Characteristics of ovarian cancer cells transduced by the bicistronic retroviral vector containing GM-CSF and HSV-TK genes.

OBJECTIVES: To explore whether HSV-TK (herpes simplex virus thymidine kinase) and GM-CSF (granulocyte-macrophage colony-stimulating factor) genes could be linked by internal ribosome entry site (IRES) in one retroviral vector and expressed by ovarian cancer cells following transfection, and to observe the characteristics of the transduced cells. METHODS: Retroviral vector pLGM-I-TK was constructed by linking the HSV-TK gene and GM-CSF gene with the IRES sequence. By using the "ping-pong" technique, pLGM-I-TK was transfected into the packaging cell line, PA317, to produce a PA317/TK-GM cell line. Using the resulting viral supernatant to infect the ovarian cancer cell line SKOV3, PCR and RT-PCR were used to explore the integration and transcription of HSV-TK and GM-CSF genes. The cytotoxicity of GCV (gancyclovir) on SKOV3/TK-GM was determined by MTT assay and the bystander effect of the HSV-TK/GCV system was also assessed. ELISA was used to measure the expression of GM-CSF by the transgene cells. RESULTS: The bicistronic retroviral vector constructed could be successfully transduced into PA317 and the titer of the retroviral vector was about 8.6 x 10(5) cfu/ml. PCR and RT-PCR demonstrated the successful integration and transcription of HSV-TK and GM-CSF genes transduced into the SKOV3 cell. SKOV3/TK-GM cells could be killed by GCV, and the IC50 was 0.7 microgram/ml. The bystander effect was demonstrated. The expression level of GM-CSF in SKOV3/TK-GM was 60.4 ng.ml-1 x 10(6) cells-1 x 2 days-1. CONCLUSION: The IRES sequence can be used to construct retroviral vectors to facilitate co-transfection of two genes. SKOV3/TK-GM cells can simultaneously express the HSV-TK and GM-CSF genes with biological activities which could be useful for enhancing the function of immune cells on the basis of suicide gene therapy.

Interaction of hypertension and diabetes on impairment of endothelial function.

OBJECTIVE: To investigate whether the interaction of hypertension and diabetes aggravates endothelial dysfunction and leads to smooth muscle dysfunction. METHODS: Noninvasive methods were used to the investigated patients with type 2 diabetes mellitus (2-DM) (Group 1), patients with hypertension (Group 2) and patients with both 2-DM and hypertension (Group 3), as well as a normal control group (Group 4) by studying a brachial artery without evidence of atherosclerotic plaque formation. RESULTS: Results showed that endothelium-dependent vasodilation decreased significantly in Group 1 (5.74% +/- 3.32%, P < 0.05), Group 2 (4.14% +/- 2.93%, P < 0.01), and Group 3 (2.78% +/- 2.08%, P < 0.001) as compared to the control (9.45% +/- 3.88%). The nitroglycerin-induced vasodilation (smooth muscle function) in Group 3 was significantly decreased as compared to the control group (14.11% +/- 4.63% vs 23.53% +/- 6.77%, P < 0.001), but there were no differences in nitroglycerin-induced vasodilation between Group 1, Group 2 and Group 4. On univariate analysis, a reduced vasodilator response to nitroglycerin was associated with endothelium-dependent vasodilation (r = 0.54, P < 0.001). CONCLUSION: Our results indicate that the interaction of 2-DM and hypertension aggravates endothelial dysfunction and further impairs the smooth muscle function.

[Clinical analysis of idiopathic thrombocytopenic purpura with pregnancy].

OBJECTIVE: To study the optimal management of pregnant women with idiopathic thrombocytopenic purpura (ITP). METHODS: The outcomes of 37 pregnancies with ITP about 10 years from (1990 to 1999) were analysed. RESULTS: Among the 37 cases, vaginal delivery and cesarean section were done in 16 and 21 cases respectively. Neonatal thrombocytopenia was diagnosed in 3 newborns. None of them had intracranial hemorrhage. The incidences of postpartum hemorrhage and infection were 8.1% and 2.7%, respectively. No maternal mortality was noted. CONCLUSIONS: In view of very low morbidity in babies of ITP mothers, we suggest that they be delivered vaginally if there is no obstetrical complications. Selected cesarean delivery should be performed in severely thrombocytopenic cases when there are sufficient fresh blood and platelet. The invasive interventions should not be used to avoid newborn thrombocytopenia.

[The bone regeneration in cleft palate after surgical repair (a prelimilnary observation)].

OBJECTIVE: To study the bone healing situation of the cleft palate after surgical procedure, and discuss the factors affecting it. METHODS: 20 patients with repaired cleft palate were examined by CT scan coronally. The result of images were observed and the position, shape and quality of bone bridge were measured. RESULTS: Formation of bone bridge was found in 13 cleft gaps out of 20 patients (65%), the sex ratio among patients with bone bridge was 1:1; there was no obvious difference between unilateral and bilateral cleft palate. Considering the operation age with the bone formation, the highest percentage of bone bridge formation fell into the group of 4-6 years old, and the most occurring regions were in premolar and anterior part of molar area. CONCLUSIONS: There can be new bone bridge regeneration in bone gap after palate surgery, and the bone formation may have some relationship with the operation age and surgical technique.

Expression of human vascular endothelial growth factor receptor Flt-1 extracellular domain 1-3 loop cDNA in Pichia pastoris, purification of the expressed product and detection of its biological activity.

OBJECTIVE: To express human vascular endothelial growth factor receptor Flt-1 extracellular domain 1-3 loop cDNA in Pichia. pastroris, and to purify the expressed product and detect its biological activity. METHODS: By inserting human Flt-1 (1-3 loop) cDNA coding 316 amino acid residues into Pichia pastoris expression vector pPIC9K containing AOX1 promoter and the sequences of alpha secreting signal peptides, a recombinant expression plasmid pPIC9K/Flt-1 (1-3) was constructed and transformed to yeast host strain GS115, then His+ Muts phenotype transformant was screened out and cultured in flasks, and Flt-1 (1-3) was expressed under the induction of 1% methanol. RESULTS: SDS-PAGE showed that after being induced with 1% methanol for 4d, the expressed product existed in supernatant in the form of soluble molecule and contained 60% of total protein expressed. Western blot showed good antigenicity and specificity of expressed product. After being purified by CM-Sepharose FF and Sephacryl S-100 chromatography, the purity of the expressed product reached above 90%. Biological assay proved that the expressed product could bind to hVEGF165 and inhibit the proliferation of HUVEC stimulated by hVEGF165. CONCLUSION: Human vascular endothelial growth factor receptor Flt-1 extracellular domain 1-3 loop was successfully expressed. The study lays a foundation for further application of the expressed product in the treatment of vasoformation related diseases, such as tumor and diabetic retinopathy.

Expression of human vascular endothelial growth factor (VEGF165) in Pichia pastoris and its biological activity.

OBJECTIVE: To express human vascular endothelial growth factor (hVEGF165) cDNA in Pichia pastroris, purify the expressed product and detect the biological activity of it. METHODS: By inserting hVEGF165 cDNA coding 165 amino acid residues into Pichia pastoris expression vector pPIC9K containing AOX1 promoter and the sequences of alpha secreting signal peptides, a recombinant expression plasmid pPIC9K/hVEGF165 was constructed and transformed to yeast host strain KM71, then multiple-copy insert transformants were screened out and cultured in flasks, and hVEGF165 was expressed under the induction of 1% methanol. RESULTS: SDS-PAGE showed that after being induced with 1% methanol for 4 d, the expressed product existed in supernatant in the form of soluble molecule and contained 60% of total protein expressed. Western blot showed good antigenicity and specificity of expressed product. After being purified by Heparin-Sepharose CL6B affinity chromatography, the purity of expressed product reached above 90%. Biological assays proved that the expressed product could stimulate the proliferation of HUVEC. CONCLUSION: hVEGF165 was successfully expressed. The study opened up a wide prospect for the application of VEGF165 in the prevention and treatment of ischemic heart disease and other tissue ischemic diseases such as secondary arterial occlusion in limbs.

[Changes in the activity of platelet L-arginine/nitric oxide pathway in hypercholesterolemia patients].

OBJECTIVE: To investigate the changes of the platelet L-arginine (L-Arg)/nitric oxide (NO) pathway in hypercholesterolemia patients. METHODS: The platelet NO production in 21 hypercholesterolemia (HC) patients and 26 normal individuals was assayed by spectrum, the NOS activity and the transportation of L-Arg were determined by (3)H-labelled L-Arg. RESULTS: The NO production in hypercholesterolemic platelets [(24.06 +/- 3.70) nmol/10(8) platelets] was decreased significantly as compared with normal controls [(28.39 +/- 4.45) nmol/10(8) platelets] (P < 0.01), and so did the NOS activity [(1.46 +/- 0.47) pmol/10(8) platelets] vs (1.81 +/- 0.50) pmol/10(8) platelets]. The transportation of L-Arg by hypercholesterolemia platelets also decreased significantly than that by normal controls. The Vmax in the former [(46.84 +/- 3.39) pmol x (10(7) platelets)(-1) x min(-1)] was significantly lower than that in the latter [(53.89 +/- 3.45) pmol x (10(7) platelets)(-1) x min(-1)]. There was no difference between the two groups in K(d) (P > 0.05). CONCLUSIONS: The activity of the L-arg/NO pathway in hypercholesterolemia platelets is decreased significantly as compared with normal controls, implying that it is probably one of the reasons for the hyperactivity of hypercholesterolemia platelets.

[The study of low dose radiation inducing hormesis effect on hematopoietic system].

OBJECTIVE: To study the hormesis effect on hematopoietic system induced by low dose radiation. METHOD: CFU-GM and BFU-E were cultured in methylcellulose semi-solid culture system, levels of GM-CSF and IL-3 were assayed by ELISA and mRNA levels of GM-CSF, G-CSF, IL-3 by in situ hybridization, narrow line hybridization and Northern blot. RESULTS: (1) The in vitro yields of CFU-GM and BFU-E from radiated mice was higher than that from the control. (2) The serum protein level of GM-CSF increased obviously than that of controls; (3) mRNA levels of GM-CSF and G-CSF were also increased. CONCLUSION: There is hormesis effect on hematopoietic system induced by low dose radiation, which may be related to the increasing of cytokines.

[Study on specificity of IgM antibody response in patients with coxsackie virus B infection].

OBJECTIVE: To study the specificity of IgM antibody response in patients with Coxsackie virus B infection. METHODS: A Coxsackie virus B specific immunoblot assay was established for detection of CVB IgM antibody response in patients with recent CVB infection. RESULTS: Of 28 patients' serum samples, positive by ELISA captured CVB IgM test, analysis results of specific IgM response defined by immunoblot assay revealed VP1 protein specific which was crossly reactive among CVB group. CONCLUSION: This finding suggested that VP1 protein in Coxsackie virus B group potentially functioned as a predominant antigen inducing detectable IgM antibody following CVB infection.

[Differential expression of cyclins D1, E and A in human breast diseases].

OBJECTIVE: To understand with greater clearness the effect of overexpression of cyclins gene and the potential implications of it for tumorgenesis in breast. METHODS: We assayed the expression of cyclins D1, E and A in different types of human breast diseases by immunohistochemical staining. RESULTS: Significant difference was seen among the malignant tumor, benign tumor and dysplasis(P < 0.001). The expressive intensity of above three kind of cyclins in malignant tumor was the highest, that in benign tumor was higher, and that in dysplasis was low. CONCLUSION: These data suggest that the expressive intensity of cyclins gene may serve as an indicator for malignant intensity of tumors. The expressive ratio difference of cyclins E and A between infiltrating ductal carcinoma and infiltrating lobular carcinoma may imply that there are different mechanisms involving the occurrence of different histological types of breast tumor.

[Effects of psychosocial factors on recurrent aphthous ulcer].

OBJECTIVE: The aim of this study is to investigate the effects of patients' psychosocial background on the recurrence of aphthous ulcer (RAU). METHODS: A total of 105 patients with RAU and 100 normal subjects of the control group were evaluated for psychosocial background. All subjects were required to complete the EPQ questionnaire, the SCL 90 questionnaire, and the enquired data were analyzed by using chi-square test. RESULTS: Special personality and social factors in RAU patients might be reasons leading to the occurrence of this disease (P < 0.01). CONCLUSION: Psychosocial disorder caused by stresses of social life events on the people with special personality could impact the occurrence of RAU.

[Cloning sequencing and expression pattern, functional analysis of nifA gene in Azospirillum brasilense Yu62].

The nifA gene of Azospirillum brasilense Yu62 was cloned and sequenced. The expression of nifA gene was investigated in wild type strain Azospirillum brasilense Yu62. The results show that expression of nifA gene is not repressed by ammounium and oxygen completely. But the expression of Yu62 nifA gene is different from that of strain Sp7 nifA gene. Expression of Yu62 nifA seems more sensitive to oxygen than that of Sp7 nifA which shows the highest expression in condition of aerobic, while the Yu62 nifA gene shows the highest expression in the condition of microaerobic. The regulation of NifA protein activity by ammonia and oxygen was investigated. Results showed that the NifA protein is repressed by ammonia, 1 mmol/L NH4Cl can inhibit activity of NifA protein completely. Oxygen concentration affects activity of NifA protein. NifA protein is highly active in 0.4%-0.5% O2.

[Cloning and identification of deltamethrin-resistance or susceptibility associated genes of Culex pipiens pallens].

OBJECTIVE: To obtain deltamethrin-resistance or susceptibility associated genes of Culex pipiens pallens. METHODS: The differentially expressed genes were obtained by suppression subtractive hybridization (SSH), and identified by cDNA microarray and reverse Northern blotting. RESULTS: 523 and 286 clones were selected respectively in the two directional SSH. 155 and 42 genes were respectively expressed 2-3 and > 3 times higher in the insecticide-resistant strain than in the susceptible strain; 15 and 9 genes were respectively expressed 2-3 and > 3 times higher in the susceptible strain than in the resistant strain. There were 2 genes only expressed in the insecticide-resistant strain. 51 three times differentially expressed clones and 2 specially expressed clones were sequenced. 44 sequences were obtained which belong to 13 new genes. There were 8 over-expressed genes in resistant strain, 7 of which were similar respectively to mitochondrion rRNA gene, 60S ribosomal protein gene, 40S ribosomal protein S4 gene, trypsin gene, chymotrypsin A gene, ospin gene, and 16S ribosomal RNA gene. There were 5 over-expressed genes in susceptible strain, 2 of them being similar with 40S ribosomal protein S29 gene and myosin regulatory light chain 2 gene. In addition, 2 genes specially expressed in resistant strain were similar respectively to glycogen branching enzyme gene and ribosomal protein 46 gene. CONCLUSION: The differentially expressed genes may be associated with deltamethrin-resistance or susceptibility of Culex pipiens pallens.

[Analysis of the mitochondria-related protein of Schistosoma japonicum and its antigen epitopes].

OBJECTIVE: To sequence the cloned gene Sj338 and to identify the encoded protein and its antigen epitopes. METHODS: The Sj338 gene fragment obtained from adult S. japonicum cDNA library amplified by PCR method was subcloned into pGEM-T vector for sequencing. The sequence of nucleotides and the characteristics of the encoded protein were analyzed by DNASIS Program and Goldkey DNA and Protein Analytical Program, and then the homology of the amino acid sequence was searched on the BLAST net. RESULTS: The cloned rSj338 gene was demonstrated to be 487 bp containing one 459 bp ORF, encoding a protein consisted of 153 amino acids with a molecular weight of 17.6 kDa. The amino acid sequence of the recombinant protein rSj338 shared 46% identity with that of the corresponding part of human mitochondrial import receptor and 44% identity with that of the Rattus sp. mitochondrial precursor receptor. The possible antigen epitopes were predicted within the peptide fragments of 26-32 aa, 37-46 aa and 147-151 aa. CONCLUSION: The protein encoded by rSj338 gene fragment might be the mitochondria-related protein of Schistosoma japonicum.

[Target transformation factor analysis for simultaneous determination of Fe3+ and Al3+ in rare earth samples].

Combined with chromazurol-S-CTMAB-acetone high sensitive system, iterative target transformation factor analysis (IT-TFA) is used to determine simultaneously the trace amount of Fe3+, Al3+ in synthetic samples and rare earth samples. Abstract factor analysis (AFA), the first stop in the method, decomposes the data matrix into abstract factors. The number of factors needed to account for the data to within experimental error is equal to the number of absorbing components in unknown samples. Having determined the number of factors, target factor analysis (TFA) is used to identify the presence of suspected components contributing to unknowns samples. The identification process involves using the normal projection matrix of row vectors to target test for suspected factors with similar coefficient (Q) as target test criterion. The normal projection matrix of column vectors is used to predict the concentration in unknown samples. The results indicate that the recoveries and accuracy are satisfactory.
