[The chemoluminescence of the primary focus of a suppurative infection and of isolated neutrophils exposed to dimephosphon].

The influence of dimephosphone at concentrations of 0.001 M-0.75 M on the chemiluminescence of tissues at the focus of purulent infection in the ear of a guinea pig, on the survival rate of the experimental animals injected with the lethal dose of Staphylococcus aureus, as well as on the spontaneous and stimulated chemiluminescence of blood neutrophils in patients with wound infection, was studied. The study showed that different concentrations of dimephosphone oppositely influenced the intensity of the chemiluminescence of neutrophil suspensions and tissues at the focus of infection: low concentrations were found to produce stimulating action and high concentrations, suppressive action. At the highest concentration used in this study (0.75 M) dimephosphone prevented the death of the animals receiving lethal doses of S. aureus.

[The role of microcirculatory factors in cancer metastasis].

Morphologic, histochemical, biochemical and morphometric procedures were employed for examination of resected regional lymph nodes obtained from 70 patients suffering various malignancies. The important role of blood microcirculation bed alterations in cancer dissemination was established, the said disorders being as follows: decreased functional activity of the vascular wall and its increased permeability matched by intravascular blood coagulation and extravascular deposition of large amounts of fibrin in the lymph node tissue. Those changes were largely determined by lymph node mast cell reaction and levels of histamine and serotonin in the lymph node tissue. They preceded lymph node involvement.

[Comparative study of dimephosphon and xidiphone efficacy in steroid-induced osteoporosis in rats].

The efficacy of dimephosphon in comparison with xydiphone was studied in rats with an osteoporosis model induced by prednisolone administration at a daily dose of 50 mg/kg over a period of 14 days. The prednisolone treatment led to an increase in the content of oxyproline (a marker of bone resorption), calcium, and inorganic phosphates in the urine. Dimephosphon (monophosphate) decreased the levels of oxyproline, calcium, and inorganic phosphate in the urine. Xydiphone (biphosphonate) equally reduced the oxyproline excretion and the calcium level in urine, while rather insignificantly affecting the phosphaturia. Dimephosphon normalized the blood calcium level, while xydiphone decreased this parameter. An additional advantage of dimephosphon over xydiphone was manifested by normalization of the body weight of test rats and the total blood protein level.

[Temperature-dependent contractions of the guinea pig vas deferens mediated by P2X receptors].

The P2X receptor agonist alpha,beta-methylene ATP and purinergic electrical stimulation induce contractility response in guinea pig vas deferens. The low-temperature response observed at 30 degrees C was markedly greater than that at 37 degrees C and 42 degrees C. The P2X receptor antagonist pyridoxal-6-phosphate-2',4'-disulfonic acid stronger inhibits the electric-field-induced contractility at 30 degrees C than at 37 or 42 degrees C, while no such difference was observed for the inhibition of contractility induced by alpha,beta-methylene ATP.

[Effect of dimephosphon and xidifon on parameters of lipid peroxidation and the antioxidant system in rats subjected to the long-term administration of prednisolone].

Experiments in rats with an osteoporosis model induced by the chronic administration of prednisolone (50 mg/kg, p.o., over 14 days) showed that the drug differently affects the lipid peroxidation (LPO) rate and the antioxidant system activity. Xydiphone (45 mg/kg, p.o.) administered over 14 days exhibited a perooxidant action. Dimephosphon (208 mg/kg, p.o.) administered over the same period of time produced an antioxidant effect as manifested by a growth in the catalase activity, an increase in the content of ceruloplasmin and glutathione, and a decrease in the content of LPO products in the blood serum and liver tissues. The results of combined administration of prednisolone and phosphonates indicate good prospects of the dimephosphon treatment as a means of normalization of the LPO and antioxidant system functioning disturbed by prolonged use of glucocorticoids.

[New approaches to prognosis of risk of development of gastropathies induced by nonsteroid anti-inflammatory agents].

A pharmacological test with indomethacin is suggested for predicting the risk of NSAID-induced gastropathy. It is shown that patients with diagnoses of rheumatoid arthritis and osteoarthrosis can be divided into two groups with respect to the indomethacin test--"vulnerable" and "resistant", characterized by a high and low risk of NAIDS-induced gastropathy development, respectively. The proposed index of resistance to this disorder, together with the glutathione redox buffer test index, can be used as reliable criteria for predicting the NSAID-induced gastropathy.

[Ximedon-electrophoresis in rehabilitation of patients with systemic scleroderma].

A double blind placebo-controlled trial performed in 56 patients with scleroderma systematica showed effectiveness of a domestic drug ximedon--a pirimidine compound--applied during electrophoresis on the affected skin, limbs. The addition of ximedon-electrophoresis to rehabilitation program for scleroderma systematica improved the condition in 77.8%, microhemocirculation in 72.2%, reduced the area of the affected skin by 9.8% (p < 0.05), skin induration in 55.6% of patients.

[Comparative study of the effect of dimephosphon monophosphonate and xydiphone biphosphonate on the histomorphometric indices of rat vertebra in a model of glucocorticosteroid osteoporosis].

The efficacy of monophosphonate dimephosphon and diphosphonate xydiphon was compared by experiments in rats with an osteoporosis model induced by the chronic administration of prednisolone. The glucocorticosteroid decreased the total density of trabecules (in both bone and cartilage tissues) in histological micropreparations of lumbar vertebrae and reduced the total bone cell count and the calcium content in the bone tissue. Dimephosphon, administered on the prednisolone background over the same period of time, normalized the total relative density of trabecules (by increasing the cartilage content), the total bone cell count, and the calcium content. Under the same conditions, xydiphone normalized the total relative density of trabecules (by increasing the bone content), but did not influence the prednisolone-decreased content of cartilage, total cell count, and calcium content in the vertebrae. Thus, the test results reveal a significant advantage of dimephosphon over xydiphone.

[Effect of dimephosphon and xydiphone on mineral metabolism and lipid peroxidation in rats subjected to "pulse-therapy" with prednisolone].

The parameters of mineral metabolism, a lipid peroxidation (LPO) process, and the efficacy of antioxidant enzyme preparations were studied in rats with an osteoporosis model induced by the administration of prednisolone in a daily dose of 100 mg/kg (i.p.) over 3 days, which is analogous to a pulsed therapy schedule in clinics. The treatment with glucocorticosteroid enhanced the excretion of hydroxyproline and inorganic phosphates with urine, increased the LPO rate and antioxidant activity of the blood serum, and decreases the level of ceruloplasmin and the catalase activity in the blood. A prophylactic treatment with dimephosphon (208 mg/kg, p.o.) and xydiphone (45 mg/kg, p.o.) prevents from the prednisolone-induced LPO activation. Dimephosphon, in contrast to xydiphone, also prevents from a prednisolone-induced increase in the excretion of hydroxyproline and inorganic phosphates with urine.

[The mechanism of action of dimephosphon].

It has been established in experiments on rats that the 7-day administration of dimephosphon per os increases blood levels of total mercapto groups and glutathione, changing the correlation of its forms in favour of the oxidized ones. This course of dimephosphon administration reduces blood concentration of lipid peroxidation products. The significance of these mechanisms in realization of the pharmacological effects of dimephosphon is under discussion.

[An experimental validation of the combined use of dimephosphon with nonsteroidal anti-inflammatory preparations].

The effects of dimephosphon combined with nonsteroidal anti-inflammatory drugs were studied in rats and mice with experimental inflammation. The influence of dimephosphon on ulcerogenic activity of these drugs was also investigated. The dependence of the combinations efficiency on the scheme of dimephosphon administration was established. Synergism in anti-inflammatory activity between dimephosphon and ortophen (sodium diclofenac), indomethacin, and acetylsalicylic acid was shown. The possibility of decreasing the ulcerogenic effects of the drugs by dimephosphon was established. The effects of dimephosphon combinations are discussed as the basis for their use in clinical practice.

[Types of biochemical response to acute pharmacological indomethacin probe in healthy volunteers].

By the type of biochemical response to acute pharmacological indomethacin probe, a group of both male and female healthy volunteers can be subdivided into two parts. The first part includes volunteers with a stability index reduced as a result of accumulation of the oxidized products and a decrease in the content of reduced glutathione (GSH). The second part includes volunteers with the stability index increased as a result of decrease in the amount of lipid peroxidation products and an increase in the GSH content. A difference between males and females with respect to the response type consisted in that females features changes in the gutathione buffer components, and males, in the activity of antioxidant enzymes (catalase and peroxidase).

[The effect of dimephosphon on exudative and proliferative processes].

The effects of dimephosphon on the models of inflammation which permitted the separation of the exudative and proliferative phases were studied on rats. The drug effect on the levels of histamine, serotonin and their precursors in blood of healthy rats was also investigated. Dimephosphon was found to exert no inhibitory influence on the proliferative reaction in contrast to the well-known non-steroidal anti-inflammatory drugs. The antihistamine activity of dimephosphon occurring on account of the intensification of histaminopexy was established. It is suggested that the antihistamine activity of dimephosphon should be used for therapeutic purposes.

[Effect of dimephosphon on the inflammatory reaction].

The effect of dimephosphon on the development of the experimental inflammatory edema (agar-, dextran- and carrageenan-induced edema) of rat and mouse paws was compared with those of voltaren and dimedrol. The anti-inflammatory activity of dimephosphon occurring on account of the antiexudative component was established. The antihistamine and antiserotonin activity of dimephosphon was shown on models of paw edema caused by subplantar administration of mediators and modulators of inflammation. The drug exerts no influence on similar effects of bradykinin and prostaglandin E2, intensifies edema-inducing action of prostaglandin F2 alpha. The involvement of the anti-inflammatory component in the therapeutic effect of dimephosphon is suggested.

[An evaluation of the skin-resorptive action of dimephosphon when used locally long term].

Experiments in rabbits and rats established that 15% dimephosphonum solution after its topical (skin) application over a long period of time (2 and 6 months) exerted no toxic skin-resorption effects, reduced blood lipid and lipid peroxidation product levels. The substance of dimephosphonum after 6-month daily skin applications caused reversible changes in renal and adrenal functions.

[Ecto-ATPases and receptors for ATP].

Although enzymes which metabolize extracellular ATP (ecto-ATPases) have been described long ago, it has only recently become clear that such enzymes play an especially important role in cells possessing ATP receptors (P2-purinoceptors). In this review the authors characterize common features of ecto-ATPases, underlining the differences between these enzymes and other ATP-metabolizing enzymes. The authors also review their own recent studies dealing with the effects of various compounds on enzyme activity and on P2-purinoceptor-mediating responses. Since most known P2-purinoceptor antagonists inhibit ecto-ATPase activity, it is important to search for selective P2-purinoceptor antagonists as well as ecto-ATPase inhibitors.
