[Rat keratinocyte primary cultures based on conductive polypyrrole primary cell culture technique].

Orthogonal experimental design has been applied to study the suitable cell culture medium and condition of conductive polypyrrole on rat keratinocyte cell cultures. The results showed that one thousand milliliters of complete minimum essential medium(CMEM) which contained 900 ml minimum essential medium(MEM), 10% fetal bovine serum(FBS), 1% NEAA, 6 ml insulin and 10 mg hydrocortisone was fit for keratinocyte cell culture. And the conductive polypyrrole obtained with 10 mA current density for 100 second switching times, given 100 mV electrical stimulation voltage for 2 hours, would stimulate the growth of rat keratinocyte cell in vitro cultures. The cell density under culture conditions described above was 1.30 times as high as that under routine culture conditions.

[The clinical value of surgical-pathological staging for endometrial carcinoma].

OBJECTIVE: To explore the practical value of surgical-pathological staging for endometrial carcinoma. METHODS: Ninety-six patients with endometrial carcinoma accepted primary surgical treatment from January 1995 to December 1999 were analyzed retrospectively in our hospital. The clinical and surgical pathologic staging of these patients were compared. RESULTS: The total differences between clinical and surgical-pathologic staging were 44 cases (45.8%), in stage I 24.0%, stage II 76.9%, in stage III 2 of 5 cases. The metastasis rate of pelvic lymph nodes was 10.3%, in stage I 1 of 16 cases, stage II 14.7%. Fourteen patients (14.6%) with extrauterine pelvic metastasis were found, of which 2 of 19 cases in stage I b, 23.1% in stage II. 9.4% with ovarian metastasis, in stage I a 9.7%, stage II 10.3%. 7.9% with positive peritoneal cytology, in stage I 4.0%, stage II 10.3%. 5.2% with omentum metastasis. 2.1% with appendix metastasis. Clinical stages, depth of myometrial invasions, pathological grades and histological types were related to the extent of pelvic and abdominal cavity by univariate analysis (P < 0.01, 0.05). The first three factors were significantly correlated to dissemination by multivariate (P < 0.05). CONCLUSIONS: The surgical-pathological staging is more accurate than the clinical staging, especial in stage II clinical stage depth of myoinvasion, patho-histological grades were closely related to the dissemination in pelvic-abdominal cavity. The surgical-pathological staging is able to assess prognosis objectively and guide therapy for endometrial carcinoma.

[Chelate-induced phytoextraction of copper contaminated upland red soil].

A copper spiked red soil was used in pot experiment to study the effects of EDTA and low molecule weight organic acids on Cu speciation in soil, and Cu uptake by Brassica juncea. The results showed that the mobility of Cu in soil was improved obviously when EDTA was added at the vigorous growth stage. Both water-extractable and exchangeable Cu concentration increased significantly following EDTA addition. Citric acid and malic acid only had an effect on water-extractable Cu, and no effect on Cu uptake by the plant. EDTA significantly increased the concentration of Cu in plant leaves and roots, and the Cu uptake by Brassica juncea. All of these indicated the chelate-induced phytoextraction.

A pilot study of a biotrickling filter for the treatment of odorous sewage air.

The nuisance impact of air pollutant emissions from wastewater treatment plants (WWTPs) is a major issue of concern to Singapore and to many developing cities worldwide. Existing chemical and physical treatment methods are efficient but costly, and may generate secondary pollutants. Biotreatment, on the other hand, is a proven control technology for effectively removing hydrogen sulphide (H2S), the principal odour component of sewage air. The biotechnology is cost-effective to remove low-concentrations of biodegradable compounds from a large flow of waste gases. A pilot-scale biotrickling filter, packed with pall rings, was set up at a WWTP in Singapore to investigate its effectiveness for treatment of odorous sewage air. A series of experiments were conducted to assess the performance of the biotrickling filter under various operating scenarios. Results indicated that even at 5 second gas retention time, the biotrickling filter could remove 95% of the inlet H2S. The behaviour of the biotrickling filter under various operating scenarios are presented and discussed in this paper.

[Sulfa-drug wastewater treatment with anaerobic/aerobic process].

Sulfa drug wastewater was treated with anaerobic/aerobic process. The removal ratios of TOC reached about 50% in anaerobic phase and about 70% in aerobic phase respectively, while volume loading rate of TOC was about 1.2 kg/(m3.d) in anaerobic phase and about 0.6 kg/(m3.d) in aerobic phase. Removal of TOC in anaerobic phase was attributed to the reduction of sulfate.

[The change of immunocompetent cells in normal human dental pulp during development of immature permanent teeth].

OBJECTIVE: To study changes of several immunocompetent cells (ICCs), namely, pulpal dendritic cells (PDCs), T lymphocytes, B lymphocytes, and endothelial cells, in normal human dental pulp according to different development stage of completely erupted but not fully developed premolar in order to provide evidence for local immune defense mechanisms during development of immature permanent teeth. METHODS: Based on the dental development status, the pulp tissue was classified into the following groups: big trumped-shaped group, small trumped-shaped group and closed group. The above mentioned cells were immunohistochemically examined by use of HLA-DR, CD45RO, CD20 mouse anti-human monoclonal antibodies. Double-staining was also done by use of the former two antibodies. The number of positively stained cells or blood vessels was counted and statistically analyzed. RESULTS: 1. Normal dental tissue of three groups all expressed HLA-DR+ PDCs. Among three groups, the number of HLA-DR+ cells in big trumped-shaped group was more than that of small trumped-shaped group and closed group. 2. There was no significant difference of positive blood vessel rate of HLA-DR expressing endothelial cells among three groups. 3. There was no significant difference of CD45RO+ T cells among three groups. 4. By use of double-labeling immunohistochemistry, HLA-DR+ PDCs and CD45RO+ T cells were found to be rarely in contact with each other. Correlation analysis between two kinds of cells did not indicate the linear relationship. CONCLUSIONS: The number of PDCs tends to decrease with increasing age in the developing dental pulp of human normal tooth, other ICCs are not related with dental development.

Computerized tomography pleurography: a new method for detecting minor pleural lesion.

OBJECTIVES: To evaluate the ability of CT pleurography (CTP) in detecting minor pleural lesions in patients with pleural effusion and to assess its value in distinguishing malignant from benign pleural lesions. METHODS: A prospective study of 50 patients with pleural effusion was conducted using conventional CT and CTP. CT scan was run after injecting an appropriate amount of contrast medium into the pleural cavity. RESULTS: In 24 patients, all lesions detected by conventional CT were demonstrated by CTP. In 13 of 24 patients, CT pleurography detected additional lesions. In 20 of 26 patients with negative findings on conventional CT, CTP was capable of demonstrating the presence of pleural lesions. The sensitivity, specificity and accuracy of detecting pleural lesions were 25%, 100% and 30% for conventional CT, 86%, 100% and 87% for CTP, respectively. CONCLUSION: CTP is superior to conventional CT in detecting and for the differential diagnosis benign and malignant pleural lesions.

[Evaluation of optic disc in open-angle glaucoma with hemifield defect by Heidelberg retinal tomography].

OBJECTIVE: To investigate whether the glaucomatous optic disc change demonstrated by Heidelberg retinal tomography (HRT) is consistent to the distribution of the visual field defects and whether Heidelberg retinal tomography, (HRT) parameters in primary open angle glaucoma (POAG) and normal tension glaucoma (NTG) are different. METHOD: Sixty-four eyes of 64 patients with open angle glaucoma (27 of POAG, 37 of NTG) and hemifield abnormality detected by Humphery Field Analyzer (HFA) were tested by HRT. The HRT parameters and the visual field results were divided into two parts (0 degree-180 degrees and 180 degrees-360 degrees) for the corresponding analysis. RESULTS: Comparing with the half disc corresponding to the intact hemifield, the half disc corresponding to the defective hemifield has significantly higher values of cup/disc area ratio and cup shape measure, but has significantly lower values of rim area, the thickness and cross-section area of the retinal nerve fiber layer. No significant differences were found between NTG and POAG eyes in the HRT parameters of the half disc corresponding to the defective hemifield. CONCLUSIONS: The optic disc changes detected by HRT are consistent to the distribution of the visual field defects, cup/disc area ratio, cup shape measure, rim area, mean thickness and cross-section area of the retinal nerve fiber layer, and they are the relatively sensitive parameters. That the HRT parameters of NTG and POAG are similar suggests they possibly have certain similar mechanisms in the development of glaucomatous neuropathy.

[Influence of a triplex superimposed treatment on HBV replication and mutation during treating chronic hepatitis B].

OBJECTIVE: To observe and evaluate the influence of a new antiviral treatment scheme on HBV replication and mutation during treating chronic hepatitis B. METHODS: In the test group, lamivudine, IFN alpha-2b, Astragalus membranaceus were chosen as a triplex superimposed treatment scheme for treating the patients who were on the state of HBV high replication and involved in the clinical condition of chronic hepatitis B. The control group was treated with lamivudine alone. The observed parameters percentage of patients in whom HBV DNA became undetectable (serum HBV DNA<1.6 ng/L), HBeAg/anti-HBe seroconversion rate; HBV DNA serum level; HBV YMDD mutation rate and pre-C region mutation rate. RESULTS: Compared with that of the control group, HBV DNA undetectable rate of the test group increased markedly at weeks 12, 36, 48(P <0.05), HBeAg negative rate of the group increased markedly at week 36, 48(P <0.05), while anti-HBe positive rate increased only at week 48 (31.58 vs.19.23%, P <0.05). After 4 weeks of treatment, HBV DNA serum level of both the test group and the control group reduced very remarkably (P <0.01), and at week 48, reduced more significantly (P <0.001). Compared with the control group, HBV DNA serum level of the test group reduced notably at week 12 (P <0.05) and very notable at week 36 and 48(P <0.01). At week 12, the pre-C region mutation occurred in the test group, and at week 24, 36,48, the pre-C region and YMDD mutations occurred in both the test group and the control group. CONCLUSIONS: As to anti-viral treatment of chronic hepatitis B, the triplex superimposed treatment had better efficacy than lamivudine alone.

[Separation of catechins and caffeine in tea polyphenol by isocratic elution high performance liquid chromatography].

The catechins, particularly in green tea, have been found to possess anti-mutagenic and anti-tumorigenic properties. As each catechin possesses distinct properties, a simple and rapid method that could be used for analysis of individual catechins in a complex mixture would be necessary. A relatively simple and rapid method for simultaneous separation of five catechins and caffeine in tea polyphenol by isocratic elution high performance liquid chromatography has been developed. The analysis could be finished within 30 min. They were measured using Resolve C18 column (at 43 degrees C) and UV detector (at 280 nm), water-85% phosphoric acid aqueous solution-acetonitrile-dimethyl formamide(DMF) (859:1:120:20, V/V) as mobile phase. There was a good linear relationship between the content of component and its peak area for catechins and caffeine, with the correlation coefficients of 0.9992-0.9999. The average recoveries (n = 5) were 83.33%-104.42%, and the relative standard deviations (n = 6) were 0.74%-1.43%. The effect of concentration of DMF in mobile phase was studied.

[A clinical analysis of cholesterol granuloma following chronic suppurative otitis media].

OBJECTIVE: To investigate the pathophysiological mechanism, clinical menifestation and radiographic diagnosis of cholesterol granuloma following chronic suppurative otitis media. METHOD: Six cases of CG following chronic suppurative otitis media, confirmed by surgery and pathology, were reviewed and analyzed. RESULT: CG frequently accompanied with other middle ear diseases, and was shown as a high signal intensity on both T1-and T2-weighted images in magnetic resonance imaging(MRI). CONCLUSION: It was postulated that the obstruction of pneumatized temporal bone air cell, caused by other middle ear diseases such as cholesteatoma and tympanosclerosis, might be the pathophysiological mechanism of CG. The evaluations of computed tomography (CT) and clinical manifestation were limited to distinguish CG from cholesteatoma or other neoplasm, while the MRI can be of great value to characteristic diagnosis.

The electroretinogram in patients with retinal vascular occlusion.

PURPOSE: To measure and analyze the electroretinogram (ERG) in patients with retinal vascular occlusion. METHOD: Fifty-eight cases (59 eyes) of retinal vascular occlusion and the fellow eyes of 47 cases were tested with Ganzfeld ERG. The scotopic and photopic ERG, and oscillatory potentials were tested according to the ERG standard of ISCEV. RESULTS: The abnormal rates of OPs and b wave were higher in retinal vascular occlusion. The abnormal ERG appeared mostly in CRVO among four types of retinal vascular occlusion. The comparisons of some amplitudes and latencies between the fellow eyes and the affected eyes showed statistically significant difference. CONCLUSION: The oscillatory potentials and b waves have important roles in evaluating the retinal function of patients with retinal vascular occlusions. There are some clinical significance for comparing ERG between the affected eye and the fellow eye.

[Character of alpha-crystallin molecular chaperone activity in selenite cataract].

PURPOSE: To evaluate the character of alpha-crystallin molecular chaperone activity in selenite cataract. METHODS: alpha-Crystallin from lenses of normal young rats and lenses with selenite cataract were separated and purified by gel filtration chromatography. The chaperone activity of alpha-crystallin was assayed by mearsuring heat-induced aggregation and scattering of catalase. RESULTS: alpha-Crystallin in both normal and cataractous lenses specifically prevented catalase against thermal aggregation compared with control proteins. The preventive ability in alpha H-crystallin was less than in alpha L-crystallin. Chaperone activity was diminished in selenite cataract. There was statistically significant difference in chaperone activity between normal and cataractous lenses in both alpha H-crystallin and alpha L-crystallin respectively. CONCLUSION: The reduction of alpha-crystallin chaperone activity in selenite cataract by preventing thermal aggregation of catalase is an important process in cataract formation.

[Study on hepatic-protective effect of "yinzhihuang" granula].

OBJECTIVE: To study hepatic-protective effect of "Yinzhihuang" granula. METHODS: By D-GalN, CCl4 induced acute hepatic damage model, ALT and AST in serum, pathological degree of hepar adopted as indexes, the potency of "Yinzhihuang" granula was observed and compared with "Yinzhihuang" oral solutions. Mice administrated ig 1-naphthyl-isothiocyanate, with bilirubin in serum used as index, the effect of "Yinzhihuang" granula was observed. RESULTS: 1.8 g/kg, 3.6 g/kg "Yinzhihuang" granula groups could decrease the rising of ALT, AST in mice serum induced by D-GalN.3.6 g/kg "Yinzhihuang" granula group could also improve pathological degree of hepar.1.0 g/kg, 2.0 g/kg "Yinzhihuang" granula groups could decrease the rising of ALT, AST in rats serum induced by CCl(4).2.0 g/kg "Yinzhihuang" granula group could also improve pathological degree of hepar.1.8 g/kg, 3.6 g/kg "Yinzhihuang" granula groups could decrease the rising of bilirubin in mice serum induced by 1-naphthyl-isothiocyanate. CONCLUSIONS: "Yinzhihuang" granula had an evident hepatic-protective effect on acute hepatic damage induced by D-GalN or CCl4, and could decrease the rising of bilirubin in mice serum induced by 1-naphthyl-isothiocyanate.

[Study on anticonvulsive effect of rhizoma pinelliae].

Anticonvulsive effect of water-soluble extract of rhizoma Pinelliae was studied. The result showed that the extract of rhizoma Pinelliae could remarkably inhibit the convulsion induced by strychnine and obviously reduce mortality rate in mice. But the extract could be destroyed easily by heat.

[A study of the relationship among interleukin-8, cortisol and idiopathic preterm labor].

OBJECTIVE: To assess the effect of interleukin-8 and cortisol on the onset of idiopathic preterm labor (PL). METHODS: In 35 women with preterm labor and 17 controls, maternal serum and urine samples were collected. Interleukin-8 concentration was measured by enzyme-linked immunosorbant assay, serum cortisol level was measured by radioimmunoassay. RESULTS: The concentration of interleukin-8 in maternal serum and urine and cortisol in serum were significantly higher in PL group than those in the control [(0.26 +/- 0.13) microg/L vs (0.16 +/- 0.08) microg/L, (0.16 +/- 0.15) x 10(-2) g/mol Cr vs (0.04 +/- 0.02) x 10(-2) g/mol Cr, (765.83 +/- 408.55) microg/L vs (512.41 +/- 142.65) microg/L; P < 0.05, P < 0.01, P < 0.01, respectively]. A strong correlation was found ammong interleukin-8 and cortisol concentration in maternal serum in PL (r = 0.448; P < 0.05). CONCLUSIONS: There is a strong relationship between interleukin-8 and cortisol in the onset of preterm labor. Immune system and endocrine system might be involved in the mechanisms of idiopathic preterm labor.

The development, maturation, and turnover rate of mouse spleen dendritic cell populations.

Three distinct subtypes of dendritic cells (DC) are present in mouse spleen, separable as CD4(-)8alpha(-), CD4(+)8alpha(-), and CD4(-)8alpha(+) DC. We have tested whether these represent stages of development or activation within one DC lineage, or whether they represent separate DC lineages. All three DC subtypes appear relatively mature by many criteria, but all retain a capacity to phagocytose particulate material in vivo. Although further maturation or activation could be induced by bacterially derived stimuli, phagocytic capacity was retained, and no DC subtype was converted to the other. Continuous elimination of CD4(+)8(-) DC by Ab depletion had no effect on the levels of the other DC subtypes. Bromodeoxyuridine labeling experiments indicated that all three DC subtypes have a rapid turnover (half-life, 1.5-2.9 days) in the spleen, with none being the precursor of another. The three DC subtypes showed different kinetics of development from bone marrow precursors. The CD8alpha(+) spleen DC, apparently the most mature, displayed an extremely rapid turnover based on bromodeoxyuridine uptake and the fastest generation from bone marrow precursors. In conclusion, the three splenic DC subtypes behave as rapidly turning over products of three independent developmental streams.

Picometer accuracy in measuring lattice displacements across planar faults by interferometry in coherent electron diffraction

We calculate the shadow image in far field below a thin crystal when a coherent electron source is placed at micrometer distances above the specimen, and note that the presence of a planar fault results in very strong oscillatory contrast. We realize these predictions experimentally using a field-emission electron source in a microscope. With this technique, we determine displacement vectors at planar faults with an accuracy down to 1 pm in studies of the Bi2Sr2CaCu2O8 superconductor containing thin intercalated layers.
