[Effects of tobacco smoke on the expression of ICAM-1 in human umbilical vein endothelial cells].

OBJECTIVE: To study the effect of tobacco smoke on the expression of intercellular adhesion molecule (ICAM)-1 in endothelial cell. METHODS: Macrophages were challenged in vitro with cigarette smog, and the supernatant was collected at 3 h, 6 h, 12 h and 24 h after tobacco treatment. The tumor necrosis factor-alpha (TNF-alpha) was measured by the ELISA method. Then the human umbilical vein endothelial cells (HUVEC) were incubated with the supernatant from the macrophages stimulated by cigarette smoke allergen. ICAM-1 in the supernatant was detected by the ELISA method. RESULTS: TNF-alpha was increased with the longer incubation time. A significant difference of TNF-alpha(t = -4.301, P < 0.01) was observed between the tobacco treated group (4.967 +/- 0.802 pg/ml) and the control group (3.181 +/- 0.214 pg/ml). Soluble ICAM-1 in HUVEC in the tobacco treated group (mean 0.068 4 ng/ml) increased 49% than the controls (mean 0.045 9 ng/ml). CONCLUSION: The results suggested that cigarette smog enhances the expression of TNF-alpha in the antigen-presenting cell(APC)-macrophage, and the latter up-regulates the secretion of the membrane glycoprotein sICAM-1 in HUVEC which can assist leukocyte immigration.

[Modification of the Photobacterium phosphoreum toxicity test method].

In order to reduce the variation of Photobacterium phosphoreum luminous intensity during testing, the Photobacterium phosphoreum toxicity test was modified by a correction factor, PIacetone, in this paper. The effects of magnetic stirring time, culture time, culture generation and culture temperature on the measured results were discussed. The modified method had good laboratory repeatability and the relative standard deviation (RSD) was between 2.1% and 13.1%. Reliable data could be obtained using the modified method. It would be beneficial to the development of the quantitative structure-activity relationships on multicomponent organic compounds.

[Atmospheric particulate mercury concentration in Changchun City and its dry deposition flux].

From July 1999 to January 2000, the particulate mercury(Hg(p)) concentration of five function sites and one contrast site were determined. In non-heating time, the particulate mercury concentration in urban atmosphere ranged from 0.022 to 0.398 ng.m-3 with an average of 0.145 ng.m-3 and the contrast site with an average of 0.084 ng.m-3. In heating time, the Hg(p) concentration in urban was in the range of 0.148-1.984 ng.m-3 with an average of 0.461 ng.m-3 and with an average of 0.211 ng.m-3 in the contrast site. The Hg(p) concentration in heating time was two times higher than that of non-heating time. Coal fire and wind-blown soil material were the two important sources of Hg(p). Wind-blown soil material contributed 7.9%-38.5% of the Hg(p) with an average of 24.1% in urban. Use theory model to estimate the dry deposition flux with the results of 43.06 micrograms.(m2.a)-1 in urban and 21.28 micrograms.(m2.a)-1 in the contrast site.

Proinflammatory cytokine expression contributes to brain injury provoked by chronic monocyte activation.

BACKGROUND: We have proposed that an increased interaction between monocyte/macrophages and blood vessel endothelium predisposes subjects to strokes. The effect of chronic monocyte activation on the development of cerebral infarcts was thus studied in rats after provocation of a modified local Swartzman reaction, in brain vasculature. MATERIALS AND METHODS: Two weeks after an IV bolus of bacillus Calmette-Guérin (BCG), we studied spontaneous superoxide production, integrin expression, endothelial adhesion of monocytes and the neurological symptoms, brain histology, and cytokine immunoreactivity after a provocative dose of LPS (30-300 microg/rat i.c.v.). RESULTS: Monocyte migration into the brain was stimulated by BCG priming. The incidence of paralysis and death in response to LPS was markedly increased in BCG-primed rats. Histological evaluation of the brains of neurologically impaired and moribund animals revealed intravascular thrombosis and pale and hemorrhagic infarcts. Infiltrates of leukocytes expressing immunoreactive IL-1:, IL-6, and TNF-alpha were found around blood vessels, cerebral ventricles, and meninges, and were accompanied by a profound microglial expression of IL1P, endothelial expression of IL-6, and expression of TNF-alpha and TNF-R 1 in glia and neurons of cortex and hippocampus. Treatment (2 x 100 microg/10 ,I, i.c.v.) with recombinant human (rh-)TNF 55kDa receptor completely prevented, and treatment with rh-IL- I receptor antagonist significantly decreased the incidence of paralysis and death in response to BCG + LPS. The improvement of neurological symptoms was accompanied by reduced histological damage and supppression of IL-1P/ expression in the brain tissue. CONCLUSIONS: The data demonstrate that chronic monocyte activation predisposes subjects to thrombosis and hemorrhage via an exaggerated release of proinflammatory cytokines.

[Relationship between methylenetetrahydrofolate reductase gene polymorphism and diabetic nephropathy].

OBJECTIVE: To investigate the relationship between methylenetetrahydrofolate reductase(MTHFR) gene polymorphism and diabetic nephropathy(DN). METHODS: The technology of polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used to detect MTHFR gene C677T mutation in 85 healthy controls, 79 type-2 diabetics without DN, and 82 type-2 diabetics with DN confirmed by urine microalbuminous detection. RESULTS: The frequencies of MTHFR gene variant genotypes and alleles in the patients with DN patients were significantly higher than those in the patients without DN and those in healthy controls (P<0.01). CONCLUSION: MTHFR gene C677T mutation is probably one of the genetic risk factors for diabetic nephropathy in Chinese Hans.

[Association between angiotensin system gene polymorphism and essential hypertension].

OBJECTIVE: To investigate whether angiotensinogen(AGT) M235T, angiotensin II type I receptor(AT(1)R) gene A1166C and angiotensin converting enzyme(ACE) gene I/D polymorphism are implicated in human essential hypertension(HT) in Chinese. METHODS: Polymerase chain reaction(PCR) and PCR combined with restriction enzyme digestion were used to detect AGT gene M235T, AT(1)R gene A1166C variations and ACE gene I/D polymorphism in 161 hypertensive patients and 134 normotensive controls. RESULTS: No statistically significant differences were found in frequencies of the ACE D allele, AGT gene 235T and AT(1)R A1166C between hypertensive patients and normotensive controls, but in hypertensive patients aged <60 years the frequencies of the ACE D allele and AGT gene 235T were significantly higher than those of the normotensive controls (P<0.05). The analysis of combined genotypes of AGT gene and ACE gene showed that the combined genotypes of DD-TT and ID-TT were significantly higher in hypertensive patients than in normotensive controls. Significant relationships between the ACE genotype and serum ACE activity were found in both groups(P<0.05). CONCLUSION: The ACE D allele and AGT 235T polymorphism may be involved in the early occurrence of HT. The combined genotypes of DD-TT and ID-TT may be a dangerous genetic factor for HT in Chinese.

hMLH1 and hMSH2 expression in human hepatocellular carcinoma.

The role of microsatellite instability (MSI) in the pathogenesis of hepatocellular carcinoma (HCC) is incompletely defined. Although high-frequency MSI (MSI-H) is infrequently seen in HCC, some studies have suggested a role for MSI in HCC development. While MSI has been clearly defined for a subset of tumors, in particular colorectal, gastric and endometrial cancers, generally accepted criteria have not been developed for other tumors. Colorectal cancers (CRC) are classified as MSI-H if >30-40% of >5 microsatellite loci analyzed show instability. The MSI-H phenotype is associated with defective DNA mismatch repair (MMR) and is observed in the majority of tumors from patients with hereditary non-polyposis colon cancer (HNPCC) and also in 15% of sporadic CRCs. Inactivating mutations of the hMLH1 or hMSH2 genes lead to defects in MMR in HNPCC. In sporadic CRCs, MMR is usually due to hypermethylation of the hMLH-1 promoter. The role of defective MMR in hepatocellular carcinogenesis is controversial. Immunohistochemistry for hMLH1 and hMSH2 reliably indicates hMLH1 or hMSH2 loss in MSI-H CRC tumors. To investigate the role of defective MMR in HCC carcinogenesis, we performed immunohistochemistry for hMLH1 and hMSH2 on 36 HCCs. BAT26, a microsatellite marker that reliably predicts MSI-H was also examined. All 36 of the tumors stained positively for both hMLH1 and hMSH2, strongly suggesting an absence of either inactivating mutations of hMLH1 and hMSH2 or promoter hypermethylation of hMLH1. None of the tumors showed MSI at the BAT26 locus. These findings suggest that defective MMR does not contribute significantly to hepatocellular carcinogenesis.

Characterization and mechanisms of chromosomal alterations induced by benzene in mice and humans.

Elevated frequencies of chromosomal aberrations have been observed in the lymphocytes of benzene-exposed workers. Similar changes occurring in the bone marrow may play an important role in the development of leukemia. The objective of this research has been to characterize chromosomal alterations induced by benzene in mice and humans and to investigate the potential role of inhibition of topoisomerase II in the myelotoxic effects of benzene. The research is presented in three sections corresponding to the specific aims of the project: genotoxicity studies in the mouse, topoisomerase II studies, and initial studies using a new fluorescence in situ hybridization (FISH) approach to detect chromosome alterations in benzene-exposed workers. The results of the mouse experiments indicate that both chromosome breakage and aneuploidy are induced in the bone marrow of B6C3F1 mice following benzene administration. Chromosome breakage is the predominant effect, and this occurs primarily in the mouse euchromatin. Significant breakage within the mouse heterochromatin was also observed, as was aneuploidy. Breakage in the mouse bone marrow erythrocytes increased as a function of both dose and duration of benzene administration. The aneuploidy resulting from benzene exposure in mice was a relatively infrequent event, with increases of both chromosome loss and hyperdiploidy being observed. In the topoisomerase studies, benzene or its metabolites were shown to inhibit topoisomerase II enzyme activity in an isolated enzyme system, in a human bone marrow-derived leukemia cell line, and in vivo in the bone marrow of treated mice. The decreased activity was probably due to the rapid degradation of the topoisomerase II protein in the treated cells. In the human biomonitoring studies, the feasibility of using FISH with tandem DNA probes to detect chromosome alterations in interphase granulocytes and lymphocytes of benzene-exposed workers was demonstrated. The results from the two worker studies were somewhat inconsistent, however. In the study of Estonian workers, characterized by lower exposures and a smaller sample size, the benzene-exposed workers exhibited elevated frequencies of breakage in the lq12 region as compared with those seen in controls. A suggestive trend toward increased hyperdiploidy was also seen, although the frequencies in the exposed workers were low and within the range of our laboratory's historical control frequencies. In the larger study of more highly exposed Chinese workers, no increase in breakage affecting the 1q12 region was seen among the exposed workers. A trend toward increased hyperdiploidy of chromosome 1 was seen in the exposed workers when the concentration of urinary benzene metabolites was used in conjunction with the frequency of hyperdiploidy observed in the lymphocytes of the individual workers. The results of these studies indicate that benzene exposure is characterized by chromosome breakage, primarily within the euchromatin, and modest increases in aneuploidy. These findings also provide the first direct evidence that benzene is capable of inhibiting the enzymatic activity of topoisomerase II in vivo, providing additional support for the hypothesis that inhibition of topoisomerase II contributes to benzene-induced toxicity and leukemogenesis.

Suppression of polyclonal immunoglobulin production by M-proteins shows isotype specificity.

Monoclonal gammopathies are B cell neoplasms that are characterized by the presence of monoclonal immunoglobulins (M-proteins) in the serum. By an unknown mechanism, the normal polyclonal immunoglobulin levels are frequently reduced in sera of these patients. To assess the role of M-protein isotype in this effect, we used serum protein electrophoresis to quantitate monoclonal and polyclonal immunoglobulins in patients and we used serum immunofixation electrophoresis to determine their M-protein isotype. When divided into populations of 30 patients with IgG M-proteins (mean 2.5 g/dl) and 19 patients with IgM or IgA M-proteins (mean 2.6 g/dl), the mean polyclonal immunoglobulin level was significantly lower in the IgG M-protein population (0.4 g/dl) than the IgM/IgA population (0.8 g/dl). Patients with IgG M-proteins also had significantly lower polyclonal immunoglobulin levels when compared separately with the patients with either IgA or IgM paraproteins. Since the polyclonal immunoglobulin fraction is comprised mostly of IgG, these results give the first direct indication that IgG M-proteins have a greater suppressive effect on polyclonal IgG levels than do M-proteins of other isotypes. These findings suggest that an isotype-specific feedback mechanism could be involved in the normal regulation of serum IgG levels.

[Clinical and pathological study of autoimmune hepatopathy].

OBJECTIVE: To study the clinical and histologic characteristics of autoimmune hepatopathy, and to explore the diagnostic criteria and therapy. METHODS: Sixty-two patients with autoimmune hepatopathy were studied between 1996 and 1998 to compare primary biliary cirrhosis (PBC) and autoimmune hepatitis (AIH) in the clinical symptoms, signs, laboratory findings and histologic features. RESULTS: PBC was 59.68% and AIH 40.32% in the autoimmune hepatopathy. The morbidity in middle and old aged female patients with autoimmune hepatopathy was 83.8%. The ratio of male and female was 1:10. Whereas, the ratio of male and female was 1:5 in AIH patients and average age was 35 years old. Seropositive autoimmune antibodies were detected in 53 serum sample (PBC 85.29%; AIH 78.95%). Positive rates of the mitochondrial antibody and subtype-M2 were 100% for PBC. IgM increasing was 80% in PBC (P<0.05) and 82.35% in AIH (P<0.025). Histologic features in PBC group consisted of cholangitis, destruction or proliferation of the bile duct and inflammation within the portal tract; while in AIH group inflammation, necrosis and fibrosis of periportal region, the aggregation of lymphocytes, mononuclear cells and plasma cells in the portal tract or periportal area were found. CONCLUSIONS: PBC and AIH are the most common two types in the patients with autoimmune hepatopathy. AIH-I is more often seen clinically. AMA-M2 is the special autoimmune antibody of PBC. The efficacy of prednisolone is better for AIH than PBC.

Do P1 and N1 evoked by the ERP task reflect primary visual processing in Parkinson's disease?

OBJECTIVES: To evaluate whether P1 and N1 evoked by ERP tasks could appropriately reflect primary visual processing in Parkinson's disease (PD). METHODS: We recorded ERPs in 13 PD patients with duration of illness less than 5 years and 18 age-matched normal control subjects. P1 and N1 from Oz were evoked by a visual oddball and a delayed matching S1-S2 task. The effect of different events on P1 and N1 was studied. All patients were given an ECD-SPECT examination, and the SPECT images were overlaid on the 3D-MRI. The correlation of P1 or N1 to the regional cerebral blood flow (rCBF) was studied. RESULTS: P1 was not influenced by different events. There was no significant P1 differences between the PD and the normal group. N1 was significantly shorter and smaller in the patients than that in the normal group. N1 amplitude after the waveform subtraction (target-frequent) in the PD group did not show significant difference with that in the normal controls, nor with the N1 before the subtraction. Nd, the subcomponent of N1 after the subtraction in the patients was significantly earlier and smaller than that in the normal controls. P1 only weakly correlated with the rCBF in the occipital lobe. N1 was correlated with the rCBF in a global region. CONCLUSIONS: The results provided some evidence that P1 might reflect the primary visual processing, and N1 might be involved in both primary and cognitive visual processing. The altered N1 in the PD patients might be due to the deformed Nd.

The study of levels of norepinephrine and dopamine-beta-hydroxylase in patients with pregnancy-induced hypertension.

BACKGROUND: The present work aimed to study the changes of both norepinephrine (NE) and dopamine-beta-hydroxylase (DbetaH) in pregnancy-induced hypertension (PIH). METHODS: NE levels and DbetaH activity in blood were determined in 116 patients with pregnancy-induced hypertension and 40 normal control subjects using high performance liquid chromatography (HPLC) and ultraviolet (UV) spectrophotometry. Among patients with PIH, 28 were graded as mild, 34 moderate and 54 severe. The normal control subjects were selected from patients at a normal term pregnancy receiving elective cesarean section. RESULTS: NE levels and DbetaH activity in blood were significantly higher in patients with severe PIH than those in control subjects (p < 0.05 for both). Both patients and control subjects had higher NE levels and DbetaH activity in the umbilical artery blood than in the umbilical vein blood (p < 0.05). NE levels and DbetaH activity in both umbilical artery and vein blood were higher in the moderate and severe PIH groups than in the mild (p < 0.05) and control (p < 0.05) groups. NE levels in the umbilical artery blood were five times higher than those in the maternal blood, and DbetaH activity was three times higher. NE levels and DbetaH activity in the umbilical artery blood were significantly higher in the PIH group with complicated intrauterine growth retardation (IUGR) or fetal distress than in the PIH group without such complication. CONCLUSIONS: The pathogenesis of PIH may relate to increase of excitability of the sympathetic nervous system in fetus.

Interactions of the antimalarial drug mefloquine with the human cardiac potassium channels KvLQT1/minK and HERG.

Mefloquine is a quinoline antimalarial drug that is structurally related to the antiarrhythmic agent quinidine. Mefloquine is widely used in both the treatment and prophylaxis of Plasmodium falciparum malaria. Mefloquine can prolong cardiac repolarization, especially when coadministered with halofantrine, an antagonist of the human ether-a-go-go-related gene (HERG) cardiac K+ channel. For these reasons we examined the effects of mefloquine on the slow delayed rectifier K+ channel (KvQT1/minK) and HERG, the K+ channels that underlie the slow (I(Ks)) and rapid (I(Kr)) components of repolarization in the human myocardium, respectively. Using patch-clamp electrophysiology we found that mefloquine inhibited KvLQT1/minK channel currents with an IC50 value of approximately 1 microM. Mefloquine slowed the activation rate of KvLQT1/minK and more block was evident at lower membrane potentials compared with higher ones. When channels were held in the closed state during drug application, block was immediate and complete with the first depolarizing step. HERG channel currents were about 6-fold less sensitive to block by mefloquine (IC50 = 5.6 microM). Block of HERG displayed a positive voltage dependence with maximal inhibition obtained at more depolarized potentials. In contrast to structurally related drugs such as quinidine, mefloquine is a more effective antagonist of KvLQT1/minK compared with HERG. Block of KvLQT1/minK by mefloquine may involve an interaction with the closed state of the channel. Inhibition by mefloquine of KvLQT1/minK in the human heart may in part explain the synergistic prolongation of QT interval observed when this drug is coadministered with the HERG antagonist halofantrine.

[Aluminum toxicity and tolerance in wheat].

The physiological and biochemical effects of different species of aluminum on the wheat Al-tolerant TAM202 and Al-sensitive TAM105 were investigated. The results showed that mononuclear Al could be absorbed by both of the two strains. Compared with TAM105, however, TAM202 absorbed less aluminum. The pH of the nutrient solution rose with the hydroponics of the two-cultivars. The content of proline in leaves of TAM105 increased obviously. The contents of protein, hexose and uronic acid in cell wall of TAM202 were higher than that in TAM105 under 75 mumol.L-1 Al. The results indicated that Al can activated the synthesis of some components relative to Al-tolerance in cell wall.

Effect of Cr(VI) exposure on sperm quality: human and animal studies.

The semen status of male workers occupationally exposed to hexavalent chromium(VI) was investigated. Sperm counts from exposed workers were 47.05+/-2.13 x 10(6)/ml and those from control group 88.96+/-3.40 x 10(6)/ml. Sperm motility decreased from 81.92+/-0.41% for the control group to 69.71+/-0.93% for the exposed workers. The levels of zinc, lactate dehydrogenase (LDH), and lactate dehydrogenase C4 isoenzyme (LDH-x) in seminal plasma for the exposed workers were 1.48+/-0.07 micromol/ml, 1.05+/-0.02 x 10(3) U, and 0.47+/-0.01 x 10(3) U, respectively, which were significantly lower than those of 5.72+/-0.15 micromol/ml, 1.49+/-0.02 x 10(3) U, and 0.78+/-0.15 x 10(3) U for the control group, respectively. Follicle stimulating hormone (FSH) (7.34+/-0.34 x 10(-3) IU/ml) in serum from the exposed workers was significantly higher than that (2.41+/-0.08 x 10(-3) IU/ml) from the control group. On the other hand, there were no significant differences in semen volume, semen liquefaction time, luteinizing hormone (LH) level in serum, and Cr concentration in both serum and seminal plasma between the exposed workers and the control group. Feeding Cr(VI) to rats significantly reduced the epididymal sperm counts from 87.40+/-3.85 x 10(6)/g epididymis in control group to 21.40+/-1.20 x 10(6)/g epididymis at a CrO(3) dose of 10 mg/kg body weight and to 17.48+/-1.04 x 10(6)/g epididymis at a CrO(3) dose of 20 mg/kg body weight. Exposure of rats to Cr(VI) also significantly increased the sperm abnormality from 2.75+/-0.06% in the control group to 6.68+/-0.32% in the exposed group at a CrO(3) dose of 10 mg/kg body and to 7.6+/-0.15% at a CrO(3) dose of 20 mg/kg body weight. In exposed rats, there was visible disruption in germ cell arrangement near the walls of the seminiferous tubules. The diameters of seminiferous tubules in exposed rats were smaller. These results suggest that occupational exposure to chromium(VI) leads to alteration of semen status and may affect the reproductive success of exposed workers.
