Characterization of the HLA-A2.2 subtype: T cell evidence for further heterogeneity.

Five blood donors were identified whose HLA-A2 is different from the common HLA-A2. Their A2 molecule (A2.2) had a more basic isoelectric point than normal A2 (A2.1). Cytotoxic T lymphocytes (CTL) restricted by HLA-A2.1, specific for influenza A and Epstein-Barr viruses, failed to lyse virus-infected target cells with HLA-A2.2. Identical patterns were obtained with both viruses. CTL from four of the A2.2-positive donors recognized target cells prepared from others in the group that shared only the HLA-A2.2 antigen. The A2.2 antigen from one donor seemed to be different in that target cells were not recognized by CTL from donors with the normal A2.1 nor with basic A2.2. There seems, therefore, to be heterogeneity within the HLA-A2.2 subtype.

Delineation of functional sites in HLA-B27 antigens. Molecular analysis of HLA-B27 variant Wewak I defined by cytolytic T lymphocytes.

An HLA-B27 positive, Epstein Barr virus-transformed cell line Wewak I was not lysed by Epstein Barr virus-specific cytolytic T lymphocytes restricted by HLA-B27. This line is weakly reactive with a B27-specific monoclonal antibody, M2, which recognizes a majority, but not all, of the HLA-B27-positive cells. To establish the molecular basis for this lack of recognition, the structure of the variant HLA-B27 antigen was compared with the known structure of HLA-B27 from the LG-2 cell line, which is representative of a major subtype of this antigen. Both molecules were almost indistinguishable by isoelectric focusing. However, comparative peptide mapping and sequencing of the difference peptides revealed two amino acid changes: At position 77, Asp in donor LG-2 had changed to Ser in the variant, and at position 152, Val in LG-2 had changed to Glu in the variant. The nature of these substitutions was consistent with the extreme similarity of the isoelectric focusing pattern. An evaluation of these findings in the context of studies on other HLA variants and H-2Kb mutants suggests that both positions 77 and 152 contribute to the determinants recognized by B27-specific cytolytic T lymphocytes. The change at position 152 adds to previous evidence suggesting that the segment 149-156 is critical for cellular recognition. In addition, it is proposed on the basis of structural comparisons that residue 77 may also participate in the epitope recognized by the B27M2 antibody.

Atrial septal defect (sinus venosus type) in a dog.

A 2-year-old Old English Sheepdog had right-side congestive heart failure characterized by pericardial effusion, pleural effusion, ascites, and increased pulmonary wedge pressure. A diagnosis of atrial septal defect was made by means of cardiac catheterization and angiography. Surgical correction initially was deferred because of pulmonary hypertension. However, when congestive heart failure could not be managed adequately, surgical correction was attempted. Postsurgically, thrombocytopenia developed and that led to bleeding, oliguira, and pulmonary edema. The bleeding was controlled by whole blood transfusion, but the oliguria and pulmonary edema were not resolved, even with intensive diuretic therapy. The dog died 24 hours after surgery.

Perceptual accuracy as a variable in marital adjustment.

This study investigated the ability of maritally-adjusted couples, couples attaining a divorce, and couples in counseling to predict the rewarding effects of their behavior on their spouse. Twenty-six couples, five obtaining a divorce, nine in counseling, and twelve adjusted couples completed a Marital Interaction Questionnaire in which they rated how rewarding each of the behaviors depicted was to them. Then each member predicted how rewarding it was to their spouse. Behaviors depicting six areas of marital interaction were included in the questionnaire. The computed perceptual accuracy scores were analyzed by a three-way classification-Analysis of Variance (ANOVA)-for unequal n's. This analysis yielded a significant main effect of marital status and a significant interaction between marital status by sex of spouse and between marital status by area of interaction. The maritally-adjusted group was always more accurate than the other two groups in their predictions, however, they were significantly more accurate only for selected areas of interaction.

Glucose-6-phosphate dehydrogenase deficiency in a dog.

After screening 3,300 dogs, one animal with a mild deficiency of erythrocyte G6PD was detected. Although it had several clinical problems for 2 months, no abnormality could be directly attributable to the reduced enzymatic activity. Biochemically the mutant was electrophoretically slower but within the normal range for Km-G6P, Km-NADP, use of 2-dG6P and deamino NADP, pH optimum, and heat stability.
