The protein kinase-C activation domain of the parathyroid hormone.

The PTH activates both adenylate cyclase and a mechanism that increases membrane-associated protein kinase-C (PKC) activity. To define the hormone's PKC activation domain we have used a panel of PTH fragments and ROS 17/2 rat osteosarcoma cells as the target cells. PTH equally and maximally increased PKC activity in ROS 17/2 cell membranes at physiological concentrations between 1-50 pM and 5-50 nM, but not at intermediate concentrations or concentrations above 50 nM. The PKC-stimulating picomolar concentrations of PTH did not stimulate adenylate cyclase in ROS 17/2 cells, while the PKC-stimulating nanomolar concentrations of the hormone did stimulate adenylate cyclase, with an EC50 of 1-2 nM. Very high concentrations of PTH, such as 100 nM, that did not increase membrane PKC activity were still able to maximally stimulate adenylate cyclase. PTH fragments lacking the N-terminal amino acids needed for adenylate cyclase activation increased membrane PKC activity, and the PKC activation domain was found to lie within the 28-34 region of the PTH molecule. This was confirmed by showing that optimally effective picomolar concentrations of the human PTH-(28-34) fragment itself were able to increase membrane-associated PKC activity to the same extent as the optimally effective picomolar concentrations of the intact PTH-(1-84) or the larger PTH-(1-34) or PTH-(3-34) fragments.

The mechanism of leukotriene B4 export from human polymorphonuclear leukocytes.

Recently, we characterized the export of leukotriene (LT) C4 from human eosinophils as a carrier-mediated process (Lam, B. K., Owen, W. F., Jr., Austen, K. F., and Soberman, R. J. (1989) J. Biol. Chem. 264, 12885-12889). To determine whether a similar mechanism regulates the release of leukotriene B4 (LTB4), human polymorphonuclear leukocytes (PMN) were preloaded with LTB4 by incubation with 25 microM leukotriene A4 (LTA4) at 0 degrees C for 60 min. PMN converted LTA4 to LTB4 in a time-dependent manner as determined by resolution of products by reverse-phase high performance liquid chromatography and quantitation by integrated optical density. When PMN preloaded with LTB4 were resuspended in buffer at 37 degrees C for 0-90 s, there occurred a time-dependent release of LTB4 but little formation or release of 20-hydroxy-LTB4 and 20-carboxy-LTB4. When PMN were preloaded with increasing amounts of intracellular LTB4 by incubation with 3.1-50.0 microM LTA4 and were then resuspended in buffer at 37 degrees C for 20 s, there occurred a concentration-dependent and saturable release of LTB4 with a Km of 798 pmol/10(7) cells and a Vmax of 383 pmol/10(7) cells/20 s. The release of LTB4 was temperature-sensitive with a Q10 of 3.0 and an energy of activation of 19.9 kcal/mol. The rate of LTB4 release at 37 degrees C is about 50 times the rate of 20-carboxy-LTB4 release. PMN preloaded with LTB4 and resuspended at 0 degree C for 1-60 min in the presence of 30 microM LTA5 progressively converted LTA5 to LTB5. The rate of LTB4 release at 0 degree C was inhibited over the entire time period, peaking at about 50% at 30 min. These results indicate that the release of LTB4 from PMN is a carrier-mediated process that is distinct from its biosynthesis.

Influence of calcitriol on prolactin and prostaglandin production by human decidua.

Receptors for calcitriol are described in human decidua. They exhibit a dissociation constant of 35 +/- 6 pM and they are at concentrations similar to those found in other tissues (0.45 +/- 0.04 fmol/micrograms DNA). They are highly specific for calcitriol since neither of the other vitamin D3 derivatives nor the steroid hormone tested displaced labeled calcitriol from the receptor. Also, calcitriol at concentrations of 10(-13) to 10(-11) M stimulates prolactin secretion by dispersed decidual cells. At these concentrations, however, the hormone has no effect on prostaglandin production. The specificity of calcitriol action was further examined by studying the effect of estrogen, progesterone, testosterone, dexamethasone and cortisol, all at 10(-7) M, on the secretion of prolactin and prostaglandins. Under the conditions used in this study, the steroids have no effect on prolactin secretion; but dexamethasone significantly inhibits prostaglandin F2 alpha output by the cells. Taken together with previous studies from our laboratory demonstrating that decidua can synthesize calcitriol the present study indicates that this hormone has an autocrine effect on human decidual cells.

Sequential bronchoalveolar lavages by endotracheal intubation in guineapigs.

We examined the feasibility of performing repeated bronchoalveolar lavage (BAL) on guineapigs. We also determined the influence of lavage volume on cell recovery in these animals, verified if the free cell populations were altered by repeated lavage, and compared the results with those obtained after euthanasia. Live animals were intubated orotracheally by direct vision laryngoscopy and lavaged, except for the last BAL which was done on isolated lungs after euthanasia. Each animal was lavaged weekly for 8 weeks: weeks 1-5 lavages were done with 5 aliquots of 2 ml of normal saline, week 6 with 3 aliquots of 1 ml, week 7 with 3 aliquots of 2 ml, and week 8 with 5 aliquots of 2 ml after euthanasia. The first 5 BAL gave similar results in volumes recovered, total number of cells, cell viability and differentials. The total cell yield of lavages 6 and 7 was less than that of the first 5 BAL and BAL 8; cell differentials with these smaller lavages were similar except for the percentage of neutrophils which was slightly higher than with the 10 ml lavage at week 3. The final lavage gave similar results as the first 5 BAL in terms of total cells recovered, cell viability, and differentials. We conclude that repeated BAL is feasible in live guineapigs, and that it gives a similar cells yield as lung lavage obtained after euthanasia.

[Quality of life in paraplegics and quadriplegics: analysis of self esteem].

This study suggests that self-esteem is a positive indicator of quality of life among persons who have sustained a spinal cord injury. Significant correlations were found between self-esteem and defensive or avoidance styles of coping. As well, self-esteem scores for quadriplegic subjects were significantly higher than scores for the paraplegic subjects. These findings should encourage nurses to evaluate their client's level of self-esteem as soon as possible during the rehabilitation process, so that they might initiate interventions that would assist the client to increase this level.

Leukotrienes and macrophage activation: augmented cytotoxic activity and enhanced interleukin 1, tumor necrosis factor and hydrogen peroxide production.

Leukotrienes (LT), and in particular LTB4, are potent inflammatory mediators and immunomodulators. In its interactions with leukocytes, LTB4 can activate numerous functions of neutrophils and modulate the activities of various lymphocyte subsets. LTB4 can also augment macrophage and monocyte cytotoxic activities and enhance their production of hydrogen peroxide and the monokines interleukin 1 and tumor necrosis factor. These observations allow a more detailed understanding of the effects of LTB4 on cellular immune and inflammatory functions.

Bronchocutaneous fistula in dogs: influence of fistula size and ventilatory mode on airleak.

Bronchocutaneous fistula (BCF) can originate at different levels, from the major airways to the peripheral lung. Little is published on the influence of the fistula origin or the ventilatory effect of the airleak. This study evaluates relative CO2 elimination via fistulas of various size and how different ventilatory modes influence both the quantity and quality, i.e., oxygen and CO2 content, of the airleak. We created BCF with five polyethylene tubings of different diameters (tube 1, 3.0 mm; tube 2, 4.0 mm; tube 3, 5.1 mm; tube 4, 6.4 mm; tube 5, 9.8 mm) in nine dogs. Six modes of ventilation were used with each tubing: spontaneous breathing (SB), pressure support (PS), high frequency (HF), assisted controlled with inspiration set at 20% (AC20) and at 67% (AC67) of the respiratory cycle, and AC20 with an end-expiratory pressure of +10 cm H2O (PEEP). For each ventilatory mode, the fistula air flow (Vf), CO2, and oxygen partial pressure of fistula air (PfCO2 and PfO2) and arterial blood were measured. Vf was measured for all tubes, while gas analysis was done for tubes 1, 3, and 5 only. As expected, Vf increased with tubing size. Vf was higher with AC67 and PEEP than with the other ventilatory modes. PfCO2 was not significantly influenced by the tube size and Vf. Fistula air alveolization was increased only with HF ventilation. Air leaked via the fistula contributed significantly to gas exchange; even when expiration was totally via the fistula, the arterial gases remained unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)

Sequential bronchoalveolar lavage in experimental extrinsic allergic alveolitis. The influence of cigarette smoking.

We studied the alterations induced by acute experimental extrinsic allergic alveolitis (EAA) on bronchoalveolar cell population in smoking and nonsmoking guinea pigs. Sixty-two animals divided into 3 groups were studied: Group 1 (17 animals), controls; Group 2 (21 animals), extrinsic alveolitis; Group 3 (24 animals), cigarette smoking and alveolitis. Bronchoalveolar lavages (BAL) were performed on Days 1, 19, and 44 for all animals. Group 3 animals had a fourth lavage before starting cigarette smoking, that is, 28 days before the beginning of the antigen injections. The other lavages were as for the other groups. BAL results on Day 1 were similar for each group. Cigarette smoking per se did not modify BAL in Group 3. EAA induction resulted in a large increase in all BAL cells, especially neutrophils of recovered fluid, which increased from 38 x 10(3) to 1,474 x 10(3) ml-1 (p less than 0.01) in Group 2 and from 58 x 10(3) to 740 x 10(3) in Group 3 (p less than 0.01). After maintenance, BAL neutrophils.ml-1 decreased to 444 x 10(3) in Group 2 (p less than 0.01), but stayed the same in Group 3: 973 x 10(3). After EAA induction, BAL neutrophils.ml-1 were higher in Group 2 than in Group 3 (p = 0.039); however, Group 2 had less neutrophils.ml-1 than Group 3 (p = 0.035) after EAA maintenance. We conclude that EAA results in a neutrophilic alveolitis and which can be evaluated by sequential BAL, and that cigarette smoking decreases the initial neutrophilic response and retards the eventual recovery during maintenance injections.
