Homologous recombination among three intragene Alu sequences causes an inversion-deletion resulting in the hereditary bleeding disorder Glanzmann thrombasthenia.

The crucial role of the human platelet fibrinogen receptor in maintaining normal hemostasis is best exemplified by the autosomal recessive bleeding disorder Glanzmann thrombasthenia (GT). The platelet fibrinogen receptor is a heterodimer composed of glycoproteins IIb (GPIIb) and IIIa (GPIIIa). Platelets from patients with GT have a quantitative or qualitative abnormality in GPIIb and GPIIIa and can neither bind fibrinogen nor aggregate. Very few genetic defects have been identified that cause this disorder. We describe a kindred with GT in which the affected individuals have a unique inversion-deletion mutation in the gene for GPIIIa. Patient platelets lacked both GPIIIa protein and mRNA. Southern blots of patient genomic DNA probed with an internal 1.0-kb GPIIIa cDNA suggested a large rearrangement of this gene but were normal when probed with small GPIIIa cDNA fragments that were outside the mutation. Cytogenetics and pulsed-field gel analysis of the GPIIIa gene were normal, making a translocation or a very large rearrangement unlikely. Additional Southern analyses suggested that the abnormality was not a small insertion. We constructed a patient genomic DNA library and isolated fragments containing the 5' and 3' breakpoints of the mutation. The nucleotide sequence from these genomic clones was determined and revealed that, relative to the normal gene, the mutant allele contained a 1-kb deletion immediately preceding a 15-kb inversion. The DNA breaks occurred in two inverted and one forward Alu sequence within the gene for GPIIIa and in the left, right, and left arms, respectively, of these sequences. There was a 5-bp repeat at the 3' terminus of the inversion.(ABSTRACT TRUNCATED AT 250 WORDS)

Sudden death due to rupture of ectopic pregnancy concurrent with therapeutic abortion.

We report three cases of sudden death resulting from internal hemorrhage associated with the rupture of an ectopic pregnancy. In each case, the woman had undergone therapeutic abortion before death. The ectopic pregnancies, however, were not suspected or identified before the women left the treatment facility. Subsequently, all three women suffered fatal rupture of a tubal pregnancy and associated massive intraperitoneal hemorrhage during the postabortive period (range, 14 to 32 days). Our investigation of these cases has identified the lack of standardized procedures for the adequate monitoring of tissue recovered during therapeutic abortion. Such monitoring could have resulted in identification of the ectopic pregnancy and possibly led to successful intervention. We discuss the causes and risks of failure to diagnose ectopic pregnancy during the postabortive period.

[Relationship between illumination and growth of the stroma of Cordyceps sinensis (Berk.) Sacc].

The growth of the stroma of Cordyceps sinensis largely depends upon the illumination in its growth period. By increasing illumination time and light intensity, its growth height can be controlled, growth rate slowed down and the corrosion time of larva body of host insect prolonged. Ultraviolet light is able to affect the growth of stroma too. The stroma also shows the strong phototaxis in its growth period. The arrangement and formation of the perithecium vary with the illumination.

[Study on albumin microspheres as drug carrier for liver targeting].

In this paper 6 factors and 12 levels of each variable were selected by Uniform Design Method and computer for preparing albumin microspheres with emulsion-chemical cross-linking. An optimal procedure for preparing albumin microspheres was established and the mean diameter of albumin microspheres is 0.41-0.47 micron. Albumin was labelled with 125I-isotope and 125I-albumin microspheres were prepared according to the optimal procedure. The suspension of 125I-albumin microspheres with 0.1% Tween-80 saline was injected via mice tail vein. The animals were sequentially killed and the radioactivity of blood, spleen, liver, kidney, stomach, heart, lung, thyroid and brain were measured. The results showed that albumin microspheres were accumulated mainly in the liver, about 68% of the injected dose at the peak concentration. The pharmacokinetics of albumin microspheres in liver was also studied and two-compartmental model can be used to describe the regulation.

[Studies on a radioligand-receptor binding assay of human chorionic gonadotropin in rat testis].

A radioligand-receptor binding assay for human chorionic gonadotropin (HCG) has been established with cell membranes preparation of rat testis. Partially purified cell membranes preparation was prepared from rat testis by sucrose density gradient ultracentrifuge. The binding assay was performed by incubating 125I-HCG with rat testis cell membranes preparation at 30 degrees C for 1.5 h in a total incubation volume of 0.5 ml. Scatchard plot analysis for the binding assay revealed a linear binding plot and yielded a bound capacity of testis cell membranes receptors of 0.36 x 10(-12) mol/micrograms protein. KD of 0.45 x 10(-10) mol/L. Hill coefficient was 0.976. Studies on the kinetics of the reaction of radioligand-receptor binding showed that the mean values for the association rate (k1) and dissociation constants (k2) were individually 0.41 x 10(-3) pmol/min and 2.12 x 10(-3) pmol/min; the dissociation plot was two-phase, with 15 x 10(-3) pmol/min of fast-phase and 1.5 x 10(-3) pmol/min of slow-phase.

Determination of human immunodeficiency virus antibody status in forensic autopsy cases using a rapid and simple FDA-licensed assay.

Evaluation of an FDA-licensed rapid assay to detect antibodies to human immunodeficiency virus (HIV) was accomplished by testing 414 serum samples collected sequentially from autopsy cases at the Office of the Chief Medical Examiner, State of Maryland. The time of postmortem collection ranged from 8 to 30 hours. All samples were tested for the presence of antibodies to HIV using a rapid peptide microfiltration assay (SUDS HIV-1 test, Murex Corporation, Norcross, GA), and an enzyme-linked immunosorbent assay (ELISA). Samples yielding repeatedly reactive results were confirmed by Western blot. Of the 414 specimens, 23 (5.6%) produced reactive results by both SUDS and ELISA, and were confirmed by Western blot. One additional sample was repeatedly reactive by ELISA but negative by the SUDS test. This sample produced an indeterminate profile by Western blot (reactivity to only p24) and was negative by several additional retroviral assays. Of the 23 HIV infected cases, 16 had risk factors for HIV infection; 19 were blacks and 18 were male. The SUDS, 10 minute test, exhibited a 100% sensitivity and 100% specificity in comparison to FDA-licensed ELISA and Western blot assays for detecting HIV antibody in autopsy serum specimens. We conclude that this rapid, simply performed assay is accurate and applicable for use in several testing situations, including autopsy rooms.

[Study of quality control on herbal drugs and preparations of daqingye and banlangen].

This paper first suggests that 2,4 (1H,3H) quinazolinedione (QZL) may be used as a quality control standard for the Chinese herbal drugs Daqingye (Folium Strobilanthis et Polygoni) and Banlangen (Radix Strobilanthis) as well as its preparations. QZL was found for the first time from the above mentioned two Chinese herbal drugs which has a representative property. Its content is stable. This made it possible to solve the difficulty of quality control for the preparations "antipyretic and common cold granules" and "Banlangan granules" and provide a reliable assay method to evaluate the quality of the herbal preparations. By changing the wavelength and using gradient elution in HPLC, 3 constituents can be determined through one injection by the present method, thus the time of determination may be shortened and the efficiency of assay increased. The method showed good linearity (r = 0.99998, n = 4) in the range of 0.2-3.2 micrograms/ml of QZL, the average recovery was above 95% with RSD < 1.5% (n = 3). HPLC condition: Column: LiChrosorb RP 18; elution gradient: methanol from 35% to 90% in water solution; determination wavelength: 226 nm and 288 nm.

[Chemical studies of Strobilanthes cusia].

Seven compounds have been isolated from the whole plant of Strobilanthes cusia (Nees) O. Ktze. Three of them are triterpenes (I-III), two are indole alkaloids (IV, V), two are quinazolinone alkaloids (VI, VII). On the basis of spectral analysis and physicochemical properties, their structures were established as lupeol (I), betulin (II), lupenone (III), indigo (IV), indirubin (V), 4(3H)-quinazolinone (VI), 2,4(1H,3H)-quinazolinedione (VII). VI and VII were found from natural plant for the first time. The results of the pharmacological tests demonstrate that compound V has anticancer activity and compound VI has hypotensive action. Compound VII can be quantitatively determined by HPLC, which may serve as a quality control standard for materia medica and its preparations. Compounds VI and VII have been confirmed by means of synthesis.

[Transarterial immuno-chemotherapy including adoptive transfer of autologous cultured lymphocytes for stage IV breast cancer patients with locally-advanced tumor].

Immuno-chemotherapy via a catheter in the subclavian artery using sequential treatment with OK-432, chemotherapeutic agents (ADM, 5-FU), and cultured autologous lymphocytes, was performed for 9 Stage IV breast cancer patients with locally-advanced primary tumor. Tumor reduction of more than 50% was observed in 8 patients including 4 whose breast tumors had disappeared. Among 11 evaluable distant metastatic lesions, 7 (1 pleural effusion, 2 lung, 2 liver, 2 bone metastases) regressed after local immunotherapy of breast or additional regional immunotherapy (1 lung, 1 liver, 1 pleural effusion). Median survival time to date is 56 months. Five patients are currently alive, although 3 of them did not undergo mastectomy. Local immuno-chemotherapy may be useful because (a) toxicity is limited, (b) low doses of anti-cancer agents during the therapy (median dose of ADM, 60 mg) do not limit subsequent systemic chemotherapy, and (c) distant metastases often regress concomitantly with the primary lesions.

Electrophysiological and pathological observations on experimental coxsackie B-3 viral myocarditis in mice.

Electrophysiological action of right ventricular myocardium examined by standard intracellular microelectrode technique and real-time microcomputer data processor system and histological and ultrastructural changes of myocardium in BALB/c mice infected with coxsackie B-3 virus from 3 days to 9 months were observed. It was found that electrophysiologic parameters of action potential changed very quickly at the early stage (3 days to 1 month) of the disease. Those abnormalities became most apparent by the 5-30th day, and 7 patterns of abnormal action potential occurred frequently within the same period. These changes were basically parallel to the myocardial lesions. At the late stage (3-9 months) the electrophysiological parameters were nearly normal, while the myocardial lesions decreased gradually. However, the abnormal patterns of action potential were still detected, even though they were improved gradually. The results suggest that myocardial damages caused by viral infection may lead to changes of cardiac electric action, which may be one of the factors in arrhythmias in the episode of viral myocarditis.

Nucleotide sequence of a cloned human HBV mutant (pDKHBV) in duck hepatoma of Qidong County.

This is the first report to describe the presence of an HBV-like DNA sequence in two hepatoma and their tumor surrounding liver tissues, one precancerous and one non-malignant liver tissue of ducks collected from Qidong County of China. The HBV-like sequences were either in an episomal form of 3.2 kb or in an integrated form of various sizes, while the DHBV DNA sequences (3.0 kb) were either present or absent in these tissues and in different size pattern. Furthermore, there was no evidence of cross-hybridization between HBV-like DNA sequences in duck and DHBV DNA. A 3.2 kb HBV-like DNA sequence has been cloned from one duck hepatoma (40 K), designated as pDKHBV. The 3218 bp full-length nucleotide sequence of this clone has been determined, which had no apparent homology with Duck Hepatitis B Virus (DHBV) genome, but was highly homologous to human HBV adw2 subtype (99.0%). The sequence was composed of four open reading frames for HBV gene Pre-S/S, X, C and P respectively. In addition to multiple sites of point mutation, one nt and two nt deletion were detected downstream the initiation codon of Pre-S1 gene and at the 3' end of C gene respectively, thereby suggesting a frameshift mutation in Pre-S gene and C gene. Based on these results, it was implicated that an unusual HBV variant might exist in Qidong County, which might have transmitted into duck through some yet unknown mechanisms. The possibility from exogenous contamination could be thus excluded.

Distribution of materno-fetus materno-suckling transfer of tritium after single injection of tritiated water in mice.

The metabolism and transfer of tritium from pregnant mice to fetus after injection of tritiated water intraperitoneally was investigated in this paper. The study was composed of three experiments. 1. Pregnant mice were injected with tritium water on the first day of pregnancy, in order to obtain a transfer coefficient of tritium from pregnant mice to fetus through placenta. 2. Pregnant mice were injected with tritiated water on the first day of parturition to study the transfer of tritium via mother milk from nursing mice to the baby-animals. 3. Pregnant mice were injected with tritiated water in different periods of gestation. The results showed that tritiated water was uniformly distributed in all of the tissues measured, including placenta, fetal membrane and amniotic fluid in experiment 1. No effect of placentas on tritium transfer from pregnant mice to fetus was found. Concentration of tritium in the baby's tissues was evidently higher than that in the pregnant mice in experiments 2 and 3, and the transfer coefficient in experiments 2 and 3 was generally higher than that in experiment 1.

Activity of neurons in anterior inferior temporal cortex during a short-term memory task.

Inferior temporal (IT) cortex of primates is known to play an important role in visual memory. Previous studies of IT neurons during performance of working memory tasks have found modulation of responses when a current stimulus matched an item in memory; however, this effect was lost if other stimuli intervened in the retention interval. To examine how IT cortex retains memories while new stimuli are activating the cells, we recorded from IT neurons while monkeys performed a delayed matching-to-sample task, with multiple intervening items between the sample and matching test stimulus. About half of the cells responded differently to a test stimulus if it matched the sample, and this difference was maintained following intervening stimuli. For most of the affected cells, the responses to matching stimuli were suppressed; however, for a few cells the opposite effect was seen. Temporal contiguity alone could not explain the results, as there was no modulation of responses when a stimulus on one trial was repeated on the next trial. Thus, an active reset mechanism appears to restrict the memory comparison to just the stimuli presented within a trial. The suppressive effects appear to be generated within or before IT cortex since the suppression of response to matching stimuli began almost immediately with the onset of the visual response. The memory of the sample stimulus affected not only the responses to matching stimuli but also those to nonmatching stimuli. There was suggestive evidence that the more similar a nonmatching stimulus to the sample, the more the response was suppressed. About a quarter of the cells showed stimulus-selective activity in the delay interval following the sample. However, this activity appeared to be eliminated by intervening stimuli. Thus, it is unlikely that delay-interval activity in IT contributed to the performance of this particular version of delayed matching to sample. To determine how much information about the match-nonmatch status of the stimulus was conveyed by individual neurons, we analyzed the responses with discriminant analysis. The responses of an individual IT neuron could be used to classify a stimulus as matching or nonmatching on about 60% of the trials. To achieve the same performance as the animal would require averaging the responses of a minimum of 25 IT neurons. There was no evidence that mnemonic information was carried by temporal variations in the spike trains. By contrast, there was a modest amount of temporal variation in sensory responses to different visual stimuli.(ABSTRACT TRUNCATED AT 400 WORDS)

Retinal pigment epithelial cell transplants in retinal degeneration slow mice do not rescue photoreceptor cells.

PURPOSE: To determine if retinal pigment epithelial cells are in any way involved in the degeneration of photoreceptor cells in the retinal dystrophy mouse model, retinal degeneration slow (rds); to determine if normal retinal pigment epithelial cell transplants can affect outer segment development in the retina. METHODS: Retinal pigment epithelial cells of neonatal normal pigmented C3H mice were isolated and transplanted into retinas of postnatal day 33 albino rds mice. Then eyes of 4-month-old rds mice, retinal pigment epithelial cell-transplanted and sham and non-treated control mice, were processed for light and electron microscopy and the thickness of the outer nuclear layer were measured and compared. RESULTS: Measurements of outer nuclear layer thickness in the transplant and control groups revealed that normal retinal pigment epithelial cell transplants did not cause photoreceptor cell rescue in rds mice. In addition, outer segments were not seen in retinal pigment epithelial cell-transplanted rds retinas. CONCLUSIONS: This study supports the conclusions of other investigators that the photoreceptor cell is the primary site of the genetic defect that results in retinal dystrophy in the rds mouse model.

Hepatitis B virus DNA detected by PCR in sera and liver tissues of Chinese patients with chronic liver diseases.

To investigate the HBV infection and its replication in Chinese patients with chronic liver disease, polymerase chain reaction (PCR) was employed to detect hepatitis B virus DNA in sera of 410 patients with chronic liver disease and liver samples from 188 patients. The HBV DNA detectability in all serum samples was 58%. Among them 100% HBeAg-positive and 58% anti-HBe cases were HBV DNA detectable respectively. However, HBV DNA was also found in 23% HBsAg-negative/anti-HBc positive chronic cases. Furthermore, 30% anti-HBs positive chronic cases who had neutralizing antibody against HBV infection, continued to contain HBV DNA. Our findings indicate that HBV infection and its replication are dominant cause of chronic liver disease and some HBV variants may escape from the protective antibody to induce chronic liver disease, even anti-HBs antibody circulated.

FGF inactivates myogenic helix-loop-helix proteins through phosphorylation of a conserved protein kinase C site in their DNA-binding domains.

Myogenin belongs to a family of myogenic helix-loop-helix (HLH) proteins that activate muscle transcription through binding to a conserved DNA sequence associated with numerous muscle-specific genes. Fibroblast growth factor (FGF) inhibits myogenesis by inactivating myogenic HLH proteins. We show that activated protein kinase C (PKC) can substitute for FGF and inhibit transcriptional activity of myogenic HLH proteins. In transfected cells, FGF induces phosphorylation of a conserved site in the DNA-binding domain of myogenin. This site is phosphorylated by PKC in vivo and in vitro and mediates repression of the myogenic program through a loss in DNA binding activity. A myogenin mutant lacking the PKC phosphorylation site is not repressed by FGF, confirming this site as a molecular target for FGF-dependent repression of muscle transcription. These results establish a direct link between the signal transduction pathways that inhibit myogenesis and the transcription factors directly activating muscle-specific genes.

Cloning, sequencing, overproduction, and purification of M. CviBI (GANTC) methyltransferase from Chlorella virus NC-1A [corrected].

We have cloned and sequenced the cvibIM gene from Chlorella virus NC-1A by selecting for the modification phenotype. The modification gene was cloned on a 7-kb BamHI fragment inserted into the BamHI site of the pUC13 plasmid. The cvibIM gene was localized at the 3' end of this fragment. Sequencing of this region revealed a large open reading frame that codes for methyltransferase (MTase; symbol M.) (predicting 260 amino acids). M.CviBI (GANTC) aa sequence is homologous to M.Dam(GATC), M.DpnII(GATC), and M.T4 (GATC), and not so to M.HinfI(GANTC), M.HhaII (GANTC), and M.DpnA(GATC). We also describe the use of the polymerase chain reaction technique to alter transcriptional and translational signals surrounding this gene so as to achieve overexpression in Escherichia coli. This construct yields M.CviBI at 2-3% of the total cellular protein. The MTase was purified by phosphocellulose, DEAE, and gel filtration chromatography. Its size by SDS-PAGE is approx. 28 kDa, in good agreement with that predicted from the nucleotide sequence.

Hepatic 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase. The role of surface loop basic residues in substrate binding to the fructose-2,6-bisphosphatase domain.

Lys-356 has been implicated as a critical residue for binding the C-6 phospho group of fructose 2,6-bisphosphate to the fructose-2,6-bisphosphatase domain of rat liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (Li, L., Lin, K., Correia, J., and Pilkis, S. J. (1992) J. Biol. Chem. 267, 16669-16675). To ascertain whether the three other basic residues (Arg-352, Arg-358, and Arg-360), which are located in a surface loop (residues 331-362) which contains Lys-356, are important in substrate binding, these arginyl residues were mutated to Ala, and each arginyl mutant was expressed in Escherichia coli and purified to homogeneity. The far UV circular dichroism spectra of the mutants were identical to that of the wild-type enzyme. The kinetic parameters of 6-phosphofructo-2-kinase of the mutants revealed only small changes. However, the Km for fructose 2,6-bisphosphate, Ki for fructose 6-phosphate, and Ka for inorganic phosphate of fructose-2,6-bisphosphatase for Arg352Ala were, respectively, 2,800-, 4,500-, and 1,500-fold higher than those for the wild-type enzyme, whereas there was no change in the maximal velocity or the Ki for inorganic phosphate. The Km for fructose 2,6-bisphosphate and Ki for inorganic phosphate of Arg360Ala were 10- and 12-fold higher, respectively, than those of the wild-type enzyme, whereas the maximal velocity and Ki for fructose 6-phosphate were unchanged. In addition, substrate inhibition was not observed with Arg352Ala and greatly reduced with Arg360Ala. The properties of the Arg358Ala mutant were identical to those of the wild-type enzyme. The results demonstrate that in addition to Lys-356, Arg-352 is another critical residue in fructose-2,6-bisphosphatase for binding the C-6 phospho group of fructose 2,6-bisphosphate and that Arg-360 binds the C-2 phospho group of fructose 2,6-bisphosphate in the phosphoenzyme.fructose 2,6-bisphosphate complex. The results also provide support for Arg-352, Lys-356, and Arg-360 constituting a specificity pocket for fructose-2,6-bisphosphatase.
