Mycobacterium tuberculosis-susceptible I/St mice develop severe disease following infection with taxonomically distant bacteria, Salmonella enterica and Chlamydia pneumoniae.

Mice of I/St strain develop severe lung inflammation and die shortly following infection with virulent mycobacteria. To find out whether tuberculosis (TB)-susceptible I/St mice are susceptible to other intracellular bacteria, we investigated two different taxonomically distant pathogens, Chlamydia pneumoniae and Salmonella enterica serovar Typhimurium. Comparison of I/St and TB-resistant A/Sn mice (both Nramp1(r)) demonstrated that the former are more susceptible to both salmonella and chlamydia, displaying a significantly shortened survival time following challenge. Lung pathology develops more rapidly in I/St compared to A/Sn mice following infection with chlamydia, despite their similar ability to control bacterial multiplication. Following infection with salmonella, substantial ( approximately 3 log) but very short (second day post-infection) interstrain differences in bacterial loads were observed, accompanied by higher levels of interleukin (IL)-6 and tumour necrosis factor (TNF)-alpha in the peritoneal cavities of I/St mice. I/St macrophages were more permissive for salmonella growth during the first 24 h following infection in vitro. Because the prominent differences in survival time did not correlate with permanent differences in bacterial multiplication, we suggest that both infections trigger fatal pathological processes whose dynamics depend strongly upon the host genetics.

[Laboratory diagnosis of toxoplasmosis using polymerase chain reaction].

Polymorphism of clinical manifestations in Toxoplasma infection and variegated disease patterns virtually rule out the diagnosis based solely on clinical symptoms, which makes modern laboratory tests particularly important. Amplification test system based on the polymerase chain reaction has been developed for the diagnosis of Toxoplasma infection. Computer analysis of nucleotide sequence of Toxoplasma gondii surface antigen gene P30 was analyzed, which helped choose and synthesize specific oligonucleotide primers. A method for biological material processing was selected, allowing sufficient DNA output. Optimal conditions for amplification reaction, ensuring absolute specificity and high (10-100 cells/sample) sensitivity, were determined.

[The detection of mycobacteria in children and adolescents by using the polymerase chain reaction].

A polymerase chain reaction (PCR) was used for determination of mycobacterial DNA in clinical samples from children and adolescents. The results were positive in 23 and 53% of cases, respectively, while standard microbiological methods failed to show presence of any bacteria in the samples. Case histories contained information on high incidence of positive Mantoux test. Microbiologically the diagnosis was confirmed only in 7 adolescents. In children mycobacteria were not found.

[Test-systems of polymerase chain reaction for determining the tuberculosis pathogen].

Two polymerase chain reaction-based test systems were used to identify Mycobacteria tuberculosis by using a clinical material for practical health purposes. The former test system strictly specifically enables M. tuberculosis to be detected. The latter also allows one to differentiate a BCG vaccine strain from the M. tuberculosis. The test systems were tested by using 11 clinical sputum samples, three of them were taken from patients with fibrocavernous tuberculosis, two from those with focal tuberculosis and one from that with infiltrative one, 5 from those with non-specific pulmonary diseases (chronic bronchitis and asthma). In addition, one serum sample from a patient with fibrocavernous tuberculosis was examined. All tests obtained from patients with tuberculosis were positive, whereas those from patients with non-specific pulmonary diseases were negative. This suggests that differentiation of BCG vaccine strain from M. tuberculosis can be used both in model experiments and in the study of clinical material lysates.

[Phage-mediated conjugative transfer of plasmids in Staphylococcus aureus].

It was shown possible to transfer nonconjugative plasmids during joint cultivation of the donor and recipient cells by transduction and phage-mediated conjugation. In the latter case it was necessary that the phage in the medium was free and the prophage was present in the recipient cells. Differences in the regularities of the transfer of the nonconjugative plasmids mobilized by the conjugative plasmid or phage were observed.

Lysogeny of methicillin-resistant Staphylococcus aureus and the role of prophages in transfer of conjugative and non-conjugative plasmids.

The lysogenicity of 49 strains of methicillin-resistant S. aureus (MRSA) isolated in Moscow clinics in the 1970s and '80s was studied by the method of mitomycin C induction. It was found that one strain had phage of serogroup B, 33 strains had serogroup F phages and 15 strains had phages of both serogroups. In the course of genetic crossing on nitrocellulose filters it was demonstrated that serogroups B and F prophages contained in recipient cells 1) increase the frequency of transfer of conjugative plasmid pG873 and 2) mobilize transfer of non-conjugative plasmids pE994 and rms7.

[Working conditions and health-related measures in the processing of waste products of phosphorus-containing mineral fertilizers].

In the processing of the waste products of phosphorus containing fertilizer production, several major health-related factors were identified depending on the industrial site's peculiarities: unfavourable microclimate conditions (both cold and heat), relative temperature differences, noise and vibration caused by the equipment, semi-product's, additives' and finished product's dusts, fluorine- and phosphorus-containing substances' affects and poor illumination. The proposed data were used for health-related improvements in the occupational conditions.

[Effect of prophages on transfer frequency of conjugative plasmid G 873].

Transfer of the conjugative plasmid G873 on filters and mixed cultivation of the donor and recipient cells in liquid media is described. In the both systems the use of the lysogenic recipient cells (phages of serogroups B and F) in the crossings increased mor than 100-fold the frequency of plasmid transfer. The conjugative transfer of the plasmid in the mixed cultivation system was proved. The conjugative transfer required the presence (while not obligatory) of calcium chloride and was restricted by the serum factors.

[The action of phage 52A on energy processes in Staphylococcus aureus].

Phage infection leads to the dissipation of the transmembrane difference of electric potentials, measured by the adsorption of lipophilic permeant ions of tetraphenyl phosphonium, in staphylococci. Besides, the reversible stimulation of cell respiration processes is observed. The duration of these effects is comparable with the period of the injection of phage DNA, equal to 2-3 minutes.

[Hemorrhage after stomach resection in ulcer disease].

Under observation were over 3 500 patients, subjected to gastric resection for the complicated ulcerous disease. In the early postoperative period hemorrhage was observed in 42 patients, including bleeding in the gastrointestinal tract--in 23, in free abdominal cavity--in 19. In cases, when early profuse bleeding following gastric resection would not respond to intensive conservative measures, then urgent relaparotomy with the aim of haemostasis, replenishment of blood loss and correction of the internal medium seem to be indicated. Bleeding from forcibly remained low duodenal ulcer in Billroth-II gastrectomy can be arrested by means of duodenotomy and suturing of the ulcer bottom with U-shape sutures. For establishing in early diagnosis of hemorrhage into the abdomen its drainage after gastric resection with a rubber strip, brought out through a separate incision in the anterior abdominal wall in the right subcostal space, seems to be rather effective.
