Selection of new Epitopes from monovalent displayed phage octapeptide library.

A library of 2 x 10(7) random octapeptides was constructed by use of phagemid-based monovalent phage display system. The randomly synthesized degenerated oligodeoxyribonucleotides (oligos) were fused to the truncated g III (p230-p403). Sequence analysis of 11 randomly chosen clones suggested that the degenerated inserts and its deduced amino acid (aa) sequences are randomly distributed. The library was used to select binding peptides to the monoclonal antibody (mAb) 9E10, which recognizes a continuous decapeptide epitope of denatured human c-myc protein. After four to five rounds of panning, most of the eluted clones could bind to 9E10. Sequence analysis of the selected positive clones indicated that the binding sequences could fall into two classes, one class (clone 1) shares a consensus motif, ISE x x L, with c-myc decapeptide; and the sequences of the other class are entirely different. The binding of both classes to 9E10 could be specifically inhibited by free c-myc decapeptide. The immunogenicity of the phage peptide was further investigated by construction of multivalent displayed phage peptides and immunization of animals with or without adjuvant. ELISA and competitive ELISA showed that anti-serum from both mice and rabbit immunized with either clone could bind to the original antigen, c-myc decapeptide. These results denote that in spite of the dissimilarity of the selected peptides with c-myc decapeptide, they are capable of inducing similar immune respones in vivo, thus actually mimicking the antigen epitope.

[Study of acidogenesis of dental plaque on cariogenesis using capillary electrophoresis].

OBJECTIVE: The aim was to investigate the relationship of acid production with caries onset. METHODS: Short-chain organic acids of dental plaque from individual subjects were analyzed by capillary electrophoresis. Dental plaques from subjects of caries free and caries active, 21 for each group, were analyzed for 7 short-chain organic acids. RESULTS: The in vivo production of lactic acid was a bit higher in caries active-group than in caries free group (but no statistic significance), however, no difference in vitro. CONCLUSION: Therefore, it is hard to relate the net quantity of acid production to the caries onset.

[Effect of recombinant human growth hormone with total parenteral nutrition on albumin synthesis in patients with peritoneal sepsis].

OBJECTIVE: To investigate the effect of recombinant human growth hormone (rhGH) in combination with total parenteral nutrition (TPN) on albumin synthesis in patients with peritoneal sepsis. METHOD: 17 patients with peritoneal sepsis were divided randomly into two groups. The control group received TPN only for 7 days, and the GH group received both rhGH (12 U/d) and TPN for 7 days. The TPN scheme and other treatment were the same in the two groups. RESULT: Serum albumin, prealbumin, and transferrin concentration were increased in patients in the GH group (P < 0.01), but no apparent effect was observed in the control group (P > 0.05). CONCLUSION: In condition of serious peritoneal sepsis, TPN can not increase albumin, prealbumin, and transferrin synthesis alone, whereas rhGH in combination with TPN significantly increase the synthesis of visceral proteins.

[Effects of IL-1 and TNF-alpha on the proliferation and cytosolic Ca2+ of lens epithelial cell in vitro].

OBJECTIVE: To study the effects of interleukin 1 (IL-1) and tumor necrosis factor alpha (TNF-alpha) on the proliferation and cytosolic Ca(2+) of bovine lens epithelial cells (BLEC) in vitro. The effects of IL-1 and TNF-alpha on posterior capsular opacification (PCO) and the mechanism of effect on the proliferation were investigated. METHODS: The effects of IL-1 and TNF-alpha on the proliferation of BLEC were detected with MTT staining colorimetry, and cytosolic Ca(2+) was measured by fluorescence determination with Fura-2. RESULTS: IL-1 10(2) - 10(5) ng/ml, TNF-alpha 10(2) - 10(4) U/ml obviously could promote BLEC proliferation (P < 0.05 - 0.01), and increase cytosolic Ca(2+) (P < 0.05 - 0.01) at the same doses. CONCLUSIONS: IL-1, TNF-alpha contribute to the PCO. One of the mechanisms of IL-1 and TNF-alpha on proliferation is by cytosolic Ca(2+).

[The choice of methods of intraocular lens implantation in traumatic cataract in the absence of capsular and zonular support].

OBJECTIVE: To study the choice between the methods of anterior chamber intraocular lens (AC-IOL) implantation and posterior chamber IOL suture fixation in traumatic cataract without posterior capsular support. METHODS: The study included 86 cases (86 eyes) with traumatic cataract. A new flexible open-loop one piece PMMA IOL was implanted in the AC-IOL group of 56 cases (56 eyes); fixation of a C-loop one piece PMMA IOL at ciliary sulcus with two sutures was performed on 30 cases (30 eyes) in the posterior chamber IOL (PC-IOL) group. RESULTS: The follow-up period was 1 - 42 months. The visual acuity 0.5 or better was in 71.4% (40/56) in AC-IOL group, in 90.0% (27/30) in PC-IOL group; 1.0 or better was in 39.3% (22/56) in AC-IOL group, in 46.7% (14/30) in PC-IOL group. CONCLUSION: Both AC-IOL implantation and PC-IOL suture fixation are the remedy methods when the PC-IOL intracapsular implantation is unsuccessful. The AC-IOL is suitable for old persons with good anterior segment (cornea, iris and the angle of anterior chamber) and poor posterior segment (vitreous, retina) situations, while the PC-IOL suture fixation is suitable for the young with relatively poor anterior segment and good posterior segment conditions.

[Study on the mitoxantrone carboxymethyl starch microspheres for hepatic artery chemoembolization].

In this paper, the technology for the preparation of plain carboxymethyl starch microsphere (CMS-MS) was optimized by the uniform design method with CMS-Na as carrier material and p-phthaloyl chloride as acrosslinker. The carboxymethyl starch microsphere loaded mitoxantrone (DHAQ-CMS-MS) was prepared by absorption method. Then, its morphology, size and size distribution, characteristics of drug loading, drug release in vitro, preparation for clinical application and its stability were studied. The pharmacokinetics of DHAQ-CMS-MS in rabbit was also studied. The results showed that the average diameter of the DHAQ-CMS-MS was 75.71 microns, drug loading was 13.21%, expansion ratio in water was 71.94%. The release of DHAQ in vitro from the microspheres was found to fit the model of single exponential function. The suspension prepared in this paper is not only convenient for clinical use, but also favorable for the improvement of the drug stability. The pharmacokinetic parameters obtained from the hepatic atery chemoembolization showed that the DHAQ-CMS-MS group, when compared with the solution group, exhibited a higher blood drug concentration in hepatic vein, its MRT was 1.96 times that of the solution group. As for the result of the peripheral vein, the MRT of the DHAQ-CMS-MS group was 1.95 times that of the solution group. This means that the drug when loaded in microsphere will be concentrated in its targeted site for a longer period, which is favorable for the treatment of hepatic cancer.

[Protective and antioxidative effect of 2(3)tert-butyl-4-hydroxyanisole against cytotoxicity induced by doxorubicin in mice].

The protective and antioxidative effects of 2(3)tert-butyl-4-hydroxyanisole (BHA) against cardiotoxicity and hepatotoxicity induced by doxorubicin in mice were investigated. After pretreatment with different oral doses of BHA, doxorubicin 30 mg.kg-1 was given i.p. Serum GPT, GOT, LDH and CK were determined, and the mortality rate of animals was observed. Quinone reductase (QR), glutathione-S-transferases (GSTs) and glutathione reductase (GR) were determined on tissue cytosols with enzyme dynamic methods. Malondialdehyde (MDA) was measured by the method of thiobarbituric acid. Compared with the doxorubicin group, the serum GPT, GOT, LDH, CK and the mortality rate of mice were significantly decreased by BHA pretreatment, and BHA was shown to inhibit the increase of MDA induced by doxorubicin (P < 0.01 and P < 0.0001). Administration of BHA resulted in increased activities of QR, GSTs and GR in the myocardium and liver (P < 0.05 or P < 0.0001). These results suggest that BHA has protective effect against the toxicity induced by doxorubicin via the induction of QR, GSTs and GR activities and anti-lipid peroxidation.

[The distribution of nitric oxide synthetase in Vcx and its relation with the expression of FOS induced by teeth movement in rats].

It was studied the central role of nitric oxide(NO) during experimental teeth movement and the relation between nitric oxide synthetase (NOS) positive neurons and FOS like immunoreactivity (FLN) with the NADPH-diaphorase histochemistry and immunocytochemical reaction method. Results indicated that NOS positive neurons and FLN showed typical distribution in Vcx and there was some overlap between them. It suggests that NO is involved in the central modulation of the stimulating message of teeth movement, and which further explains the central modulation mechanism of experimental teeth movement in rats.

[Study on relationship of the expression of platelet-derived growth factor and its receptor-beta in liver tissues with liver fibrosis].

The expression of platelet-derived growth factor(PDGF)-BB and PDGF receptor-beta(PDGFR-beta) in liver tissues from viral hepatitis was detected by immunohistochemistry. The results showed that the expression of PDGF-BB and PDGFR-beta in the liver tissues correlated closely with the degree of liver fibrosis. The levels of their expression in the liver cirrhosis and fibrosis stage S3-4 of chronic viral hepatitis(CH) were much higher than those in acute viral hepatitis and fibrosis stage S0-2 of CH. Meanwhile, the expression of PDGF and its receptor was associated with the expression of desmin-positive cells pro-collagen III peptide in liver tissues and tissue inhibitor of metalloprotenase-1 in serum. It is suggested that PDGF-BB and its receptor-beta may promote the progress of liver fibrosis by means of the activation, proliferation and differentiation of satellite cells.

[Study on expression of transforming growth factor-beta 3 in the liver tissues].

33 specimens of liver biopsy from patients with different types of viral hepatitis were detected for transforming growth factor-beta 3(TGF-beta 3) by immunohistochemistry. The results showed that TGF-beta 3 mainly distributed in the sinusal walls cells in sinus portal, areas fibrous, septum, hepatocytes in pseudolobes and necrotic areas of severe hepatitis. The expressing intensity of TGF-beta 3, was stronger in the cirrhosis, chronic severe hepatitis than that in the chronic and acute hepatitis. As the expression of TGF-beta 3 increased, the level of serum bilirubin ascended and plasmozyme activity was prolonged. It may be concluded that the expression of TGF-beta 3 is correlated with severe hepatitis and the progress of the liver cirrhosis.

[Study on expression of P16 protein in the liver tissue of viral hepatitis].

P16 protein is a cell cycle regulator involved in the genesis of many types of tumors. In order to study the significance and expression of P16 protein in liver tissue of viral hepatitis, 48 patients with viral hepatitis and 7 cases with hepatocarcinoma were investigated by immunohistochemistry. The results showed that the positive rate of P16 protein expressed in the liver tissues of mild (85.0%) and moderate (91.66%) chronic viral hepatitis were significantly higher than that in liver cirrhosis and hepatocarcinoma (P < 0.01, 0.001), this suggest that the expression of P16 plays an important role in the carcinogenesis.

[Detection of hepatitis C virus-C, E, NS3 and NS4 regions' antigens in hepatocellular carcinoma and liver cirrhosis].

Using 4 McAbs to HCV-C, E, NS3 and NS4 regions' antigens and PcAb to HBsAg, 59 cases of hepatocellular carcinoma(HCC) and 35 cases of liver cirrhosis(LC) were tested by immunohistochemistry technique. Positive reactions for hepatitis C virus were mainly present in the cytoplasm of hepatocytes and tumor cells with fine granules. The positive rates of HCV were 17.2% in HCC(29 cases) of Beijing, 26.7% in HCC(30 cases) of Shenyang and 14.3% in LC(35 cases) of Shenyang. C region's McAb had the highest positive rate of detection, which suggested that C region's protein had a high level expression. The positive rates of hepatitis B virus surface antigen were 63.0% in HCC(29 cases) of Beijing, 73.3% in HCC(30 cases) of Shenyang and 54.3% in LC(35 cases) of Shenyang, all of which were higher than their positive rate for HCV detected. In HCC and LC, HBV and HCV inclined to suppress the opposite side.

[An in-vitro evaluation of the Sichuan Union Univ-I membrane lung and mathematical modeling calculation].

A new ELF hollow fibre lung, Sichuan Union Univ-I(SUU-I), constructed in our laboratory was tested in vitro to evaluate its oxygen transfer performance before it undergoes expensive animal trial. The test rig, conditions, procedure and the modelling method are introduced in the present paper. The results demonstrate that the Sichuan Union Univ-I membrane lung possesses the gas exchange ability claimed in the AAMI standards, and approaches the levels of the widely accepted membrane lungs, MAXIMA (Medtronic, US), SARNS(3M, US) and SAFE-II (Polystan, Denmark). The in vitro technique described in this paper proved practical, economical, and simple in the design and manufacturing of a new type membrane lung, as well as in research and development of membrane lungs.
