Conformational free energy of armadillo metmyoglobin.

The conformational free energy of armadillo metmyoglobin was examined over a pH range of 4.4-8.0 and a guanidinium chloride concentration of 0-2.3 M. For isothermal unfolding at 25 degrees essentially the same value was obtained for the conformational free energy from all the data: 27 +/- 2 kJ/mol. These data suggest that the armadillo has the least stable metmyoglobin of any mammal thus far examined. The cooperativity of the unfolding with respect to denaturant is considerably less than for other mammalian myoglobins. On unfolding only three to four side chains with a pKA of 6 in the unfolded protein are protonated instead of the six found for horse and sperm whale myoglobins.

Facial sensibility testing in the normal and posttraumatic population.

A reliable, reproducible, simple examination of facial sensibility is described. Evaluation of 60 healthy subjects established normal values, trends, and variations of facial sensibility. Comparison of these normal values with 20 posttraumatic patients revealed that postfacial fracture sensibility testing was abnormal. Abnormalities in pressure threshold testing was most consistently associated with functional sensory complaints. Twelve of the 20 patients had significant sensory complaints at one year following the trauma.

Eikenella corrodens keratitis: case report.

Eikenella corrodens is a Gram-negative facultative anaerobe which is part of the normal human oropharyngeal flora and an opportunistic pathogen of mucous membrane tissues. We report a case of secondary E. corrodens ulcerative keratitis with hypopyon in a 39-year-old male with herpes simplex keratitis. To the best of our knowledge this is the first reported case of E. corrodens as a pathogen in bacterial keratitis.

Determination of fitness in children with asthma. Use of standardized tests for functional endurance, body fat composition, flexibility, and abdominal strength.

Children with asthma frequently have exercise-induced disease that can limit their participation in both organized sports and vigorous free play. We measured fitness in a group of children with moderately severe to severe asthma with an instrument that is used widely and is available to clinicians for assessment of the physical capability of their patients. Abnormalities in physical fitness were present primarily in the area of endurance, with performance in the nine-minute run frequently found to be low. Increased skin-fold thickness was also present more frequently than in the normal population. Abdominal strength and flexibility were normal. Abnormalities in fitness were not well explained by measures indicating the presence of poorly controlled disease. All children with significant asthma probably should be tested to document the level of fitness so that appropriate intervention can be initiated.

Comparison of regeneration across a vascularized versus conventional nerve graft: case report.

A histological and functional comparison of regeneration across a vascularized and conventional nerve graft was carried out in a clinical case. The two branches of the medial antebrachial cutaneous nerve were used to neurotize the median nerve at the wrist. The superficial branch of the radial nerve (vascularized nerve graft) and a conventional sural nerve graft were used as the interposition grafts between the two branches of the medial antebrachial cutaneous nerve and the median nerve. This nerve grafting procedure was carried out in two stages. The proximal neurorrhaphy was carried out 7 months prior to the distal nerve repair. Biopsies were taken from the distal portions of the vascularized radial sensory nerve and the conventional sural nerve at the time of the second procedure. Histological evaluation demonstrated superior regeneration across the vascularized nerve graft. Subjectively, the patient described better sensory recovery in the territory innervated by the vascularized nerve graft. Sensory testing in the two territories demonstrated better sensibility in the territory innervated by the vascularized nerve graft.

Conformational free energies of myoglobins of small mammals.

Myoglobins from three small placental mammals and one small marsupial were isolated from cardiac or skeletal muscle. The conformational free energies of these four myoglobins were estimated from guanidinium chloride unfolding data at pH 8 and 25 degrees. The myoglobins from rat and rabbit are more stable than that of the most stable myoglobin previously studied, that of the sperm whale. In addition, these two myoglobins unfold with greater cooperativity than previously characterized myoglobins. The data obtained herein demonstrate unequivocally for the first time that the stability of homeotherm myoglobins correlates with neither the size of the organism nor its metabolic rate.

Thermodynamics of alligator metmyoglobin unfolding.

The conformational free energy of alligator metmyoglobin was examined over a pH range of 4.4-8.0, a temperature range of 18-48 degrees C, and a guanidinium chloride concentration of 0-2.3 M. For isothermal unfolding at 25 degrees C essentially the same value was obtained for the conformational free energy from all the data: 7.0 +/- 0.5 kcal/mol. The cooperativity of the unfolding with respect to denaturant is considerably less than for mammalian myoglobins. On unfolding three to four side chains with a pKa of 6.3 in the unfolded protein are protonated instead of the six expected. The temperature at which delta H (unfolding) is zero is much lower than for previously characterized myoglobins. Alligator metmyoglobin, considerably less stable than other previously characterized myoglobins, may not be as compactly folded.

Bronchial blood flow affects recovery from constriction in dog lung periphery.

We investigated the effect of eliminating the bronchial circulation on recovery time from intravenous histamine challenge in canine lung periphery. Results from animals with intact bronchial circulations were compared with a second group in which the left lower lobe was isolated in situ. The pulmonary artery to this lobe was perfused and a bronchoscope was wedged in a small airway, which provided an index of resistance to airflow through the collateral system. The lobe was challenged with intravenous histamine, and the time constant of recovery (tau) from bronchoconstriction was measured. With or without pulmonary blood flow, elimination of the bronchial circulation increased tau 44.4 and 48.5%, respectively. This increase was similar to that found by stopping pulmonary blood flow alone (56.5%). Histamine challenges were also performed in sympathectomized or vagotomized animals with intact bronchial circulations. Neither of these conditions increased tau. We conclude that blood flow through the bronchial circulation affects the recovery time from intravenous histamine challenge in the lung periphery to a degree similar to that of the pulmonary circulation.

Immunological comparison of heat-shock proteins and alpha-crystallin.

It has been shown using ELISA and affinity chromatography, that extracts from heat-shocked Drosophila melanogaster larvae and cultured cells react with monoclonal antibodies raised against bovine alpha-crystallin. It is suggested that this cross-reactivity is associated with the region of the sequence corresponding to residues 98-108 of the bovine alpha A-crystallin chain. This region may be important for the functions of both alpha-crystallin and the heat-shock proteins.

Local vagal responses in the lung periphery.

In previous work we studied responses to ozone (O3) in the lung periphery of anesthetized male mongrel dogs. O3 (0.1 ppm) delivered locally to the lung periphery through a bronchoscope wedged in a segmental airway increased collateral resistance (Rcs) 31.5 +/- 5%. Bilateral cervical vagotomy or pretreatment with atropine aerosol prevented these responses to O3. In the present study we asked two questions. First, is the vagus necessary for responses to 0.1 ppm O3 because it maintains base-line tone? Second, are physiological responses to O3 administered through a bronchoscope localized to the challenged region? To answer the first question, we increased base-line Rcs (38.4 +/- 11.8%) by administering an acetylcholinesterase inhibitor, neostigmine (aerosol), through the bronchoscope. Following neostigmine, O3 exposure increased Rcs (55.8 +/- 18.4%) only if the vagi were intact. To answer the second question, we introduced two bronchoscopes simultaneously into different regions of the lung. When O3 was delivered through one bronchoscope, responses were detected only in the exposed region. However, when the whole left lung was exposed to O3, the responses were also detected in the right lung, but only when the vagi were intact. We conclude that the lung has the capacity to respond to localized oxidant insult with vagally mediated responses limited to the region that is challenged. However, when a larger area is exposed, vagally mediated responses become generalized and affect both lungs.

The pubococcygens and female orgasm: a correlational study with normal subjects.

The relationship of pubococcygeal condition to orgasmic responsiveness in 102 women from a university community was examined in a controlled, prospective investigation. Pubococcygeal strength was measured with a perineometer while responsiveness was assessed by a standardized interview yielding reliable measures of self-reported orgasmic response. Subjects whose responsiveness might have been impaired by such factors as alcohol consumption, inadequate stimulation, and high sexual anxiety were excluded from analyses. Excellent within-session but poor across-session test-retest reliability of the perineometer measures was noted. Initial Strength Contracting proved to be the most reliable measure. Though the majority of the parous women had performed Kegel exercises after delivery, parity was negatively related to pubococcygeal strength on most measures. Contrary to experimental hypotheses, pubococcygeal strength was not found to be positively related to frequency or self-reported intensity of orgasm. Furthermore, women with higher pubococcygeal strength did not report that vaginal stimulation contributed more to attainment of orgasm, nor did they rate vaginal sensations during coitus as more pleasurable. Only in the case of pleasurability of orgasm through clitoral stimulation was a significant, though low, relationship obtained. Possible factors contributing to the discrepancy between these findings and previous uncontrolled investigations are discussed, as are the implications of these findings for the use of Kegel exercises in the treatment of orgasmic dysfunction.

Preparation and characterization of a novel group of pituitary-derived peptides stimulatory for DNA synthesis in fibroblasts.

Using [3H]thymidine incorporation into Balb/c 3T3 fibroblasts as an index of mitotic activity, several cationic growth-promoting peptide fractions of relatively low potency were identified and partially purified from bovine pituitaries. The most completely characterized fraction exhibited an isoelectric pH of 10.4, and gel exclusion chromatograms under non-denaturing conditions were consistent with a self-associating system. Gel exclusion chromatography in the presence of 6 M guanidine hydrochloride demonstrated that the major reduced and S-alkylated peptides comprising this fraction had molecular weights of 8400 and 9000, and there was no evidence of disulfides between these or other minor peptides. This was confirmed by polyacrylamide gel electrophoresis in the presence of sodium lauryl sulfate and urea. Two other active fractions, each with an isoelectric pH of 9.9, were found to have major reduced and S-alkylated peptides with molecular weights between 6400 and 13 000, and the chromatographic results suggested the presence of intramolecular disulfides. The peptides from all fractions had a relatively high percentage of basic amino acids (approx. 20%); lysine was predominant (10-17%) and histidine was relatively low (less than 1%). Based on various criteria including amino acid composition, molecular weight, isoelectric point, and biological assays, the mitogenic activity in these fractions does not appear to result from fibroblast growth factor. Thus, the pituitary seems to contain multiple cationic mitogenic peptides of low potency for fibroblasts in vitro and their target cells in vivo remain to be defined.
