[Lipid composition of the placenta of rats with protein restriction and deficiency of essential fatty acids].

The influence of diet restriction and EFA deficiency during pregnancy in the rat on the lipid and phospholipids composition of the placenta was investigated. Female virgin albino Wistar rats weighing 130 +/- 4 g, were assigned to three equivalent groups. Prior mating and during pregnancy each group of rats received the following regimen: Animals in the Control Group (C) were fed a 25% casein diet in ad libitum quantities; the dietary Restricted Group (D) received the same control diet in amounts calculated to approximate 50% (g/100 g rat) of the intake of group C; the Deficient and Restricted Group (DD) rats were fed a restricted amount of EFA deficient diet. On the 21st day of gestation pregnant animals were sacrificed. The foetuses and placentae obtained by caesarium section were isolated and weighed. A 50% food restriction before and during pregnancy resulted in a significant decrease in phospholipid contents (p less than 0.05); severe EFA deficiency superimposed to 50% food restriction, moreover induced significant changes in the fatty acid pattern of phospholipids, decreasing n3 and n6 fatty acids and increasing eicosatrienoic acid. There was an accumulation of triglycerides in the placenta of rats fed on the EFA deficient diet. In the two restricted groups fetal weight was reduced, but although in the DD group, placental weight was not affected, litter size was dramatically reduced.

Influence of nutrient limitation and low pH on serogroup B Neisseria meningitidis capsular polysaccharide levels: correlation with virulence for mice.

Neisseria meningitidis strain M986, which possesses a polyanionic sialic acid capsular polysaccharide, was resistant to the bactericidal effects of normal rabbit serum, but sensitive when immune serum and complement were present. An isogenic strain PRM102, deficient in the ability to produce capsular polysaccharide, was sensitive to normal serum. Strain M986, when grown under conditions of low pH or nutrient limitation, synthesized increased levels of capsular polysaccharide. This was accompanied by an increase in cell surface hydrophilicity and virulence for mice. Cells grown in low-pH, iron-limited medium synthesized the highest concentration of polysaccharide and exhibited the highest cell surface hydrophilicity and virulence among the cases examined. The increase in capsular polysaccharide was partly explained by a decrease in the specific activity of a membrane-bound cytidine monophosphate-N acetylneuraminic acid hydrolase. The results suggest that conditions of nutrient limitation and low pH exert profound effects on the physicochemical nature of the meningococcal cell surface which, in turn, cumulate in enhanced virulence of this organism for mice.

Role of lipid intermediate(s) in the synthesis of serogroup B Neisseria meningitidis capsular polysaccharide.

Neisseria meningitidis serogroup B strain M986 was examined for the involvement of lipid intermediate(s) participating in the biosynthesis of the sialic acid capsular polysaccharide. The addition of exogenous undecaprenyl phosphate, phosphatidylethanolamine, or phosphatidylglycerol to particulate membranes, in the presence of cytidine 5'-monophosphosialic acid, resulted in the stimulation of sialyltransferase activity specifically by undecaprenyl phosphate. Sialyltransferase activity, after delipidation of particulate membrane proteins, was specifically reconstituted by undecaprenyl phosphate. After the addition of 14C-labeled cytidine 5'-monophosphosialic acid to particulate membranes, the level of labeled lipid intermediate(s), extracted by chloroform-methanol (2:1), increased up to a maximum level between 3.75 and 5.0 min, which subsequently decreased to a lower steady-state level. Pulse-chase experiments revealed a transient, solvent-extractable, lipid-linked component. The extracted N-acetylneuraminic acid was in polymeric form. Sequential oxidation and reduction of the extracted radioactivity followed by neuraminidase treatment revealed an average degree of polymerization of four or five N-acetylneuraminic acid residues. Bacitracin-sensitive peptidoglycan was synthesized in vitro by particulate membranes. Cross-competition experiments between peptidoglycan and capsular polysaccharide synthesis by preincubation of precursors of one pathway during synthesis of the other revealed a competitive effect for a common component. This component was believed to be a common pool of undecaprenyl phosphate. A model for the production and regulation of the capsular polysaccharide is proposed.

Physiology of sialic acid capsular polysaccharide synthesis in serogroup B Neisseria meningitidis.

The pathway for biosynthesis of sialic acid capsular polysaccharide was examined in Neisseria meningitidis serogroup B strain M986 and in strain PRM102, an isogenic mutant defective in polysaccharide production. Strain PRM102 was found to possess only 25% of the level of sialyltransferase activity that was found in strain M986, but it had wild-type levels of both the N-acetylneuraminic acid (NANA) condensing enzyme and the CMP-NANA synthetase. A new meningococcal enzyme, a CMP-NANA hydrolase, was found in both meningococcal strains. This enzyme generated CMP and NANA from CMP-NANA, had a Km of 0.88 microM, had a Vmax of 10.75 nmol of NANA produced per h per mg of protein, and was completely inhibited by 45.3 microM CMP. The sialyltransferase, which also had CMP-NANA as substrate, was insensitive to CMP addition. Subcellular fractionation and purification of cytoplasmic and outer membranes on sucrose density gradients revealed that both the sialyltransferase and the CMP-NANA hydrolase were cytoplasmic membrane associated. The NANA condensing enzyme and the CMP-NANA synthetase were found to be cytosolic. A working hypothesis for the regulation of sialic acid polysaccharide synthesis was developed. The CMP-NANA hydrolase with its high affinity for CMP-NANA regulates polysaccharide formation by the sialyltransferase, whereas CMP, a product of both the sialyltransferase and the CMP-NANA hydrolase, modulates the activity of the hydrolase on the cytoplasmic membrane.

[Chemical composition of human milk. Influence of the nutritional status of the nursing mother].

The chemical composition of human milk obtained from 16 underweight women and 25 controls collected at four, eight and 12 weeks of lactation, was determined in order to assess the effect of maternal nutrition upon milk quality. No meaningful differences were observed for protein, lactose, energy, calcium, iron and copper contents. The values obtained are comparable to those reported in the literature from developed countries. As to Zn content, both groups exhibited decreased values with time, with significant higher values for the controls at the 12th week of observation. Magnesium showed exactly the opposite behavior. On the basis of the present study, it is concluded that maternal nutrition does not condition important changes in the chemical composition of human milk. The authors propose the application of comparable methodologic and analytical criteria, in order to achieve a more complete understanding of the relationships between maternal nutrition and lactation.

[Sodium and potassium content of various Chilean foods].

Sodium and potassium contents of 40 high-protein dietary products were determined in order to complete the Table de Composición Química se Alimentos Chilenos (Chemical Composition Table of Chilean Foods). These cations' level must be strictly controlled in diets of many renal and heart patients. In Chile, Nutritionists who are in charge of preparing these diets, do not have a national composition table related to the sodium and potassium content for most of the food products. Samples of fluid cow's milk, dried milk with different fat contents, some cheeses, hen eggs, bovine entrails, some meat derivates and several meat cuts, including bovine, pork, lamb and chicken were studied.

The heliothis virescens 170 kDa aminopeptidase functions as "receptor A" by mediating specific Bacillus thuringiensis Cry1A delta-endotoxin binding and pore formation.

The relationship between Bacillus thuringiensis Cry1Aa, Cry1Ab and Cry1Ac delta-endotoxin binding and pore formation was investigated using a purified 170 kDa aminopeptidase N (APN) from Heliothis virescens brush border membranes. Aminopeptidases with molecular sizes of 110, 140 and 170 kDa were eluted from a Cry1Ac toxin affinity column using N-acetylgalactosamine. The 140 kDa aminopeptidase has a cross-reacting determinant typical of a cleaved glycosyl-phosphatidylinositol anchor. After mild base treatment to de-acylate the glycosyl-phosphatidylinositol linkage and incubation in phosphatidyl inositol phospholipase C, anti-cross-reacting determinant antibody recognized the 170 kDa protein. Kinetic binding characteristics of Cry1A toxins to purified 170 kDa APN were determined using surface plasmon resonance. Cry1Aa, Cry1Ab and Cry1Ac, but not Cry1C and Cry1E toxins recognized 170 kDa APN. Each Cry1A toxin recognized two binding sites: a high affinity site with KD ranging from 41 to 95 nM and a lower affinity site with KD in the 325 to 623 nM range. N-acetylgalactosamine inhibited Cry1Ac but not Cry1Aa and Cry1Ab binding to 170 kDa APN. When reconstituted into phospholipid vesicles, the 170 kDa APN promoted toxin-induced 86Rb+ release for Cry1A toxins, but not Cry1C toxin. Furthermore Cry1Ac, the Cry protein most toxic to H. virescens larvae, caused 86Rb+ release at lower concentrations, and to a greater extent than Cry1Aa and Cry1Ab toxins. The correlation between toxin-binding specificity and 86Rb+ release strongly suggests that the purified 170 kDa APN is the functional receptor A in the H. virescens midgut epithelial cell brush border membranes.
