The activity of 7S immunoglobulins (Ig) antibody to RNA, obtained in rabbits after a prolonged immunization with RNA-methylated bovine serum albumin complex was evaluated in vitro on normal (3T3) and simian virus 40-transformed (SV 3T3) mouse fibroblasts. The presence of anti-RNA antibody in the culture medium inhibited both the SV 3T3 cell proliferation and the [3H]thymidine incorporation. In contrast, an increased [3H]uridine incorporation was evident within 48 and 96 hr of culture. No significant modification in these 3 parameters was observed in 3T3 cultures treated in the same manner. Both 3T3 and SV 3T3 showed cytoplasmic fluorescence when cultured in the presence of fluoresceinated anti-RNA Ig. However, with the indirect fluorescence technique anti-RNA Ig were detected in SV 3T3 cytoplasm only. These data suggest that anti-RNA Ig were taken up by both 3T3 and SV 3T3, but only in SV 3T3 did the anti-RNA Ig retain their antigenic properties and block cellular proliferation.

Myeloid precursors can be grown in vitro in the presence of specific growth factors; however, their expansion is limited by a competing process of terminal differentiation. Proto-oncogenes seem to be involved in cellular proliferation and/or differentiation and may also play a role in the myelopoietic process. Murine myeloid precursors which are grown in vitro with growth factors respond with augmented self-renewal upon infection with recombinant retroviruses carrying the v-myc or v-src oncogenes, suggesting a synergism or complementation between some viral oncogenes (v-onc) and certain growth factors. We now show that the combination of two v-onc genes (raf and myc) induces the selective proliferation of monocytic cells from fresh murine bone marrow (BM) in the absence of a specific growth factor supplement. Depending on the culture conditions these cells can either differentiate and cease to proliferate or grow continuously, thus mimicking the alternative pathways that can be followed by committed BM stem cells in vivo.

Oral vaccination of rabbits with killed polivalent E. coli vaccines (enteropathogenic strains) induced the production of circulating and fecal antibodies to the large majority of serotypes present in the vaccines. The production of specific IgA immunoglobulins in the intestine was also detected. The titres were in general of the same height as those obtained when individual vaccines were administered singularly.
The induction of a state of immune tolerance is a complex event which can take place at different stages of the immune response since it depends both upon the chemical structure and the experimental manipulation of the antigen. This paper deals mainly with the relationship between antigen structure and the kinds of tolerance induced.
