Localization of mRNAs representing interstitial collagenase, 72-kda gelatinase, and TIMP in healing porcine burn wounds.

The process of wound healing sets in motion a complex and dynamic series of events, which includes the remodeling of the extracellular matrix. Degradation of matrix macromolecules is mediated through the actions of the matrix metalloproteinase family. Conversely, the actions of this enzyme family are regulated by tissue inhibitors of metalloproteinases (TIMPs). In this study, we have developed riboprobes derived from human cDNAs representing collagenase, 72-kDa gelatinase, and TIMP and have found them to be sufficiently specific and sensitive for use in in situ hybridization studies of porcine burn wounds. Expression of these mRNAs, although not seen in uninjured skin, was found to be a predictable and locally distinct event in wound repair. Transcripts for collagenase and TIMP but not 72-kDa gelatinase were detected at the resurfacing epithelial margin; label was also detected in and around follicular epithelium within the wound bed. Transcripts for both metalloenzymes and TIMP were found throughout the viable dermis and subcutaneous tissues underlying the wound bed. However, expression of 72-kDa gelatinase was most prominent in the superficial dermis adjacent to the resurfacing epidermis at the wound margin. Collagenase and TIMP transcripts were particularly prominent in a perivascular pattern in the dermis and in the connective tissue network surrounding adipocytes in the subcutaneous zone. Numerous cell types appeared to be involved, including keratinocytes, fibroblasts, macrophages, and endothelial cells. Future exploitation of this porcine thermal injury model is likely to provide information about the spatial and temporal patterns of matrix metalloproteinase and TIMP expression in cutaneous wound healing.

Protein tyrosyl phosphatase-1B is expressed by normal human epidermis, keratinocytes, and A-431 cells and dephosphorylates substrates of the epidermal growth factor receptor.

In the epidermis tyrosine kinases such as those found in the epidermal growth factor receptor (EGF-R) phosphorylate regulatory molecules on tyrosine and play an important role in controlling epidermal growth. Phosphotyrosyl phosphatases (PTPase) that dephosphorylate EGF-R and other proteins phosphorylated on tyrosine must also play an important role in controlling epidermal growth. The presence and metabolism of one such PTPase, PTP-1B, was detected and studied in human skin using biochemical, immunochemical, and molecular biologic methods. The message for PTP-1B was expressed in human epidermis, in keratinocytes cultured from human epidermis, and in human keratinocyte cell lines. The 49-kDa but not the 37-kDa form of PTP-1B was identified in membranes prepared from these cells and tissues by immunodetection on Western blots. Nearly all of the labeled proteins identified by gel electrophoresis of an A-431 particulate fraction phosphorylated with [gamma-32P] ATP in the presence of epidermal growth factor are substrates for PTP-1B because their labeling decreased after incubation with a catalytically active and purified PTP-1B fusion protein. Immunohistochemical methods were used to show that PTP-1B was primarily localized to the basal cell layers in normal thick epidermis. The presence of PTP-1B in intact human epidermis suggests that this molecule is not an artifact limited to cultured cells but is an important molecule in the in vivo regulation of epidermal functions.

Plasminogen with type-I mutation in the Chinese Han population.

A functionally inactive plasminogen (PLG) variant, PLG M5, is polymorphic in the Japanese population and has a codon 601 mis-sense mutation (GCT for Ala to ACT for Thr), designated type-I mutation. The present study aimed to reveal whether the plasminogen with type-I mutation is present in the Chinese Han population. Among 104 healthy Chinese students, phenotype PLG AM5 was found in three subjects (2.9%), while 100 subjects were phenotype A and one was phenotype AA3. In the three subjects with PLG AM5, plasma concentrations of immunoreactive PLG were normal but PLG activities were decreased. Analysis using PCR and dot-blot hybridization with allele-specific oligonucleotide probes revealed the presence of the type-I mutation in the PLG gene of all three subjects with PLG AM5. The data indicate that PLG with type-I mutation is present in the Chinese Han population, possibly at a polymorphic frequency.

Interaction between heat shock factor and hsp70 is insufficient to suppress induction of DNA-binding activity in vivo.

The intracellular level of free heat shock proteins, in particular the 70-kDa stress protein family, has been suggested to be the basis of an autoregulatory mechanism by which the cell measures the level of thermal stress and regulates the synthesis of heat shock proteins. It has been proposed that the DNA-binding and oligomeric state of the heat shock transcription factor (HSF) is a principal step in the induction pathway that is responsive to the level of 70-kDa stress protein. To test this hypothesis, we investigated the association between HSF and 70-kDa stress protein by means of a coimmunoprecipitation assay. We found that 70-kDa stress proteins associate to similar extents with both latent and active forms of HSF, although unlike other 70-kDa stress protein substrates, the association with HSF was not significantly disrupted in the presence of ATP. Gel mobility shift assays indicated that active HSF trimers purified from a bacterial expression system could not be substantially deactivated in vitro with purified 70-kDa stress protein and ATP. In addition, elevated concentrations of hsp70 alone could not significantly inhibit induction of the DNA-binding activity of endogenous HSF in cultured rat cells, and the induction was also not inhibited in cultured rat cells or Drosophila cells containing elevated levels of all members of the heat shock protein family. However, the deactivation of HSF to the non-DNA-binding state after prolonged heat stress or during recovery could be accelerated by increased levels of heat shock proteins. Hence, the level of heat shock proteins may affect the rate of disassembly of HSF trimers, but another mechanism, as yet undefined, appears to control the onset of the oligomeric transitions.

Effect of luminal angiotensin II on proximal tubule fluid transport: role of apical phospholipase A2.

Recent micropuncture studies showed the existence of high concentrations of angiotensin II (ANG II) in proximal tubular fluid. In the present study, we have examined the effect of luminal ANG II, alone and in combination with peritubular ANG II, on fluid transport (JV) in the isolated perfused rabbit proximal convoluted tubule. In comparison with peritubular ANG II, luminal ANG II caused a similar but more potent biphasic effect on JV. At 10(-11) M, luminal ANG II maximally increased JV to 204 +/- 22% of the baseline compared with 142 +/- 10% by peritubular ANG II at 10(-10) M. At 10(-8) M, luminal ANG II suppressed JV to 9.7 +/- 16% of the baseline compared with 64 +/- 14% by peritubular ANG II. When luminal and peritubular ANG II were combined at concentrations that impose similar effect on JV, the effects of luminal and peritubular ANG II were not additive. However, when combined at concentrations that would otherwise impose opposing effects on JV, the stimulatory effect predominated. In support of the role of apical phospholipase A2 (PLA2) on the effect of luminal ANG II, ANG II stimulated PLA2 activity in isolated brush-border membrane vesicles, and addition of PLA2 inhibitor, mepacrine or dibucaine, to the luminal perfusate attenuated the effect of luminal ANG II on JV. In summary, these studies show a potent effect of luminal ANG II on proximal tubule JV involving activation of brush-border membrane PLA2. When combined, luminal and peritubular ANG II exert their effects in concert on proximal tubule JV.

Fast glucocorticoid feedback inhibition of ACTH secretion in the ovariectomized rat: effect of chronic estrogen and progesterone.

The purpose of this study was to determine whether estrogen and progesterone influence fast glucocorticoid negative feedback regulation of the ACTH and corticosterone (CORT) responses to stress. Mature rats were ovariectomized and 6 weeks later implanted with 17 beta-estradiol (E2, 0.5 mg), E2 and progesterone (P, 100 mg; E2 + P group) or placebo pellets (OVX). Seven days later rats were subjected to a single or repeated intermittent footshock stress (0.2 mA, 15 s duration, 0.5 s on). The repeated stress was of the same intensity and duration, and was applied either during the time domain of the rate-sensitive fast glucocorticoid feedback when plasma CORT levels are rising (5 min after the onset of the first stress), or at the time of peak CORT response (15 min) to the initial stress. Plasma ACTH and CORT were measured from serial samples. Estrogen replacement alone or in combination with progesterone lowered the immediate (t = 5) ACTH and CORT response to a single stress in ovariectomized animals. The second stress applied 5 min after the initial stress produced net ACTH responses similar to those obtained after a single stress in the OVX and E2 + P-replaced hormone groups, while total ACTH responses were lower in the E2-treated group. In ovariectomized animals, a facilitation of ACTH response by a prior stress is apparent in response to a footshock 15 min later, when the integrated ACTH secretion is significantly greater than the response measured after a single shock, or after a repeated shock 5 min apart. Anterior pituitary proopiomelanocortin (POMC) mRNA levels were lower in groups with E2 or E2 + P replacement compared to OVX animals. In contrast, hypothalamic corticotropin-releasing factor (CRF) mRNA levels did not increase significantly. However, hypothalamic glucocorticoid receptor (GR) mRNA levels increased after 17 beta-estradiol treatment, and this increase was reversed by progesterone. These results suggest that prior stress leads to both a fast-feedback inhibition and a facilitation of the subsequent stress response. In the absence of gonadal hormones this facilitation is balanced by fast-feedback inhibition during the glucocorticoid fast-feedback time domain, and is unmasked outside of this time domain. Estrogen suppresses POMC mRNA synthesis leading to a decrease in the availability of releasable ACTH, thereby reducing the facilitation. Progesterone may counter this effect of estrogen by decreasing the efficacy of the fast rate-sensitive glucocorticoid negative feedback.

Mechanism of apical and basolateral Na(+)-independent Cl-/base exchange in the rabbit superficial proximal straight tubule.

The present study was undertaken to determine the magnitude and mechanism of base transport via the apical and basolateral Na(+)-independent Cl-/base exchangers in rabbit isolated perfused superficial S2 proximal tubules. The results demonstrate that there is an apical Na(+)-independent Cl-/base exchanger on both membranes. HCO3- fails to stimulate apical Cl-/base exchange in contrast to the basolateral exchanger. Inhibition of endogenous HCO3- production does not alter the rate of apical Cl-/base exchange in Hepes-buffered solutions. Both exchangers are inhibited by H2DIDS and furosemide; however, the basolateral anion exchanger is more sensitive to these inhibitors. The results indicate that the apical and basolateral Cl-/base exchangers differ in their transport properties and are able to transport base equivalents in the absence of formate. The formate concentration in rabbit arterial serum is approximately 6 microM and in vitro tubule formate production is < 0.6 pmol/min per mm. Formate in the micromolar range stimulates Jv in a dose-dependent manner in the absence of a transepithelial Na+ and Cl- gradient and without a measurable effect on Cl(-)-induced equivalent base flux. Apical formic acid recycling cannot be an important component of any cell model, which accounts for formic acid stimulation of transcellular NaCl transport in the rabbit superficial S2 proximal tubule. We propose that transcellular NaCl transport in this nephron segment is mediated by an apical Na+/H+ exchanger in parallel with a Cl-/OH- exchanger and that the secreted H+ and OH- ions form H2O in the tubule lumen.

Radioimmunoassay detects the frequent occurrence of autoantibodies to the Mr 65,000 isoform of glutamic acid decarboxylase in Japanese insulin-dependent diabetes.

Glutamic acid decarboxylase antibodies (GAD65Ab) are common in new onset Caucasian insulin-dependent diabetic (IDDM) patients but it is unclear if this marker is also prevalent in patients of other ethnic backgrounds. We determined antibodies against human recombinant GAD in Japanese diabetic patients using a radioimmunoassay with competition between in vitro translated 35S-GAD65 and non-labelled recombinant human GAD65 (rhGAD65). GAD67 antibodies (GAD67Ab) were similarly analyzed but without antigen competition. In 73 Japanese diabetic patients, GAD65Ab were found in 11/16 (69%) of patients with short-duration (less than 5 yrs) IDDM, 6/23 (26%) with long-duration (5 or more yrs) IDDM and 10/20 (50%) with slowly progressive diabetes. High GAD65Ab levels were associated with concomitant autoimmune diseases (p = 0.021). GAD67Ab were found in 4/16 (25%) of patients with short-duration IDDM, 3/23 (13%) with long-duration IDDM and 2/20 (10%) with slowly progressive diabetes. In 14 non-insulin dependent diabetic (NIDDM) patients, GAD65Ab and GAD67Ab were not found (0/14) and 1/50 (2%) healthy controls were positive in either assay. Among the GAD67Ab-positive samples, 8/9 (88%) were also high level GAD65Ab positive, 7/9 (77%) were displaced by an excess of rhGAD65 and the antibody levels correlated (r2 = 0.573; p = 0.003). Our data are consistent with a strong association of GAD65Ab also in Japanese IDDM, and suggest that, when present, GAD67Ab are frequently directed to epitope(s) common to GAD65 and GAD67.

[4-Aminopyridine induced histamine release and its antagonism by certain drugs].

4-Aminopyridine (4-AP), a known potassium channel blocker, was shown to induce histamine release from mast cells in mice. After ip 4-AP 5 mg.kg-1 the histamine content increased in blood, but decreased in the lung tissue. Calcium antagonists nifedipine (NIF) 500 mg.kg-1 ig, TMB-8 300 mumol.L-1 in vitro and potassium channel opener minoxidil (MIN) 100 mg.kg-1 inhibited the histamine release induced by 4-AP from mouse peritoneal mast cells (PMC). These results provide evidence that potassium channels are present in mouse mast cell membranes and indicate that the mechanism of histamine release by 4-AP may be related to the potassium channel blocking effect. As the result of this effect, the calcium channels open and the Ca2+ influx to the mast cells increases, thus eliciting histamine release.

[Primary hepatic carcinoma treated by chemoembolization--a clinical analysis of 20 cases with survival period longer than 3 years].

Advanced primary hepatic carcinoma was treated in 110 patients by percutaneous transhepatic arterial chemoembolization (TAE) from January 1987 to October 1989 according to Seldinger's protocol with mitomycin C, Adriamycin or carboplatin and 40% iodized oil. The treatment was given once every 4-12 weeks and the number of treatment for each patient varied from 2 to 19 depending on patients' condition and response. Of the 110 patients, 20 survived > 3 years, among which 7 survived > 4 years and 4 survived > 5 years. Following treatment, a greater than 50% reduction of tumor size was observed in 13 cases and a decrease in AFP to normal level in 12 cases. Seven patients were treated with TAE followed by tumor resection and, 2 patients were treated with TAE due to recurrence after tumor resection 11 patients were treated with TAE alone. The mean survival period was 53.1 and 43.5 months, respectively. TAE is considered as the first choice of palliative treatment for patients with advanced hepatic carcinoma. Factors that might influence the result of TAE treatment are discussed.

[Epidemiology of melioidosis in China].

From 1975 to 1989, a total of 73 strains of P. pseudomallei was isolated from the water samples and the pathological samples of human and domestic animals in 13 counties and cities located different latitude from four provinces Qiong, Yue, Gui and Xiang in China. Serological investigation demonstrated that the geographical distribution of the organism had a significant correlation with the positive rate of antibodies against P. pseudomallei and the native foci of the organisms distributed over the southern subtropical zone and the edge of tropical zone in Qiong, Yue and Gui. In endemic areas, the positive rates of antibodies against P. pseudomallei in human-beings, horses, oxen and pigs are 3.8%-15.2%, 9.1%-18.4%, 6.6%-33.0% and 35% respectively. The investigation results showed the horses and mules infected by the organism would interfere with quarantine of the animals, meanwhile, the meat contaminated by the bacterium would endanger the public health. In Sept. and Oct. of 1989, three cases in Zhanjiang and Sanya of Hainan were reported, two cases died of acute melioidosis with septicemia, another case was the chronic leg ulcers. So, it was predicated that there could have some cases of melioidosis which were misdiagnosed or missed out.

Bond strengths of dentinal bonding systems to enamel and dentin.

Contemporary, third-generation dentinal bonding products have become highly specialized in producing high bond strengths to dentin. This investigation compared in vitro bond strengths of six dentinal bonding systems and their matched composite resins to human enamel and dentin. The effects of treatment by dentinal primers on enamel bond strengths as well as the effects of phosphoric acid on the strengths of dentinal bonds were measured. The use of dentinal primer on enamel improved the bond strengths of Prisma Universal Bond 3/Prisma APH and XR Bond/Herculite systems and had no effect on Denthesive/Charisma, Scotchbond 2/Silux, and Tenure/Perfection, while the enamel bond strengths of Gluma/Pekalux declined. Pretreatment of dentin with phosphoric acid improved the bond strengths of Denthesive/Charisma, Prisma Universal Bond 3/Prisma APH and XR Bond/Herculite, but had no effect on Gluma/Pekalux, Scotchbond 2/Silux and Tenure/Perfection.

Thymic peptides increase glutathione level and glutathione disulfide reductase activity in vascular endothelial cells.

The glutathione redox cycle plays an important role in antioxidant and detoxification mechanisms. We recently reported that a calf thymic peptide (TP) protected vascular endothelial cells from oxidant injury induced by hydrogen peroxide. Using electrophoresis and amino acid sequencing analysis, we have now shown that TP consists of two peptides. The fast-moving peptide has 9 amino acid residues at the NH2 terminal and accounts for 92% of total quantity, while the other peptide has 18 amino acid residues at the NH2 terminal and amounts to 8%. The present study investigated the effect of TP on glutathione redox cycle. Confluent monolayers of bovine pulmonary artery endothelial cells (PAEC) were incubated with TP (12.5-100 micrograms/mL) for 24-48 h. TP caused a dose-dependent increase in glutathione (GSH) level and glutathione disulfide reductase activity but no significant change in GSH peroxidase activity. Exposure of PAEC to an organic oxidant t-butyl hydroperoxide (tBHP) resulted in decreased GSH level, increased lipid peroxidation, and elevated leakage of intracellular lactate dehydrogenase. Preincubation of PAEC with TP prevented these changes induced by tBHP. The data suggest that the antioxidant effect of TP may be due, at least in part, to its modulation of the GSH redox cycle in vascular endothelial cells. TP may thus be considered a new antioxidant with novel activities in addition to being an immune regulator.

[Immunoelectronmicroscopic study of fibronectin in Kupffer's cell in rats with multiple organ failure].

This essay studied the content and distribution of Fibronectin (Fn) in Kupffer's cells in rats with multiple organ failure (MOF) with the immunoelectronmicroscopy technique. The results revealed that there was a significant increase in Fn level in plasma and the amount of immunogold particles in Kupffer's cells during the early stage, but the changes were not evident in the late stage. Our findings indicated that the changes in content of Fn was closely related to the function of MPS and might play an important role in the pathogenic mechanism of MOF.

[Comparison of the sensitivity of retinoic acid sensitive and resistant HL-60 cell lines to several anti-leukemia drugs].

By using MTT and trypan blue exclusion assay, the sensitivity of retinoic acid sensitive HL-60 and resistant HL-60/RA cell to six anti-leukemia drugs such as RA, Ara-c, harringtonine was compared. It was found that all five drugs except RA exhibited an approximately equivalent IC50 to HL-60 and HL-60/RA cells. The results suggest that tumor cells may not develop resistance to differentiation inducer and cytotoxic anti-tumor agent parallel. It also suggests that it is reasonable to use a combination of differentiation and cytotoxic anti-leukemia agents concomitantly or sequentially.

[Pharmacokinetics and bioavailability study on acetaminophen oral drop in healthy volunteers].

The concentrations of acetaminophen in human plasma were determined by RP-HPLC after administration of a single oral dose of 1000 mg acetaminophen oral drop or tablet to eight normal male volunteers in a crossover study. The concentration vs time curves of acetaminophen all fitted the two compartment model. The Cmax in oral drop and tablet were 11.23 +/- 0.93 and 8.79 +/- 0.93mg/L, respectively; Tmax were 0.86 +/- 0.08 and 1.48 +/- 0.39h; T1/2 beta 3.03 +/- 0.79 and 2.51 +/- 1.10 h; AUC 42.35 +/- 6.20 and 40.60 +/- 6.79mg/L.h. When AUC in tablet was 100%, the relative bioavailability of acetaminophen oral drop was 105.58%. The results showed that the oral drop and tablet are bioequivalent.

[Therapy of experimental autoimmune myasthenia gravis in rabbits with 4-aminopyridine and 3,4-diaminopyridine].

The autoimmune myasthenia gravis (AMG) in rabbits was produced by intradermal injection of N-AChR-rich membrane vesicles isolated from the electric organ of Narcine limlei. After i.v. 4-aminopyridine (4-AP) 0.8 mg.kg-1 to 8 AMG rabbits, their general posture improved promptly, the features of gastrocnemius compound action potentials and toe twitches elicited by 4-Hz stimuli applied to the sciatic nerve returned to normal, and the tetanic plateau evoked by 50-Hz indirect stimulation was again well sustained. This improved condition lasted 9.1 +/- 2.5 h. Other 8 AMG rabbits given 3,4-diaminopyridine (3,4-DAP) 0.4 mg.kg-1 showed a similar improvement for 9.3 +/- 3.1 h. These results indicated that 4-AP and 3,4-DAP were effective in treating the AMG in rabbits, they may be useful in the clinical treatment of myasthenia gravis patients.

[Measuring of immune complexes and antinuclear antibodies in effusion of secretory otitis media].

Immune complexes (IC) and antinuclear antibodies (ANA) in middle ear effusions from 40 cases (45 ears) of secretory otitis media were measured using PEG precipitation method and fluorescent microscopy. The range of absorbence for IC varies between 0.124-0.512 in patients and is higher than those (0.132) in normal human sera. Thirty-two ears (71.1%) are ANA positive. The result indicates that secretory otitis media is related with autoimmunity. The deposit of IC may be one of the causes of effusion in otitis media.
