Longitudinal studies of blood pressure in children.

A longitudinal study of six years was conducted to find out the pattern of longitudinal changes of blood pressure and to affirm the "tracking phenomenon" of blood pressure in children in China. We initially measured blood pressure and related parameters of 2,946 children (aged 4-14 years) in 1981 at Fanshan county, Beijing, and then two follow-up remeasurements were conducted in 1985 and 1987, respectively. The results indicated that: the average level of blood pressure increases with age even after adjusting for height and weight; tracking coefficients of systolic blood pressure range from weak to moderate levels, increasing with age. Only 30% of the children whose systolic blood pressure was beyond the 90th percentile of the systolic pressure distribution at the first examination remained at the same region after four years. Multiple stepwise regression was used to determine factors correlated with blood pressure. Our results indicate that systolic blood pressure in children is correlated with body weight, pulse rate, serum glucose and HDL-C, while diastolic blood pressure is correlated only with pulse rate and serum glucose.

Differential detection of rat islet and brain glutamic acid decarboxylase (GAD) isoforms with sequence-specific peptide antibodies.

We studied the distribution of the M(r) 65,000 and M(r) 67,000 isoforms of glutamic acid decarboxylase, GAD65 and GAD67, in rat islets and brain by immunocytochemistry. Synthetic peptides representing selected GAD65 or GAD67 sequences were used to produce sequence-specific antibodies, allowing differential immunocytochemical detection of the two isoforms. GAD-specific reactivity of each peptide antiserum was confirmed by ELISA, immunoblotting, and immunoprecipitation. Immunostaining specificity was verified by displacement with either immunizing or irrelevant peptide. Dual immunostaining with GAD isoform-specific antibodies and polyclonal antibodies to glucagon showed that GAD65 was primarily detected in rat pancreatic islet beta-cells, whereas alpha-cells had weak GAD65 staining. In contrast, GAD67 was detected primarily in alpha-cells. In rat brain, GAD65 and GAD67 were present in neuron cell bodies and processes. These data demonstrate that antibodies raised against the N-terminus of GAD allow differential immunocytochemical identification of GAD67 and GAD65. Differential expression of GAD isoforms within islet alpha- and beta-cells supports the role of GAD65 in autoimmune diabetes and stiff-man syndrome.

Regulation of cell proliferation and differentiation by Myc.

Myc is a nuclear phosphoprotein which controls cellular proliferation, most likely by regulating gene activity. The finding that the neuronal model cell line PC12 lacks the Myc DNA binding partner, the Max protein, and the demonstration that Myc is a repressor of gene activity as well as a transactivator, lead to models for Myc action in regulating cell growth.

A toluene-tolerant mutant of Pseudomonas aeruginosa lacking the outer membrane protein F.

The outer membrane protein profiles of a toluene-tolerant mutant, Pseudomonas aeruginosa, strain PAK103, were compared with those of its parent strain PAO1161. Protein F (OprF), the most abundant outer membrane protein in the parental strain PAO1161, was missing in the toluene-tolerant strain PAK103. The absence of OprF may lead to the loss of toluene diffusion across in the outer membrane of the mutant cells.

Comparison of the values of basic fibroblast growth factor determined by an immunoassay in the sera of patients with traumatic brain injury and enhanced osteogenesis and the effects of the same sera on the fibroblast growth in vitro.

In patients with severe traumatic brain injury, the early healing of fractures is accompanied by hypertrophic callus formation or heterotopic ossifications, which might even result in ankylosis of the affected joints. Analysis of the sera of patients with traumatic brain injury revealed post-traumatic dynamic changes of basic fibroblast growth factor immunoreactivity, similar to those observed during fracture healing associated with enhanced osteogenesis. The aim of this study was to determine whether such changes in basic fibroblast growth factor concentrations could be related to the phenomenon of enhanced osteogenesis. Basic fibroblast growth factor immunoreactivity was determined (using an IEMA kit) in the sera of patients with traumatic brain injury and bone fractures (n = 8) and in the sera of patients with either traumatic brain injury alone (n = 10) or bone fractures alone (n = 7), and the effects of these sera on L929 fibroblast growth were analysed in vitro. The results did not prove a causative relationship between the changes of basic fibroblast growth factor immunoreactivity and in vitro growth promoting effects of the sera. However, it is apparent that, in addition to changes in the growth-promoting activity and basic fibroblast growth factor concentration of serum, other as yet unknown post-traumatic changes can cause enhanced osteogenesis.

[Hypomagnesemia in patients of gynecologic neoplasms following chemotherapy with cisplatin].

Serum magnesium in 79 patients of gynecologic neoplasms treated with cisplatin and their controls was measured. The results showed: (1) the average value of serum magnesium in patients following chemotherapy with cisplatin was significantly lower than in the controls; (2) the incidence of hypomagnesemia was positively correlated with the number of the courses and the total dosages of chemotherapy, being 52.9% after one to three courses of treatment and 92.0% after more than six courses of treatment; (3) the incidence of hypomagnesemia was directly related with the severity of gastrointestinal disorders; (4) serum magnesium following cisplatin chemotherapy was not correlated with serum blood urea nitrogen (BUN). Hypomagnesemia is clinically characterized by symptoms of the nervous system which are found in 14.0% of hypomagnesemic patients.

Repeated radial keratotomy.

38 cases 59 eyes received repeated radial keratotomy in which the secondary incisions were performed along the scars of old incisions. The average follow-up after the reoperation was 13 months. Before reoperation, the mean spherical equivalent of the refraction was -3.78D and the mean keratometry was 40.15D. 13 months after the reoperation, the mean spherical equivalent of the refraction decreased by 2.30D, and the mean keratometry decreased by 2.04D. After the reoperation, the mean uncorrected visual acuity increased by 5 lines. There was no relationship between the therapeutic effect of the reoperation and the degree of initial myopia or pre-reoperative keratometry. The repeated radial keratotomy can decrease the degree of astigmatism and its complications are few.

[Analysis of factors causing visual disability and its preventive measures in China].

Analysis of sampling data from Chinese people in 1987 showed prevalence of visual disability was 10.08/1000. Major factors causing visual disability were congenital or hereditary defects, ametropia, amblyopia and corneal disease in children aged 0-14 years; senile cataract, trachoma and corneal disease in the elderly people aged 60 and over; and ametropia, amblyopia, cataract and corneal disease in the other people. Glaucoma and eye trauma were two major causes leading to severe visual disability. Suggestion of three-level prevention for visual disability in the country was put forward by the authors with focus on primary prevention, including genetic counselling and health care in pregnancy, preventing from eye infection and trauma, and active treatment of primary diseases, such as diabetes.

[VM-26 plus CCNU in the prevention of brain metastasis in lung cancer patients].

Brain metastasis is of frequent occurrence in patients with cancer of the lung. To ascertain if brain metastasis is preventable, 34 lung cancer patients without brain metastasis were treated with 4 cycles of VM-26 plus CCNU at 4-week intervals. As a control for comparison, an equal number of lung cancer patients free of brain metastasis were treated with chemotherapy protocol which did not include VM-26 and CCNU. The patients were monitored by clinical symptoms, signs and CT or MRI scan and were followed up for 4-12 months. The results indicate that none of the VM-26 plus CCNU-treated patients developed brain metastasis while brain metastasis occurred in 7 (20.5%) of 34 control patients. The difference was statistically significant (P < 0.05).

[The reverse-flow lateral sural cutaneous artery island flap: anatomical study].

The circulatory pattern of a skin flap was studied by microsurgical dissection, dye injection and angiography in 50 lower legs of fresh cadavers. The conclusions are as following: (1) The lateral sural cutaneors artery descends constantly and superficially accompanied by concomitant nerves. The upper post-lateral area of the lower leg in the best donor site of the flap. (2) The lateral sural cutaneous artery and the peroneal perforating artery form a anastomotic arcus around the lateral malleolus in 88.6% of the lower legs. The mean diameter of the anastomotic arcus is 0.63 +/- 0.19 mm. (3) The peroneal perforating vessel is 3.5 +/- 1.4 cm in length. Based on it the pedicle of the flap can be prolonged 6-8 cm on the main vessel of the lateral sural cutaneous artery. (4) Creation of the flap has no influence to the blood supply and function of the lower leg. (5) By anterograde transfer, the lateral sural island flap could be used to repair the defects around the knee and upper tibia. However, retrograde transfer of the flap has a higher clinical values in repiring the defects on the lower one third of lower leg and medial malleolus.

[Effect of xueguantong on inhibiting experimental atherosclerosis in rabbits].

In this experiment, the model of experimental atherosclerosis in rabbits was established by feeding the cholesterol contained food for 12 weeks. The effect of Xueguantong (XGT) in inhibiting atherosclerotic formation in rabbits has been studied. It was found that XGT could decrease serum total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), platelet aggregation and platelet adhesion. The aortic atherosclerotic plaque intimal coverage and ratio of atherosclerotic plaque of aortic intimal coverage to aortic medium coverage in XGT groups were significantly smaller than that in the model group (P < 0.05). The results showed that XGT could inhibit experimental atherosclerotic formation in rabbits, it might be relevant to that XGT could reduce the TC and inhibit platelet aggregation.

[Study on economic NPK(nitrogen-phosphorus-potassium)-requirements for medicinal safflower].

A design of second order orthogonal rotative regression was developed and field tested, and a model for obtaining 18kg/mu dry flower yield was simulated and built according to several better combinations based on the model of total safflower dry flower yield. The optimal NPK-requirements for obtaining the above 18kg/mu dry flower yield are N:6-6.5kg/mu, P2O5:10.0-11.0kg/mu, K2O:6.9-10.0kg/mu. These requirements may be adjusted rationally according to soil fertility, manure sources, and so on.

[Influence of radiotherapy and chemotherapy on the function of malignant tumor patients and regulation function of acupuncture].

The observation on the indexes of cortisol, estradiol, estriol and testosterone showed that incretory function of malignant tumor patients had different extent of pathologic changes, after radiotherapy and chemotherapy, making the change strengthened. Acupuncture can regularize this disorder of incretory function of patients treated with radiotherapy and chemotherapy to some extent.

[Hepatic albumin's mRNA in nephrotic syndrome rats treated with Chinese herbs].

It was found in our previous studies that two Chinese herbs, Astragali and Angelica (A & A), with a high protein diet could protect the decrease of serum albumin and markedly increase the synthesis rate of protein by 18N-glycine metabolic study in nephrotic rats. A further experiment was designed to investigate the effects of A&A and high protein intake on the level of hepatic albumin's mRNA in nephrotic rats. Male S. D. rats were divided into 3 groups: high protein diet alone (39.7% HP); HP with nephrotic syndrome (HPN); and HPN with therapy (HPNT). Nephrotic syndrome in rat was induced by accelerated nephrotoxic nephritis, withfed A&A (2 g of each in 3 ml) daily for 14 days. At the 15th day, the serum and liver were collected and the level of serum albumin and mRNA were measured with biochemistry, Northern blot, dot blot hybridization and quantity in laser density screening. The level of serum albumin in HPNT was higher than that of HPN (3.7 +/- 0.1 vs 2.9 +/- 0.29, P < 0.01) with an unchanged urinary output. The level of hepatic albumin mRNA in HPNT was higher than HPN (2.56 +/- 0.24 vs 1.7 +/- 0.12, P < 0.01) and HP (2.56 +/- 0.24 vs 1.25 +/- 0.24, P < 0.01). We conclude that in a molecular biologic level A&A combined with high protein intake could increase the albumin synthesis in the liver of nephrotic rat.

[The distribution of abnormal hemoglobins in the silk road region of China].

215,785 individuals living in the Silk Road region of China were surveyed for abnormal hemoglobins (Hbs). Among them, 695 carriers were detected. The average incidence of abnormal Hbs was 3/22 and the frequencies of abnormal Hbs among 12 nationalities reached 0.15/1000-12.17/1000. The primary structural analyses were made in 271 persons, and 24 variants [13 alpha-chain variants and 11 beta-chain variants] were identified. HbJ Tashikuergan and Hb Tianshui were discovered the first time in the world. HbS, Hb Bunbury, Hb Setif and HbI Philadelphia were not found previously in the Chinese population. Three variants, i.e. HbD Punjab, HbG Taipei and HbG Coushatta occurred at the highest frequencies and showed a gradient distribution along the Silk Road, suggesting that they may originate from Caucasians in Central Asia, Han in the Yellow River valley, and the ancient nomadic minorities of China in Mongolia Plateau respectively. The frequencies of other variants were quite low. Some were probably imported from other countries and areas, and some arose from independent mutations. The data from many variants support the movements of various populations in this area, as reported in numerous historical documents. It is indicated that the Silk Road promoted the national fusion among the Chinese nationalities, Mongolian and Caucasian races.
