Mutagenic studies on the hair dye 2-(2',4'-diaminophenoxy)ethanol with different genetic systems.

A new hair-dye coupler, 2-(2',4'-diaminophenoxy)ethanol was analyzed for its potential mutagenic activity in different genotoxic assays, namely gene reverse mutations in Salmonella typhimurium, forward mutations in the yeast Schizosaccharomyces pombe, and in the V79 Chinese hamster cell line grown in vitro (HGPRT forward mutation system). Two other genetic test systems, measuring the mitotic gene conversion in Saccharomyces cerevisiae (strain D4) and the unscheduled DNA-repair synthesis in a HeLa cell line grown in vitro, were also used. 2,4-Diaminoanisole, a mutagenic/carcinogenic structurally related hair-dye coupler, and a group of well-known mutagens, namely methyl methanesulfonate, ethyl methanesulfonate, cychlophosphamide, hycanthone and N-nitrosodimethylamine, were used as positive controls. The new aromatic amine, 2-(2',4'-diaminophenoxy)ethanol, was negative in all the assays performed, under the same treatment conditions as in the case of all the positive controls.

Selection in HAT medium is not a reliable method for the study of reversion from 6-thioguanine resistance to sensitivity.

6-Thioguanine-resistant mutants were induced in V79 Chinese hamster cells with 2-aminopurine, ICR-170 and hycanthone. Samples of mutants of different origin were treated with EMS or 1CR-170 and plated in HAT medium for selection of revertants. In the result, a significant fraction of HAT-resistant clones was not 6-thioguanine-sensitive as would have been expected from a genuine reversion to wild type.

Embryotoxic effects of 5-hydroxytryptamine during the peri-implantation period in the rat.

Administration of 5-hydroxytryptamine creatinine sulphate (5-HT) 5-10-20 mg/kg s.c. to pregnant rats during the preimplantation period (1200 h, Days 1 through 5) did not inhibit implantation. Experiment 2: Administration to pregnant rats during the peri-implantation period (0900 h, Day 6) of 5-HT 5 mg/kg s.c. did not affect implantation or embryo-fetal development. 5-HT 10 mg/kg s.c. produced a significant increase (P greater than 0.01) in the resorption rate (31.8%) and severe cardiovascular or ophthalmic malformations in 5.3% of viable fetuses. 5-HT 20 mg/kg produced a resorption rate (97.2%) virtually incompatible with the continuance of pregnancy. Experiment 3: Histological examination of uterine preparations made from Day 6 pregnant rats sacrificed 6, 24, and 30 h after receiving 5-HT 20 mg/kg s.c. showed, in 30 h post-injection preparations, toxic effects at implantation sites (uterine lumen completely deprived of epithelial layer and filled with cellular debris, and complete degeneration of implanted embryos) but no toxic effects between implantation sites. Experiment 4: Administration of 5-HT 10 mg/kg s.c. to pregnant rats during the postimplantation period (Days 10 and 11) produced a 63.2% resorption rate, a reduction in the mean weight of viable fetuses, and severe malformations in 24% of viable fetuses. The embryotoxic activity of 5-HT may be attributed to its vasoconstrictive action which renders the uterine mucosa or trophoblast ischemic, thus causing irreversible damage to the luminal epithelium at the implantation sites.

Computer simulation of the dynamics of cell populations.

A modified version of the program of Valleron and Friendel is presented for the computer simulation of growing populations of cells. The method effects a sizable reduction of the storage requirement and allows one to follow the growth and division of cell clones for a large number of generations. Moreover, it is particularly useful to simulate specific biological events assumed to occur in the cell under various hypotheses and to compare them. Examples of this are also given.

Analysis of a cell cycle model for Escherichia coli.

Ribosome and protein synthesis, DNA replication and cell division in Escherichia coli cells are described by a mathematical model that integrates previous descriptions in quantitative terms and proposes a new formalization to relate ribosome net synthesis to cell growth. The model assumes a cell size control of DNA replication and therefore is structurally divided into two subsystems: the first, whose state variables are ribosomes and protein, and the second, which is activated when the protein level reaches a threshold and which is comprised of DNA replication and cell division. The dynamics of the entire system is set only by the first subsystem: the values of its parameters determine whether the cells will be in a resting condition or will grow exponentially and in the latter case the resulting duplication time, while the structure and the parameter values of the second subsystem determine the size and the composition of the cell and the timing of DNA replication during the cycle. Relationships are derived that allow a simple determination of the time of initiation and of termination of DNA replication and the number of chromosome origins involved in any possible cell cycle as well as the macromolecular levels at the beginning of a cycle and on the average in a population of cells in balanced exponential growth.

Experiments on the effect of the medium in mutation tests with the HGPRT system in cultured mammalian cells.

Higher mutation frequencies were observed on 8AG than on 6TG medium when HGPRT-deficient mutants were being selected in V79 Chinese hamster cells. 2 alternative explanations for the effect of the medium were considered, namely (1), that mechanisms are present that cause resistance to 8AG only, in addition to that (or those) causing resistance to both drugs, and (2), that mutants with low HGPRT content survive on 8AG but not on 6TG medium, owing to lower affinity of 8AG for the enzyme. The second explanation was favoured as a result of various experimental approaches, including kinetics of expression on the 2 media, cross-resistance at different expression times and serial selection on the 2 media.

The extent of fetal ossification as an index of delayed development in teratogenic studies on the rat.

In teratogenic studies toxic effects may manifest themselves in retarded fetal development, such as a reduction in fetal weight. In searching for an additional index, the number of centers of ossification in seven skeletal districts (sternum, metacarpus, metatarsus, cervical and caudal vertebrae, anterior and posterior proximal phalanges) of rat fetuses delivered on days 19, 20 and 21 of gestation were counted and compared. Results showed uneven ossification in day-19 and -20 fetuses, but sufficiently advanced, homogeneous and uniform ossification in day-21 fetuses to provide a reliable quantitative index for evaluating retarded fetal development. It is therefore proposed that the stage of skeletal ossification in day-21 fetuses be used in teratogenic studies in the rat to evaluate retarded fetal development.
