Effects of morphine on rheological properties of rat red blood cells.

To evaluate the effect of morphine on red blood cells, in vivo and in vitro rat models of morphine dependence were established. Rheological properties of rat red cells were measured by ektacytometry; the biophysical changes in the membrane of rat red cells were measured by the Fourier-Transformed Infrared technique (FT-IR) and the fluorescence depolarization method. The results show that the membrane fluidity of red cells was greatly reduced by morphine and the secondary structure of membrane proteins was changed. This suggests that morphine affects the rat red cell membrane directly, rather than through opioids-receptors.

Suberoylanilide hydroxamic acid as a potential therapeutic agent for human breast cancer treatment.

BACKGROUND: Suberoylanilide hydroxamic acid (SAHA) is a prototype of the newly developed, second-generation, hybrid polar compounds. It is a novel histone deacetylase inhibitor with high potency for inducing cell differentiation of cultured murine erythroleukemia cells. Studies with SAHA have primarily been performed with hematopoietic tumor cells. Here we extent these studies with SAHA to human breast cancer cell lines in an attempt to find better therapeutic agents for breast cancer treatment. MATERIALS AND METHODS: Human breast cancer cell lines, MCF7, MDA-MB-231, and MDA-MB-435, as well as normal cells, including the normal breast epithelial cell line MCF-10A, and fibroblasts, were treated with SAHA. Cells assayed for cell survival by using trypan blue exclusion assay, colony formation assay, and cell cycle and apoptosis analysis. The effects of SAHA on cell cycle and apoptosis regulatory proteins were examined by Western blots analysis. The identification of additional target genes was carried out by differential display (DD) and reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: SAHA inhibited clonogenic growth of MCF7, MDA-MB-231, and MDA-MB-435 breast cancer cells. These cells were more sensitive to SAHA-mediated cytotoxic effects than normal breast epithelial cells and fibroblasts. The cytotoxic effects of SAHA on breast cancer cells were manifested by G1 and G2/M cell cycle arrest and eventual apoptosis. The pan-caspase inhibitor, Z-VAD.fmk, blocked SAHA-induced cell death, DNA laddering, and cleavage of poly(ADP-ribose) polymerase, indicating the involvement of caspases in SAHA-mediated apoptosis. In addition, SAHA modulated cell cycle and apoptosis regulatory proteins. For example, cyclin-dependent kinase (CDK) inhibitors p21WAF1/Cip1 and p27Kip1 were induced, and retinoblastoma protein pRb was hypophosphorylated. Moreover, SAHA induced several genes associated with differentiation and/ or growth inhibition. These genes encode gelsolin, isopentenyl-diphosphate delta isomerase (IDI1), and 1,25-dihydroxyvitamin D-3 up-regulated protein 1 (VDUP1), the last two of which were identified by DD. Induction of these genes may contribute to SAHA-mediated pro-differentiating and antiproliferative effects. CONCLUSIONS: SAHA induced growth inhibition, cell cycle arrest, and eventual apoptosis in human breast cancer cells, possibly by modulating cell cycle and apoptosis regulatory proteins, such as CDK inhibitors p21 and p27, pRb, and other differentiation and/or growth inhibition-associated genes, including gelsolin, IDI1 and VDUP1. This, together with the low toxicity in normal cells, suggests that SAHA might have therapeutic potential for the treatment of human breast cancers.

Decreased bone mineral density in female patients with systemic lupus erythematosus after long-term administration of Tripterygium Wilfordii Hook. F.

OBJECTIVE: To investigate the possible bone changes in female patients with systemic lupus erythematosus (SLE) induced by long-term administration of Tripterygium Wilfordii Hook. F (TW). METHODS: 70 female SLE patients were divided into 4 groups according to their drug history: SLE disease control group, corticosteroids treatment group, TW treatment group, and both corticosteroids and TW treatment group. Bone mineral density (BMD) of the lumbar spine 2-4 and biochemical markers of bone turnover were studied. RESULTS: Long-term administration of TW could significantly decrease BMD levels in female SLE patients (P < 0.05). The patients receiving TW for more than 5 years had significantly lower BMD levels compared with those for less than 5 years. The degree of decreased BMD induced by TW was less severe compared with that of prednisone. No significant differences were observed in the biochemical markers of bone turnover among four groups (P > 0.05). CONCLUSION: Long-term administration of TW could decrease BMD levels in women. Osteoporosis may be an important problem for SLE patients treated with TW.

[Clinical and pathological analysis of 114 patients with postoperative wound infections by M. abscessus].

OBJECTIVE: To evaluate the pathological findings of postoperative wound infections by M. abscessus. METHODS: Proven sources of infections in 114 patients with postoperative wound infections were contaminated instrument disinfection liquid. According to the wound exudates cultivation and bacteriologic identification, the pathogen of postoperative infections was proved M. abscessus. Pathological features of postoperative wound infections were analyzed retrospectively. RESULTS: The postoperative wound infections in 114 cases involved cutaneous and subcutaneous tissue 95(83.3%), deep muscular tissue 21(18.4%), local lymph nodes 13(11.4%) and visceral organ, peritoneum and mesentery 3(2.6%). The main pathological appearances were granulomatous lesions and nonspecific chronic pyogenic inflammations. The granulomatous lesions include pyogenic tuberculosis-like granuloma, atypical tuberculosis-like granuloma and tuberculosis-like granuloma. The three types of granuloma could be seen in the same case and the percentage of three types was 75.4%, 28.1%, 59.6% respectively. 38 cases (33.3%) were showed red mycobacterium at acid-fast stain. CONCLUSIONS: The pathological appearances of most postoperative wound infections by M. abscessus were more or less different from those of tuberculosis. Awareness of the pathologic features of the postoperative wound M. abscessus infections will facilitate the diagnosis so that appropriate therapy may be initiated before the disease is far advanced and prevent infection dissemination.

[The diagnostic value of telomerase activity in bronchial biopsy specimen for lung cancer].

OBJECTIVE: To explore the diagnostic value of telomerase activity for lung cancer. METHODS: Telomerase activity was measured by PCR-TRAP in cancerous lesions and contralateral bronchial tissues from patients with lung cancer and in the bronchial tissues from patients with noncancerous pulmonary disorders, in which the tissue specimens were obtained by fiberoptic bronchoscopy. The result of telomerase detection was compared with the positive rates of pathologic study and bronchial brush cytology. RESULTS: The positive rate of telomerase activity in lesions of lung cancer was significantly higher than that in tissues of the contralateral bronchi from patients of lung cancer (P < 0.01) and that in tissues from patients of noncancerous pulmonary disorders (P < 0.01). There was no significant difference between the positive rate of telomerase activity expression and the positive rates of pathology and bronchial brush cytology (P > 0.05). CONCLUSION: Examination of telomerase activity was of significance in the diagnosis of lung cancer.

[Effects of serum and decidual tissue fluid in early pregnant women on natural killer cells activity and lymphocyte proliferation].

OBJECTIVE: To study the effects of serum and decidual tissue fluid in early pregnant women on natural killer cells (NK cells) activity and lymphocyte proliferation. METHOD: The effects of the serum and decidual tissue fluid obtained from 32 normal early pregnant women on NK cells activity and lymphocyte proliferation were tested by isotope incorporation methods. Sera of 25 nonpregnant women were chosen as controls. RESULTS: The NK cells activity and lymphocyte proliferation of sera and decidual tissue fluids from early pregnant women were lower than those of sera from nonpregnant women (P < 0.01). So were those of decidual tissue fluid as compared with sera during early pregnancy (P < 0.005). CONCLUSIONS: Serum and decidual tissue fluid from early pregnant women expressed immunosuppressive effects. It is possible that these effects of decidual tissue fluid play an important role in regulating local immune status of pregnant uterus.

[Molecular cytogenetics of bone tumors].

OBJECTIVE: To study alterations of molecular cytogenetics of bone tumor serially. METHODS: Chromosomal analysis, immunohistochemistry, molecular hybridization, nested-PCR/SSCP, DNA sequence analysis, and gene transfection techniques were used to investigate the common bone tumors. RESULTS: Chromosomal aberrations were found in 50 cases of aggressive bone tumors, with a chromosome number aberration rate of 86.96% and a structure aberration rate of 93.94%. However, there were differences among different kinds of bone tumors. The incidence of chromosomal instability in 53 patients with bone tumors was higher than that in normal controls. Meanwhile, the fragile sites of chromosomes of the patients were closely correlated with the high nonrandom chromosome breakpoints of tumor cells and related oncogene loci. The data on p53 and Rb from 305 cases and 89 cases, respectively, indicated that abnormalities of p53 and Rb were common events in bone tumors. The accumulation of p53 protein and loss of Rb protein were more common in aggressive bone tumors. Structures and their products of related proto-oncogenes (ras, myc, fos, bcl-2, met) were studied in 124 cases. Bone tumors presented multi-oncogenes abnormalities with differences in different pathological kinds. Exogenous wild-type p53 gene transfected into osteosarcoma cell line induced growth inhibition and apoptosis of tumor cells. CONCLUSIONS: Abnormalities of chromosomes, proto-oncogenes, and tumor suppressor genes frequently exist in bone tumor cells and are correlated with oncogenesis.

[The toxicity of busulphan and cyclophosphamide as the preparative regimen for allogeneic peripheral blood stem cell transplantation].

OBJECTIVE: To analyse the treatment-related complications of busulphan and cyclophosphamide (BU-CY) as the conditioning regimen for allogeneic peripheral blood stem cell transplantation (allo-PBSCT). METHODS: The clinical data of 40 leukemia patients undergoing allo-PBSCT between June 1997 and May 1999 in our BMT center were retrospectively analysed. RESULTS: Recovery of neutrophil and platelet was achieved at a median of day +13 (9 - 28) and day +12 (7 - 60) respectively. Acute GVHD occurred in 17 of 40 patients (42.5%) with grade II-IV in 10 patients (25%). Chronic GVHD developed in 21 out of 30 evaluable patients (70%). Mild to severe mucositis occurred in 30 patients (75%), and 4 of them had severe esophagitis with bleeding. Haemorrhagic cystitis developed in 8/40 (20%) patients, the median time of its onset was day +100 (+7 to +165). Six of 40 patients (15%) developed interstitial pneumonia (IP), 5 of them were due to cytomegalovirus infection, and the remaining one due to pneumocystis carinii infection. No hepatic veno-occlusive disease was observed and no seizure occurred. During the median follow-up of 480 (300 - 1000) days, 4 (10%) patients relapsed and 8 (20%) patients died of the transplant-related complications. The 3 year leukemia-free survival rate was 70%. CONCLUSION: BU (domestic busulfan)-CY regimen is relatively easy to administer and well tolerated, with low extramedullary toxicities.

[Effects of different cryoprotectants on biological properties of hematopoietic cells derived from cord blood].

OBJECTIVE: To search for effective cryoprotectants that can be used in cord blood bank to cryopreserve hematopoietic cells. METHODS: Four kinds of combined reagents were used to cryopreserve cord blood cells. The effects of these cryoprotectants on biological properties of hematopoietic cells were monitored and compared after the cells were refrigerated for 1 to 4 months. The detected biological properties included the number of total nucleated cells (TNC), CD(34)(+) cells and CFCs. RESULTS: The cryoprotectants of Dextran-40 + 10%DMSO resulted in the highest recovery rates, TNC, CD(34)(+) cells and CFCs were 77.8%, 78.5% and 87.2%, respectively. The combination of 10%DMSO + 6% AB serum + 5%HES led to the lowest recovery rates. CONCLUSION: Dextran-40 + 10%DMSO is an ideal cryoprotectant combination for cryopreserving hematopoietic cells in the cord blood bank.

[Isolation and phenotypic analysis of lamina propria mononuclear cells from colonoscopic biopsy specimens].

This study was directed to the method of isolation of lamina propria mononuclear cells (LPMC) from mucosal biopsy specimens and the phenotypic analysis of the cells. Ten biopsy specimens, taken individually from 8 patients with ulcerative colitis (UC), 5 normal controls and 22 patients with colorectal cancer, were collected to compare the factors that influence isolation and cell yield and to analyze the lymphocyte subsets by means of two-color flow cytometry. LPMC yield averaged 10(6) with a viability of more than 95% in UC, twice the yields in other two groups. The percentages of total T, B cells in three groups were similar (P > 0.05). When the UC group was compared with the other two groups, a significantly higher proportion of CD3(+)-CD4(+)-T cell (49.98% vs 37.54% and 37.25%, P < 0.01) was noted, whereas a lower proportion of CD3(+)-CD8(+)-T cell (23.64% vs 31.52% and 31.07%, P < 0.01) was observed. These data showed that the yield and viability of LPMC isolated from biopsy specimens were good enough for a phenotypic or functional analysis, which might be helpful to mucosal immune research on gut disorders, such as UC.

[Establishment and application of a computerized radiography cephalometrics system].

We have developed a new-typed computerized radiography cephalometrics and prediction system. It includes cephalometrics, growth prediction, orthopedics/orthodontics prediction and profile prediction. In this study, 47 lateral cephalograms were randomly selected to process a reliability test. The results showed the two groups were in agreement on all of the 25 measurement index. The system is easier, quicker and less prone to error than traditional cephalometrics approach. It is convenient for orthodontists.

[The study of occlusal contact areas of upper premolars and molars in the unilateral cleft lip and palate].

OBJECTIVE: Occlusal contact plays an important roll in masticatory procedure. It infers to biting force and masticatory performance. In order to analysis the oral function in UCLP, the author tested the occlusal contact areas of upper premolars and molars in UCLP. METHODS: 22 UCLP patients (male 13, female 9, permanent teeth) were selected, which had no temporomandibular joint dysfunction. All of them had received plastic surgery treatment and never received orthodontic therapy. Their occlusal contact areas of upper premolars and molars in intercaspid position were recorded by performed soft wax, and the results were managed by computer with the software of micro image analysis system. All the data were input to computer and be analyzed by statistics. RESULTS: 1. In normal people, the occlusal contact areas of upper premolars and molars had no differences in bilateral and in sexual. 2. there was no sexual difference in UCLP as that in normal group, and the occlusal contact areas in cleft side or non-cleft side had no difference yet. 3. The occlusal contact areas in UCLP were lower than that in normal people. 4. The decrease of occlusal contact areas was the most important factor to low masticatory performance in UCLP patients. CONCLUSION: After plastic surgery in UCLP, orthodontic therapy will improve masticatory contact and contribute to craniofacial esthetic and masticatory function.

[Overproduction and partial characterization of the Ssh7a and Ssh7b proteins from Sulfolobus shibatae].

The Sulfolobus shibatae Ssh7a and Ssh7b proteins have been separately overproduced in Escherichia coli and purified using a simple procedure which includes a heat treatment step. The recombinant and native Ssh7 proteins are similar in the ability to bind both negatively supercoiled and relaxed DNAs. In addition, the recombinant Ssh7a resembles the native Ssh7 protein in constraining negative DNA supercoils. Our data suggest that the two isoforms of Ssh7 interact with DNA in a similar fashion, and the methylation state of Ssh7 interact with DNA in a similar fashion, and the methylation state of Ssh7 does not affect DNA binding and supercoil constraining by the protein.

[Effects of phytohormones on growth and content of depsides in Salvia miltiorrhiza suspension cells].

This paper deals with the effects of 2,4-D, BA and GA3 on the growth and content of two depsides (rosmarinic acid and lithospermic acid B) in suspension cells of Salvia miltiorrhiza. The results showed cell growth and rosmarinic acid content reached the maximum on the 12th day and lithospermic acid B content on the 16th day after incubating cells in the subculture medium MS + 2, 4-D 1 mg/L + KT 0.1 mg/L. This cell line was a growth-product-associated. With the same concentration (1 mg/L), 2, 4-D stimulated the cell growth but prohibited the formation of lithospermic acid B; GA3 inhabited the cell growth but stimulated the formation of two depsides; The effects of BA is between 2, 4-D and GA3. The concentration optimum of phytohormones tested displayed 3 mg/L for BA and 1 mg/L for GA3. For the optimum time of adding BA and GA3 was in med-term (the 8th day) and early term (at the beginning) of culture period respectively. The synergiatic function of BA and GA3 on the depside formation was also showed that adding GA3 (1 mg/L) was favour for the formation of lithospermic acid B of the suspension cell cultured in the medium containing BA 3 mg/L. The suitable time of adding GA3 was the 6the day after incubating cells in the medium of MS + BA 3 mg/L.

[Spectra and crystallographic sites of Ce3+ in Ca3MgSi2O8].

An efficiency purplish blue phosphor Ca3MgSi2O8:Ce3+ has been synthesized. The emission and excitation spectra of Ca3MgSi2O8:Ce3+ under 260 nm and 360 nm excitations show that they have their common broad band emission characters, but the maximums and the band widths of the emission spectra are different. The excitation spectra of Ce3+ 350 nm emission and 460 nm emission clearly show that there are remarkable differences. These results indicate that there are two kinds of Ce3+ emission centers in the host of Ca3MgSi2O8. According the analysis of the crystal structure and the Van Uitert's empirical equation, we have a conclusion that two centers occupy two kinds of octacoordinated Ca sites, respectively.

HCPTPA, a protein tyrosine phosphatase that regulates vascular endothelial growth factor receptor-mediated signal transduction and biological activity.

Angiogenesis is a tightly controlled process in which signaling by the receptors for vascular endothelial growth factor (VEGF) plays a key role. In order to define signaling pathways downstream of VEGF receptors (VEGFR), the kinase domain of VEGFR2 (Flk-1) was used as a bait to screen a human fetal heart library in the yeast two-hybrid system. One of the signaling molecules identified in this effort was HCPTPA, a low molecular weight, cytoplasmic protein tyrosine phosphatase. Although HCPTPA possesses no identifiable phosphotyrosine binding domains (i.e. SH2 or phosphotyrosine binding domains), it bound specifically to active, autophosphorylated VEGFR2 but not to a mutated, kinase-inactive VEGFR2. Recombinant VEGFR2 and endogenous VEGFR2 were substrates for recombinant HCPTPA, and HCPTPA was co-expressed with VEGFR2 in endothelial cell lines, suggesting that HCPTPA may be a negative regulator of VEGFR2 signal transduction. To pursue this possibility, an adenovirus directing the expression of HCPTPA was constructed. When used to infect cultured endothelial cells, this adenovirus directed high level expression of HCPTPA that resulted in impairment of VEGF-mediated VEGFR2 autophosphorylation and mitogen-activated protein kinase activation. Adenovirus-mediated overexpression of HCPTPA also inhibited VEGF-induced cellular responses (endothelial cell migration and proliferation) and inhibited angiogenesis in the rat aortic ring assay. Taken together, these findings indicate that HCPTPA may be an important regulator of VEGF-mediated signaling and biological activity. Potential interactions with other signaling pathways and possible therapeutic implications are discussed.

Comparison of amperometric methods for detection of exocytosis from single pancreatic beta-cells of different species.

Two methods for amperometric detection of exocytosis at single pancreatic beta-cells were compared. In the first, direct detection of insulin was accomplished using an insulin-sensitive chemically modified electrode. In the second, 5-hydroxytryptamine (5-HT) that had been allowed to accumulate within the beta-cell secretory vesicles was detected with a bare carbon electrode. The goal of the comparison was to determine whether 5-HT secretion was a valid marker of insulin secretion in single beta-cells. To aid in this comparison, some experiments involved simultaneous measurement of insulin and 5-HT at cells previously allowed to accumulate 5-HT. Upon application of common insulin secretagogues, current spikes resulting from detection of 5-HT, insulin, or both compounds were obtained indicative of secretion via exocytosis. The mean area of current spikes obtained from simultaneous measurements equaled the sum of the mean area of insulin and 5-HT measured independently. Additionally, analyses of the number of spikes obtained for detection of insulin, 5-HT, or both compounds were similar for several common secretagogues. These data support the hypothesis that accumulated 5-HT is released from insulin containing secretory vesicles, exclusively. In addition, measurement of insulin and 5-HT from beta-cells of different species was compared to determine whether a species dependence exists for the two methods compared here. Detection of 5-HT results in a similar number of spikes that are equivalent to insulin in frequency and amplitude in human, porcine, and canine beta-cells; however, in mouse and INS-1 beta-cells, 5-HT is more readily detected than insulin.

Long-term effects of neonatal seizures: a behavioral, electrophysiological, and histological study.

Previous studies have demonstrated that recurrent seizures during the neonatal period lead to permanent changes in seizure threshold and learning and memory. The pathophysiological mechanisms for these changes are not clear. To determine if neonatal seizures cause changes in hippocampal excitability or inhibition, we subjected rats to 50 flurothyl-induced seizures during the first 10 days of life (five seizures per day). When the rats were adults, we examined seizure threshold using flurothyl inhalation, and learning and memory in the water maze. In separate groups of animals, we evaluated in vivo paired-pulse facilitation and inhibition in either CA1 with stimulation of the Schaffer collaterals or dentate gyrus with stimulation of the perforant path. Following these studies, the animals were sacrificed and the brains evaluated for mossy fiber sprouting with the Timm stain. Compared to control animals, rats with 50 flurothyl seizures had a reduced seizure threshold, impaired learning and memory in the water maze, and sprouting of mossy fibers in the CA3 pyramidal cell layer and molecular layer of the dentate gyrus. No significant differences in impaired paired-pulse inhibition was noted between the flurothyl-treated and control rats. This study demonstrates that recurrent neonatal seizures result in changes of neuronal connectivity and alterations in seizure susceptibility, learning and memory. However, the degree of impairment following 50 seizures was modest, demonstrating that the immature brain is remarkably resilient to seizure-induced damage.
