[Slot blot analysis of metastasis suppression gene nm23 expression in human lung cancer].

To investigate the role of metastasis suppression gene (nm23) in the development and progression of human lung cancer, the mRNA expressions of nm23-H1 and nm23-H2 genes in a series of pulmonary tissues collected at various sites and with different properties were studied with slot blot hybridization. According to the observations on the normal pulmonary tissues, benign lesions, para- and non-cancer tissues, primary cancer and metastatic lymph nodes, there was a tendency of mRNA expression reduction of nm23-H1 and nm23-H2 genes. Among them, nm23-H2 mRNA expressions in lung cancer tissues were significantly decreased when compared with normal pulmonary tissues (P < 0.05), and nm23-H1 and nm23-H2 gene expressions in metastatic lymph nodes were both reduced, compared with those in normal pulmonary tissues (P < 0.05). There was no significant correlation between nm23 gene expression and lymph node metastasis in lung cancer. The results implied that reduction of nm23 gene expression might be associated with the development of lung cancer, but no evidence of metastasis suppression by nm23 gene was revealed in this study.

[An analysis of antenatal psychological status and its related factors].

OBJECTIVE: To provide basis for antenatal psychological counselling by investigating psychological status and related factors of parturients. METHODS: Analysing psychological status of 100 primigravida by self-made questionnaires. Symptom checklist go (Scl-go), HAMD and HAMA. RESULTS: Single-factor analysis showed: six factors, that were social behavior, mood controlling, history of abortion, relation with parents, psychological preparation for delivery and family roles, are correlated with psychological status of pregnant women. Multi-factor analysis indicated: 3 factors i.e. relation with parents, psychological preparation for delivery and social behavior were positively correlated with psychological status of parturients. CONCLUSIONS: Relation with parents, preparation for delivery and social behavior are the most important factors to influence antenatal psychological status and useful references for antenatal psychological counselling.

[Changes of hemodynamics, oxygen delivery and oxygen consumption during anesthesia induction with propofol in patients with rheumatic mitral stenosis].

Changes of hemodynamics, oxygen delivery and consumption were studied in 20 patients with rheumatic mitral stenosis, undergoing mitral valve replacement, anesthesied induction with propofol(Group P) or midazolam(Group M). The results showed that hemodynamic values were decreased after intravenous propofol, and recovery after tracheal intubation. Hemodynamic values also decreased and last to 2 min after tracheal intubation in Group M. The reduction degree of HR in Group P were more, the degree of SVRI,. LVSWI, RVSWI less than those in Group M respectively. Oxygen consumption decreased more than delivery. It is suggested that induction of anesthesia with propofol in patients with rheumatic mitral stenosis is safe, and may be advantageous than midazolam.

[Effects of propofol and etomidate on cerebral oxygen supply and demand balance during middle hypothermia cardiopulmonary bypass].

UNLABELLED: Effects of propofol(Group P) and etomidate(Group E) on cerebral oxygen supply and demand balance during middle hypothermia cardiopulmonary bypass(CPB) were studied in twenty patients undergoing cardiac valve replacement. Arterial blood and jugular bulb blood were taken to analyse blood gas, and cerebral oxygen content were caculated. RESULT: During hypothermia, jugular bulb oxygen saturation(SjO2), jugular bulb blood oxygen content(CjO2) increased; cerebral oxygen extraction rate(CEO2) decreased significantly in two groups(P < 0.05). During rewarming SjO2, CjO2 were significantly decreased, CEO2 increased compared with that before rewarming and hypothermia in two gbroups (P < 0.05). In Group P, SjO2 and CjO2 were higher, CEO2 lower than those in Group E respectively during rewarming (P < 0.05). CONCULUSION: The activity of propofol reduced cerebral metabolism, cerebral oxygen consumption and CEO2 is better than that of etomidate.

[Treatment of malignant germ cell tumors of the ovary: a clinical analysis of 233 cases].

OBJECTIVE: To evaluate the treatment methods of malignant germ cell tumors of the ovary. METHODS: Two hundred thirty three patients with malignant germ cell tumors of the ovary treated during 1958 through 1993 were retrospectively analyzed. The number of patients in different stage of the disease was as follows: stage I, 94; stage II-IV, 43; recurrence or metastasis, 96. Unilateral salpingo-oophorectomy was performed in 78 patients, or total abdominal hysterectomy and bilateral salpingo-oophorectomy with or without omentectomy and appendectomy was performed in 151 cases, and biopsy in 4 cases. Operation alone was done in 17 cases, operation plus radiotherapy with or without chemotherapy in 65, operation plus chemotherapy in 151 cases. RESULTS: Of the 233 patients 127 were disease-free. The number of survivors in stage I patients was 78, in stage II-IV was 17. There were 32 cases with recurrence and metastasis. The overall 5-year survival rate was 55.6%. The 5-year survival rate differed between dysgerminoma and nondysgerminomatous tumurs being 84.2% for the former and 44.6% for the latter. In patients with nondysgerminomatous tumors, chemotherapy with PVB and BEP regimens gave a 5-year survival rate of 66.0% and 73.3%, respectively. CONCLUSION: Malignant germ cell tumors of the ovary is very sensitive to chemotherapy. The PVB and BEP regimens result in excellent response in these patients. Complete remission and preservation of fertility can be obtained in patients with early tumors. Preoperative chemotherapy and postoperative adjuvant chemotherapy can improve survival in patients with advanced tumors.

[Northern blot analysis of nm23 gene expression in human lung cancer].

OBJECTIVE: To investigate the role of nm23 gene expression in lung cancer. METHODS: Forty human lung cancer tissues and 19 non-cancerous pulmonary tissues were studied for their nm23-H1 and nm23-H2 mRNA expression with nonradioactive Northern blot hybridization. Correlation of nm23 mRNA expression with clinical features of lung cancer was analyzed. RESULTS: The mRNA expression of nm23-H2 gene in poorly differentiated squamous-cell carcinoma was significantly decreased compared to that in moderately differentiated squamous-cell carcinoma. The mRNA expression of nm23-H1 and nm23-H2 genes in small cell lung cancer was decreased compared to that in squamous-cell carcinoma. No significant difference in nm23 mRNA expression was observed in lung cancers with and without lymph node metastasis, nor was there significant difference in patients with lung cancers in different stages. CONCLUSION: The mRNA expression of nm23 gene is correlated with the degree of differentiation of lung cancer, but there is no evidence of metastasis suppression effect by nm23 gene.

[Cardiovascular protective effects and NO-mediated cerebrovasorelaxant effects of extract of ginkgo biloba leaves].

OBJECTIVE: To examine the anti-free radical related cardiovascular protective effects and NO-mediated cerebrovasorelaxant effects of extract of the leaves of Ginkgo biloba(EGb) in isolated preparations. METHODS: Experiments on the anti-free radical related cardiovascular protective effects were performed in rabbit Langendorff heart and isolated aortic rings damaged by diphenyl-picryl hydyazyl(DPPH). Protective effects of EGb were determined by comparing the results of EGb pretreated group with the DPPH-injured controls. Cerebrovasorelaxant effects of EGb were examined in ring preparations of isolated porcine basilar artery in vitro using tissue bath techniques. RESULTS: EGb protected the isolated rabbit heart from the DPPH-injury to the cardiac contractility and the aortic endothelium from DPPH-attenuated ACh-induced relaxation. EGb concentrationde dependently relaxed the basilar artery and this effect was more significant in endothelium intact rings than those endothelium denuded rings. EGb enhanced the TNS-induced relaxation in basilar artery and this effect was abolished by pretreatment of N-nitro-L-arginine or tetrodotoxin. CONCLUSIONS: EGb exerts cardiovascular protection against DPPH-impaired cardiac contraction in rabbit isolated heart and endothelium-dependent relaxation in the aortic ring of rabbit in vitro. EGb relaxes porcine basilar artery and enhances the TNS-induced relaxation via a NO pathway.

[Detection of TCR gamma gene in acute lymphoblastic leukemia after remission and its clinical significance].

OBJECTIVES: To investigate the quantitative evaluation of chemotherapy efficiency in patients with acute leukemia and its clinical significance. METHODS: Clonal rearrangement of T-cell receptor gamma gene in ALL was detected quantitatively by using competitive PCR. RESULTS: One handred and fifty-eight samples were collected from 54 childhood ALL. The median follow-up duration was 33.5 months. At remission, the amount of leukemic cells in 36 cases was less than 1 x 10(-5), and in 18, was (354 +/- 327) x 10(-5). The efficiency of induction therapy was related significantly to the amount of the detectable residual leukemic cells in the early remission phase and to clinical relapse. In some patients in remission, the amount of leukemic cells persistently under 1 x 10(-5) without clinical relapse. CONCLUSION: Quantitative evaluation of the chemotherapy efficiency is helpful for individualized treatment.

[The role of mast cells in myocardial fibrosis in SHRsp].

OBJECTIVE: To study the role of mast cells in hypertensive myocardial fibrosis. METHODS: The myocardial collagen and the degree of myocardial fibrosis were assessed using Masson's trichrome staining and image analysis plus toluidine blue as well as proliferating cell nuclear antigen (PCNA) staining and transmission electronic microscopy for SHRsp, aged 34 weeks (S group, n = 6). Besides, age and sex-matched WKY (W group, n = 5) were used as the controls. RESULTS: MC count increased 3.9 folds and PCNA immunostaining positive cells (PCNAP) increased 3.9 fold (both P < 0.0001) in S vs W group. MC were positively correlated with the interstitial collagen volume fraction (ICVF), perivascular collagen area/vascular lumenal cross area (PVCA/VLCA), and PCNAP (r = 0.47, P < 0.05; 0.59, P < 0.001; 0.68, P < 0.001 respectively). Electromicroscopy showed specific piecemeal degranulation of the mast cells, locating between the praliferative interstitial collagen fibers. CONCLUSION: The findings suggested that MCs play a role in certain degree to the myocardial fibrosis present in SHRsp.

[Study on endothelin-1 positive expression and quantitative analysis in lung cancer].

OBJECTIVE: In order to study endothelin-1 (ET-1) positive expression in lung cancer and the relationship between the ET-1 quantitative analysis and the types, grades of lung cancer. METHOD: ET-1 positive expression and quantitative analysis were detected using avidin-biotin-peroxidase complex method and image analysis technology. RESULT: The ET-1 positive expression were mainly located in the cytoplasm in 104 cases with various types of lung cancer. The positive rate of ET-1 in adenocarcinoma, squamous-cell carcinoma and large-cell-lung cancer were 71.4%, 57.1% and 40.0% respectively. The positive rate of ET-1 in small-cell-lung cancer was 21.4%, significantly lower than others. The results of image analysis on adenocarcinoma and squamous-cell carcinoma showed that the lower lung cancer differentiation, the higher ET-1 quantitative. CONCLUSION: There is ET-1 positive expression in all of types of lung cancer, but there is a high ET-1 positive expression in adenocarcinoma and squamous-cell carcinoma. The results of image analysis indicated that ET-1 quantitive expression was somehow related to the differentiation degree of neoplasm, so ET-1 quantitative analysis may be as a monitoring index of adenocarcinoma and squamous-cell carcinoma growing.

[Effects of stimulation at different areas of nucleus raphe dorsalis on genioglossus and diaphragm activities].

Effects of electrical and chemical stimulation of the dorsal and ventral areas of the nucleus raphe dorsalis (dNRD and vNRD) on genioglossus and diaphragm activities were observed in 48 urethane-anaesthetized and vagotomized rabbits. (1) Long train electrical stimulation at the dNRD facilitated genioglossus and diaphragm activities. (2) Long train electrical stimulation delivered to the vNRD excited genioglossus activity but inhibited diaphragm activity. (3) Effects of microinjection of glutamate at the dNRD and vNRD were similar to the effects of electrical stimulation. The above results suggest that excitation of nucleus raphe dorsalis (NRD) increases genioglossus activity and reduces upper airway resistance. The dNRD and vNRD play different roles in modulating diaphragm activity.

[Effects of glycine and strychnine microinjected into unilateral and bilateral Bötzinger complex on phrenic nerve discharges in rabbits].

The effects of microinjection of glycine and strychnine into unilateral and bilateral Bötzinger complex (Böt. C) on phrenic nerve discharges were observed in 34 urethane anaesthetized, vagotomized, paralyzed and artificially ventilated rabbits. The results are as follows: (1) microinjection of glycine into unilateral Böt. C induced an increase of respiratory rate and expiratory phrenic nerve discharges; (2) microinjection of glycine into bilateral Böt. C induced non-rhythmic tonic phrenic nerve discharges with an amplitude 40%-70% of control; strychnine could competitively antagonize the effects of glycine; and (3) microinjection of strychnine into bilateral Böt. C induced a decrease of respiratory rate and the amplitude of phrenic nerve discharges, in addition to prolongation of expiratory duration while inspiratory duration remained unchanged. The above results suggest that the Böt. C plays a crucial role in initiating and sustaining expiration.

[Application of three demensional reconstruction of CT in lateral cranial base surgery].

OBJECTIVE: To study the characteristics of 3D-CT of the lateral cranial base and its application in lateral cranial base surgery. METHODS: Five normal adult subjects and 10 with lateral cranial base lesions were scanned with Philips Tomoscan SR 7000 CT scanner. The scanning ranges extended from the top to the bottom of the temporal bone, including whole lesions, with bone window algorithm and contiguous serial 1.5 mm thin-cut scans in succession in axial plane. The 3D-CT reconstruction was processed in the Easy Viewing CT/MRI workstation. RESULTS: The 3D-CT reconstruction could clearly delineate the foraminan, fissures and important bony marks. The ranges of the lesions, the relationships between the lesions and the structures around them were demonstrated more clearly than 2D-CT. It was of important significance in differential diagnosis for jugular foramen lesions. 3D-CT could be used to imitate operation and design operative plan. CONCLUSION: 3D-CT was of important value in diagnosis of lateral cranial base tumors, and could be used to imitate operation and design operative plan.

[A study on the nutrient physiology of Ophiopogon japonicus (Thunb.) Ker-Gawl].

By means of observing at regular intervals and on tixed plants, the growth stages, organogenetic habits, content of nutrient elements and its changing regularity, storage habit of dry matter, relationship between aerial and subterraneous parts, and fertilizer requirements have been studied for Ophiopogen japonicus.

[Observation on inhibitory effect of Coptis alone and its combination with Scutellaria and Liquorice on the growth of Staphylococcus aureus].

Using MIC as relative index, an observation was made on the growth inhibition of Staphylococcus aureus by Coptis alone or in combination with Scutellaria and or liquorice. The result shows that when Coptis is combined with Scutellaria, the bacteriostatic effect gets weaker for Scutellaria, but remains unchanged for Coptis, and when Scutellaria takes greater percentages in the combination, the effect of Coptis seems to be stronger. When Coptis is combined with liquorice, whatever the respective percentages may be, the effect tends to get weaker. When the three drugs are used in combination, the effect remains unchanged or even gets stronger for Coptis or liquorice, but tends to decrease for Scutellaria. Coptis used singly or in combination with Scutellaria or liquorice is inferior in bacteriostatic effect to the combination of the three drugs. The mechanism is yet to be further studied.
