Phosphatidylinositol 4,5-bisphosphate binding to the pleckstrin homology domain of phospholipase C-delta1 enhances enzyme activity.

The pleckstrin homology (PH) domain is a newly recognized protein module believed to play an important role in signal transduction. While the tertiary structures of several PH domains have been determined, some co-complexed with ligands, the function of this domain remains elusive. In this report, the PH domain located in the N terminus of human phospholipase C-delta1 (PLCdelta1) was found to regulate enzyme activity. The hydrolysis of phosphatidylinositol (PI) was stimulated by phosphatidylinositol 4,5-bisphosphate (PIP2) in a dose-dependent manner with an EC50 = 1 microM (0.3 mol%), up to 9-fold higher when 5 microM (1.5 mol%) of PIP2 was incorporated into the PI/phosphatidylserine (PS)/phosphatidylcholine (PC) vesicles (30 microM of PI with a molar ratio of PI:PS:PC = 1:5:5). Stimulation was specific for PIP2, since other anionic phospholipids including phosphatidylinositol 4-phosphate had no stimulatory effect. PIP2-mediated stimulation was, however, inhibited by inositol 1,4, 5-triphosphate (IP3) in a dose-dependent manner, suggesting a modulatory role for this inositol. When a nested set of PH domain deletions up to 70 amino acids from the N terminus of PLCdelta1 were constructed, the deletion mutant enzymes all catalyzed the hydrolysis of the micelle forms of PI and PIP2 with specific activities comparable with those of the wild type enzyme. However, the stimulatory effect of PIP2 was greatly diminished when more than 20 amino acid residues were deleted from the N terminus. To identify the specific residues involved in PIP2-mediated enzyme activation, amino acids with functional side chains between residues 20 and 40 were individually changed to glycine. While all these mutations had little effect on the ability of the enzyme to catalyze the hydrolysis of PI or PIP2 micelles, the catalytic activity of mutants K24G, K30G, K32G, R38G, or W36G was markedly unresponsive to PIP2. Analysis of PIP2-stimulated PI hydrolysis by a dual substrate binding model of catalysis revealed that the micellar dissociation constant (Ks) of PLCdelta1 for the PI/PS/PC vesicles was reduced from 558 microM to 53 microM, and the interfacial Michaelis constant (Km) was reduced from 0.21 to 0.06 by PIP2. The maximum rate of PI hydrolysis (Vmax) was not affected by PIP2. These results demonstrate that a major function of the PH domain of PLCdelta1 is to modulate enzyme activity. Further, our results identify PIP2 as a functional ligand for a PH domain and suggest a general mechanism for the regulation of other proteins by PIP2.

Binding Sites

Positive charge at position 549 is essential for phosphatidylinositol 4,5-bisphosphate-hydrolyzing but not phosphatidylinositol-hydrolyzing activities of human phospholipase C delta1.

Point mutagenesis, phosphatidylinositol (PI), and phosphatidylinositol 4,5-bisphosphate (PIP2) hydrolysis assays and equilibrium centrifugation PIP2 assays were used to study the functional roles of four highly conserved arginine residues in the Y region of human phospholipase C delta1 (PLCdelta1) (Arg-527, -549, -556, -701). Most of the mutant enzymes were either partially defective or fully active in their abilities to catalyze the hydrolysis of PI or PIP2. However, upon substitution of Arg-549 by glycine or histidine, the mutant enzyme was defective in its ability to catalyze the hydrolysis of PIP2, but it is still able to hydrolyze PI. Replacing Arg-549 with lysine had little effect on the level of PI and PIP2 hydrolytic activities of the mutant enzyme. The residual PIP2 hydrolyzing activity of R549H is highly dependent on pH. R549H showed 5-10% of the PIP2-hydrolyzing activity of the native enzyme between pH 5 and 7 and nondetectable PIP2-hydrolyzing activity at pH 8. The PIP2-hydrolyzing activity of R549G was not detectable at all pH values. Kinetic analysis of PLCdelta1-catalyzed PIP2 hydrolysis revealed that the micellar dissociation constant Ks and interfacial Michaelis constant Km were similar in the native, R549K, and R549H enzymes; but the specific activity at the saturated substrate mole fraction and infinite level of substrate (Vmax) of the R549H mutant were reduced by a factor of 15. PIP2 competitively inhibits the native enzyme to hydrolyze PI at both pH 7 and 8. However, PIP2 inhibits R549H only at pH 7.0 and does not inhibit R549G at either pH. Taken together, these results suggest that positive charge at position 549 of PLCdelta1 protein is essential for the enzyme to recognize and catalyze the hydrolysis of PIP2 but not PI.

Thiopentone requirements in adults after varying pre-induction doses of fentanyl.

The thiopentone dose needed for satisfactory induction of anaesthesia was determined in 60 adult patients using a double-blinded study design. No premedication was given. The effect of pretreatment with four different doses of fentanyl on ED50 for thiopentone was determined and compared with a control group not receiving fentanyl. The cardiovascular effects of the combinations were also studied. ED50 for thiopentone was reduced from 4.0 mg.kg-1 in the control group to 2.2 mg.kg-1 in patients receiving fentanyl 5.0 micrograms.kg-1. Only in the group of patients receiving fentanyl 5.0 micrograms.kg-1 was there no significant change in systolic arterial blood pressure. An increased heart rate was noted in patients induced with thiopentone in combination with fentanyl 0.625 micrograms.kg-1. No adverse effects of thiopentone, fentanyl or the combinations were noted.

[Investigation on invasion of preepiglottic space by supraglottic carcinoma].

This study, based on analysis of 100 whole-organ serial sections of the larynx, concluded that preepiglottic space invasion is involved in 60% of supraglottic carcinoma. The preepiglottic space is involved either by invasion through the adjacent structures of the epiglottis, or more frequently by destroying the epiglottic cartilage and the thyro-epiglottic ligament. The preepiglottic space is involved mainly by epiglottic and epiglottic-ventricular carcinomas, accounting for 95% of carcinomas which invade the space. The involvement of preepiglottic space by supraglottic carcinoma is intimately related to the morphologic features, marginal growth patterns, and T classification. Ulcerative carcinoma is the commonest, accounting for 65%. The incidence of the infiltrative carcinomas is the highest. With increase in T stage, the percentage of invasion of preepiglottic space is increased. We believe that the easy invasion of the preepiglottic space by the supraglottic carcinoma is due to the fact that the epiglottic and epiglttic-ventricular carcinomas which account for 83% of supraglottic carcinomas are located in front of the larynx. In addition, the carcinoma easily destroys the epiglottic, cartilage and enters the preepiglottic space because the embryonic barrier which exists below the glottis and the perichondrium of the epiglottic cartilage is quite fragile.

Adult

Influence of hearing of 22 G Whitacre and 22 G Quincke needles.

Audiograms were performed pre-operatively and 2 days postoperatively in 48 patients given spinal anaesthesia for transurethral resection of the prostate. Hearing levels were examined at 1000 Hz and below. Either 22 G standard design (Quincke) needles (n = 25) or 22 G pencil-point design (Whitacre) needles (n = 23) were used. Hearing loss of 10 dB or more at two or more frequencies were observed in six of 25 patients in the Quincke group and in two of 23 patients in the Whitacre group. The mean hearing level was more reduced in the Quincke group. The shape of the tip of the spinal needle seems to be of some importance to the effects on hearing level that may occur after spinal anaesthesia.

Aged

Reported anaesthetic complications during an 11-year period. A retrospective study.

Retrospectively 120 anaesthetic complications with potential medicolegal consequences were reviewed. Seven deaths occurred. Eighty percent of the complications were potentially avoidable. Of the complications 4.4% resulted in major morbidity and 82.3% in intermediate morbidity. Dental injuries were reported with an annual frequency of six. The use of pulse oximetry and/or capnography could possibly have prevented 18% of all complications apart from dental injuries. Ten cases were reported to the National Swedish Board of Health and Welfare; four cases resulted in disciplinary consequences. The influence of human factors on the development of complications is stressed.

Anesthesia

[Flow cytometric analysis of colorectal cancer and polyps].

Thirty-four cases of colorectal cancer, 24 cases of polyps of various types and 30 cases of control colorectal mucosa were analysed by flow cytometry for DNA content in order to study their relation to histopathology and prognosis. The result showed that the DNA content of 6 out of 24 polyps (25%) were aneuploid, and all of these 6 cases with aneuploid DNA content were adenomas accompanied with mild to severe dysplasia, and six cases of juvenile polyps were diploid. There was a correlation between DNA aneuploid and severe dysplasia (P less than 0.05), but not with the histopathologic type. Twenty-three out of 34 cancers were aneuploid (68%), and correlation was noticed between the DNA Index (DI) and Dukes' classification (F = 17.37), but not with the differentiation of tumors. Patients with DNA aneuploid tumours had a worse prognosis than those with only DNA diploid tumours. Therefore, DNA ploidy of colorectal cancers detected by flow cytometry is of prognostic value for colorectal cancer.

Adenoma

Interactions of neural networks: models for distraction and concentration.

We present a model of neural group interactions, which are projections from one neural network (network B) of McCulloch-Pitts neurons connected via a Hebbian rule, to another network (network A) of the same structure. We first consider the case in which the projecting network B is in a pattern different from the initial attracting state of network A. A critical projecting strength lambda c is found such that for lambda below this value there exists a noise threshold sigma lambda corresponding to each lambda. For the case where lambda less than lambda c and the noise level sigma less than sigma lambda, there are two possible retrievals, with different probabilities: the initial attracting state of network A and the projecting pattern. If lambda less than lambda c and sigma greater than sigma lambda, stable states of network A disappear. In the case lambda greater than lambda c, network A is pulled out of its initial basin of attraction and into that of the projecting pattern. This analysis provides a model for distraction. Second-order interactions reduce the distraction. When the projecting network B is in the same pattern as the initial attracting state of network A, the projection acts as an external reinforcement, which enables network A to retrieve in highly noisy conditions. Sharp noise thresholds for nonzero retrievals are shown to be eliminated by the projection. Higher-order connectivity improves the retrieval ability of the network. The second case serves as a model of concentration. We discuss the model of distraction and concentration (i) in connection with common experience of expectation of recognition and (ii) in connection with recent T-maze experiments on infant rats; finally, we suggest a refined version of the Bruner-Potter experiment to test our prediction of the disappearance of hysteresis.

Oscillations and chaos in neural networks: an exactly solvable model.

We consider a randomly diluted higher-order network with noise, consisting of McCulloch-Pitts neurons that interact by Hebbian-type connections. For this model, exact dynamical equations are derived and solved for both parallel and random sequential updating algorithms. For parallel dynamics, we find a rich spectrum of different behaviors including static retrieving and oscillatory and chaotic phenomena in different parts of the parameter space. The bifurcation parameters include first- and second-order neuronal interaction coefficients and a rescaled noise level, which represents the combined effects of the random synaptic dilution, interference between stored patterns, and additional background noise. We show that a marked difference in terms of the occurrence of oscillations or chaos exists between neural networks with parallel and random sequential dynamics.

Hearing loss after spinal anesthesia is related to needle size.

Audiograms were performed preoperatively and 2 days postoperatively in 28 patients given spinal anesthesia for transurethral resection of the prostate. In 14 patients 22-gauge and in 14 patients 26-gauge spinal needles were used. Hearing loss of 10 dB or more at any frequency was observed in 13 of 14 patients in the 22-gauge group and in 4 of 14 patients in the 26-gauge group. There was a statistically significant reduction in hearing level in the low-frequency range in patients in whom the 22-gauge needle was used. Hearing loss was unilateral at five frequencies and bilateral at one frequency. No cases of postspinal headache occurred. Audiometry may be a more sensitive indication of cerebrospinal fluid leak than postspinal headache.

A human tRNA gene cluster encoding the major and minor valine tRNAs and a lysine tRNA.

A human genomic DNA clone hybridizing to mammalian valine tRNA(IAC) contained a cluster of three tRNA genes. Two valine tRNA genes with anticodons of AAC and CAC, encoding the major and minor cytoplasmic valine tRNA isoacceptors, respectively, and a lysine tRNA(CUU) gene were identified by Southern blot hybridization and DNA sequence analysis of a 7.1-kb region. At least nine Alu family members were interspersed throughout the 18.5-kb human DNA fragment, with three Alu elements in the intergenic region between the valine tRNA(AAC) gene and the lysine tRNA gene. Each of the five Alu family members in the sequenced region can be categorized into one of the four Alu subfamilies. The coding regions of all three tRNA genes contain characteristic internal split promoter sequences and typical RNA polymerase III termination signals in the 3'-flanking regions. The tRNA genes are accurately transcribed by RNA polymerase III in a HeLa cell extract, since the RNase T1 fingerprints of the mature-sized tRNA transcription products are consistent with the structural genes. The lysine tRNA(CUU) gene was transcribed only slightly more efficiently than the valine tRNA(CAC) gene in the homologous in vitro transcription system. Surprisingly, the valine tRNA(CAC) gene was transcribed about eightfold more efficiently than the valine tRNA(AAC) gene, implicating the presence of a modulatory element in the upstream region flanking the tRNA(CAC) gene.

[A study on completely defined agar media for mycobacteria].

A serial of completely defined agar media suitable for growth of the common mycobacteria which are pathogenic was reported in this paper. A tool was supplied for the study of mycobacteria on nutrition and physiology, biochemical feature, genetics and variation and on the mechanism of resistance and susceptibility to antimicrobial drug, and with it would be in favour of developing more satisfactory media for mycobacteria. The majority of strains of the mycobacteria tested grew more rapidly on the completely defined agar media than on Lowenstein-Jensen medium and the final yield of the microbes on the agar media was less than or equal to those on Lowenstein-Jensen medium. There were only small amounts of growing cells on the completely defined agar media among the strain of mycobacterium bovis. Tcolonies with the above mentioned twodifferent patterns were found to grow on the completely defined media from mycobacterium avium. M. intracclluare and M. fortuitum.

[Electron microscopic observations on Bacteroides thetaiotaomicron].

Three strains of B. thetaiotaomicron have been isolated from caeca of BALB/c-nu/+ mice of SPF. These Bacteroides are obligately anaerobic, Gram-negative, non-spore-forming and non-motil without flagella rods. A characteristic of staining is deep at ends and stainless at the medium of a rod. The isolation and identification of these strains has been reported in 1987. This paper introduced only the results of EM observations. Under the SEM, the unstained area of rods is always showing a concavity, which is just a nucleoid in sections under the TEM. Many lamellar corpuscles have been found in cell plasma. Some of them have been secreted out of the cells. The chemical properties and physiological functions of them are unknown.

[Studies on synthesis of baogongteng A--a new myotic agent].

Baogongteng A, isolated from Erycibe obtusifolia Benth., is a new tropane alkaloid used for treating glaucoma. Using synthetic 6 beta-acetoxy-tropinone as starting material, racemic Baogongteng A(8) was synthesized. Synthetic Baogongteng A also shows myotic activities in rabbits, but the potency is half of that of the natural product.
