Epithelial-cell architecture during involution of the human thymus.

One hundred thymus glands were assessed histologically as to their degree of involution. Epithelial cells were demonstrated by an immunoperoxidase method using a monoclonal antibody against cytokeratin. The distribution of these cells was studied in the medulla, the cortico-medullary junction, the cortical parenchyma and the subcapsular cortex. As involution proceeds, the loss of cells from the thymus is almost totally confined to the lymphoid-cell elements. The architecture of the epithelial-cell network remains largely intact although there is extensive collapse of the structure due to the loss of the intervening lymphocytes. Even when involution is apparently complete; sheets of epithelial cells can be demonstrated in the thymic remnant.

A hybridization and enzyme blocking method to enrich minor cDNA sequences.

A method is presented which allows for the enrichment of low frequency cDNA sequences. The crucial step in the procedure is the hybridization of a pool of cDNA to homologous or heterologous RNA to a Rot value which will leave minor sequences in a single strand cDNA form while the major sequences form cDNA:RNA hybrids. This allows subsequent enzymatic differentiation between major and minor sequences resulting ultimately in the degradation of the major sequences. The procedure is general and simple as it requires no column chromatography step. The method is designed to integrate into a widely used cDNA cloning protocol and results either in double-stranded cDNA which can be used for molecular cloning or as a source of probes for hybridization.

Impotence induced by a single gene mutation.

Inbred male mice homozygous for the stubby gene mutation were reported to be sterile nineteen years ago. However, the basis of the sterility has not been identified since testicular steroidogenesis and spermatogenesis are normal. In the present studies, the sexual behavior of stubby mice and their coisogenic, normal siblings was compared quantitatively. The experimental observations provided unequivocal evidence that the phenotypic basis of the sterility of stubby mice is impotency. Stubby mice represent the first animal model for the study of impotence and provide the first evidence of an autosomal gene mutation that has a primary effect on male sexual behavior.

Regulated expression of cloned human fetal A gamma-globin genes introduced into murine erythroleukemia cells.

We have analysed the expression of cloned human fetal gamma-globin genes introduced into murine erythroleukemia cells by a protoplast fusion procedure. Both the wild-type a gamma-globin gene and a mutant derivative related to a phenotype of hereditary persistence of fetal hemoglobin were studied. In both cases the level of gamma-globin mRNA increased by a factor of approximately sevenfold when erythroid cell differentiation was induced by treatment with hexamethylenebisacetamide. Thus, the regulation of the expression of the cloned fetal A gamma-globin gene in murine erythroleukemia cells resembled that of cloned adult beta-globin genes.

Clinical experience with 910 carbon tip leads: comparison with polished platinum leads.

Since 1980, we have implanted 910 carbon tip leads from three different manufacturers. The pacing thresholds from these leads were compared to those of standard polished platinum electrode configurations. Our major findings are that while acute voltage thresholds are not significantly different, two of the three carbon tip leads that we used in the study have statistically lower chronic thresholds then polished platinum leads (p less than or equal to 0.01).

The clinical utility of pacemaker follow-up visits.

Because of the increased complexity of modern pacemakers, pacemaker follow-up visits in specialized centers become more and more indispensable. In this study, the results of 15,000 outpatient visits to the cardiac pacing center between the years 1983-1985 are presented. In most cases (92.8%), verification of normal function was made; however, mandatory reprogramming was required in 1.2%, and hospitalization for various reasons was required in 6% of visits. Careful outpatient monitoring of pacemakers is therefore very important for detecting early or late pacemaker complications.

Survival and mortality in 3,701 pacemaker patients: arguments in favor of pacemaker reuse.

This study analyzed the results of 3,701 patients implanted with cardiac pacemakers at the Centre Chirurgical du Val d'Or between 1976 and 1981. Two pacemaker populations were compared: those having a new pacemaker and those implanted with a reused pacemaker. There were no statistically significant differences between the two groups, either in terms of indications for implantation or in terms of actuarial survival of patients. In addition, there was no significant change in survival of the pulse generator. The reutilization of pacemakers appeared to be in no way detrimental to patients.

Increase in the concentration of major boar sperm surface proteins during maturation in the epididymis.

High-resolution two-dimensional polyacrylamide gel electrophoresis analyses have indicated that several major boar sperm plasma membrane polypeptides (PMPs) increased in concentration during maturation in the epididymis. To investigate this further, monoclonal antibodies (MAbs) to two of these PMPs/glycoproteins referenced as 4.85 and 5.0 and polyclonal antisera (PCA) raised against PMP 5.0 were used to quantify these changes. ELISA assays for both PMPs, using solubilized plasma membrane (PM) and intact PM as antigens, indicated that both PMPs were present in greater concentration in cauda PM than in caput PM. ELISA assay using PCA against 5.0 and whole sperm from the cauda and caput epididymis also showed an increase in the concentration of this protein during transit through the epididymis. Indirect FITC fluorescence microscopy showed that MAbs reacted specifically with the luminal aspect of secretory cells of the corpus epididymis and, with greater intensity, with secretory cells of the cauda segment. When MAb tags were used the fluorescence technique indicated that PMP 5.0 was restricted to a narrow area of PM overlying the principal segment of the head of caput sperm but fluorescence extended further into the principal segment and into the flagellum of cauda PM. Since MAb binding sites appear to be partially masked, PCA were also used to localize antigenic sites. PCA to PMP 5.0 showed antigen localization in both head and flagellar PM of cauda and caput sperm. The fluorescence of cauda sperm PM, however, was significantly more intense than that of caput sperm PM. MAb to PMP 4.85 also bound to antigen more extensively on cauda sperm. The sum of these analyses suggests that major sperm PMPs are secreted by the epididymis and absorbed by sperm. The magnitude of surface protein changes occurring during epididymal transit in boar sperm appears to be greater than heretofore recognized.

The gene for human fibroblast interferon (IFB) maps to 9p21.

The interferons have been classified into alpha, beta, and gamma (leukocyte, fibroblast, and immune). We used a human genomic clone for beta 1 interferon IFB to determine the gene copy number in two patients with unbalanced rearrangements of 9p. Our results provide evidence for regional assignment of this gene to 9p21.

A histological and immunohistochemical study of the changes induced in the brains of white mice by infection with Toxoplasma gondii.

The topography and the severity of the brain lesions induced in male and female white mice 1-6 weeks following inoculation with Toxoplasma gondii were studied by conventional histological and peroxidase-antiperoxidase (PAP) immunohistochemical techniques. The medulla oblongata and fourth ventricle were the most severely damaged areas of the brain, while the cerebellum was only minimally involved in this only in the later stages of the disease. Toxoplasma cysts were revealed by the PAP method as early as 2 weeks after infection. The number of cysts and the severity of the lesions increased gradually each week following inoculation. The lymphoplasmacytic element in the lesions was less intense in male than in female mice. The significance of these findings in respect to the possible immunological pathogenesis of CNS toxoplasmic changes is discussed. A correlation of the experimental findings with the known neurologic manifestations of the disease is also attempted. The advantage of the PAP method in the study of toxoplasmic infection is stressed.

Osmiophilic cytoplasmic inclusions in the lymph node post-capillary venules of the guinea pig.

A hitherto unknown pleomorphic osmiophilic inclusion was found in te endothelial cells of the guinea pig lymph node post-capillary venules; its diameter ranges from 0.4 to 2.2 micrometers, it is bound by a single membrane and consists of heavily dense granular area interrupted by irregularly spaced light bands. Occasionally, irregularly arranged membranous profiles in the granular matrix are also seen. It appears that the formation of this unique inclusion is related to the sex hormonal environment but its significance is yet unknown.

BCG-induced changes in the post capillary venules of the guinea pig lymph node.

The post capillary venules (PCV) of the regional lymph nodes of BCG-injected male and female guinea pigs were examined under light and electron microscopy. The endothelial cells showed increased height and pyroninophilia. Their cytoplasm contained increased numbers of ribosomes and polyribosomes and prominent rough endoplasmic reticulum with occasionally dilated cisternae. The number of lymphocytes migrating through the PCV wall was also increased. The activation of the endothelial cells of PCV induced by BCG may increase the recirculation of lymphocytes from the blood stream. These may be of importance in the antitumour activity of BCG. In control guinea pigs the height of the endothelial cells of PCV was significantly greater in the female than in the male. After BCG injection this sex difference was still apparent.
