Behavioural and convulsant actions of two methyl esters of beta-carboline-3-carboxylic acid in photosensitive baboons and in DBA/2 mice.

The proconvulsant and convulsant actions of methyl 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM) and of methyl-beta-carboline-3-carboxylate (beta-CCM) have been evaluated in two animal models of reflex epilepsy, the photosensitive baboon, Papio papio, and the audiogenic seizure prone DBA/2 mouse. In baboons, myoclonic responses to photic stimulation are markedly enhanced 1 min after DMCM, 0.25 mg/kg i.v. In the absence of photic stimulation DMCM, 0.5 mg/kg i.v. induces a single brief tonic clonic seizure within 10-90 s. beta-CCM, 0.025-0.05 mg/kg i.v. similarly enhances myoclonic responses to photic stimulation. Generalised seizures occur without photic stimulation 0.5-3 min after beta-CCM, 0.1-0.2 mg/kg. Pretreatment with the excitatory amino acid antagonist, 2-amino-7-phosphonoheptanoic acid (2-APH), 110 mg/kg i.v., prevents the generalised seizures induced by DMCM, 0.5 mg/kg, but not those induced by beta-CCM, 0.1-0.2 mg/kg. In DBA/2 mice beta-CCM and DMCM are indistinguishable in potency as convulsants (ED50 values for clonic seizures: 4.4 and 4.6 mg/kg i.p. respectively) and as proconvulsants (ED50 values for facilitation of clonic seizure responses to an 83 dB sound stimulus: 0.25 and 0.23 mg/kg). Pretreatment with 2-APH gives equipotent protection against audiogenic seizures induced by beta-CCM, 1 mg/kg or DMCM, 1 mg/kg. The differences in relative potency of beta-CCM and DMCM in the two species are probably accountable for in terms of differing metabolism. A differential action of the two beta-carbolines on receptor subtypes, with enhancement of excitatory amino acid release playing a more important role in epileptogenesis after DMCM, is proposed.

Ambient monitoring of airborne radioactivity near a former thorium processing plant.

Twenty-four hour sampling for airborne radioactivity near a former thorium and rare-earth extraction facility was conducted for approx. 2 months with high-volume and dichotomous air samplers. Thoron ( 220Rn ) daughters were identified in the air and confirmed to be originating from the waste storage site. High-volume samplers near the facility measured average 212Pb concentrations of 177, 43, and 237 pCi/m3 with corresponding ranges of (1.9-1351), (1.5-301) and (0.73-2201)pCi/m3. Simultaneous measurements with dichotomous samplers at the same sites measured average 212Pb concentrations on coarse particulates (2.5-15 microns dia.) of 14, 4 and 10 pCi/m3 and on fine particulates (less than 2.5 microns dia.) of 94, 9 and 214 pCi/m3, respectively. Corresponding ranges were (0.2-109), (0.1-63) and (0.1-94) pCi/m3 for coarse particulates and (0.7-1094), (0.4-101) and (0.5-2685) pCi/m3 for fine particulates. Uranium, thorium and radium radionuclides were not identified as being present in significant concentrations in the particulate samples.

Perceived and objective conditions as predictors of the life satisfaction of urban and non urban elderly.

The effects of objective and subjective conditions on the life satisfaction of urban and non urban elderly are examined in a sample consisting of 1405 randomly selected elderly Vermonters in a four-county area of Northwestern Vermont. Results confirm that there are few significant objective differences between urban and non urban elderly. However, the non urban feel subjectively that they are better off on most conditions. Perceived evaluations of status are found to be better predictors of life satisfaction than objective measures. Furthermore, most disadvantaged conditions' are better predictors of life satisfaction for urban residents than for non urban residents.

Transformation of group F streptococci by plasmid DNA.

When the Challis strain of Streptococcus sanguis was transformed by the 17 megadalton beta plasmid from Streptococcus faecalis strain DS5, the plasmid underwent a 1.5 megadalton deletion (LeBlanc & Hassell, 1976). Furthermore, the covalently closed circular (CCC) plasmid DNA isolated from Challis transformants was rapidly converted to a linear form which did not possess any detectable transforming activity. To obtain stable CCC plasmid DNA a competent culture of a Lancefield group F streptococcus, strain DL8 (ATCC 12393), was used as a recipient of beta plasmid DNA. The plasmid DNA isolated from group F transformants exhibited the same configuration and size characteristics as the DS5 beta plasmid, and the CCC configuration was stable upon storage. CCC plasmid DNA from a group F transformant was biologically active and, when added to competent cultures of strain DL8, transformed them at frequencies about 100-fold greater than did beta plasmid DNA from DS5. This suggests the existence of a restriction--modification system in strain DL8.
