In vivo enhancement of mitogen-induced lymphocyte DNA synthesis by sodium diethyl-dithiocarbamate (DTC).

Sodium diethyl dithiocarbamate (DTC), a low molecular weight sulphur compound, has been previously shown to be endowed of immunomodulatory properties. To get a better understanding of the mechanisms whereby DTC regulates the immune response, mitogen-induced lymphoproliferation was studied in spleen from guinea pigs treated with a single i.v. injection of DTC. The results clearly show that DTC enhances, after a lag period, DNA synthesis in T and B lymphocytes. DTC which appears not to act at the level of immuno-competent cells seems to modulate the immune responses through indirect pathways.

Yersinia enterocolitica gastroenteritis: a prospective study of clinical, bacteriologic, and epidemiologic features.

A prospective study was performed to estimate the frequency of gastroenteritis due to Yersinia enterocolitica in Montreal children and their families. Evidence of bacterial infection was correlated with clinical features and serologic responses. YE was isolated from the stools of 181 (index cases) of 6,364 children with gastroenteritis over a 15-month period; Salmonella was isolated from 280 and Shigella from 68. Median ages were 24, 30, and 41 months, respectively. All but 18 YE isolates were biotype 4, serotype 0:3. YE was not found in the stools of 545 children without gastrointestinal symptoms. Clinical manifestations of the index cases with YE biotype 4, serotype 0:3 (n = 57) included diarrhea (98%), fever (88%), abdominal pain (64.5%), and vomiting (38.5%) with mean durations of 14, 3.9, 7.7, and 2.4 days, respectively. The duration of excretion of YE in the stool ranged from 14 to 97 days (mean 42). Spread of YE occurred in 27 of 57 families studied, involving 15 of 41 children and 19 of 117 adult contacts; approximately one-third of infected contacts developed diarrhea. Agglutination titers of greater than or equal to 200 were demonstrated in all index cases infected with serotype 0:3, with the exception of two very young infants. YE is a frequent cause of bacterial gastroenteritis in Montreal children. The illness is characterized by persistent diarrhea and abdominal pain, prolonged YE excretion in the stools, and moderate communicability.

Characteristics of the interaction of a treponemal hemolysin with rabbit erythrocytes.

The mechanism of action on rabbit red cells of Treponema hyodysenteriae hemolysin was studied using volume analysis and release of hemoglobin. While fixation of the hemolysin on the erythrocytes is temperature independent, it appears that hemolysis is temperature dependent. The kinetics of hemolysis proceed according to a sigmoid curve characterized by a prelytic lag. The duration of the prelytic lag varies inversely with the quantity of hemolysin but the rate and the maximum value of hemolysis are directly proportional to the quantity of hemolysin. The effect of sucrose and trypan blue on the hemolysin and the the red cells suggest that erythrocyte lysis is likely to be induced by the hemolysin in a way different from that known for other hemolytic agents.

Size separation and polyclonal activation to immunoglobulin secretion of early precursors of B lymphocytes.

In adult mammals the bone marrow is a major site of B lymphocyte production, which occurs through the differentiation of pluripotent stem cells. In vivo studies have permitted the description of intermediate stages of differentiation. Following the recent development of culture conditions and the discovery of many polyclonal activators, it is now possible to carry out in vitro a finer analysis of B lymphocyte precursors differentiation. Because in vivo studies have shown that precursors vary in size, adult murine bone marrow cells were separated according to size. Fractions were cultured with polyclonal mitogens and tested for their capacity to develop into Ig-secreting cells. With this assay system, up to six populations were detected in adult bone marrow that can differentiate along the B cell lineage. The characterization of those populations was made according to size, mitogen(s) responsiveness, and kinetics of maturation. Precursor frequencies estimated by limiting dilution analysis further suggest that in cultures of some fractions of the gradient, new lymphocytes are produced through differentiation of immature precursors, although in cultures of some other fractions B lymphocytes may arise by self-renewal of mature cells.

Changes in mixed lymphocyte culture-reactive lymphocytes following alloimmunization of single lymph nodes in sheep.

When an "isolated" single lymph node is challenged with irradiated allogeneic lymphocytes, there is a change in the reactivity of the lymphocytes flowing out of the node when they are cultured in vitro in unidirectional mixed lymphocyte cultures (MLC) against the immunizing lymphocytes. These changes in the reactivity of the recipient lymphocytes are shortlived, follow a set time sequence in relation to the cell traffic changes accompanying the immune response, are the property of small lymphocytes and not blast cells, are exhibited by surface Ig-negative cells, and they are specific for the donor lymphocytes. It is suggested that antigen causes the selective retention of antigen-specific lymphocytes within the stimulated node followed by the proliferation and differentiation of large numbers of antigen-specific cells, which then leave the lymph node as small lymphocytes.

Efficacy of cold enrichment techniques for recovery of Yersinia enterocolitica from human stools.

Stool specimens from children with gastroenteritis and their household contacts were cultured for Yersinia enterocolitica by direct plating onto routine laboratory media. These stools were also inoculated into phosphate-buffered saline and subcultured to the same media after 1 day or 3 weeks of incubation at 4 degrees C. Y. enterocolitica was isolated from 174 index cases and 34 household contacts. One hundred eighty-one isolates were of serotype O:3, and the remaining 21 belonged to other serotypes. Eighty-one percent (147/181) of O:3 isolates were recovered by direct plating, and 6.1% (11/181) and 13% (23/181) were recovered by 1-day and 3-week cold enrichment, respectively. For other serotypes, 26% (7/27), 0%, and 74% (20/27) were isolated by direct plating, 1-day cold enrichment, and 3-week cold enrichment, respectively. The efficacy of the cold enrichment for the patients were still symptomatic, 94 and 6% of Y. enterocolitica were identified by direct plating and cold enrichment, respectively. Isolation rates were 66% by direct plating and 34% by cold enrichment when stools were obtained from asymptomatic carriers or from those convalescing from Y. enterocolitica gastroenteritis. These results indicate that the cold enrichment methods increase the sensitivity of Y. entercolitica culture methods considerably in convalescent and asymptomatic subjects but only minimally in patients with diarrhea caused by serotype O:3.

Canadian experience with Yersinia enterocolitica (1966--1977).

Data pertaining to 1,219 cultures of Yersinia enterocolitica isolated in Canada are summarized. Of the 977 cultures isolated from humans, Y. enterocolitica serotype 0:3, biotype 4, phage type IXb was the most predominant type in Ontario, Quebec and the four Eastern Provinces. In the Western Provinces the predominant strains were indol-positive, serotypes 0:8; 5,27 and 4,32. Most of the 242 cultures of nonhuman origin, except those isolated from swine and a few isolated from wild animals, were indol-positive, biochemically atypical and serologically nontypable or belonging to different serotypes which have seldom been found in human infections.

Yersinia enterocolitica infection in children: 4 years experience in the Montreal urban community.

From 1973 to 1976, a total of 146 strains of Yersinia enterocolitica was isolated from children at the Ste. Justine Hospital and Montreal Children's Hospital. All strains tested were of serotype 0:3, biotype 4. The majority (80%) of children from whom Y. enterocolitica was isolated were under 5 years of age and in 74 of 93 cases reviewed diarrhea was the predominant symptom. Case reports on 2 patients with unusual clinical features are also presented.

Opposite effects of BCG on spleen and lymph node cells: lymphocyte proliferation and immunoglobulin synthesis.

C57BL/6 mice were immunized intravenously (i.v.), intraperitoneally (i.p.), or subcutaneously with one dose of Bacillus Calmette-Guérin (BCG). At various time intervals after injection, the lymphocyte response, as measured by thymidine incorporation into DNA, and the number of immunoglobulin-secreting cells were determined in vitro before and after mitogenic stimulation with phytohemagglutinin, concanavalin A, or lipopolysaccharide. In unstimulated cultures, the spontaneous thymidine incorporation and immunoglobulin synthesis of spleen cells were increased to some extent in mice infected i.p. or i.v. with BCG, as compared with noninfected mice. In contrast, after mitogenic stimulation, a marked depression of the proliferative response of spleen cells to both T- and B-cell mitogens and a marked inhibition of LPS-induced immunoglobulin secretion were observed in mice infected i.v. and to a lesser extent in those infected i.p. The depression of lymphoblastogenesis in spleens was fully established 15 days after infection and persisted for a long period of time. When unfractionated or plastic-adherent spleen cells from BCG-infected mice were cultured with normal spleen cells, a strong depression of their reactivity to phytohemagglutinin, concanavalin A, and lipopolysaccharide was observed. After the removal of cells adherent to plastic, the response was partially restored in the nonadherent population from mice infected i.p., but not in that from mice infected i.v. After mitogenic stimulation, lymph node cells of mice inoculated subcutaneously showed a response to mitogen higher than that of normal cells. These results thus demonstrate that, depending on the route of administration, BCG exerts very different effects.

Surface membrane differentiation of hemopoietic cells as observed by radioactive labeling.

Bone marrow cells from normal rats were separated by velocity sedimentation into three distinct populations corresponding to granulocytes, lymphocytes and reticulocytes. Cells from each population were surface labelled via the lactoperoxidase radioiodination or the tritiated borohydride reduction. Distinct labeling patterns were observed on SDS-PAGE for each cell populations. Separation of bone marrow cells from anemic rats injected with TAB vaccine led to four populations corresponding to successive stages of erythroid cell maturation. Labelled protein patterns were not in this case as different from one population to the other, except for one species which increased in intensity with the degree of maturation. The tritiated glycoprotein profiles show a shift from high to low molecular weight species during the process of maturation.

Suppressor cells in rabbit peripheral blood.

Rabbit peripheral blood leukocytes (PBL) added to cultures of autologous spleen cells, primed in vivo to sheep red cells, are able to suppress with high efficacy the secondary in vitro response of the spleen cells to that antigen. Removal of nylon wool adherent cells from PBL abolishes the suppressive effect. When the PBL are fractionated by velocity sedimentation the suppressor cells can be separated from the responding cells. The circulating lymphocytes, freed from the inhibiting effect, either by nylon wool absorption or by velocity sedimentation fractionation, are able to give a strong secondary in vitro anti-SRC response, in which the long latency period, usually observed when PBL are stimulated with antigen in culture, is abolished or at least reduced. The suppressor effect present in the PBL is not due to granulocytes, platelets or erythrocytes.

Differences in the immunological activities of antibody-secreting cell precursors in mouse spleen selected on the basis of antigen-binding capacity.

Mouse spleen cells transfer a retarded and lower than normal IgG antibody response after removal of cells with a high antigen-binding capacity. The population enriched in these latter cells transfers a response equal to or, more often, higher than the response of the unfractionated controls and does not appear to have originated from previous priming of the mice. These cells are present in athymic nude mice and appear to be more susceptible to inactivation by an anti-Ig antiserum and complement than precursors of IgM antibody-secreting cells.
