Social support, reciprocity, and well-being.

Systematic random-sampling procedures were used to gather a sample of 191 community residents in Kaohsiung city, Taiwan, and survey them regarding (a) amount of social support given and received; (b) perceived reciprocity of support in relationships with family members, friends, and colleagues; (c) negative affect; and (d) psychological symptoms. Extraversion and social desirability were also measured. Both receiving and giving support were related to negative affect after controlling for the effects of extraversion and social desirability. These two personality factors also substantially masked the negative impact of support on psychological symptoms. Reciprocity of support within the family domain was related to well-being. Individual differences in support exchanges were noted, and women received more support than men.

Calcium signaling in transgenic mice overexpressing cardiac Na(+)-Ca2+ exchanger.

We have produced transgenic mice which overexpress cardiac Na(+)-Ca2+ exchange activity. Overexpression has been assessed by Western blot, Northern blot, and immunofluorescence. Functional overexpression was analyzed using membrane vesicles and isolated ventricular myocytes. In whole cell clamped myocytes dialyzed with 0.1-0.2 mM Fura-2, the magnitude of ICa and Ca2+i-transient triggered by ICa or caffeine were not significantly different in transgenic vs. control myocytes. In transgenic myocytes, activation of ICa, however, was followed by a large slowly inactivating transient inward current representing INa-Ca. This current depended on Ca2+ release as it was abolished when sarcoplasmic reticulum (SR) Ca2+ was depleted using thapsigargin. Cai-transients triggered by rapid application of 5 mM caffeine, even though equivalent in control and transgenic myocytes, activated larger INa-Ca (approximately 5 pA/pF at -90 mV) in transgenic vs. control myocytes (1.5 pA/pF). The decay rate of caffeine-induced Ca2+i-transient and INa-Ca was 2.5 times faster in transgenic than in control myocytes. 5 mM Ni2+ was equally effective in blocking INa-Ca in control or transgenic myocytes. In 9 out of 26 transgenic myocytes, but none of the controls, Ca2+ influx via the exchanger measured at +80 mV caused a slow rise in [Ca2+]i triggering rapid release of Ca2+ from the SR, SR Ca2+ release triggered by the exchanger at such potentials was accompanied by activation of transient current in the inward direction. In 2 mM Fura-2-dialyzed transgenic myocytes caffeine-triggered Cai-transients failed to activate INa-Ca even though the kinetics of inactivation of ICa slowed significantly in caffeine-treated myocytes. In 0.1 mM Fura-2-dialyzed transgenic myocytes 100 microM Cd2+ effectively blocked ICa and suppressed Cai-transients at -10 or +50 mV. Our data suggests that in myocytes overexpressing the exchanger, the content of intracellular Ca2+ pools and the signaling of its release by the Ca2+ channel vis-à-vis the Na(+)-Ca2+ exchanger were not significantly altered despite an up to ninefold increase in the exchanger activity. We conclude that the exchanger remains functionally excluded from the Ca2+ microdomains surrounding the DHP/ryanodine receptor complex.

Fat embolism with the use of intraosseous infusion during cardiopulmonary resuscitation.

The objective of this prospective study was to assess the incidence and magnitude of fat emboli after cardiopulmonary resuscitation and intraosseous infusions. An animal laboratory at a university center was used to study 33 mixed-breed piglets. The piglets underwent hypoxic cardiac arrest followed by chest compressions and mechanical ventilation for a minimum of 30 minutes. The animals were divided in groups: group 1 (n = 5), which had no intraosseous cannulas, group 2 (n = 6), which had intraosseous cannulas with infusion, groups 3 (n = 6), 4 (n = 6), and 5 (n = 8), which had intraosseous cannulas with infusion of epinephrine, normal saline, and sodium bicarbonate respectively, and group 6 (n = 2), which was a sham group with no intraosseous cannulas and no cardiopulmonary resuscitation. At cessation of cardiopulmonary resuscitation, representative lung samples were collected from upper and lower lobes of each lung and observed for fat globules and bone marrow elements. Fat globules were seen in the peribronchial blood vessels and intravascular areas throughout all lung fields of groups 1 through 5. There was no difference in appearance or distribution of fat globules among the 5 treatment groups. Analysis of variance showed no statistical significance (P < 0.05) within or among groups 1 through 5. The use of the intraosseous cannula for infusion of emergency drugs and fluids did not increase the magnitude of fat embolization over cardiopulmonary resuscitation alone in this animal model. The benefits of using this procedure in critically ill children as a means of rapid vascular access for resuscitation is well established. However, the risk of fat embolism in this population needs further study.

Carbamazepine inhibition of neuronal Na+ currents: quantitative distinction from phenytoin and possible therapeutic implications.

Carbamazepine and phenytoin, two of the most commonly prescribed antiepileptic drugs, have been proposed to share a similar mechanism of action by use-dependent inhibition of Na+ channels. The proposed similar mechanism of action, however, cannot explain the common clinical experiences that the two drugs are different; in some patients, one drug may be more effective than the other. This may occur even when optimal therapeutic concentrations are reached with both medications in plasma or the cerebrospinal fluid. In this study, we show that the action of the two drugs on Na+ channels are quantitatively very different. The affinity between inactivated Na+ channels and carbamazepine (apparent dissociation constant approximately 25 microM) is approximately 3 times lower than that of phenytoin, yet the binding rate constant of carbamazepine onto the inactivated Na+ channels is approximately 38,000 M(-1)/sec(-1), or approximately 5 times faster than that of phenytoin. It is speculated that carbamazepine may be more effective than phenytoin in treating seizures whose ictal depolarization shift is relatively short, whereas a better response to phenytoin may imply abnormal discharges characterized by more prolonged depolarization.

A transcriptional insulator element, the su(Hw) binding site, protects a chromosomal DNA replication origin from position effects.

Eukaryotic chromosomes are organized into domains of activity for both transcription and DNA replication. Transcriptional "border," or "insulator," elements have been implicated in mediating the organization of transcriptional domains. However, the DNA sequence elements which might demarcate domains of DNA replication activity are unknown. su(Hw) protein binding sites [su(Hw)BSs] are potent transcriptional insulator elements which can block enhancer action, as well as positive and negative chromosomal position effects. Here we report that flanking su(Hw)BSs can also create a chromosomal domain permissible for activity of the chorion gene DNA replication origin. During Drosophila oogenesis the chorion (eggshell) gene loci are amplified approximately 80-fold through repeated initiation of DNA replication. The cis-acting amplification control element, on the third chromosome (ACE3), is required for high levels of amplification initiating at the nearby major origin of replication, Ori-beta. A transgenic chorion locus construct containing ACE3 and Ori-beta was able to amplify but was extremely sensitive to position effects: only 7 of 21 independent insertions amplified >10-fold. The inclusion of flanking su(Hw)BSs in the construct dramatically protected DNA replication from position effects: 31 of 31 insertions now amplified >10-fold, and this protection was reduced in a su(Hw) mutant background. Amplification was equal on both sides of the su(Hw)BS, demonstrating that replication fork passage is not significantly impeded by these sites. Inclusion of only a single su(Hw)BS in the construct did not detectably protect the chorion gene DNA replication origin from position effects.

Growth factor-mediated K+ channel activity associated with human myeloblastic ML-1 cell proliferation.

ML-1 cell proliferation is dependent on the presence of serum growth factors. Removing serum from the culture medium results in growth arrest and promotes differentiation. In this study, we found that a 4-aminopyridine-sensitive K+ channel was highly expressed in proliferating ML-1 cells and significantly diminished in G1-arrested ML-1 cells induced by serum deprivation but was restored within 30 min in these cells with addition of 10% fetal bovine serum (FBS) or 5 ng/ml epidermal growth factor (EGF). Intracellular adenosine 3',5'-cyclic monophosphate (cAMP) levels, but not guanosine 3',5'-cyclic monophosphate, were significantly increased in serum-deprived cells stimulated by FBS or EGF, and the effects of FBS and EGF on the channel activation were mimicked by exogenous cAMP. In inside-out patches, K+ channel activity was significantly increased by the cAMP-dependent protein kinase catalytic subunit, whereas the effect of EGF on K+ channel activation was blocked by Rp-8-(4-chlorophenylthio)adenosine 3',5'-cyclic monophosphothioate. Together, our results demonstrate that serum growth factors stimulate K+ channel activity in proliferation of ML-1 cells through protein kinase-induced phosphorylation and suggest an important molecular mechanism for serum growth factor-stimulated mitogenesis in ML-1 cells.

Estrogen relaxation of coronary artery smooth muscle is mediated by nitric oxide and cGMP.

Estrogens are proposed to exert protection against cardiovascular disease, and evidence now suggests that this protection involves a direct vasodilatory effect. We have shown previously that estrogen relaxes endothelium-denuded porcine coronary arteries by opening the large-conductance calcium- and voltage-activated potassium (BKCa) channel of myocytes through guanosine 3',5'-cyclic monophosphate (cGMP)-dependent phosphorylation (35). The present study confirms these results and now demonstrates that this mechanism involves production of nitric oxide (NO). S-nitroso-N-acetylpenicillamine (SNAP), an NO donor, or 8-bromo-cGMP mimicked the effect of estrogen on BKCa channels. Furthermore, inhibition of NO synthase (NOS) attenuated estrogen- or tamoxifen-induced BKCa-channel activity, and this effect was disinhibited by L-arginine. Inhibition of guanylyl cyclase activity blocked the stimulatory effect of estrogen, SNAP, or L-arginine on BKCa channels. Furthermore, 17 beta-estradiol stimulated accumulation of nitrite and cGMP in coronary myocytes. Therefore, we propose that the vasodilatory effect of estrogen on the coronary circulation is mediated by NO. A portion of the beneficial cardiovascular effects of estrogen may be attributed to relaxation of vascular smooth muscle by a process that involves NO- and cGMP-dependent stimulation of BKCa channels.

Transesophageal echocardiography in rats using an intravascular ultrasound catheter.

In vivo assessment of cardiac structure and function in small animals is an important experimental goal, but currently available techniques have significant limitations. A commercially available intravascular ultrasound (IVUS) system was adapted to perform transesophageal echocardiography (TEE) in rats. Twelve Sprague-Dawley rats (270-370 g) were anesthetized with intraperitoneal pentobarbital sodium. A 4.3-Fr, 30-MHz or an 8-Fr, 20-MHz IVUS catheter was inserted into the esophagus to obtain long-axis views of the aortic arch, short-axis views of the ascending aorta, and long-axis views of the pulmonary artery. A preshaped, 8-Fr, 20-MHz catheter was used to obtain short-axis images of the left ventricle (LV) at the midpapillary muscle level, which were used to measure LV diastolic and systolic dimensions (diameters) and to calculate LV mass and fractional shortening. Measurements by TEE were compared with those obtained by transthoracic echocardiography in 6 of 12 rats. Postmortem, the LV was weighed to determine actual LV mass. The correlation coefficients between TEE- and transthoracic echocardiography-calculated LV mass and actual LV mass were 0.94 and 0.88, respectively, and had a good agreement with actual LV mass. Inter- and intraobserver variability of TEE measurements was <10%. IVUS instrumentation may offer an alternative technique for the accurate, serial assessment of LV dimensions, mass, and systolic function and a means of imaging the great vessels in small laboratory animals.

Design of a haptic stick interface as a pilot's assistant in a high turbulence task environment.

A study involving 8 Air Force pilots was conducted to examine the efficacy of a force-reflecting joystick to improve performance during a simulated landing task in wind turbulence. By adding certain force characteristics to a joystick, it was of interest to see if performance may change, different control effort may be utilized, and workload measures may be altered based on the joystick utilized. The main results show that certain performance measures significantly improved by having the force reflection condition on. The implications of this study are that in certain types of precision tracking tasks, subjected to external disturbances, the addition of the force characteristics to the joystick can significantly improve performance, result in less effort for control, and lower subjective workload.

[Pathological analysis and clinical importance of invasion and neck nodal metastasis in squamous cell carcinoma of the tongue].

To determine the absolute and relative of histopathological parameters and conventional indicators in correlation with invasion and nodal metastasis, we performed both univariate and multivariate statistical analysis in 92 patients with squamous cell carcinoma of the tongue by spss/pc+ statistical system. Mode of invasion (MI) and peritumoral lymphocytic infiltrating density (PLID) were significant and independent factors by multivariate analysis (P = 0.000, P = 0.009 respectively), although univariate analysis showed that MI, PLID, T-stage and Grading were significantly associated with nodal metastasis. A correlation exists only between T-stage and MI by multivariate statistical test (P = 0.045), while, PLID, T-stage and Grading were significant parameters by univariate analysis. A highly dense PLI around cancer nest was observed which blocked nest from tissue and capillary lymph vessels.

[Investigation on hepatitis G virus (HGV) infection among different populations in Shenzhen].

A reverse transcription polymerase chain reaction assay with nested primers (RT-nested PCR) deduced from the 5' untranslated region of HGV genome was established to detect HGV RNA. By using this assay, we tested the serum samples of 106 paid blood donors, 80 intravenous drug users and 168 cases with viral hepatitis in Shenzhen, China. The positive rates of HGV RNA were 8.5%, 46.3% and 7.7%, respectively. The positive rates of HGV RNA in 61 cases with chronic hepatitis B and 33 cases with chronic hepatitis C were 8.2% and 21.2%. Among the 33 cases with chronic hepatitis C, the positive rate of HGV RNA in 15 cases infected with HCV by blood or blood products was 40.0%, compared with 1.7% in 18 cases without exposure to blood or blood products (P<0.05). The results of this study suggest that HGV infection is common in paid blood donors; the intravenous drug user is the population at high risk for HGV infection; the patients with post-transfusion chronic hepatitis C are often supperinfected with HGV.

[The method for studying kinetics of formation reaction of complex in dimeric system].

The method for studying kinetics of formation reaction of complex in aggregation system was reported in this paper. The concentration of more than one species was calculated by improved method for calculating dimeration constants. Kinetics of reaction between Cu (II) and tri-sulfophthalocyanine (S3Pc) was studied in this way. It shows that the rate of reaction between Cu(II) and S3Pc can be expressed as follows: v = K[Cu][S3Pc]

[The aggregation and kinetics of decomposition reaction of Ga-sulfophthalocyanine].

The aggregation and kinetics of decomposition reaction of Ga-sulfophthalocyanine has been studied by electronic spectroscopy. The effects of pH on the aggregation are discussed. The experimental results show that the rate of decomposition reaction of Ga-sulfophthalocyanine can be expressed as follow: -dcM/dt = 12.279cDcOH-. Where cD and cOH- concentration of GA-sulfophthalocyanine and OH- respectively.

[A resonance Raman spectrometer with UV-visible continuously tunable excitation lines].

A set of resonance Raman spectrometer with excitation lines continuously tunable in 220-970nm, which is a first set in China, has been assembled. The factors influenced on the signal noise ratio (SNR) of spectra were analyzed in detail. The methods to improve the SNR were suggested and the parameters of the spectrometer were optimized. A satisfied Raman spectra can be obtained.

Retroviral mediated gene transfer of Flt3 ligand enhances proliferation and MAP kinase activity of AML5 cells.

Flt3/flk-2 ligand (Flt3-L) co-stimulates and synergizes with cytokines such as granulocyte-macrophage colony stimulating factor, granulocyte colony stimulating factor (G-CSF), interleukin-3 (IL-3), and erythropoietin in the proliferation of bone marrow and cord blood hematopoietic stem and progenitor cells. To study the biological effects of Flt3-L on the Flt3-L responsive AML5 cell line, the retroviral vector L(Flt3-L)SN was constructed based on the vector LXSN, but containing the human Flt3-L cDNA transcriptionally regulated by the Mo-MLV LTR. High-titer amphotropic producer cells that generated 10(6) cfu/mL after shuttle packaging through ecotropic packaging cells were isolated. AML5 cells were cultured overnight with L(Flt3-L)SN retroviral supernatant, 8 micrograms/mL polybrene, and 100 U/mL G-CSF, and expanded 1 week in medium with G-CSF. Transduced cells were selected in medium containing 0.4 mg/mL G418 and then in medium with 1.0 mg/mL G418. Retroviral mediated gene transfer in G418-resistant cells was confirmed after amplification by PCR of neo-specific sequences in genomic DNA. Northern blot analysis demonstrated L(Flt3-L)SN mRNA expression. Soluble Flt3-L was undetectable (< 100 pg/mL) by ELISA assay of conditioned medium from transduced cells, but Flt3-L was detected on the surface of AML5 cells by FACS analysis. Cells were plated in colony assay with and without 100 ng/mL Flt3-L, 100 U/mL G-CSF, and the combination. Gene transfer or growth factor treatment increased somewhat the clonogenicity of the nontransduced AML5 cells. More strikingly, L(Flt3-L)SN and each growth factor combination greatly increased the size of the resultant colonies such that the size of colonies from AML5/Flt3-L cells without added growth factor approximated that of the AML5 cells stimulated by exogenous soluble Flt3-L. Moreover, MAP kinase activity in L(Flt3-L)SN-transduced cells cultured without soluble Flt3-L was increased to the level induced in control cells by soluble Flt3-L. These results indicate that retroviral mediated gene transfer and autologous expression of the Flt3-L enhances growth and intracellular signaling of AML5 cells, information that should be of value for studying the effects of Flt3-L on immature subsets of primary hematopoietic stem and progenitor cells.

Early life experiences linked to diabetes mellitus: a study of African-American migration.

African Americans experienced massive internal migrations that shifted more than 6 million Southern-born blacks to other sections of the United States over the past century, a trend that only recently has been reversed. Whenever mass migration takes place, there is an opportunity to examine the role of the native and relocated environments in the development of disease. This article examines those relationships for diabetes mellitus, a group of diseases that disproportionately affect African-Americans relative to other racial and ethnic groups in the United States. Age-specific and age-adjusted rates with 95% confidence intervals were calculated for males and females for combinations of five regions of birth and four regions of residence at time of death. Southern-born males had statistically significantly higher death rates from diabetes than did their counterparts who died in the same regions in 9 of 16 comparisons. For females, those born in the South had statistically significantly higher rates in 15 of 16 comparisons. The results of this study indicate that place of birth and early life experiences are statistically associated with diabetes mortality among African Americans regardless of place of residence at time of death.

Hassles, appraisals, coping and distress: a closer look at the stress process.

In this exploratory study, important components of the stress process were under close scrutiny. Subjects self-administered a computer programmed interview, which started with a self-selected daily hassle and went on to examine the person's appraisals of the incident, actions taken towards it and its impact on the person. Its Personal evaluations of coping efficacy and personal decisions of coping consistency were also probed. Results suggested that daily hassles were recurrent and minor in nature; and people tended to make mixed and combined appraisals of these hassles. Perceived coping efficacy affected perceived stress impact and future coping consistency. It seemed that there was a self-monitoring mechanism in the coping process, and people's coping efforts tended to be rather economical.
