomic absorption spectrometry and determination of lead traces in waters.

Fundamentals of derivative hydride-generation atomic absorption spectrometry (DHGAAS) are described. A linear relationship was obtained between the derivative absorbance and the concentration of analysis in a sample. The new DHGAAS method was applied to the determination of traces of lead in water. The conditions affecting the derivative absorbance of lead were evaluated and optimized. The detection limit and sensitivity of the proposed method were 26 times and 8.8 times better, respectively, than those of conventional hydride-genera-tion atomic absorption spectrometry. The characteristic concentration (for a derivative absorbance of 0.0044) and detection limit (3sigma) for lead were 0.017 and 0.096 ng mL(-1), respectively, for a 2 mV min(-1) sensitivity range setting. The recovery range was 92.5-103%.

No association between dopamine D2 receptor gene (DRD2) and human intelligence.

Significantly diminished intellectual functioning, as indicated by appropriately administered IQ tests with scores below 70, is a frequent mental handicap leading to severe social disadvantages and serves as a paradigm for molecular genetic research of complex disorders and traits due to its multitude of known and unknown, genetic as well as environmental causes. Since the number of confounding variables is expected to be considerably reduced in the normal population at the opposite ends of the IQ distribution, we employed a contrast of extremes approach by comparing adults of high (N = 71) and average IQ (N = 78) in association studies to search for genes involved in the multigenic forms of familial mental retardation. The dopamine D2 receptor gene (DRD2) was chosen as a candidate gene for general cognitive ability (g) since it has been found to be associated with visuospatial ability which in turn is highly correlated with g. Confirming two similar studies in children, however, no significant differences were obtained. Given three negative studies, the DRD2 gene is unlikely to pay a major role in g.

Evidence for specific ceramidase present in the intestinal contents of rats and humans.

A neutral ceramidase activity stimulated by bile salt was previously identified in the intestinal content. Recently, bile salt stimulated lipase (BSSL) was found to have ceramidase activity. It is unknown whether the ceramidase activity previously found is attributable to BSSL. To address this question, we compared the behaviors of high quaternary aminoethyl (HQ) anion exchange chromatography, the distributions, the stability, and the responses to lipase inhibitor between ceramidase and pancreatic BSSL. The proteins from whole small intestinal contents of humans and rats were precipitated by acetone and dissolved in 20 mM Tris buffer pH 8.2. These proteins had neutral ceramidase activity but not BSSL activity against p-nitrophenyl acetate. When the proteins were subject to HQ chromatography, two peaks of ceramidase activity were identified, which had acid and neutral pH optima, respectively. Neither of them had BSSL activity against p-nitrophenyl acetate. Western blot using BSSL antiserum failed to identify BSSL protein in the fractions with high neutral ceramidase activity. In rat intestinal tract, pancreatic BSSL activity was high in the duodenum and declined rapidly in the small intestine, whereas neutral ceramidase activity was low in the duodenum and maintained a high level until the distal part of the small intestine. In addition, orlistat, the inhibitor of lipase, abolished human BSSL activity against p-nitrophenyl acetate and slightly reduced its activity against ceramide but had no inhibitory effect on ceramidase activity isolated by HQ chromatography. In conclusion, we provide the evidence for a specific ceramidase other than pancreatic BSSL present in the intestinal content. The enzyme may play important roles in digestion of dietary sphingolipids.

Healing of burn wounds in transgenic mice overexpressing transforming growth factor-beta 1 in the epidermis.

Transforming growth factor-beta (TGF-beta) isoforms are multifunctional cytokines that play an important role in wound healing. Transgenic mice overexpressing TGF-beta in the skin under control of epidermal-specific promoters have provided models to study the effects of increased TGF-beta on epidermal cell growth and cutaneous wound repair. To date, most of these studies used transgenic mice that overexpress active TGF-beta in the skin by modulating the latency-associated-peptide to prevent its association with active TGF-beta. The present study is the first to use transgenic mice that overexpress the natural form of latent TGF-beta 1 in the epidermis, driven by the keratin 14 gene promoter to investigate the effects of locally elevated TGF-beta 1 on the healing of partial-thickness burn wounds made on the back of the mice using a CO(2) laser. Using this model, we demonstrated activation of latent TGF-beta after wounding and determined the phenotypes of burn wound healing. We found that introduction of the latent TGF-beta1 gene into keratinocytes markedly increases the release and activation of TGF-beta after burn injury. Elevated local TGF-beta significantly inhibited wound re-epithelialization in heterozygous (42% closed versus 92% in controls, P < 0.05) and homozygous (25% versus 92%, P < 0.01) animals at day 12 after wounding. Interestingly, expression of type I collagen mRNA and hydroxyproline significantly increased in the wounds of transgenic mice, probably as a result of a paracrine effect of the transgene.

Barrel-stave model or toroidal model? A case study on melittin pores.

Transmembrane pores induced by amphiphilic peptides, including melittin, are often modeled with the barrel-stave model after the alamethicin pore. We examine this assumption on melittin by using two methods, oriented circular dichroism (OCD) for detecting the orientation of melittin helix and neutron scattering for detecting transmembrane pores. OCD spectra of melittin were systematically measured. Melittin can orient either perpendicularly or parallel to a lipid bilayer, depending on the physical condition and the composition of the bilayer. Transmembrane pores were detected when the helices oriented perpendicularly to the plane of the bilayers, not when the helices oriented parallel to the bilayers. The evidence that led to the barrel-stave model for alamethicin and that to the toroidal model for magainin were reviewed. The properties of melittin pores are closely similar to that of magainin but unlike that of alamethicin. We conclude that, among naturally produced peptides that we have investigated, only alamethicin conforms to the barrel-stave model. Other peptides, including magainins, melittin and protegrins, all appear to induce transmembrane pores that conform to the toroidal model in which the lipid monolayer bends continuously through the pore so that the water core is lined by both the peptides and the lipid headgroups.

Bicondylar tibial plateau fractures managed with the Sheffield Hybrid Fixator. Biomechanical study and operative technique.

The two main challenges in the management of bicondylar tibial plateau fractures are: Firstly, the compromised skin and soft tissue envelope which invite a high rate of complications following attempted open reduction and dual plating. Secondly, poor bone quality and comminuted fracture patterns, which create difficulty in achieving stable fixation. Although dual plating is considered to be the best mechanical method of stabilizing these complex fractures, there remains concern regarding the high rate of complications associated with extensive soft tissue dissection, required for the insertion of these plates in an already compromised knee. The Sheffield Hybrid fixator (SHF) technique offers a solution to the two main problems of these difficult fractures by minimizing soft tissue dissection, since bone fragments are reduced and fixed percutaneously, and providing superior cancellous bone purchase with beam loading stabilization for comminuted fractures. Our biomechanical testing showed the SHF with four tensioned wires to be as strong as dual plating and able to provide adequate mechanical stability in the fixation of bicondylar tibial plateau fractures. This was confirmed clinically by a prospective review of the use of the SHF at our centre, for managing complex and high-energy tibial plateau fractures with a good final outcome and no cases of deep infection or septic arthritis.

The Sheffield hybrid fixator--a clinical and biomechanical review.

The Sheffield hybrid fixator (SHF) was developed to complement and extend the use of current monolateral fixation systems. Unlike other hybrid designs, it relies on rings for the diaphyseal as well as the metaphyseal support. Since it is an all ring system it may be used to cross joints and perform progressive deformity and contracture correction. Unlike the Ilizarov system only one ring is required at each level and diaphyseal transfixation is avoided. Initial biomechanical testing has demonstrated similar mechanical characteristics to an all wire Ilizarov fixator, however, increased shear motion was noted due to the stiffness asymmetry between the segments controlled by wires and screws. The first 100 cases were reviewed with good patient compliance and satisfactory results. Minor component problems were identified and corrected during this period and no significant complications occurred. The device is recommended for a wide range of complex trauma and limb reconstruction uses.

Glyceroglycolipids from Serratula strangulata.

The rhizomes of Serratula strangulata yielded three glyceroglycolipids, i.e. 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-(6-amine-6-deoxy-alpha-D-glucosyl)-glycerol, 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-(6-p-hydroxy-phenyl-propionamido-6-deoxy-alpha-D-glucosyl)-glycerol and 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-[alpha-D-glucose(1-6)-beta-D-allose]-glycerol, as well as a known sesquiterpene lactone and three known phytoecdysones. Their structures were elucidated on the basis of spectral data, especially by 2D NMR spectroscopic methods and chemical conversion. These compounds exhibited significant antibacterial and antitumor activities.

Taraxastane-type triterpenoids from Saussurea petrovii.

Two taraxastane triterpenoids, i.e. taraxast-20-ene-3beta,30-diol (1) and 20alpha,21alpha-epoxy-taraxastane-3beta,22alpha-diol (2), as well as four known triterpenes taraxast-20(30)ene-3beta,21alpha-diol (3), taraxast-20(30)-ene-3beta-ol (4), taraxast-20-ene-3beta-ol (5) and taraxastane-3beta,20alpha-diol (6) were isolated from the Chinese medicinal plant Saussurea petrovii. Their structures were elucidated by spectroscopic methods. These compounds, especially 1 and 2, exhibit significant antitumor and antibacterial activity.

Biofilter treatment of aquaculture water for reuse applications.

Biotreatment of aquaculture water for recirculation purposes is a sensible mean to support the further growth of aquaculture industry without excessive water demands that are environmentally unsustainable. This study evaluates the efficacy of biofilter treatment of an eel (Anguilla japonica) culture pond water using different filter media and flow scheme arrangements. The experimental results demonstrate that biofilter systems packed with suitable filter media are capable of improving the quality of effluents for recirculation applications. The characteristics of the filter media appear to be more critical than biofilter flow scheme arrangements in affecting the efficacy of the biofilter treatment. Filter media with surface and structural characteristics are conducive to the development of biofilms and the capture of organic suspended matter are desirable to ensure good and consistent biofilter performance. Under such circumstances the bacterial "consortia" in the biofilter are capable of utilizing the captured organic suspended matter as an alternative substrate to support their metabolic activities when the concentration of the primary substrate (i.e., BOD) is low. For the eel pond water, a biofilter packed with filter media having cross-link structures and a high bed porosity, followed by another biofilter packed with a type of filter media having rough surfaces, produced the best results under the conditions tested. Moreover, a preliminary cost-benefit analysis confirms its cost advantages.
