Expression of dominant-negative Ikaros isoforms in T-cell acute lymphoblastic leukemia.

Ikaros, a zinc finger-containing DNA-binding protein, is required for normal lymphocyte development. Germ-line mutant mice that express only non-DNA binding dominant-negative "leukemogenic" Ikaros isoforms lacking critical NH2-terminal zinc fingers develop an aggressive form of T-cell leukemia. We studied Ikaros gene expression in leukemic cells from 18 children with T-cell acute lymphoblastic leukemia (T-ALL). In each of the 18 T-ALL cases as well as JK-E6-1 and MOLT-3 cell lines, we found high-level expression of dominant-negative isoforms of Ikaros with abnormal subcellular compartmentalization patterns. Nuclear extracts from these cells failed to bind to the IKAROS-specific binding sequence in DNA. PCR cloning and sequencing confirmed that JK-E6-1 and MOLT-3 cell lines as well as leukemic cells from 9 of 10 patients with T-ALL expressed dominant-negative Ikaros isoforms Ik-4, Ik-7, and Ik-8 that lack critical NH2-terminal zinc fingers. In 6 of 10 patients, we detected a specific mutation leading to an in-frame deletion of 10 amino acids (delta KSSMPQKFLG) upstream to the transcription activation domain and adjacent to the COOH-terminal zinc fingers of Ik-2, Ik-4, Ik-7, and Ik-8. Thus, children with T-ALL express high levels of dysfunctional dominant-negative Ikaros isoforms.

[Genetic complementation group analysis of xeroderma pigmentosum patients in China].

OBJECTIVE: To establish skin fibroblast cell strains derived from Chinese xeroderma pigmentosum(XP) patients and to analyze the genetic complementation group and distribution. METHODS: From four XP patients, skin fibroblast cell strains were derived and used for the complementation group analysis by autoradiography and cell-fusion technique. RESULTS: Of the four XP patients from apparently independent families, three were assigned to group C, and one belonged to group E. This is the first report on XP-E patient in China. CONCLUSION: Based on the data from the previous 7 cases and the present 4 cases, the distribution of complementation group in Chinese XP patients known at present is: nine XP-C, one XP-F and one XP-E. Although the number of cases is still limited, XP-C appears to be more frequent in China. This forms a marked contrast to Japanese XP patients, who are dominated by XP-A with infrequent presence of XP-C.

[Evaluation on the establishment and operation on neonatal tetanus surveillance program in Hainan Province].

OBJECTIVE: In order to learn the actual morbidity and influencing factors of neonatal tetanus(NNT) and to provide scientific basis for NNT elimination. METHODS: A surveillance program was set up in 1997 in Hainan Province and to operate at the same time. RESULTS: 216 NNT cases were investigated in 1997. The number was higher than reported 78 cases. The result from surveillance showed that parturition at home and the old delivering method were the main influencing factors 88.89% of mothers of NNT cases had no TT immunization history. The surveillance data of Sanya City and Qiongzhong County indicated the NNT morbidity in 1997 had declined 61.29%, 82.05% than that in 1996 respectively due to the improvement of TT immunization rate. CONCLUSION: The study result proved that the most economical and effective measure to eliminate NNT is to increase TT immunization rate of women at child-bearing-age. It was practical integrate the NNT surveillance program into AFP surveillance system and operate along with it.

[Effects of mifepristone on choriocarcinoma cell line JAR in vitro].

OBJECTIVE: To investigate the effects of mifepristone on cell proliferation and hCG secretion of human choriocarcinoma cell line JAR and the cytotoxic effects of mifepristone on this cell line. METHODS: The cytotoxic effects of four different concentration of mifepristone (1,5,10,20 mumol/L) were assessed with methyl thiazolyl tetrazolium (MTT) colorimetric assay. The expression of nuclear proliferative antigen Ki-67 and the amount of hCG secreation in JAR cells were detected with avidin-biotin complex method, immuno-histochemical method and enzyme-linked immunosorbent assay (ELISA) respectively. RESULTS: There were dose-dependent cytotoxic effects on JAR cells in four different concentrations of mifepristone. This effect plateaued at 20 mumol/L. When the JAR cells were exposed to 5 mumol/L mifepristone for 72 h, the expression of Ki-67 antigen was declined and the amount of hCG secretion was decreased significantly at the third day. CONCLUSION: In vitro, mifepristone has cytostatic and cytotoxic effects on JAR cell line and may inhibit the secretion of hCG in JAR cells.

[Orbital fat preservation and orbicularis muscle flap suspension for lower eyelid pouches].

OBJECTIVE: This is to introduce the experience in the treatment of lower eyelid pouches by orbital fat preservation and orbicularis oculi muscle flap suspension techniques. METHODS: From Sept. 1996 to May 1997, thirty patients with lower eyelid pouches were treated using Hamra's procedure. During the operation, the arcus marginalis was incised. The orbital fat was released and advanced beyond the infraorbital rim and sutured. A lateral-based orbicularis oculi muscle flap was created, and its pedicle was repositioned to the periosteum of the upper lateral orbital rim in a superiomedial direction. RESULTS: A total of 26 patients were available for postoperative follow-up for 3 to 8 months (mean 5.6 months). Of them one patient developed unilateral lower eyelid ectropion due to flabby reposition of the orbicularis oculi muscle flap. The rest obtained excellent cosmetic results without complications. CONCLUSION: This new method has following advantages: 1. The lower eyelid pouches and the infraorbital rim show that may be corrected simultaneously: 2. Sunken eyelids and scleral show that may follow fat removal in conventional lower blepharoplasty can be effectively avoided. 3. "Cheek lift" and a youthful eyelid-cheek complex contour can be obtained.

Surgical treatment of DeBakey type I aortic dissection using the "elephant trunk technique".

OBJECTIVES: To review the indications, operative methods and postoperative management of the "elephant trunk technique", and to report two cases of DeBakey type I aortic dissection treated with the "elephant trunk technique". METHODS: Two cases of DeBakey type I aortic dissection were operated with selective cerebral perfusion via the right subclavian artery. At the first stage, a tubular dangling aortic graft prosthesis ("elephant trunk") was inserted into the distal aorta while replacing the ascending aorta and aortic arch. The distal elephant trunk prosthesis was then used at the second stage involving the replacement of the sections of the distal aorta via a left-sided thoracotomy. RESULTS: The two operations were successful. Ultrafast computed tomograph (UFCT) showed that the two patients were cured after the first stage operation, and the second stage procedure could have been avoided. CONCLUSIONS: The "elephant trunk technique" is a multiple stage approach in the treatment of extensive aneurysmal diseases of the aorta. The procedure is indicated for patients who have combined diseases of both ascending aorta plus aortic arch segments and descending aortic aneurysm. It can also be used for patients with DeBakey type I aortic dissection. Some patients can be cured after the first stage operation.

[A clinical retrospective review of stage IB1 and IB2 cervical cancer].

OBJECTIVE: To investigate the prognostic factors and treatment of cervical cancer in stage IB1 and IB2. METHODS: Three hundred fifty-six patients with stage IB cervical cancer were treated with redical hysterectomy. Seventy two patients received preoperative radiation (IB1 n = 39, IB2 n = 33), 83 patients received postoperative radiation (IB1 n = 61, IB2 n = 22), 11 patients received radiation before and after operation (IB1 n = 5, IB2 n = 6). RESULTS: Two hundred forty nine patients had been followed up for more than 5 years. The overall 5-year survival rate was significantly higher in patients in stage IB1 than in stage IB2. In patients with deep tumor invasion of the cervical matrix, the difference in 5-year survival was also significantly different between patients in stage IB1 and IB2. However, no significant difference was observed between the 2 groups of patients with positive nodes, nor was it related to tumor cell differentiation. Patients treated with surgery alone had similar 5-year survival rates but it was significantly higher in stage IB1 than in stage IB2 patients who received preoperative radiotherapy. Post-operative radiotherapy did not help survival in both groups of patients. CONCLUSION: The survival rate of cervical cancer in stage IB1 significantly differs from that in stage IB2. However, postoperative radiotherapy does not help improve survival of stage IB1 and IB2 patients with high risk factors.

[Expression and characteristics of laryngeal carcinoma-associated antigen].

OBJECTIVE: To study the characteristics of laryngeal carcinoma-associated antigen (LCAA) and its expression on laryngeal carcinoma tissue. METHODS: The binding of a cocktail monoclonal antibodies Lc9, Lc11, Lc12 to human laryngeal squamous-cell carcinoma, laryngeal precancerous lesions and normal laryngeal mucosa was examined by immunohistochemical method. LCAA isolated and purified by gel-filtration, and chromotography from 3 laryngeal squamous-cell carcinomas. The reactivity of the purified antigen was determined after treatment with trypsin, NaIO4, methanol and heating. RESULTS: The positive rate of LCAA expression in 90 cases of laryngeal carcinoma, 14 cases of precancerous lesions and in 10 cases of normal laryngeal mucosa was 97.7%, 50.0% and 0, respectively. LCAA was a glycoprotein, moderately heat stable. The purified antigen gave two bands of apparent molecular weights of 61,400 and 56,500 on SDS-PAGE and Western blot. CONCLUSION: The results provide a basis for immunoimaging diagnosis and targeting chemotherapy for laryngeal cancer.

[Experimental study of yishou tiaozhi tablet on free radical metabolism in aging model mice].

OBJECTIVE: To observe the effect of Yishou Tiaozhi tablet (YSTZT) on the metabolism of free radical in mice of aging model. METHODS: Using the aging model induced by hypodermic injection of 5% D-Galactose in mice for forty days. The content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) in serum, activity of monoamine oxidase B (MAO-B), Na(+)-K(+)-ATPase in brain were investigated after the treatment. RESULTS: In comparing to the aging model group, it showed that YSTZT could obviously inhibit the increase of level of MDA in serum and the activities of MAO-B in brain; and enhance the activities of SOD, GSH-Px in serum, and the activities of Na(+)-K(+)-ATPase in brain. Furthermore, YSTZT could obviously elevate the tolerance to fatigue and hypoxia in mice. CONCLUSIONS: YSTZT can effectively improve the free radical metabolism of senile body, it is an ideal and effective medicine in preventing aging, which is also one of the experimental basis in proving the theory of "Spleen and Kidney deficiency complicated with stasis and phlegm cause aging".

[Influence of Helicobacter pylori lipopolysaccharide on the function of enterochromaffin-like cells in vitro].

OBJECTIVE: To investigate the relationship between Helicobacter pylori (Hp) infection and pathophysiology of abnormal gastric acid secretion and mucosal proliferation by studying the effect of Hp lipopolysaccharide (LPS) on the function of enterochromaffin-like (ECL) cells in vitro. METHODS: Pure isolated ECL cell preparation with a purity of +/- 95% was developed by proteinase digestion, elutriation and density gradient centrifugation with viability of > 95% as determined by trypan blue exclusion. After a short duration of culture basal and stimulated histamine secretion and DNA synthesis of ECL cells were measured by enzyme immunoassay and bromodeoxyuridine (BrdU) uptake, respectively. RESULTS: Hp LPS stimulated basal and gastrin-stimulated histamine secretion (EC(50)4 x 10(-11) mol/L and EC(50) 10(-10) mol/L respectively). These effects were completely inhibited by somatostatin (10(-10) mol/L) but not by gastrin receptor antagonist L(365 260). Hp LPS did not stimulate basal DNA synthesis but significantly increased gastrin stimulated BrdU uptake with EC(50) of 10(-10) mol/L. Escherichia coil LPS had no significant effect on both histamine secretion and DNA synthesis. CONCLUSION: Hp stimulates both ECL cell proliferation and secretion in vitro. An interaction between Hp LPS and ECL cells may contribute to the reported abnormalities in acid secretion and gastric pathophysiology noted in Hp infection.

[Detection of human immunodeficiency virus infection in tuberculosis patients].

OBJECTIVE: Combined infection of human immunodeficiency virus (HIV) and Mycobacterium tuberculosis (MTB) has attracted wide attention. This study was to find out infection of HIV and occurrence of AIDS in tuberculosis patients. METHODS: Primary screening was conducted in 2 973 inpatients with pulmonary tuberculosis by gelatin particle agglutination assay or rapid gold-labeled HIV 1 + 2 antibody assay. The HIV positive cases were sent to Beijing Municipal Center Laboratory for verification. RESULTS: Three HIV positive cases were detected among 2,973 pulmonary tuberculosis patients with the positive rate of 1.01 per thousand. Two were male and 1 female with average age of 30 years (23, 27, and 40 years respectively) accounting for 2.86 per thousand of the 21 - 41 age group, among which 1 infected through intravenous drug injection, 2 by heterosexual behavior. And all the 3 HIV positive cases had developed to be AIDS. CONCLUSIONS: Enough knowledge of the risk of combined HIV/MTB infection and high-degree alert should be emphasized in medical professionals involving in diagnosis, treatment and management of tuberculosis. Routine HIV antibody detection in high-risk tuberculosis patients for HIV infection, especially in young and middle-age tuberculosis patients, is helpful for early diagnosis and treatment of AIDS and avoiding its spreading.

[Clinical and immunological studies on transplantation of total cornea with circular lamellar sclera].

OBJECTIVE: To observe the clinical results of total cornea with circular lamellar sclera transplantation (CST) and study its immunological mechanisms by animal experiments. METHODS: 109 patients with corneal transplantation were retrospectively analyzed, and according to the methods of operation they were divided into groups to compare their rate of rejection and the time of occurrence of corneal neovascularization. The methods of grouping and operation in animal experiments were similar to that in the clinical practice. RESULTS: The time of occurrence of neovascularization and rejection of CST were later than that in the total corneal transplantation (TCT), and the postoperative rate of rejection at 3 to 6 months was obviously increased in comparison with that in the partial penetrating keratoplasty (PKP). CONCLUSION: CST is an important method for rebuilding ocular anterior segment, and the donor limbus is active in the immunological of rejection.

[A case-control study on relationship between body mass index and lung cancer in non-smoking women].

OBJECTIVE: To investigate the relationship between body mass index (BMI) and lung cancer in non-smoking women. METHODS: A population-based case-control study on lung cancer in non-smoking women was conducted during February 1992 to December 1993 in Shanghai. Totally, 504 cases of lung cancer of non-smoking women and 601 population controls were studied. RESULTS: The risk of lung cancer increased with the decrease of BMI. The odds ratio of lung cancer in non-smoking women was 1.95, as compared to controls with the highest quartile of their BMI, with a dose-response pattern (P < 0.0002 with chi(2) test for trend), adjusted for age, schooling and income. Non-conditional logistic regression analysis was used to adjust for some main risk factors found in other studies carried out at home, such as passive smoking, tea drinking, eye irritation by cooking oil fume, kinds of cooking oils, pulmonary tuberculosis, family history of lung cancer, age of menarche, menstrual cycle and dietary nutrition (including intake of vitamins C and E, beta-carotene, total fat and calorie), was used to adjust. There still existed relationship between BMI and lung cancer in non-smoking women after adjustment for those factors. Stratification analysis showed that there was relationship mainly between BMI and lung adenocarcinoma. CONCLUSION: BMI may be a risk factor for lung cancer in non-smoking women.

[Analysis for spatial distribution of Oncomelania snail in mainland China by geographic information system(GIS) database].

OBJECTIVE: To analyze spatial distribution of Oncomelania snail populations in mainland China by geographic information system (GIS) database. METHODS: Genetic variation experiment data and experimental data of susceptibility of 34 snail populations were collected from nine provinces in China, to set up a database. A world digital map was linked with the database in a software ArcView Release 3.0 a to be used to divide zones with ArcView spatial analyze function and GIS overlaying function. AVHRR NDVI satellite images were collected to form a new one for prevalent season, which were overlaid with distribution maps of heterozygous indices of snail populations, polymorphic locus percentage and infection rates of snail population with schistosome for classifying the images. RESULTS: Spatial analysis showed that distribution maps of genetic heterozygous indices of snail, percentage of polymophic loci and infection rate of snail can be divided into two large zones, i.e., east zone and west zone, both with varied pictures. Analysis for overlaying three-stratum distribution map with AVHRR NDVI satellite image showed four distinctly different spatial zones, including west Sichuan zone, west Yunnan zone, middle zone of river, lake and marshland, and southeast coastal zone. CONCLUSION: It is the first time using GIS to analyze data of Oncomelania population genetics and confirmed the population structure of Oncomelania snails presented discrete sub-population model, which supports the theory of sub-species of Oncomelania spp. Exited in mainland China.

[The progress in research on changes of central venous pressure under simulated weightlessness and microgravity].

One of the most profound circulatory changes that occur in man during exposure to microgravity is a cephalad redistribution of fluid. Central venous pressure (CVP) is not only an important standard for determining the cephalad redistribution of fluid but also the unique valuable means for measuring continuously cardiac filling pressure under simulated weightlessness (SW) and microgravity, and has a relationship with neurohumoral [correction of nerohumoral] regulation. With the development of manned spaceflight, the researches in space suggested that CVP did not increase, which is directly contradictory to the previous results under SW. This paper mainly discusses the changes of CVP and the effects of countermeasures on CVP under SW and microgravity, and the mechanisms of decreased CVP under microgravity. It is postulated that more suitable SW models, more studies about the area of low pressure including especially pulmonary circulation, and comprehensive studies in multiple systems are needed.

[Effects of 17 beta-estradiol on intracellular free calcium, inositol-1,4,5-trisphophate and calmodulin in human osteoblast-like osteosarcoma cell line TE85].

OBJECTIVE: To study the effects of 17 beta-estradiol (E2) on intracellular free calcium ([Ca2+]i), inositol-1,4,5-trisphophate (IP3) and calmodulin (CaM) in human osteoblast-like cell line TE85. METHODS: Using Fluo-3/AM as fluorescent indicator, the [Ca2+]i was measured by laser confocal microscopy system. The IP3 content was determined by anion-exchange chromatography. CaM content was detected by a high sensitive assay based on stimulation of Ca(2+)-dependent phosphodiesterase activity. RESULTS: E2 at dose of 0.1 and 1.0 nmol/L increased fluorescent level by 4.7 and 6.1 times. Pretreatment with thapsigargin (100 nmol/L), the E2 caused only 1.5 times elevation in fluorescence. E2 induced a concommitant bi-peak increase in IP3 content. At the presence of E2(1.0 nmol/L), the CaM content increased by 85.2%. Tamoxifen did not affect the effect of E2 on [Ca2+]i, IP3 and CaM content. But, the inhibitor of phospholipase C (neomycin) and pertusis toxin depressed them partly or completely. CONCLUSIONS: E2 regulate bone cells function by way of Ca2+/CaM activation.

[Molecular epidemiology of respiratory syncytial virus infection in infants].

OBJECTIVE: To search for the molecular epidemiology of respiratory syncytial virus (RSV) infection in infants. METHODS: The randomly amplified polymorphism DNA (RAPD) technology was used to study 96 RSV strains isolated and identified from the patients in Changchun State Hospital of Pediatrics during 1992-1994 and 1 reference strain of M type. RESULTS: All strains were typable, four RAPD patterns were observed, different diseases have different types. 87.14% of the patients' RSV strains had the same RAPD pattern(R1). CONCLUSION: RAPD method is a rapid, easy and high resolution technique, with which we can know on the molecular level the microbial infection state. There was epidemic of R1 type infection during the period surveyed.

[The experimental treatment of laryngeal carcinoma with all-trans retinoic acid].

OBJECTIVE: To investigate whether all-trans retinoic acid (RA) has antitumor effect in vivo. METHODS: Sixteen nude mice were transplanted with human laryngeal cell carcinoma PHC3. Eight of them were injected with RA intraperitoneally and others with solvent as the control. Response was measured and tumor histopathological features were studied. RESULTS: The growth and weight of RA-treated tumors were significantly reduced in comparison with those of the control. The growth inhibition rate reached more than 50% in the RA group. Studies also showed that RA-treated cells became well-differentiated. There was a significant decrease of c-myc protein level in the RA treated tumors as compared with the control. CONCLUSION: It is suggested that RA could induce differentiation of human laryngeal carcinoma cells and suppress tumor growth in vivo. Regulation of c-myc gene or protein may be related to those happening.
