Functional immobilization of a DNA-binding protein at a membrane interface via histidine tag and synthetic chelator lipids.

The coupling of a DNA-binding protein to self-organized lipid monolayers is examined at the air-water interface by means of film balance techniques and epifluorescence microscopy. We used two recombinant species of the heat shock factor HSF24 which differ only in a carboxy-terminal histidine tag that interacts specifically with the nickel-chelating head group of a synthetic chelator lipid. As key function, HSF24 binds to DNA that contains heat-shock responsible promoter elements. In solution, DNA-protein complex formation is demonstrated for the wild type and fusion protein. Substantial questions of these studies are whether protein function is affected after adsorption to lipid layers and whether a specific docking via histidine tag to the chelator lipid leads to functional immobilization. Using lipid mixtures that allow a lateral organization of chelator lipids within the lipid film, specific binding and unspecific adsorption can be distinguished by pattern formation of DNA-protein complexes. At the lipid interface, functional DNA-protein complexes are only detected, when the histidine-tagged protein was immobilized specifically to a chelator lipid containing monolayer. These results demonstrate that the immobilization of histidine-tagged biomolecules to membranes via chelator lipids is a promising approach to achieve a highly defined deposition of these molecules at an interface maintaining their function.

[Cognitive therapy and psychoanalytical therapy of limited personalities: a critical consideration].

Comparative studies of psychoanalytical and cognitive approaches to psychotherapy for borderline personalities reveal not only convergences of opinion, particularly concerning the importance of the relationship with the therapist, but also noteworthy differences. Different types of cognitive therapy are to be distinguished by their highly structural nature and by the learning of cognitive capacity with a view to reducing the severity of emotional and behavioral disorders. The different types of analytical therapy attempt a reorganisation of the personality by referring to a complex psychopathological model, allowing an overview of mental functions and of transferral interaction, and attempt to modify them.

Molecular organization of histidine-tagged biomolecules at self-assembled lipid interfaces using a novel class of chelator lipids.

In molecular biology, the expression of fusion proteins is a very useful and well-established technique for the identification and one-step purification of gene products. Even a short fused sequence of five or six histidines enables proteins to bind to an immobilized metal ion chelate complex. By synthesis of a class of chelator lipids, we have transferred this approach to the concept of self-assembly. The specific interaction and lateral organization of a fluorescent fusion molecule containing a C-terminal oligohistidine sequence was studied by film balance techniques in combination with epifluorescence microscopy. Due to the phase behavior of the various lipid mixtures used, the chelator lipids can be laterally structured, generating two-dimensional arrays of histidine-tagged biomolecules. Because of the large variety of fusion proteins already available, this concept represents a powerful technique for orientation and organization of proteins at lipid interfaces with applications in biosensing, biofunctionalization of nanostructured interfaces, two-dimensional crystallization, and studies of lipid-anchored proteins.

Engineered fusion molecules at chelator lipid interfaces imaged by reflection interference contrast microscopy (RICM).

In molecular biology, biotechnology, and protein-engineering, the expression of histidine fusion proteins is a very powerful technique for the identification and one-step purification based on the interaction of the histidine stretch with immobilized metal complexes. By synthesis of a novel class of chelator lipids, this technique was combined with the concept of self-assembly leading to interfaces for immobilization and orientation of histidine-tagged biomolecules (Schmitt et al., 1994). Here, the chelator lipid layers were transferred onto solid substrate by vesicle fusion and Langmuir-Blodgett-techniques. Specific binding of a peptide containing an oligohistidine sequence to these functionalized interfaces was demonstrated by reflection interference contrast microscopy (RICM). Due to the phase separation behaviour of lipid mixtures, the chelator lipid interface could be further structured in two dimensions. Binding and organization of histidine-tagged molecules at these two-dimensional recognition arrays was imaged by RICM with a layer thickness resolution of 0.2 nm, and 0.5 microm laterally. Specific docking can be triggered by adding nickel ions and disrupted by EDTA. This concept opens up possibilities for reversible immobilization, enrichment and organization of histidine fusion proteins at interfaces and their application in biosensing.

[The intraoral determination of the sagittal condyle path inclination during face-bow recording].

In a clinical trial involving 40 fully dentate persons, three different intraoral procedures to determine the sagittal condyle path inclination were checked for accuracy. In each test person the condyle path inclination was determined three times, i.e. by the central bearing point method, the protrusive check-bite method and by a protrusive bite record integrated in the bite fork. At 5 mm of protrusion the reproducibility of the results obtained with the central bearing point method (mean value 2.1 +/- 2.0 degrees) were far better than that with either of the two other methods; the difference was statistically significant. The protrusive check-bite records and the protrusive bite fork records did not differ from each other as far as their accuracy was concerned (mean values 3.6 +/- 2.1 degrees and 3.4 +/- 1.6 degrees, respectively). The mean value of the condyle path inclination to the hinge axis infra-orbital plane in the sample was about 50 +/- 8 degrees regardless of the method used. Thus, protrusion records can be integrated in the face bow transfer.

[Schizophrenias in DSM-IV and ICD-10 and French psychiatry].

DSM-IV and ICD-10 show the convergence of international nomenclatures of psychotic disorders. Their emphasis on antheorism and the definition of operational diagnostic criteria converge with one major trend in traditional French psychiatry which has been illustrated by Chaslin. The principal discrepancy which concerns chronic hallucinatory psychosis has been reduced by the restrictive conception of schizophrenic disorders in DSM-IV and ICD-10. Despite the attempt to define French empirical criteria, chronic hallucinatory psychosis has not been accepted by international classifications and is in pronounced decline in French scientific works. On the other had, the "bouffées délirantes aiguës", taken up by ICD-10, are acknowledged internationally.

Self assembly of covalently anchored phospholipid supported membranes by use of DODA-Suc-NHS-lipids.

We present a novel preparation method for the self assembly of covalently anchored phospholipid supported membranes. The surface is gold covered by cysteamine. Vesicles containing DMPC and activated DODA-Suc-NHS-lipids assembled on this surface. The whole self-assembly process is monitored conveniently by Near Infrared Surface Plasmon Resonance (NIR-SPR). Comparing the data to those obtained by Ca(2+)-mediated vesicle fusion, confirmed this interpretation.

[Sleep disorders in the elderly].

Insomnia dominates the sleep disorders of the elderly. Sleep apnea, the restless legs syndrome and nocturnal agitation represent other facets. After eliminating the possibility of painful or degenerative organic disease, iatrogenic cause and mode of life, other symptoms should be sought, namely mental and personality-related. Investigation requires polysomnogram recording, supported by careful history-taking, including objective and subjective factors. Treatment includes notions of diet, biological rhythm and desynchronisation. It should take into consideration the deleterious effects of prolonged use of certain psychotherapeutic agents.

Pharmacokinetic study of yohimbine and its pharmacodynamic effects on salivary secretion in patients treated with tricyclic antidepressants.

The pharmacokinetic parameters and the time course of the effect after acute oral administration of yohimbine on salivary secretion in patients treated with tricyclic antidepressants were investigated. Yohimbine (10 mg) increased both salivary outflow and plasma noradrenaline levels for 4 h. Pharmacokinetic parameters (t1/2, tmax, Cmax and AUCexp) and plasma concentrations of noradrenaline were higher in patients treated with tricyclic antidepressants than in controls. At this dose, yohimbine induced a relatively large number of side effects. A lower dose (4 mg) increased salivary secretion for 3 h without any side effects in patients treated with tricyclic antidepressants but not in healthy volunteers. These data describe an interaction between yohimbine and tricyclic antidepressants and thus show that a relatively low dose (4 mg) of yohimbine could be useful in the treatment of dry mouth due to tricyclic antidepressants.

Gastroenteropancreatic neuroendocrine tumours contain a common set of synaptic vesicle proteins and amino acid neurotransmitters.

Human neuroendocrine tumours of the gastroenteropancreatic system contain major integral membrane proteins of small synaptic vesicles of neurons, together with characteristic membrane polypeptides of large dense-core vesicles of neurons and neuroendocrine cells. The membrane polypeptides characteristic for small synaptic and large dense-core vesicles are detected in pheochromocytomas (n = 6), functional (n = 6) and non-functional (n = 6) foregut, and midgut carcinoids (n = 17). All gastroenteropancreatic tumours contain large amounts of amino acid neurotransmitters, i.e. glycine and glutamate. gamma-Aminobutyric acid, however, is only found in some foregut carcinoids. Thus, neuroendocrine gastroenteropancreatic tumours possess a vesicle type with a content and membrane composition similar to small synaptic vesicles of neurons.

Complexation studies on inositol-phosphates: IV. Ca(II) complexes of myo-inositol 1,4,5-trisphosphate.

The stability constants of the complexes formed between Ca2+ and the myo-inositol 1,4,5-triphosphate (Ins(1,4,5)P3) were determined by potentiometric titration in two different media and temperature conditions (medium 1: I = 0.1 M But4NBr, 25 degrees C; medium 2: I = 0.2 M KCl, 37 degrees C). Mainly because of the presence of potassium the results obtained in these media show large differences in both the nature and the stability of the complexes. In medium 1, MH2L and M2L species are formed along with the ML and MHL species which also exist in medium 2. In addition, the stability of the latter species decreases by more than one log unit in going from medium 1 to medium 2. In an attempt to assess the biological significance of the metal binding to Ins(1,4,5)P3, the results were compared to the Ca2+-ATP complexes that form in the same media conditions. Taking into account the relative stability of the complexes of both systems, it is likely that the action or metabolism of Ins(1,4,5)P3 may be influenced by coordination of either alkali or alkali-earth cations.

Effect of 3 weeks treatment with yohimbine on salivary secretion in healthy volunteers and in depressed patients treated with tricyclic antidepressants.

The effect of yohimbine treatment (4 mg three times daily) for 3 weeks on salivary secretion was investigated. In healthy volunteers, acute administration of yohimbine increased salivary volume within 1 h to a similar extent before and at the end of the treatment period. In depressed patients treated with tricyclic antidepressants (and exhibiting a reduced salivary flow), yohimbine also acutely increased salivary volume. In contrast, the alpha 2-adrenoceptor antagonist failed to modify resting values measured in the morning (i.e. 10 and 14 h after the last administration in healthy volunteers and depressed patients respectively). This result indicates that alpha 2-adrenoceptor antagonists may have a potential therapeutic use in the treatment of dry mouth caused by tricyclic antidepressant drugs.
