Comparison of the use of bulk to micro culture of cell preparations for lymphokine-activated cytotoxicity assays.

Different assay systems have been used to quantitate lymphokine-induced natural cytotoxic activity as a measure of immune status. This study compares the effects of inducing cytotoxicity in a bulk culture system, where effector cells are transferred to a micro culture well for assay, to a micro culture system where the effector cells are not transferred. The effector/target ratio for both the bulk and micro culture systems was calculated using the number of viable effector cells present at the time of target cell addition. After overnight incubation with interleukin-2 (IL-2), the lytic activity of murine spleen cells to targets using a micro culture system was increased two-fold over the bulk culture method. This increase was amplified further after 5 days of activation with IL-2, in that the micro culture system resulted in a four-fold increase in cytotoxic activity. The loss of some adherent cells in the bulk culture system did not explain the overall decrease in recovered cytotoxicity. The difference appeared to be related to cell loss during centrifugation. Therefore, the E/T ratios are different in the two systems if not corrected for the number of viable cells.

Anthropometric, psychomotor, and hemodynamic changes during twenty-one continuous days of eating only meals, ready-to-eat (MREs).

This study investigated anthropometric, psychomotor, and hemodynamic changes in young adult men during 21 continuous days of eating only meals, ready-to-eat (MREs). The MRE diet group lost more weight and bodyfat (significant at abdominal site, total skinfolds, and total bodyfat percent), and had significantly greater gains in hand grip strength than did controls. Results support the contention that, when eaten as designed, MREs are nutritionally adequate for maintaining body weight and muscle strength without adverse effects.

Paradoxical instructions in relaxation training.

The effects of instructions (paradoxical versus standard), treatment technique (EMG biofeedback, progressive muscle relaxation and self-relaxation) and subjects' sex were investigated in a 2 by 3 by 2 between-subjects design. Dependent measures were the changes in EMG and skin temperature over one 40-min. training session for 72 volunteers. For EMG change, the main effect of training was significant, with muscle relaxation producing less reduction than other techniques. This finding reflected an increase in EMG in the 5-min. posttreatment interval for the muscle relaxation group. For temperature change, the interaction of instructions and sex was significant, indicating that paradoxical instructions produced greater increases in temperature than standard instructions for men but not for women.

Regional pulmonary blood flow in mitral disease studied by xenon radiospirometry.

Regional lung perfusion was measured in the sitting position by 4 external detectors after intravenous injection of 133Xe in 24 patients with mitral valve disease and in 8 people with no cardiopulmonary disease acting as normal controls. Right- and left-sided heart catheterization was carried out on the patients on the following day. Mitral valve stenosis was found in 9, mitral valve regurgitation in 8, and both stenosis and regurgitation in the remaining 7. Regional lung perfusion in the normal people fell linearly from the basal to the apical sections of the lungs. The perfusion distribution in patients with mitral valve disease and a pulmonary capillary vein (PCV) pressure lower than 15 mmHg (2-0 kPa) did not differ significantly from that of the controls. A redistribution of the regional perfusion, with an increase in the apical perfusion and a fall in the basal perfusion of the lungs, was seen in patients with a raised PCV pressure. The hyperperfusion of the apical lung sections correlated with the mean pressure in the pulmonary artery (r=+0-795, P less than 0-001), while the basal hypoperfusion correlated with the PCV pressure (r=0-842, P less than 0-001). The PCV pressure can be predicted with an exactitude of +/- 7 mmHg (0-9 kPa) (95% confidence limits). Neither the cardiac index nor the pulmonary vascular resistance correlated with the changes in perfusion. Xenon radiospirometry is a rapid and reliable method for evaluating PCV pressure before or after operation in patients with mitral valve disease.

Extraskeletal Ewing's sarcoma. Diagnosis of a case by fine needle aspiration cytology.

A retroperitoneal mass was subjected to fine needle aspiration biopsy for cytologic evaluation. The aspirate consisted of a monotonous population of undifferentiated tumor cells whose cytologic appearance was consistent with a small-cell (Ewing's) sarcoma. The differential diagnosis of extraskeletal Ewing's sarcoma from other small-cell neoplasms, such as lymphoma, neuroblastoma and other soft-tissue sarcoma, cannot be made solely on the basis of morphologic studies. As in this case, histochemical studies and consideration of the clinical features are needed to make the final diagnosis.
