[Analysis of telomerase activity and telomere length in acute myelogenous leukemia].

OBJECTIVE: To study the changes and significance of telomerase activity and telomere length in acute myelogenous leukemia. METHODS: TRAP-ELISA-PAGE was used to detect telomerase activity, Southern blot to estimate the length of telomere. RESULTS: Telomerase activity was significantly higher in AML(Absorption(A): 2.298 +/- 1.059) than in normal control(A: 0.387 +/- 0.598) and the mean telomere length of AML [(7.6 +/- 2.1) kb] was significantly shorter than that of normal control[(9.3 +/- 1.9) kb]. The alteration of the telomere length was detected mainly in telomerase-positive AML. CONCLUSIONS: It suggested that there was a close relationship between telomerase activity and telomere length in AML. The activation of telomerase might play an important role in the genesis and development of AML and telomere length changes may correlate with the activation of telomerase.

[Leaching and vertical transport of Zirconium-95 in soils].

The leaching and vertical transport of Zirconium-95 in three representational soils (paddy soil, red-yellow earth and sea mud) in Zhejiang province and the influence of salinity on the activities of Zirconium-95 in sea mud were studied with soil-pole installation. The results show that: 1. The content of Zirconium-95 in collected leachate was low after passing through soil column with 250 ml, 500 ml, 750 ml leaching amount. The amount of Zirconium-95 in collected leachate of paddy soil, sea mud and red-yellow earth was about 5.33%-7.68%, 0.77%-1.32% and almost 0 of labelled amount respectively. And with increasing the total amount of leaching water, the content of Zirconium-95 in collected leachate varied indistinctively, which indicated that once Zirconium-95 was absorbed by soils, it was very difficult to desorb. 2. In paddy soil, red-yellow earth and sea mud, 45.99%-50.02%, 96.02%-97.16% and 95.94%-98.01% radioactivity of Zirconium-95 was concentrated in the 0.4 cm upper layer soils respectively, which illustrated that most of Zirconium-95 remained in the surface soils. 3. With the increase of salinity in leaching water, the amount of Zirconium-95 in surface soils (0-0.2 cm) presented obviously increased, which demonstrated that increasing salinity could raise the adsorption rate of Zirconium-95 in soils.

[The influence of HOXB2 anti-sense oligodeoxynucleotides on the proliferation and expression of human umbilical vein endothelial cells].

OBJECTIVE: To explore the effects of HOXB2 anti-sense oligodeoxynucleotides (asodn) on the proliferation and the expression of human umbilical vein endothelial cells (HUVECs). METHODS: Various concentrations of HOXB2 ASODN modified by thiophosphate were transfected into HUVECs by liposome mediation. MTT and RT-PCR methods were employed to determine the influence of different concentrations of ASODN on endothelial proliferation and the expression level of HOXB2 mRNA. RESULTS: After the transfection of HOXB2 ASODN, the endothelial proliferation was inhibited in dose-dependent manner. Simultaneously, the expression level of HOXB2 mRNA decreased significantly. CONCLUSION: HOXB2 might play important roles in the proliferation of endothelial cells.

[Single topotecan or in combination with other chemotherapeutic agents for 18 recurrent advanced ovarian cancer patients].

OBJECTIVE: This study was designed to evaluate the efficacy and toxicity of topotecan hydrochloride, used singly or combined with other drugs in patients with recurrent advanced ovarian cancer. METHODS: Eighteen ovarian cancer patients who had received cytoreductive operation 1 to 3 times and 8 to 18 courses of chemotherapy but still failed in DDP-based(86.3%) and Taxol-based(32.7%) as well as combined DDP and Taxol-based(30.7%) chemotherapy, were divided into 3 groups: 1. Topotecan 1.2 mg/m2 q.d.i.v. for 5 consecutive days every 3 weeks for a total of 6 patients and 14 courses. 2. Topotecan 0.7 mg/m2 q.d.i.v. for 5 consecutive days and cis-platin 20 mg/m2 q.d.i.v. for 3 consecutive days over 4 weeks and 3. Topotecan 0.7 mg/m2 q.d.i.v. for 4 consecutive days and taxol 100 mg/m2 q.d.i.v. for one day as well as cis-platin 20 mg/m2 q.d.i.v. for 3 consecutive days totalling 12 patients and 27 courses. All together 18 courses, median 2.7 courses, were given. RESULTS: All 18 patients were evaluable. PR was achieved in 3 patients with a response rate of 16.7%. The main adverse effects were hematological toxicity, lasting for 3-27 days. Ten (55.6%) patients developed leucocytopenia Grade III-IV which occurred in 16(33.3%) courses. Six(33.3%) patients developed thrombocytopenia Grade III-IV which occurred in 8(16.7%) courses. Six(33.3%) patients developed RBC reduction Grade III which occurred in 8(16.7%) courses. Peripheral nerve inflammation was most serious in the non-hematologic toxicity. CONCLUSION: Topotecan is effective in recurrent advanced ovarian cancer patients who have failed in DDP or/and taxol-based regimes. Topotecan combined with cisplatin and taxol would be effective even if single topotecan has failed. Careful evaluation before treatment is indicated.

[The retinal nerve fiber layer defect and its related clinical features in early primary open-angle glaucoma].

OBJECTIVE: To investigate the distribution of diffuse and local retinal-nerve-fiber-layer defects (RNFLD) and their clinical features in early primary open-angle glaucoma (POAG) and normal-tension glaucoma (NTG). METHOD: The types of RNFLD in 81 cases with POAG and 70 cases with NTG were observed, the distribution of diffuse and local RNFLD were analyzed, and the maximum untreated intraocular pressure (IOP) and the history of optic disc hemorrhage between the two types of RNFLD in the two glaucomatous groups were compared. RESULTS: RNFLD appeared in 78 of 81 cases with POAG, 50 of them were diffuse and 28 were local; 27 of diffuse and 43 of local RNFLD were found in 70 cases with NTG. The IOP was (28.78 +/- 5.41) mm Hg in the diffuse and (26.2 +/- 5.2) mm Hg in the local type in POAG (t = 2.04, P < 0.05), and (19.2 +/- 1.9) mm Hg and (17.8 +/- 2.0) mm Hg, respectively in NTG (t = 2.94, P < 0.05). The incidence of disc hemorrhage was 10/78 and 18/70 in POAG and NTG, respectively (chi(2) = 4, P < 0.05). That was 3/50 in the diffuse and 7/28 in the local type in POAG (chi(2) = 4.22, P < 0.05), and 3/27 and 15/43, in NTG respectively (chi(2) = 4.91, P < 0.05). There were no statistic differences in the diffuse type between the POAG and NTG groups (chi(2) = 0.15, P > 0.05), and neither were in the local type (chi(2) = 1.31, P > 0.05). CONCLUSION: The diffuse RNFLD tends to be distributed in POAG, while the local RNFLD tends to be distributed in NTG. The maximum IOP in the eye with diffuse RNFTD is higher than that in the eye with local RNFLD, either in POAG or NTG group. The occurrence of disc hemorrhage is higher in the eye with local RNFLD than that in the eye with diffuse RNFLD in the two groups respectively. There are differences in clinical features between the two types of RNFLD in early POAG and NTG.

[DNA ploidy analysis and Ki-67 examination on renal neoplasms: investigation of degree of malignancy and biological behavior of small renal tumors].

OBJECTIVE: To investigate the degree of malignancy and the biological behavior of small renal tumors and provide a pathological basis for clinicians in determination of further treatment and prognosis. METHODS: Flow cytometry and immunohistochemical studies were performed on 48 small tumors with diameters less than 3 cm. and 39 renal cell carcinomas with diameters larger than 3 cm in comparison to assess their DNA ploidy status and Ki-67 PR (proliferation rate). RESULTS: Of the 48 small renal tumors, 6 adenoma were all diploid with low PR (< 1%). Aneuploid rate of 42 small renal tumors with clear and granular cell type was 16.7%, showing no statistical difference with the 33.3% aneuploid rate of 39 large renal cell carcinomas. 42 small renal tumors had a lower mean PR than large renal cell carcinoma. DNA ploidy correlated closely with nuclear grade, stage and histological type. There existed a close relation between PR and grade. CONCLUSION: Small renal tumors consist of both adenoma and carcinoma, therefore tumor size can not be used as a criterion in their differential diagnosis. DNA ploidy and Ki-67 PR are important indicators for predicting the biological behavior and prognosis of renal parenchymal tumors.

[Protective effect of huyin decoction on acute hepatic injury induced by CCl4 in mice].

OBJECTIVE: To observe the protective effect of Huyin decoction on acute hepatic injury induced by CCl4 in mice. METHODS: The model of acute hepatic injury induced by CCl4 was used. The levels of ALT and AST in serum were detected. And histopathology of liver was observed. RESULTS: Huyin decoction could decrease ALT and AST in serum and hepatic histopathological injury of the acute hepatic injury mice. CONCLUSION: Huyin decoction possessed protective effect on acute hepatic injury in mice.

[An exploration of animal behavior screen platform for novel gene function in central nervous system].

For the purpose of large-scale screening of novel gene functions in mammalian nervous system, we have developed an animal behavior-monitoring platform employing antisense-oligo technology. Twenty genes of different categories were chosen from a low abundant gene (c)DNA sub-library of rat brain. Antisense oligo-nucleotides of these genes were designed and synthesized according to the homologues of the genes in mouse for mouse behavior tests. These antisense oligos were injected into the lateral ventricles of mouse brain using a Hamilton micro-syringe, with saline and oligos of scramble sequences as controls. These mice were tested with the following behavior model paradigms: metabolism, open field behavior, tail flick latency, and step-down test. Out of the 20 genes tested, 14 genes showed significant behavioral differences from the control groups at the level of P value less than 0.05 or 0.001 in different behavior animal models.

[The effects of otitis media with effusion and hearing loss on the speech outcome after cleft palate surgery].

OBJECTIVE: To find out the exact effect of OME and hearing loss on the speech outcome post palatoplasty in cleft palate patients. METHODS: Otoscopy and tympanometry were performed in 18 cleft palate patients whose ages ranged from 5 to 23 years old and hearing thresholds were also tested before cleft palates repair. All patients were followed up from 3 to 18 months postoperatively. Articulation scores were evaluated and linear correlation analysis was performed between these results and their mean hearing thresholds for air conduction in speech frequencies before the palate closure. The incidences of the OME were compared among the groups with different articulation scores. RESULTS: The postoperative articulation scores correlate negatively (r = -0.504, P = 0.032 < 0.05) to the degrees of hearing loss. But the incidences of OME had no significant differences among the groups with different articulation scores. CONCLUSIONS: The hearing level preoperation is more important than tympanometric data on the foresight of the speech result postoperation in cleft palate patients.

[Orthotopic detenia cecal-ascending colon urinary reservoir: report of 18 cases].

OBJECTIVE: To improve the technique of detenia cecal-ascending colon continent urinary reservoir for farther improving the life quality of the patient. METHODS: Orthotopic detenia cecal-ascending colon urinary reservoir was carried out by complete resection of all the tenia and multiple transverse incision of colonic circular muscular layer for 18 patients with bladder cancer underwent radical cystectomy. RESULTS: The patients have been followed up for 6-20 months. All were completely continent during the day and 13 patients got complete continence at the night. Only 3 patients had nocturnal incontinence. The capacity of the urine reservoir was 410-520 ml. The maximum intrareservoir pressure was 39-60 cmH2O, while the post-micturition residual volume was 15-46 ml and the maximum flow rate was 12-28 ml/per second. No evidence of ureter reflux and ureteral obstruction was observed and serum electrolytes were normal in all patients. CONCLUSIONS: Orthotopic detenia cecal-ascending colon urinary reservoir shows good clinical and functional results, and the simple technique, exerts low pressure in the reservoir and produces minimal complications.

[Study on growth rhythm in populations of Pinellia ternata (Thunb.) Breit].

OBJECTIVE: To study the genetic diversity of Pinellia ternata. the growth rhythm. METHOD: Sixteen populations of P. ternata. originated mostly from middle and lower reaches of Yangtze River valley were collected and cultivated in Weigang Experimental Farm of Nanjing Agricultural University with same cultural conditions, whose differences in growth rhythm have been observed and compared continuously for two years. RESULT AND CONCLUSION: There are obvious differences in sprouting, lamina spreading, bolting and "sprout tumble", Three times of "sprouting" and so-called "sprout tumble" and two fast-growing periods for plants in most of the populations in spring and autumn respectively were observed within a year, including mass bolting in May. But the starting time and duration are very different from population to population, with flowering being prior to sprouting in the population originated from Jurong county.

[A study of bone development and metabolism in childhood].

OBJECTIVE: To explore the regularity of bone development and metabolism in childhood. METHODS: A survey was made to collect and analyze the data on bone biochemistry, the density of lumbar vertebrae and the X-ray examination of calcaneus in 224 healthy children aged 3-15 years in the urban area of Chengdu. RESULTS: Comparative analysis of serum and urine calcium and phosphate concentrations showed no significant difference between the age groups. The serum alkaline phosphatase (ALP) and bone alkaline phosphatase (BALP) were found to increase with age and reach their peak values in children aged 7-12 years, and their values were significantly higher than that of adults (P < 0.05). The density of lumbar vertebrae and the trabecular bone of calcanesus also developed with age. CONCLUSION: The measurements of the bone biochemistry, bone density and the trabecular bone of calcaneus which reflect the bone development and metabolism have their special regularity in childhood. The criteria in use for adulthood is not fit for the evaluation in childhood. The data obtained can be used in clinical diagnosis and treatment of the bone metabolic diseases of children.

[A study on the relationship between postmortem interval and the changes of DNA content in the kidney cellule of rat].

OBJECTIVE: To study changes of DNA content in the kidney cellule of rats and relationship with the postmortem interval. METHODS: This experiment chose seven parameter of cell nuclear, including the area and integral optical density, determined the changes of DNA content in the kidney cellule of 15 rats at different intervals between 0 and 48 h postmortem with auto-TV-image system. RESULTS: The degradation rate of DNA in nuclear has a certainty relationship to early PMI(in 48 h) of rat, and get binomial regress equation. CONCLUSION: Determining the quantity of DNA in nuclear should be an objective and exact way to estimate the PMI.

[The forensic determination of post-traumatic epilepsy in 21 patients].

UNLABELLED: To evaluate the forensic determination of post cerebral traumatic epilepsy. METHODS: In 21 patients, traumatic history and previous history were analysied combined with the demonstrations of electroencephalogram(EEG), X-ray, CT and MRI. RESULTS: Post-traumatic epilepsy, manily in late stage, usually occurred following serious cerebral trauma. The type of traumatic epilepsy was determined by the traumatic location and extent. Abnormal epileptic wave in scalp EEG and 24 h dynamic EEG and medical image examinations were helpful for qualitative analysis. CONCLUSION: The forensic determination of post traumatic epilepsy must be on the basis of traumatic and previous history combined with EEG, CT and MRI analysis.

[A study on mRNA expression levels of integrin beta 1 during the healing process of mandibular fractures].

OBJECTIVE: The aim of this study was to investigate the spatial and temporal mRNA expression changes of integrin beta 1 during the healing process of mandibular fractures. METHODS: The in situ hybridization was performed using digoxigenin-labeled integrin beta 1 mRNA oligonucleotide probe. During the healing process of mandibular fractures, the staining pattern of beta 1 mRNA was examined both spatially and time-dependently. RESULTS: Integrin beta 1 was widely present in the cells of the fractured bone, especially in osteoblasts. The positive staining was observed in the cytoplasm and nucleolus of osteoblasts. After 7 days of fracture, the expression level of integrin beta 1 increased. The peak appeared between the 14th and the 30th day, and almost recovered to normal between the 60th and 90th day. CONCLUSION: During the healing process of mandibular fracture, the expression level of integrin beta 1 in bone, especially in osteoblasts rose. It is suggested that integrin beta 1 may play an important role in the fracture healing process.

[Expression of bone morphogenetic protein (BMP)-2, 4 in mouse embryonic tooth during the bud stage].

OBJECTIVE: The purpose of this paper was to investigate the distributions of bone morphogenic protein (BMP)-2, 4 in the developing molars of mice during the bud stage and, the functional relationship between BMP-2 and BMP-4. METHODS: The distribution of BMP-2, 4 were examined in developing mouse molars on the 14th (the 14th embryonic day), using monoclonal antibodies and immunohistochemistry methods. RESULTS: BMP-2, 4 were detected both in the epithelial cells and dental mesenchyme cells. CONCLUSION: There is a similar distribution pattern between BMP-2 and BMP-4. Therefore, there may be a functional relationship between BMP-2 and BMP-4 during the development of mouse embryonic teeth in the bud stage.

[Establishment of human neuroectodermal tumor cell line (PFSK) with stable expression of xanthine oxidoreductase].

Xanthine Oxidoreductase (XOR) is the key enzyme in purine metabolism and also produces oxygen free radicals. As a continuation of our previous work, in this study, we constructed a retrovirus expression vector (pLNCX2-XOR) containing full length of XOR cDNA. Retrovirus was produced by the virus package cell line PT-67 following the transfection of PT-67 with pLNCX2-XOR and used to infect the human primitive neuroectodermal tumor cell line (PFSK). Infected PFSK cells were selected by G418 to establish cell line with stable expression of XOR. The expression of XOR in the cell line we established was confirmed by RT-PCR, Immunocytochemistry and XOR activity assay.
