Escherichia coli cells exposed to streptomycin display a mutator phenotype.

Mistranslation mediated by the mutA and mutC tRNA alleles elicits a strong mutator phenotype (H. S. Murphy and M. Z. Humayun, J. Bacteriol. 179:7507-7514, 1997; M. M. Slupska, C. Baikalov, R. Lloyd, and J. H. Miller, Proc. Natl. Acad. Sci. USA 93:4380-4385, 1996). Here, we show that exposure to streptomycin, an antibiotic known to promote mistranslation, induces a recA- and umuDC-independent mutator phenotype detected as enhanced mutagenesis at a 3, N4-ethenocytosine lesion borne on transfected M13 single-stranded DNA.

Surgical treatment of trigeminal neurinomas.

OBJECTIVE: To investigate the best surgical approach for the removal of trigeminal neurinomas (TNs). METHODS: A retrospective analysis of 75' patients with TNs in Huashan Hospital was carried out. RESULTS: In the early group (1978-1984), a series of conventional intradural approaches were used; in the late group (1985-1995), an epidural approach via the skull-base craniotomy was used. Total tumor removal was achieved in 58% (20/35) of patients in the early group and 80% (32/40) in the late group (P < 0.025). Temporary and permanent cranial nerve morbidity were 62.7% and 37% in the early group and 28.1% and 10% in the late group (P < 0.001). CONCLUSION: The best microsurgical approach for the removal of trigeminal neurinomas except those confined to the posterior fossa is epidural approach or epidurotransduro-transtentorial approach via the skull-base craniotomy.

[Chemical constituents of Saururus chinensis (Lour.) Bail].

OBJECTIVE: To search for the active chemical constituents of Saururus chinensis. METHOD: 95% ethanol extracts from the aerial part of Saururus chinensis was mixed with silica gel, and then eluted with petroleum ether, ethyl acetate and methanol. On the basis of physicochemical properties and spectroscopic analysis, structures of the compounds obtained were identified. RESULTS: Eight compounds were isolated from the ethyl acetate and methanol parts, and identified as aristolactam A II(I), daucosterol(II), hyperoside(III), isoquercitrin(IV), quercitrin(V), quercitin-3-O-beta-D-glucopyranosyl(1-->4)-alpha-L-rhamnopyranoside(VI), ellagic acid(VII) and corilagin(VIII) respectively. CONCLUSION: Compounds I, II, VI, VII and VIII were found from this plant for the first time.

[Effect of arginine on deep partial-thickness burn in rats].

To investigate the therapeutic effect of arginine on burn, a randomized controlled trial in 20% TBSA deep partial-thickness burned rats was designed. The rats' wound healing, cellular immunity and nitrogen balance were evaluated at appointed postburn days. The results showed that the arginine group got better wound healing and faster recovery of cellular immunity, and achieved positive nitrogen balance. These suggest that supplement of exogenous arginine after burn may be beneficial to the victim in promoting would healing, reducing postburn immune suppression and enhancing anabolism of protein.

Detection of bcr/abl fusion transcripts by semiquantitative multiplex RT-PCR combined with a colormetric assay in Ph positive leukemia.

We studied the feasibility of the clinical application of a new bcr/abl analysis system, C-TRAK t(9;22), consisting of a multiplex RT-PCR and a colormetric assay. With this system, bcr/abl transcripts could be detected in all of 24 cytogenetic Philadelphia chromosome (Ph) positive leukemia patients and in none of eight Ph negative patients. Multiple bcr/abl transcripts could be detected in three of the 24 Ph positive patients, the fusion of bcr exon 1 to abl exon 2 (e1a2 junction) dominated that of bcr exon 13 to abl exon 2 (b2a2 junction) in two cases and that of bcr exon 14 to abl exon 2 (b3a2 junction) and b2a2 dominated e1a2 in one case. This system was sensitive enough to be able to detect even one bcr/abl transcript-producing cell in 50000 bcr/abl negative background cells, thus making it suitable for semiquantitative evaluation. Minimal residual disease (MRD) was monitored in one Ph positive leukemia patient who underwent allogenic bone marrow transplantation (allo-BMT). After allo-BMT, a weak positivity of the bcr/abl transcript continued with no clinical relapse; this result was consistent with that of a conventional nested PCR assay using ethidium bromide staining. Including all the procedures for RNA extraction, it took only about 10 h to detect the bcr/abl transcripts. Our findings indicate that this bcr/abl analysis system provides a quick and sensitive method for screening bcr/abl transcripts and possibly for monitoring MRD in Ph positive leukemia patients.

Expression of the mouse metallothionein-I gene conferring cadmium resistance in a transgenic cyanobacterium.

This paper reports the construction of a transgenic strain of cyanobacterium aimed at removing heavy metal pollution in waters. The mouse metallothionein-I (mMT-I) gene was inserted in the vector pRL-439 downstream of the strong psbA promoter. The resulting plasmid pRL-MT was ligated at the EcoRI site of the shuttle vector pKT-210 to generate the shuttle expression vector pKT-MT. This recombinant plasmid was introduced into Anabaena sp. PCC 7120 by triparental conjugative transfer. After selection on streptomycin, a stable transgenic Anabaena strain was obtained. The presence of the mMT-I gene was confirmed by DNA/DNA hybridization and its expression was demonstrated by immunodetection with specific antibodies. A metal tolerance experiment showed that this transgenic Anabaena strain had acquired higher metal resistance.

Cadmium decreases SGLT1 messenger RNA in mouse kidney cells.

Mouse renal cortical tubule cells in primary culture exposed to cadmium (Cd2+) develop decreased Na(+)-glucose cotransport activity as measured by uptake of the glucose analogue alpha-methyl-glucoside. RNA was isolated from kidney cell cultures, and after reversed transcription, the DNA was amplified with primers to rat SGLT1 (the high affinity isoform of the sodium glucose cotransporter) and mouse beta-actin. Only one product was identified after amplification with the rat SGLT1 primers, which on sequencing was 96% identical to rat SGLT1. Compared to beta-actin, the intensity of the SGLT1 message declined progressively as CdCl2 concentration in the medium increased from 0 to 10 microM. Similar decreases in SGLT1 mRNA were also observed as media zinc (Zn2+) concentrations rose from 0 to 75 microM or as copper (Cu) concentrations increased from 0 to 150 microM. Exposure to 8 microM Cd as Cd-metallothionein (Cd7-MT) also caused a fall in relative SGLT1 mRNA abundance, and at nearly identical internal Cd concentrations of 40-43 pmol/microgram DNA, both Cd7-MT and CdCl2 reduced SGLT1 mRNA to 33% of control. In general, the fall in SGLT1 mRNA was more rapid than the decline in Na(+)-dependent glucose uptake after cells were exposed to Cd2+. These findings suggest that the effects of Cd2+ and other metals on renal glucose transport are related to decreased expression of SGLT1 message.

[Experimental study of the effect of danshen on the viability of burned skin].

In order to investigate the influence of Danshen on the viability of skin from II degree burned and the effect of Danshen on the wound healing, 20 rabbits having a wound from second degree burn were divided into two groups randomly. The experimental group was treated with Danshen and the control was treated with normal saline. Then succinate dehydrogenase, SDH activity, malonyl dialdehyde, MDA concentration, water content and morphological change of wound tissue were observed dynamically. The results showed that the levels of SDH activity in the experimental group were significantly higher than that of the control group (P < 0.05), the MDA concentration and water content of the former were lower than that of the latter (P < 0.05) and the morphological examination showed that the wound healing process of former were better than that of the latter. The conclusion was Danshen had a beneficial effect on the viability of the burned skin and this effect was due to its protecting the residual epithelial cells of the burned skin.

[Observation of cicatricial fibroblasts in culture and its biological properties].

In order to study the biological properties of fibroblasts isolated from different tissues. The fibroblasts from normal skin, hypertrophic scar and keloid were cultured, respectively, in vitro, and their morphologies and growth kinetics were compared. The results revealed that although fibroblasts in keloid were irregularly arranged, crisscross and overlapping with loss of polarization, there was no significant difference in the 3 groups so far the cellular morphology of fibroblast itself, cellular growth curve, cellular mitotic index, cloning efficiency and DNA content provided those cultures were in the same cellular density and culture conditions. It was concluded that fibroblasts isolated from culture of normal skin, hypertrophic scar and keloid in vitro showed no significant difference in morphology and growth kinetics, on the contrary, their biological behaviors were quite similar.

[Determination of organic pollutants in water by off-line supercritical fluid extraction-Gas Chromatography/Infrared Spectrometry/Mass Spectrometry].

A supercritical fluid extraction (SFE) method using carbon dioxide for the determination of organic pollutants in Yellow River water was developed. In this paper, organic contaminants in 3 L Yellow River water were concentrated on 1 g GDX-301 adsorbent and then eluted from the adsorbent with supercritical CO2. The analytes extracted by SFE were collected into 1 mL alcohol collecting solvent through a SiO2 restrictor (35 cm x 50 microns i.d.). Finally, the collecting solvent was analyzed by Gas Chromatography/Infrared Spectrometry/Mass Spectrometry. The SFE conditions which were optimized for removal of the analytes from adsorbent were 20 MPa, 60 degrees C and 40 min. The extraction efficiencies of SFE were directly compared to those obtained by using 15 mL dichloromethane elution. Efficiencies of SFE under conditions of 20 MPa, 60 degrees C and 40 min were generally higher than those of solvent elution.

[Immobilization of N-acetyl-1,6-diaminohexane(NADAH) on diol-silica and study of mechanism of NADAH and hexamethylene bisacetamide(HMBA) inducing tumor cells differentiation].

A method for separating NADAH and its immobilization onto diol-silica have been developed. HMBA and its meabolite NADAH were used as the inducers for many types of tumor cells' differentiation. The inducing mechanism of HMBA is not clear yet. Experiments show that HMBA and NADAH have a relatively strong hydrophobic reaction and hardly ion-exchange with some DNA and some proteins of the cytosolic fraction of HL-60 human acute progranulocytic leukemia cells, BIU-87 human bladder carcinoma cells and the human erythrocyte membrane, when utilizing immobilization of NADAH. The retention time of the proteins and DNA was longer than that of the phosphatides. These results show that the adsorption of HMBA and NADAH with some proteins and DNA is higher than that with the phosphatides. The expected receptor binding biospecifically with HMBA has not been found.

Detailed Analysis of Determination of Contact Angle Using Sphere Tensiometry

The configurations of the interfacial menisci around a sphere during the pulling process were analyzed and the methods for the simultaneous determination of surface and interfacial tension and contact angle were discussed. Furthermore, a method for the determination of contact angle using sphere tensiometry, which is independent of interfacial tension, was developed and tested with experiments. Studies on rLMP7, a beta-subunit of the multicatalytic proteinase.

Subunit rLMP7 of the multicatalytic proteinase (MCP), which has been associated with chymotrypsin-like proteinase activity, was examined in rat liver and hepatocyte-derived cell lines. rLMP7 was detected in both nucleus and cytosol in liver by immunohistochemistry and immunoblotting, using a peptide-specific anti-rLMP7 antibody. A M(r) 30,000 precursor protein was present only in cytosol, as was a minor component of M(r) 25,000. Mature rLMP7 (M(r) 23,000) was present in MCP in both nucleus and cytosol, although it was not detectable in the nuclear scaffold. Two rLMP7 cDNAs (designated rLMP7.1 and rLMP7.s) were identified by rapid amplification of 5' ends using RT/PCR, a result which was confirmed by Northern blot analysis and RNase protection assays. rLMP7.1 is 3-4x more abundant than rLMP7.s; it is 50 nt longer than the previously reported cDNA sequence and includes an upstream in-frame ATG within a consensus translation initiation sequence, which encodes the M(r) 30,000 rLMP7 precursor protein identified in vivo. rLMP7.s is 100 nt shorter than rLMP7.1 and does not contain the most 5' ATG. Transient transfection analyses with rLMP7.1 and rLMP7.s constructs coupled to green fluorescent protein showed that both transcripts were efficiently expressed in vivo. In vitro expression of these two rLMP7 cDNAs showed that rLMP7.1 produces the M(r) 30,000 precursor protein, whereas rLMP7.s produces two smaller peptides of M(r) 25,000 and 23,000. Purified 20S MCP preparations were able to proteolytically process the M(r) 30,000 precursor to the M(r) 25,000 product but not to the mature rLMP7 form. However, incorporation of this processed M(r) 25,000 product (or of either M(r) form produced from rLMP7.s) did not occur in vitro. In vitro processing and pulse-chase experiments suggested that the mature M(r) 23,000 subunit is derived, at least in part, from the M(r) 30,000 precursor. The M(r) 25,000 form may be a stable product produced directly from rLMP7.s.

Estrogenic effects of nonylphenol on pS2, ER and MUC1 gene expression in human breast cancer cells-MCF-7.

Many chemicals have recently been discovered to have estrogenic activity, including the surfactant intermediate nonylphenol (NP). It has been well documented that estrogen is a facilitator of human breast cancer development under certain conditions, and environmental estrogens such as NP are currently under intense investigation. Using the expression of pS2 (a trefoil peptide expressed in breast cancer cells), MUC1 (a member of the mucin family) and ER (the human estrogen receptor) genes as estrogen-responsive reporter genes, the effects of estradiol and NP on human breast cancer cells-MCF-7 were studied. In the time course study, the mRNA expressions were detected after NP (10 microM) or estradiol (E2, 0.1 microM) treatments using the RT-PCR technique. The results indicated: (1) NP and E2 induced pS2 mRNA expression after a 2-h exposure and (2) NP induction produced the highest level of MUC1 mRNA after 2 h, which was reduced to only 42% of control at 48 h. E2 treatment resulted in a gradual increase in MUC1 expression over the course of the exposure. The highest level of MUC1 mRNA was at 48 h. This indicates that NP may stimulate MUC1 expression by a different mechanism than E2. (3) NP affected ER expression in the same manner as MUC1. In contrast, E2 stimulated ER expression in a similar manner as pS2; the highest level was at 2 h and expression remained elevated through the 48-h point. NP is an estrogenic compound that alters pS2, MUC1 and ER gene expression in MCF-7 cells. NP may affect MUC1 expression through a different mechanism than E2. The link between aberrant MUC1, PS2 and ER expression and the development of breast cancer also needs to be elucidated through further investigation.

Hydrophobic chromatography of the HL-60 cellular fraction co-binding with hexamethylene bisacetamide.

Methods of separating N-acetyl-1,6-diaminohexane (NADAH) and its immobilization to diol-silica have been developed. Hexamethylene bisacetamide (HMBA) and its metabolite NADAH are used as inducers of leukemia cell differentiation. The inducing mechanism of HMBA is still not clear. Experiments show that HMBA and NADAH undergo relatively strong hydrophobic reactions and do not readily undergo ion-exchange with the proteins of the cytosolic fraction of HL-60 cells during immobilization of NADAH; the retention time of the proteins was longer than that of the phosphatides. These results show that the adsorption of HMBA and NADAH to proteins was higher than that to phosphatides. The expected biospecific receptor binding with HMBA has not been found.

[Effects of excess fluoride on bone turnover under conditions of diet with different calcium contents].

OBJECTIVE: To study the effects of excess fluoride on bone turnover under conditions of diet containing different amount of calcium. METHODS: The experiment was performed on rats raised on a balanced diet with adequate calcium or a monotonous diet with low calcium and given amount of fluoride in their drinking water (F, 100 mg/L) for 2 months or 1 year. RESULTS: Osteomalacia, osteoporosis and accelerated bone turnover were observed with elevated serum alkaline phosphatase (ALP), osteocalcin (BGP) and parathyroid hormone (PTH) in rats fed on low calcium diet and fluoridized water for 2 months. In the rats fed on adequate calcium diet and fluoridized water for 2 months, only slightly increased osteoblastic activity was found while the average width of trabecular bone was increased with elevated serum ALP activity in rats raised on the same diet and water for 1 year. CONCLUSIONS: The basic effect of excess fluoride on bone is the causation of a high bone turnover state which can also be induced to a milder extent by low calcium diet itself. Therefore, the formation of a high bone turnover state is the pathogenetic basis for low dietary calcium intake to exacerbate the severity of skeletal fluorosis.
