Defective endothelium-dependent relaxation of vascular smooth muscle and endothelial cell Ca2+ signaling in mice lacking sarco(endo)plasmic reticulum Ca2+-ATPase isoform 3.

Sarco(endo)plasmic reticulum Ca2+ ATPase isoform 3 (SERCA3) is one of two Ca2+ pumps serving intracellular Ca2+ signaling pools in non-muscle tissues; however, unlike the ubiquitous SERCA2b, it exhibits a restricted cell-type distribution. Gene targeting was used to generate a mouse with a null mutation in the SERCA3 gene. Homozygous mutant mice were viable, fertile, and did not exhibit an overt disease phenotype. Because SERCA3 is expressed in arterial endothelial cells, aortic ring preparations were analyzed to determine whether it is involved in the regulation of vascular tone. Contraction-isometric force relations in response to phenylephrine or KCl, as well as relaxation produced by exposure to a nitric oxide donor, were similar in wild-type and null mutant aortas. Acetylcholine-induced endothelium-dependent relaxation of aortas after precontraction with phenylephrine was significantly reduced in homozygous mutants (61.3 +/- 5.6% in wild type, 35.4 +/- 7.3% in mutants). Ca2+ imaging of cultured aortic endothelial cells demonstrated that the acetylcholine-induced intracellular Ca2+ signal is sharply diminished in SERCA3-deficient cells and also indicated that replenishment of the acetylcholine-responsive Ca2+ stores is severely impaired. These results indicate that SERCA3 plays a critical role in endothelial cell Ca2+ signaling events involved in nitric oxide-mediated relaxation of vascular smooth muscle.

Effectiveness of low-dose ASA in prevention of secondary ischemic stroke, the ASA Study Group in Taiwan.

This randomized double-blind controlled study was carried out to investigate the effect of 100 mg acetylsalicylic acid (ASA) per day on the secondary prevention of ischemic stroke. Patients who suffered a first ischemic stroke from 13 participating hospitals were enrolled. They were independent or only partially dependent in activities of daily living and all had received brain CT for diagnosis. Eligible patients were randomly allocated to the 100 mg ASA or the nicametate citrate (a vasodilator) groups, and trial medications were started within three to six weeks after the onset of stroke. The primary end point was cerebral reinfarction, and intracranial hemorrhage was classified as an adverse event. Four hundred and sixty-six patients participated in this study; and 222 cases (136 males and 86 females) were allocated to the ASA group while 244 cases (150 males and 94 females) were assigned to the nicametate group. No significant difference in baseline characteristics between the two groups was observed. Cerebral reinfarction developed 6.3% (14/222) in the ASA group and 11.9% (29/244) in the nicametate group. According to the Cox's proportional hazards model, the estimated risk ratio (ASA group vs. nicametate group) was 0.538, with a 95% confidence interval of 0.284-1.019. The result was of borderline statistical significance. The risk for cerebral reinfarction was reduced by almost 50% among those who took 100 mg ASA versus those who took nicametate.

Anti-allergic effect of an aqueous extract of wu-hu-tang.

Wu-Hu-Tang (WHT), a Chinese formulation which consists of seven crude drugs, has been used for the treatment of asthma for hundreds of years. In this paper, an investigation on the anti-allergic activity of an aqueous extract of WHT was undertaken to find the pharmacological basis for the ethnomedical use of the formulation. WHT produced a significant inhibition on the homologous passive cutaneous anaphylaxis (PCA) in rats and the heterologous PCA in mice, decreased the degranulation of mast cells of calvarial periosteum in rats, inhibited the release of anaphylactic mediators from sensitized lung tissues of guinea pigs and the contraction of isolated guinea pigs ileum induced by histamine. These results indicated that the therapeutic activity of WHT for asthma may be related to its inhibitory effects on immediate hypersensitivity.

The physical location of genes cdc2 and prh1 in maize (Zea mays L.).

A biotin-labelling in situ hybridization technique was first used to physically map two single copy genes, cdc2 and prh1, in maize. These two genes are metabolically interrelated genes. The full-length cDNA clones cdc2ZmA and ZmPPI of genes cdc2 and prh1 were adopted as the probes. They are 1.3 and 1.6 kb in size, respectively. Clone cdc2ZmA was physically mapped on the long arm of chromosomes 4, 8, and 9. The percent distances from centromere to detection site were 57.9 +/- 2.7, 28.4 +/- 1.5, and 88.2 +/- 3.3. The detection rate was 19.2%. Clone ZmPPI was physically mapped on the long arm of chromosomes 4, 6, and 8. The percent distances were 53.6 +/- 1.2, 60.8 +/- 2.9 and 17.1 +/- 1.6. The detection ratio was 18.5%. The technique of chromosome ISH and the relationship between the location and function of these two genes have been discussed.

[A serologic study of legionellas in the patients with corpulmonle infection].

By MAT, the serum antibodies to Lp1-Lp14 were measured in fifty patients with infection of corpulmonale admitted in three hospitals in Datong. The antibody positive rate was 38% and the infection rate was 18%. The result indicated that some of the patients with corpulmonle infection were caused by Lp. and the seriousness of Corpulmonle was related to the infection of Lp. An alternative method for the therapy of the infection of corpulmonle was offered.

Fibronectin and hyaluronan in bronchoalveolar lavage fluid from young patients with chronic obstructive pulmonary diseases.

Fibronectin (FN) and hyaluronan (HA) in bronchoalveolar lavage fluid (BALF) and FN released by alveolar macrophages (AM) were examined in 7 nonsmoking healthy control subjects, and 20 smoking patients with chronic obstructive pulmonary diseases (COPD). All patients and subjects were no more than 40 years old. According to the 95% confidence limits of HA and FN in BALF from nonsmoking healthy control subjects, the smoking patients were divided into two groups, those who had HA and FN levels within the limits for nonsmoking controls were classified as the first group (group 1) and those with levels above the control limits were classified as the second group (group 2). Our data showed that the concentrations of HA and FN in BALF and FN released by AM were significantly higher in group 2 than in group 1 patients. There were also significant differences between the two groups in pulmonary function measurements (DLco, FEV1, and FEV1/FVC) which were lower in group 2. HA levels in group 2 patients correlated directly with counts of inflammatory cells in BALF (BALF cells/ml, and numbers/ml of total cells, macrophages, neutrophils and lymphocytes), and the concentration of FN released by AM, and showed an inverse correlation with pulmonary function (DLco and FEV1/FVC). Our results suggest that the inflammatory repair response and fibrosis may play a role in the development of emphysema in young patients with COPD.

A ten-year observation on experimental infection of periodic Brugia malayi in man.

This paper reports the results of 10 years of observations on the clinical manifestations, pathology and immunity to filariasis and aetiological biology of filariae in three volunteers (first author and his family members) who were inoculated experimentally with infective larvae of periodic Brugia malayi in 1981. The changes in clinical symptoms and signs were recorded systematically. Microfilariae were first detected at 41 and 46 weeks after inoculation in two subjects and remained detectable in small numbers until 8-8.5 years after infection. The microfilarial density fluctuated at 1-2 mf 120 microliters-1. Thereafter no microfilariae were detected in 12 blood sample examinations, suggesting that the adult reproductive period of periodic B. malayi could last up to 8-9 years in the human body. Eosinophilia occurred mainly before and at the initial stage of microfilaraemia. An increase in the lymphocytes was observed to some extent at 2-156 weeks after infection. Biopsy at the inoculation site 6 weeks after inoculation showed infiltration of the lymph node by inflammatory cells, mainly eosinocytes, lymphocytes and monocytes. Lymphangiectasis and lymphostasis were observed in both limbs and pelvic regions by lymphangiogram taken 11 weeks after inoculation. Antibodies against B. malayi first appeared at 2-5 weeks after infection, peaked at 12-56 weeks and thereafter declined gradually. Subjects A and C became antibody free but subject B remained positive to antibody against B. malayi 10 years after infection. E-rosette forming lymphocytes became lower than normal at 11 weeks and recovered to normal within 10 years after infection.

Ultrastructural features of penile tissue in impotent men.

Penile erection is related to the integrity of penile tissue. In order to study the ultrastructural histopathological changes in penile erectile tissue in impotent men, biopsies were taken from 29 impotent and 5 normal patients during surgical treatment. Light microscopy revealed insignificant histological changes and various degrees of fibrotic change in the corpora cavernosa of impotent men, and electron microscopy revealed severe myopathic, fibrotic changes in the penile tissue. These non-specific changes could not be used to determine the aetiology of impotence, and the relationship between the ultrastructural appearances, the causes of impotence and the results of treatment was unclear. Further studies are required to establish the value of intracorporeal biopsy in selecting treatment for impotent patients.

The causes of ischemic stroke in patients under 45 years of age.

From November 1982 to June 1991, a total of 81 cases of ischemic stroke in persons under 45 years of age were analyzed retrospectively. There were 44 males and 37 females, aged from 2 to 45 years (average 34.6 years). The modes of onset were immediately completed in 55 cases, progressed over a period of 1 hour to 5 days in 23 cases, and fluctuated over a period of 1 to 2 days in 3 cases. Fifty-six cases had one stroke episode; the others had transient ischemic attack or/and several episodes of cerebral infarction. Sixty-five cases had symptoms arising from the region of the carotid artery, more than 3.4 times those arising from the basilar artery territory. Probable causes of stroke were atherosclerosis (54.3%), embolism (28.5%), arteropathy (8.6%), hematological disease and coagulopathy (3.7%) and undetermined (4.9%).

Parkinson's disease in Taiwan: an analysis of 215 patients.

215 Chinese Parkinson's disease (PD) patients on levodopa therapy were followed up between 1982 and 1991. The ratio of males to females was 2.4 to 1. The mean durations from onset of the illness to stages I and II of Hoehn and Yahr (mild disability) were 4.0 and 6.5 years, to stage III (moderate) 7.9 years, and to stages IV and V (severe) 9.8 and 11.8 years. The mean duration of illness for living patients was 8.6 years. The mean duration of illness before death for the 46 patients who died was 8.9 years. The mean age at death was 68 years (4.4 years less than the normal life expectancy in Taiwan). The fate of this disease showed that patients with unilateral symptoms initially had a better prognosis than those with bilateral symptoms. The symptoms at onset were unilateral in 70% of the patients, of whom 91% had a spread of symptoms to the opposite side after a mean of 3.4 years. Familial PD occurred in 2.8% of our patients. The occurrence of blood ABO groups was not significantly different between the PD patients and the general population of Taiwan. Our findings differed from those in Western series by having a predominance of males, and a relatively shorter duration of unilateral symptoms before spread to the opposite side. In addition, the duration of illness and the survival time under levodopa treatment were shorter in Taiwanese and Japanese than in Western series.

Dural arteriovenous malformation of the transverse sinus with sinus occlusion: report of a case.

We report a case of dural arteriovenous malformation (AVM) of the transverse sinus with sinus occlusion. This 49-year-old man developed right parietal lobe dysfunction with acute onset. Computed tomography and magnetic resonance imaging (MRI) showed a non-hemorrhagic venous infarct in the subcortical white matter of the right parietal lobe, and diffusely dilated subcortical veins. Thrombosis of the right internal jugular bulb was also revealed on MRI. Cerebral angiography showed a dural AVM in the posterior fossa with occlusion of the right transverse sinus and retrograde venous drainage into the superior sagittal sinus, causing diffuse engorgement of the superficial cortical and the deep intramedullary veins. The focal neurologic deficits in this case were due to a non-hemorrhagic venous infarct in the subcortical white matter of the right parietal lobe secondary to retrograde cortical venous drainage.

[Effects of PGE1 on penile blood flow].

From February to July in 1989, 47 patients came to our O.P.D. with the chief complaint of impotence. The average age was 48.3 +/- 10.7 y/o. We applied intracoporeal injection of prostaglandin PGE1 (20 mg); and evaluated its penile blood flow effect by color duplex scanning (Acuson 128). The erectile responses of the test showed that: 6 patients (12.8%) had normal response: and 16 patients (34%) had imperfect response. Altogether, the total positive response rate was 46.8%, and 25 patients (53.2%) showed impaired response. The onset of response was 9.1 +/- 3.6 minutes and the duration of erection was 59.2 +/- 24.7 minutes. The percentage of diameter change of both deep arteries after injection was Rt: 58.1 +/- 41.5%; left: 52.3 +/- 35.6%. The peak velocity of right cavernosal arterial flow after intracoporeal injection was 35.5 +/- 15.9 cm/sec; and that of the left side was 33.2 +/- 16.9 cm/sec. There was no correlation between the increment of peak velocity of the deep arterial flow and the erection grade. The same phenomenon was also found between the increased change in the diameter of the deep artery and the erection grade. 16 patients (34.1%) experienced tolerable pain during the procedure. Two patients (4.3%) experienced dizzines and discomfort due to venous leakage. No priapism was found. This study suggests PGE1 may be an excellent potential alternative to other vasoactive drug with less complication in the diagnosis and treatment of impotence. But the cost and stability were its shortcoming.(ABSTRACT TRUNCATED AT 250 WORDS)

[A comparative study on the metabolism of nucleic acids during the subculture and differentiation of tobacco callus].

Changes in the contents of DNA and RNA, RNA species, the synthesis rates of DNA and RNA, and the activity of DNase and RNase were investigated in the callus of tobacco (Nicotiana tabacum L. cv. Willow Leaf) during subculture and differentiation. The contents of DNA and RNA were higher in differentiating callus than that in subcultured callus. After day 12, the contents of DNA and RNA in differentiating callus rose continuously while the contents of DNA and RNA in subcultured callus remained constant. Changes in RNA species and its relationship to total RNA level were also analyzed. At the stage of shoot primordium formation in differentiating callus, the activity of RNase increased markedly and the synthesis rate of RNA increased continuously; while the RNase activity and the synthesis rate of RNA in subcultured callus were much lower during the same period. During the period of shoot growth, the synthesis rate of DNA in differentiating callus was elevated compared to that in subcultured callus. The results above suggested that the metabolism of nucleic acids in differentiating callus was more active than that in subcultured callus.

Effect of aging on phosphate metabolites of rat brain as revealed by the in vivo and in vitro 31P NMR measurements.

Changes of phosphate metabolism in brains of neonate, weaning and adult rats were compared using both in vivo and in vitro nuclear magnetic resonance spectra. Ratios of phosphocreatine/nucleoside triphosphate (PCr/NTP) were the same in neonatal brain in both in vivo and in vitro studies, but not in weaning and adult brains. This discrepancy may have resulted from extended cerebral hypoxia due to slowed freezing of the brain by the increased skull thickness and brain mass in the weaning and adult rats. Variations in in vitro extraction condition for this age-related study may lead to systematic errors in the adult rats. Nevertheless, the phosphomonoester/nucleoside triphosphate (PME/NTP) ratios in extracts of brain from neonatal rats were higher than those obtained in vivo. In addition, the glycerophosphorylethanolamine plus glycerophosphorylcholine/nucleoside triphosphate (GPE+GPC/NTP) ratios, which were not measurable in vivo, showed age-dependent increase in extracts of rat brain. Some of the phosphomonoester and phosphodiester molecules in rat brain may be undetectable in in vivo NMR analysis because of their interaction with cellular components. The total in vitro GPE and GPC concentration in brain from neonatal rat was estimated to be 0.34 mmole/g wet tissue.

Thrombospondin and a 140 kd fragment promote adhesion and neurite outgrowth from embryonic central and peripheral neurons and from PC12 cells.

The ability of thrombospondin (TSP), an extracellular matrix glycoprotein, and two proteolytic fragments to support adhesion and neurite outgrowth from embryonic dorsal root ganglia, spinal cord neurons, and PC12 cells was examined. Anti-TSP antibodies or a synthetic peptide (GRGDS) containing an RGD cell-binding region was also added to cells plated on TSP. TSP and its 140 kd fragment were more efficient than laminin controls in supporting adhesion. Neurites formed on laminin, on varying concentrations of TSP, and particularly the 140 kd fragment. The amino-terminal heparin-binding domain supported little adhesion and outgrowth. Both adhesion and process outgrowth on TSP were inhibited by addition of anti-TSP antibodies, but not GRGDS.

An epidemiological study on occupational acute pyrethroid poisoning in cotton farmers.

A cross sectional survey on the prevalence of acute pyrethroid poisoning in cotton farmers was conducted in 1987 and 1988. A total of 3113 pyrethroid spraymen (2230 men (71.6%) and 883 women (28.4%] were interviewed after spraying and followed up for 72 hours. Adverse effects of pyrethroid exposure were found in 834 of them (26.8%) manifested as abnormal facial sensations, dizziness, headache, fatigue, nausea, or loss of appetite. Only 10 subjects, who developed significant systemic symptoms and had signs of listlessness or muscular fasciculation, were diagnosed as having mild occupational acute pyrethroid poisoning with a prevalence of 0.31% in subjects exposed to pure pyrethroids and 0.38% in subjects exposed to pyrethroid organophosphate mixtures. Measurements of pyrethroid concentrations in the air of the breathing zone, in skin pads, and in urine samples showed that dermal contamination is the main route of exposure to pyrethroids in cotton growers. Preventive measures are recommended.

Comparison of the effects of papaverine versus prostaglandin E1 on penile blood flow by color duplex sonography.

To compare the effects of papaverine (60 mg) and prostaglandin E1 (20 microgram: PGE1) on penile blood flow, the vascular response detected with color duplex ultrasound scanning and the clinical response in 34 impotent men were observed and recorded after they had received an intracorporeal injection of the respective drugs on separate days. The results demonstrate that PGE1 has the advantage of a slower onset, longer maintenance, less chance of priapism and is at least as effective as papaverine in inducing an increment of penile blood flow. In conclusion, PGE1 may be a better alternative to papaverine in the diagnosis and treatment of impotence.

[The effects of hypertension, diabetes mellitus, atrial fibrillation, transient ischemic attack and smoking on stroke in Chinese people].

Clinical findings and cranial CT scan identified stroke in 315 admitted patients at Taichung Veterans General Hospital from January 1, 1989 to December 31, 1989. Hemorrhage was found in 137 cases and infarction in 178 cases (thrombosis in 124 cases, embolism in 54 cases). Most cases with stroke (72.4%) were distributed in ages between 55 and 74 years. The mean age of patient with cerebral infarction was 64.8 years, which was about 3.3 years higher than those with cerebral hemorrhage (mean age 61.5 years). The control group consisted of 117 persons (matched in age and sex) who had no history of stroke. In comparison of the five risk factors (hypertension, diabetes mellitus, atrial fibrillation, transient ischemic attack, and smoking) between the patients and the controls, we found that cerebral thrombosis was significantly associated with hypertension, diabetes mellitus, and smoking; cerebral embolism with hypertension, atrial fibrillation, and transient ischemic attack; and cerebral hemorrhage with hypertension only.
